Phosphorylation and dephosphorylation of tau protein by the catalytic subunit of PKA, as probed by electrophoretic mobility retard
Background: Tau is a microtubule associated protein that regulates the stability of microtubules and the microtubule-dependent axonal transport. Its hyperphosphorylated form is one of the hallmarks of Alzheimer's disease and other tauopathies and the major component of the paired helical filaments that form the abnormal proteinaceous tangles found in these neurodegenerative diseases. It is generally accepted that the phosphorylation extent of tau is the result of an equilibrium in the activity of protein kinases and phosphatases. Disruption of the balance between both types of enzyme activities has been assumed to be at the origin of tau hyperphosphorylation and the subsequent toxicity and progress of the disease.
Objective: We explore the possibility that, beside the phosphatase action on phosphorylated tau, the catalytic subunit of PKA catalyzes both tau phosphorylation and also tau dephosphorylation, depending on the ATP/ADP ratio.
Methods: We use the shift in the relative electrophoretic mobility suffered by different phosphorylated forms of tau, as a sensor of the catalytic action of the enzyme. Results: The results are in agreement with the long-known thermodynamic reversibility of the phosphorylation reaction (ATP + Protein = ADP+Phospho-Protein) catalyzed by PKA and many other protein kinases.
Conclusion: The results contribute to put the compartmentalized energy state of the neuron and the mitochondrial-functions disruption upstream of tau-related pathologiesFinancial support has been provided by a grant from the Spanish Government PGC2018 096177-B-10
Specific Profile of Tau Isoforms in Argyrophylic Grain Disease
Argyrophylic grain disease (AGD) is a neurodegenerative condition that has been classified among the sporadic tauopathies. Entities in this group present intracellular aggregates of hyperphosphorylated tau, giving rise to characteristic neuronal and glial inclusions. In different tauopathies, the proportion of several tau isoforms present in the aggregates shows specific patterns. AGD has been tentatively classified in the 4R group (predominance of 4R tau isoforms) together with progressive supranuclear palsy and corticobasal degeneration. Pick's disease is included in the 3R group (predominance of 3R isoforms), whereas tau pathology of Alzheimer's disease represents and intermediate group (3 or 4 repeats [3R plus 4R, respectively] isoforms). In this work, we have analyzed tau present in aggregates isolated from brain samples of patients with argyrophylic grain disease. Our results indicate that the main tau isoform present in aggregates obtained from patients with AGD is a hyperphosphorylated isoform containing exons 2 and 10 but lacking exon 3.This work was supported by grants from Spanish Plan Nacional (SAF2011-24841), CIBERNED and Reina Sofia Foundation.S
Tridimensional Structural Analysis of Tau Isoforms Generated by Intronic Retention.
Background: Tauopathies are a subset of neurodegenerative diseases characterized by abnormal tau inclusions. Recently, we
have discovered a new, human specific, tau isoform termedW-tau that originates by intron 12 retention. Our preliminary data
suggests this newly discovered W-tau isoform might prevent aberrant aggregation of other tau isoforms but is significantly
downregulated in tauopathies such as Alzheimer´s disease.
Objective: To accurately predict, examine, and understand tau protein structure and the conformational basis for the
neuroprotective role of W-tau.
Methods: A tridimensional deep learning-based approach and in vitro polymerization assay was included to accurately
predict, analyze, and understand tau protein structure and the conformational basis for the neuroprotective role of W-tau.
Results: Our findings demonstrate: a) the predicted protein tridimensionality structure of the tau isoforms raised by intron
retention and their comparison with the other tau isoforms; b) the interaction of W-tau peptide (from W-tau isoform) with
other tau isoforms; c) the effect of W-tau peptide in the polymerization of those tau isoforms.
Conclusions: This study supports the importance of the structure-function relationship on the neuroprotective behavior of
W-tau inhibiting tau fibrillization in vitro.post-print436 K

Effects of self-efficacy, catastrophizing, fear of movement and joint position sense in the maintenance of wrist unspecific pain in athletes. An observational study.
Wrist pain is a highly prevalent condition in racket sport population. Although
different wrist injuries have been described in the literature as more prevalent in racket sport,
unspecific chronic wrist pain is common problem on amateur population.
Material and methods: A cross-sectional study were conducted between October 2021 to June 2022.
Participants over 18 th suffered for wrist pain were recruited from different sports centers where
racket sports were practiced.
Objective: The aims of this research was to study the relationships and the predictive power of
Self-Efficacy, Catastrophizing, Fear of Movement and proprioception on the persistence of
unspecific wrist pain in athlete population.
Results: One hundred and four athletes participated in the study (54 men and 50 women), most of
them tennis and paddle players. Psychological variables were correlated (P 0.01) with wrist pain
persistence, suggestive of complex interactions between cognitive-affective processes and wrist
unspecific pain in athletes.
Conclusion: The results suggest that targeting high levels of Self-Efficacy, Catastrophizing and Fear
of Movement could be a prognostic factor of wrist unspecific maintained pain in athlete population.Universidad de Málaga. Campus de Excelencia Internacional Andalucía Tech
Specific Peptide from the Novel W-Tau Isoform Inhibits Tau and Amyloid β Peptide Aggregation in Vitro
W-Tau, a new tau human-specific splicing isoform generated by intron retention, has been recently described. This isoform contains an 18-residue unique sequence corresponding to the translation of the retained region of intron 12. In this work, we have described that such 18-amino-acid peptide from the retained intron 12 can inhibit tau and β amyloid peptides aggregation under in vitro conditions. This inhibitory function is also present in smaller fragments of the 18-residue peptide. This research was funded by the Spanish Ministry of Science

Predictive Biomarkers to Chemoradiation in Locally Advanced Rectal Cancer
There has been a high local recurrence rate in rectal cancer. Besides improvements in surgical techniques, both neoadjuvant short-course radiotherapy and long-course chemoradiation improve oncological results. Approximately 40–60% of rectal cancer patients treated with neoadjuvant chemoradiation achieve some degree of pathologic response. However, there is no effective method of predicting which patients will respond to neoadjuvant treatment. Recent studies have evaluated the potential of genetic biomarkers to predict outcome in locally advanced rectal adenocarcinoma treated with neoadjuvant chemoradiation. The articles produced by the PubMed search were reviewed for those specifically addressing a genetic profile’s ability to predict response to neoadjuvant treatment in rectal cancer. Although tissue gene microarray profiling has led to promising data in cancer, to date, none of the identified signatures or molecular markers in locally advanced rectal cancer has been successfully validated as a diagnostic or prognostic tool applicable to routine clinical practice
