    A Window Into Eastern Mediterranean Productivity Conditions Over Three Pliocene Precession-Forced Climate Cycles

    Here, we explore the importance of export productivity versus anoxia in the formation of sedimentary layers with enhanced total organic carbon (TOC) content. We use geochemical, sedimentological and micropaleontological records from two SW Sicily outcropping successions, Lido Rossello (LR) and Punta di Maiata (PM), over three Early Pliocene precession-forced climate cycles (4.7–4.6 million years ago [Ma]). Gray marls, deposited during precession minima, show enhanced TOC in both records. We suggest that basin-wide, low-oxygenated bottom-waters, resulting from freshwater-induced stratification during precession minimum, was integral to preserving gray marl TOC. Furthermore, prolonged eastern Mediterranean stratification may have produced a deep chlorophyll maximum (DCM), leading to “shade-flora” dominated productivity. The LR succession displays two unique laminated layers containing enhanced TOC. These laminations do not occur at specific times in the precession cycle or in time-equivalent PM samples. They are likely to have been produced by an intermittent dysoxic/anoxic pool at LR, caused by a local depression, which enhanced TOC preservation. Consequently, the laminations provide a rare window into “true” eastern Mediterranean productivity conditions during precession maxima, as organic matter is typically poorly preserved during these period due to enhanced ventilation. The laminated “windows” indicate that eastern Mediterranean export productivity may not have been significantly lower during precession maxima compared to precession minima, as previously thought. During these periods, productivity conditions are likely to have been comparable to the modern eastern Mediterranean, with a spring-bloom caused by enhanced winter/spring deep-water mixing preceding a summer “shade-flora” bloom caused by a summer-stratification induced DCM

    The force of events: the 'Brexit interval' and popular aspirations for Gibraltarian diplomacy

    This paper highlights both an overreliance on legal perspectives in the study of paradiplomacy at the expense of more dynamic understandings of agency, and also the affective force of waiting and other temporal states on political subject formation. Empirically, it reports the results of a longitudinal study on Gibraltarians’ concerns over the Gibraltar–Spain frontier. By comparing data from two identical surveys conducted a year apart during the period between the Brexit referendum and the (as yet incomplete) legal withdrawal, we trace the force of the incomplete event on political subjectivities. Conceptualizing our findings through assemblage theory and paradiplomacy, we highlight that the intensity of the event has heightened Gibraltarians’ dissatisfaction with their constitutional reliance on the UK to resolve Brexit in a way advantageous for Gibraltar. A minor shift occurred in the year studied towards more agentic proscriptions of what the Government of Gibraltar ought to do to resolve Brexit. Quantitative analysis reveals that younger respondents tend to emphasize this more agentic view, while older respondents tend to advocate further lobbying of the UK or feel Gibraltar has a complete lack of agency. Qualitative analysis of the respondents’ policy proscriptions reveals a complex set of views within each perspective on agency

    Molecular approaches to trematode systematics: 'best practice' and implications for future study

    To date, morphological analysis has been the cornerstone to trematode systematics. However, since the late-1980s we have seen an increased integration of genetic data to overcome problems encountered when morphological data are considered in isolation. Here, we provide advice regarding the ‘best molecular practice’ for trematode taxonomy and systematic studies, in an attempt to help unify the field and provide a solid foundation to underpin future work. Emphasis is placed on defining the study goals and recommendations are made regarding sample preservation, extraction methods, and the submission of molecular vouchers. We advocate generating sequence data from all parasite species/host species/geographic location combinations and stress the importance of selecting two independently evolving loci (one ribosomal and one mitochondrial marker). We recommend that loci should be chosen to provide genetic variation suitable to address the question at hand and for which sufficient ‘useful’ comparative sequence data already exist. Quality control of the molecular data via using proof-reading Taq polymerase, sequencing PCR amplicons using both forward and reverse primers, ensuring that a minimum of 85% overlap exists when constructing consensus sequences, and checking electropherograms by eye is stressed. We advise that all genetic results are best interpreted using a holistic biological approach, which considers morphology, host identity, collection locality, and ecology. Finally, we consider what advances next-generation sequencing holds for trematode taxonomy and systematics

    Identifying a Window of Vulnerability during Fetal Development in a Maternal Iron Restriction Model

    It is well acknowledged from observations in humans that iron deficiency during pregnancy can be associated with a number of developmental problems in the newborn and developing child. Due to the obvious limitations of human studies, the stage during gestation at which maternal iron deficiency causes an apparent impairment in the offspring remains elusive. In order to begin to understand the time window(s) during pregnancy that is/are especially susceptible to suboptimal iron levels, which may result in negative effects on the development of the fetus, we developed a rat model in which we were able to manipulate and monitor the dietary iron intake during specific stages of pregnancy and analyzed the developing fetuses. We established four different dietary-feeding protocols that were designed to render the fetuses iron deficient at different gestational stages. Based on a functional analysis that employed Auditory Brainstem Response measurements, we found that maternal iron restriction initiated prior to conception and during the first trimester were associated with profound changes in the developing fetus compared to iron restriction initiated later in pregnancy. We also showed that the presence of iron deficiency anemia, low body weight, and changes in core body temperature were not defining factors in the establishment of neural impairment in the rodent offspring

    Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries

    A ray of venom: Combined proteomic and transcriptomic investigation of fish venom composition using barb tissue from the blue-spotted stingray (Neotrygon kuhlii)

    Fish venoms remain almost completely unstudied despite the large number of species. In part this is due to the inherent nature of fish venoms, in that they are highly sensitive to heat, pH, lyophilisation, storage and repeated freeze-thawing. They are also heavily contaminated with mucus, which makes proteomic study difficult. Here we describe a novel protein-handling protocol to remove mucus contamination, utilising ammonium sulphate and acetone precipitation. We validated this approach using barb venom gland tissue protein extract from the blue-spotted stingray Neotrygon kuhlii. We analysed the protein extract using 1D and 2D gels with LC-MS/MS sequencing. Protein annotation was underpinned by a venom gland transcriptome. The composition of our N. kuhlii venom sample revealed a variety of protein types that are completely novel to animal venom systems. Notably, none of the detected proteins exhibited similarity to the few toxin components previously characterised from fish venoms, including those found in other stingrays. Putative venom toxins identified here included cystatin, peroxiredoxin and galectin. Our study represents the first combined survey of gene and protein composition from the venom apparatus of any fish and our novel protein handling method will aid the future characterisation of toxins from other unstudied venomous fish lineages.     The cardiovascular and neurotoxic effects of the venoms of six bony and cartilaginous fish species

    Fish venoms are often poorly studied, in part due to the difficulty in obtaining, extracting, and storing them. In this study, we characterize the cardiovascular and neurotoxic effects of the venoms from the following six species of fish: the cartilaginous stingrays Neotrygon kuhlii and Himantura toshi, and the bony fish Platycephalus fucus, Girella tricuspidata, Mugil cephalus, and Dentex tumifrons. All venoms (10–100 μg/kg, i.v.), except G. tricuspidata and P. fuscus, induced a biphasic response on mean arterial pressure (MAP) in the anesthetised rat. P. fucus venom exhibited a hypotensive response, while venom from G. tricuspidata displayed a single depressor response. All venoms induced cardiovascular collapse at 200 μg/kg, i.v. The in vitro neurotoxic effects of venom were examined using the chick biventer cervicis nerve-muscle (CBCNM) preparation. N. kuhlii, H. toshi, and P. fucus venoms caused concentration-dependent inhibition of indirect twitches in the CBCNM preparation. These three venoms also inhibited responses to exogenous acetylcholine (ACh) and carbachol (CCh), but not potassium chloride (KCl), indicating a post-synaptic mode of action. Venom from G. tricuspidata, M. cephalus, and D. tumifrons had no significant effect on indirect twitches or agonist responses in the CBCNM. Our results demonstrate that envenoming by these species of fish may result in moderate cardiovascular and/or neurotoxic effects. Future studies aimed at identifying the molecules responsible for these effects could uncover potentially novel lead compounds for future pharmaceuticals, in addition to generating new knowledge about the evolutionary relationships between venomous animals.</p
