    The development and application of an expert marker system to support ‘High Stakes’ examinations

    been used as the benchmark assessment for vocational IT skills ever since. It currently attracts over 300,000 entries per year across 9 different application areas. This equates to over 1m scripts per year, all of which require marking, moderation and processing to award of result. Over the last 5 years OCR has worked with the University of East Anglia to develop an automated ‘expert’ marking system for four of the application areas within CLAIT (Core, Word Processing, Spreadsheet, Database). This system is now operational and during the first pilot phase successfully supported 30 Centres and processed over 2000 script submissions. From initial concept the system set out to extend the scope of computer assisted assessment beyond simple skill tests as it was not desirable to reduce the CLAIT assessment to series of atomised functions tests or to place software simulations within testing Centres (over 3500 in the UK). It was also important that the candidate experience of the assessment remained relatively untouched and so an off-line delivery/on-line processing solution was proposed. This paper provides an overview of the modular approach taken by OCR and reflects on the successes and challenges faced during implementation. The presentation will provide a demonstration of the system with a brief explanation of the intelligent workflow processing and the application of expert marking rules to ensure valid and reliable assessment outcomes

    Linear chemically sensitive electron tomography using DualEELS and dictionary-based compressed sensing

    We have investigated the use of DualEELS in elementally sensitive tilt series tomography in the scanning transmission electron microscope. A procedure is implemented using deconvolution to remove the effects of multiple scattering, followed by normalisation by the zero loss peak intensity. This is performed to produce a signal that is linearly dependent on the projected density of the element in each pixel. This method is compared with one that does not include deconvolution (although normalisation by the zero loss peak intensity is still performed). Additionaly, we compare the 3D reconstruction using a new compressed sensing algorithm, DLET, with the well-established SIRT algorithm. VC precipitates, which are extracted from a steel on a carbon replica, are used in this study. It is found that the use of this linear signal results in a very even density throughout the precipitates. However, when deconvolution is omitted, a slight density reduction is observed in the cores of the precipitates (a so-called cupping artefact). Additionally, it is clearly demonstrated that the 3D morphology is much better reproduced using the DLET algorithm, with very little elongation in the missing wedge direction. It is therefore concluded that reliable elementally sensitive tilt tomography using EELS requires the appropriate use of DualEELS together with a suitable reconstruction algorithm, such as the compressed sensing based reconstruction algorithm used here, to make the best use of the limited data volume and signal to noise inherent in core-loss EELS

    Effect of glyphosate application time on yield parameters of South African glyphosate-resistant maize cultivars

    Glyphosate is the most used herbicide in South Africa. Due to observations by some South African maize producers that the application of glyphosate to glyphosate-resistant (GR) maize cultivars resulted in reduced yield, we conducted an in-depth study under local conditions. Through field trials, over two seasons (2017/2018 and 2018/2019), we investigated whether the application time of glyphosate would impact maize yields negatively. Various yield parameters were measured subsequent to glyphosate application to the local GR maize cultivars DKC74-74BR, DKC78-79BR, KKS4581, KKS8408, BG5785BR, PAN6R-710BR, P1814R and P2880WBR. Four glyphosate products were included (Roundup PowerMax®; Slash Plus 540 SL; TouchdownForte® and MambaTM DMA 480 SL), resulting in 32 cultivar x glyphosate product combinations. Each product was applied at V4, V4+V6, V6 and V8 growth stages together with an untreated control. Yield parameters measured (ears per plant, rows per ear, kernels per row, thousand kernel mass and yield) were expressed as a percentage of the control. The trials were planted as randomised complete block designs with three replicates. Limited response was observed with all the parameters investigated, with a significant negative yield response, greater than the untreated control, observed in only 3.1% of the cultivar x glyphosate product combinations evaluated. No clear trends or discernible and consistent impacts on yield and yield parameters could be established based on the application time of glyphosate (within label recommendations) across seasons. The findings contribute significantly to the knowledge base and current understanding of the international community and local producers alike regarding the effective use of glyphosate and generic variations thereof in crops of diverse genetic backgrounds. Significance: Limited response in the yield parameters evaluated were obtained in response to the application time (V4, V4+V6, V6 and V8) of the four glyphosate products on eight GR maize cultivars tested (p=0.1). Inconsistent patterns or trends were detected in cases where significance was obtained, implying that it would not be possible to draw accurate conclusions or formulate recommendations. Application time of glyphosate did not result in a significant reduction in yield compared to the untreated control, in the majority of the cultivar x glyphosate product combinations investigated, confirming that glyphosate application conducted within label specifications would not reduce yield, irrespective of the glyphosate product or genetic background of maize

    Assembly of functional diversity in an oceanic island flora

    Oceanic island floras are well known for their morphological peculiarities and exhibit striking examples of trait evolution1–3. These morphological shifts are commonly attributed to insularity and are thought to be shaped by the biogeographical processes and evolutionary histories of oceanic islands2,4. However, the mechanisms through which biogeography and evolution have shaped the distribution and diversity of plant functional traits remain unclear5. Here we describe the functional trait space of the native flora of an oceanic island (Tenerife, Canary Islands, Spain) using extensive field and laboratory measurements, and relate it to global trade-offs in ecological strategies. We find that the island trait space exhibits a remarkable functional richness but that most plants are concentrated around a functional hotspot dominated by shrubs with a conservative life-history strategy. By dividing the island flora into species groups associated with distinct biogeographical distributions and diversification histories, our results also suggest that colonization via long-distance dispersal and the interplay between inter-island dispersal and archipelago-level speciation processes drive functional divergence and trait space expansion. Contrary to our expectations, speciation via cladogenesis has led to functional convergence, and therefore only contributes marginally to functional diversity by densely packing trait space around shrubs. By combining biogeography, ecology and evolution, our approach opens new avenues for trait-based insights into how dispersal, speciation and persistence shape the assembly of entire native island floras.Fil: Barajas Barbosa, Martha Paola.     The effects of nitroxyl (HNO) on soluble guanylate cyclase activity: interactions at ferrous heme and cysteine thiols

    It has been previously proposed that nitric oxide (NO) is the only biologically relevant nitrogen oxide capable of activating the enzyme soluble guanylate cyclase (sGC). However, recent reports implicate HNO as another possible activator of sGC. Herein, we examine the affect of HNO donors on the activity of purified bovine lung sGC and find that, indeed, HNO is capable of activating this enzyme. Like NO, HNO activation appears to occur via interaction with the regulatory ferrous heme on sGC. Somewhat unexpectedly, HNO does not activate the ferric form of the enzyme. Finally, HNO-mediated cysteine thiol modification appears to also affect enzyme activity leading to inhibition. Thus, sGC activity can be regulated by HNO via interactions at both the regulatory heme and cysteine thiols

    Stratocumulus Cloud Clearings and Notable Thermodynamic and Aerosol Contrasts across the Clear–Cloudy Interface

    Data from three research flights, conducted over water near the California coast, are used to investigate the boundary between stratocumulus cloud decks and clearings of different sizes. Large clearings exhibit a diurnal cycle with growth during the day and contraction overnight and a multiday life cycle that can include oscillations between growth and decay, whereas a small coastal clearing was observed to be locally confined with a subdiurnal lifetime. Subcloud aerosol characteristics are similar on both sides of the clear–cloudy boundary in the three cases, while meteorological properties exhibit subtle, yet important, gradients, implying that dynamics, and not microphysics, is the primary driver for the clearing characteristics. Transects, made at multiple levels across the cloud boundary during one flight, highlight the importance of microscale (~1 km) structure in thermodynamic properties near the cloud edge, suggesting that dynamic forcing at length scales comparable to the convective eddy scale may be influential to the larger-scale characteristics of the clearing. These results have implications for modeling and observational studies of marine boundary layer clouds, especially in relation to aerosol–cloud interactions and scales of variability responsible for the evolution of stratocumulus clearings

    Phylogenetic composition of native island floras influences naturalized alien species richness

    Islands are hotspots of plant endemism and are particularly vulnerable to the establishment (naturalization) of alien plant species. Naturalized species richness on islands depends on several biogeographical and socioeconomic factors, but especially on remoteness. One potential explanation for this is that the phylogenetically imbalanced composition of native floras on remote islands leaves unoccupied niche space for alien species to colonize. Here, we tested whether the species richness of naturalized seed plants on 249 islands worldwide is related to the phylogenetic composition of their native floras. To this end, we calculated standardized effect size (ses) accounting for species richness for three phylogenetic assemblage metrics (Faith's phylogenetic diversity (PD), PDses; mean pairwise distance (MPD), MPDses; and mean nearest taxon distance (MNTD), MNTDses) based on a phylogeny of 42 135 native island plant species and related them to naturalized species richness. As covariates in generalized linear mixed models, we included native species richness and biogeographical, climatic and socioeconomic island characteristics known to affect naturalized species richness. Our analysis showed an increase in naturalized species richness with increasing phylogenetic clustering of the native assemblages (i.e. native species more closely related than expected by chance), most prominently with MPDses. This effect, however, was smaller than the influence of native species richness and biogeographical factors, e.g. remoteness. Further, the effect of native phylogenetic structure (MPDses) on naturalized species richness was stronger for smaller islands, but this pattern was not consistent across all phylogenetic assemblage metrics. This finding suggests that the phylogenetic composition of native island floras may affect naturalized species richness, particularly on small islands where species are more likely to co-occur locally. Overall, we conclude that the composition of native island assemblages affects their susceptibility to plant naturalizations in addition to other socioeconomic and biogeographical factors, and should be considered when assessing invasion risks on islands

    The Children's Respiratory and Environmental Workgroup (CREW) birth cohort consortium: design, methods, and study population

    Background: Single birth cohort studies have been the basis for many discoveries about early life risk factors for childhood asthma but are limited in scope by sample size and characteristics of the local environment and population. The Children’s Respiratory and Environmental Workgroup (CREW) was established to integrate multiple established asthma birth cohorts and to investigate asthma phenotypes and associated causal pathways (endotypes), focusing on how they are influenced by interactions between genetics, lifestyle, and environmental exposures during the prenatal period and early childhood. Methods and results: CREW is funded by the NIH Environmental influences on Child Health Outcomes (ECHO) program, and consists of 12 individual cohorts and three additional scientific centers. The CREW study population is diverse in terms of race, ethnicity, geographical distribution, and year of recruitment. We hypothesize that there are phenotypes in childhood asthma that differ based on clinical characteristics and underlying molecular mechanisms. Furthermore, we propose that asthma endotypes and their defining biomarkers can be identified based on personal and early life environmental risk factors. CREW has three phases: 1) to pool and harmonize existing data from each cohort, 2) to collect new data using standardized procedures, and 3) to enroll new families during the prenatal period to supplement and enrich extant data and enable unified systems approaches for identifying asthma phenotypes and endotypes.
