The binomial “autophagy-cancer” is intricate and methylomic studies can help to understand it by changing point of view from a gene level to an -omic one. Recently, autophagy-modulating properties of several phytochemicals have attracted attention in anticancer research. We evaluated whether Indicaxanthin (IND), the peculiar known beneficial phytochemical of prickly pear, seasonally available in the southern Italy, could induce autophagy in Caco2 cells, and whether it results from an epigenomic modification and/or a direct molecular interaction. IND increased autophagy in Caco-2 cells; the methylomic signature, obtained by Reduced Representation Bisulfite Sequencing (15 million of clusters) reported that 14 main genes of autophagy, showed a different methylation consistent with the induction of this phenomenon. Among these: MTOR, ATG13, BECN1, TFEB, ATG3, WIPI2, TECPR1, SNAP29, VPS11, VPS16. By traditional approaches we confirmed the demethylation of BECN1 gene and the increase of Beclin1 levels. By in-silico molecular modelling, we displayed a possible interference of IND, by competitive mechanisms, in the Beclin1-Bcl2 interaction. Methylomic signature and molecular modelling has been helpful to understand autophagy IND-induced in intestinal epithelial tumour cells. Our results suggest that the pro-autophagic action promoted by this phytochemical involves both epigenomic modulation and post-translation mechanisms by direct interaction with key targets of autophagy pathway

Discovering Knowledge from a Residential Building Stock through Data Mining Analysis for Engineering Sustainability

In this paper, a dataset of 92,906 dwellings was analysed adopting data mining techniques for the classification of heating and domestic hot water primary energy demand and for the evaluation of the most influencing factors. The sample was classified in three energy demand categorical variables (Low, Medium, High) considering different geometrical and physical attributes. The output of the model made it possible to set reference threshold values among the physical variables. Moreover, high energy demand dwellings were analysed in depth using a k-means algorithm in order to evaluate the design variables which need to be considered in a refurbishment process

Antiproliferative activity of synthetic fatty acid amides from renewable resources

AbstractIn the work, the in vitro antiproliferative activity of a series of synthetic fatty acid amides were investigated in seven cancer cell lines. The study revealed that most of the compounds showed antiproliferative activity against tested tumor cell lines, mainly on human glioma cells (U251) and human ovarian cancer cells with a multiple drug-resistant phenotype (NCI-ADR/RES). In addition, the fatty methyl benzylamide derived from ricinoleic acid (with the fatty acid obtained from castor oil, a renewable resource) showed a high selectivity with potent growth inhibition and cell death for the glioma cell line—the most aggressive CNS cancer

Exploitation of Business Models for Deep Renovation

In this workshop, we discussed the exploitation experiences gathered in some relevant H2020 projects on deep renovation: TripleA-reno, ProGETonE, P2Endure, 4rinEU, STUNNING, and TURNkey Retrofit. Directives and practical insights that support the experimentation with exploitation strategies in EU-funded projects will be elaborated as outcome of the workshop. The ability to envision user needs and integrate them into a project value proposition is a vehicle towards more effective and sustainable business models for deep renovation practices. However, many projects still struggle to involve the user in their business model design method. In spite of the clarified potential for business models underpinning energy efficiency measures to generate profit in the clean energy market, the uptake of deep renovation practices is not flourishing to its full potential. Several barriers have been highlighted that still distress the market uptake of energy efficiency measures, among which the need for long-term capital for often comparatively small investments, the strain of measuring and distributing the energy savings due to deep renovation practices, as well as general lack of familiarity among consumers with renovation concepts. The dispute here, with even the economically sound business models, is that, for the user, there is an apparent lack of attractiveness for deep renovations, that goes beyond the mere financial savings or economical profitability offered by the investment. A conceivable resolution to upsurge the market uptake of renovation practices, is therefore the design of business models, which focus on the multiple environmental, social and financial benefits deep renovation practices might deliver for the different stakeholders involved in the process. Benefits, that are sometimes hard to monetise and generalize among the involved user groups, as they do not always resonate to the provider/receiver of these values equally, but on the contrary can develop contrasting interests. Therefore, in this report, we are summarizing the contributions of several H2020 projects on deep renovation, including: TripleA-reno [1], ProGETonE [2], P2Endure [3], 4rinEU [4], STUNNING [5], and TURNkey Retrofit [6]. This, in a way to explore what type of deep renovation business models exist in practice, and how they actually are impacting the EU market uptake of deep renovation practices

Quality of Life of Sardinian Immigrants in Buenos Aires and of People Living in Italy and Sardinia: Does the Kind of Care have a Role for People with Depression?

Background / Objectives: The aim of the study was to compare the Quality of Life (QoL) of Sardinian immigrants to Argentina with Sardinians residing in Sardinia. The hypothesis was that a different availability of effective treatments for mood disorders may impact the well being of persons with these disorders. Methods: One out of five families of Sardinian origin was randomly selected. An Italian study (including Sardinia) was adopted as the control. The Mood Disorder Questionnaire was used for screening mania/hypomania; the diagnosis of Current Major Depressive Disorder was conducted by means of the Patient Health Questionnaire in immigrants and by means of a clinical interview in the control study and in an immigrant subsample (to verify comparability); the Short-Form Health Survey-12 was applied to measure QoL. Results: The Sardinian immigrants showed a higher QoL than Italians in Italy (but not with Sardinians residing in Sardinia). On the contrary, the attributable burden worsening QoL due to lifetime manic/hypomanic episodes, as well as to current depressive episodes, was found higher among Sardinian immigrants with respect to both Sardinian residents in Sardinia and the total Italian sample. The use of effective treatment for mood disorder was higher in Italy. Conclusion: The study found that in a sample of Sardinian immigrants in Buenos Aires the impact of a mood disorder affects QoL more incisively than in Sardinians residing in Sardinia. The suggested hypothesis of a possible role of beliefs guiding the search for treatments will be verified in future studies

A social engagement Fast Track on Energy Communities-Key Lesson Learned from H2020 EU Projects

    Get PDF
Energy communities organise collective and citizen-driven energy actions that will help pave the way for a clean energy transition, while moving citizens to the forefront. The energy market is rapidly transforming and so is the role of the consumer. Yesterday’s passive consumers become central actors in today’s energy markets. Today, as prosumers, citizens can benefit from their generation, consumption, and storage capabilities. Moreover, by supporting social engagement and citizen participation, energy communities can help provide flexibility to the electricity system through demand response, storage, and peer-to-peer energy exchange. Based on the collective debate from nine H2020 running projects (Renaissance, COMETS, Sender, eCREW, Lightness, ReDream, HESTIA, UP-STAIRS and NRG2peers), several challenges and key lessons learned can be identified for just social engagement. These challenges and lessons are relevant for the present and future development of EU energy communities

Electron Spin Resonance and Thermoluminescence dating of shells and sediments from Sambaqui (shell mound) Santa Marta II, Brazil

    Get PDF
In Tupi, the word Sambaqui means “mound of shells”. These archaeological sites are cultural vestiges left by the prehistoric occupation of the Brazilian coast from five to six thousand years ago. Mollusks, fishes, and other marine edible foods were important for the survival of this population. The remains of foods, mainly shells, were heaped up, giving a mound of different proportions, which became part of the landscape of the Brazilian coastal plain. Due to the large number of Sambaquis in Brazil and considering that Sambaqui Santa Marta II, Laguna, SC, has not yet been dated, Electron Spin Resonance (ESR) measurements were performed in aragonite shells collected from different layers of Sambaqui Santa Marta II, starting from the base to the center of the Sambaqui. Radiocarbon analysis by Accelerator Mass Spectrometry (AMS) at Beta Analytic laboratory were also performed for comparison with ESR results. Before measurements, shells were chemically etched, after drying, were pulverized and sieved. The sediments were separated into small portions which were irradiated with gamma radiation from a 60Co source with doses from 5 up to 120 Gy. Ages around 2,000 to 4,000 years have been obtained. The results obtained are consistent with the dates of others Sambaquis of the region, possibly were built at the same time
