    Variation of Basal EROD Activities in Ten Passerine Bird Species – Relationships with Diet and Migration Status

    Inter-specific differences in animal defence mechanisms against toxic substances are currently poorly understood. The ethoxyresorufin-O-deethylase (EROD) enzyme plays an important role in defence against toxic chemicals in a wide variety of animals, and it is an important biomarker for environmental contamination. We compared basal hepatic EROD activity levels among ten passerine species to see if there is inter-specific variation in enzyme activity, especially in relation to their diet and migration status. Migratory insectivores showed higher EROD activity compared to granivores. We hypothesize that the variable invertebrate diet of migratory insectivores contains a wider range of natural toxins than the narrower diet of granivores. This may have affected the evolution of mixed function oxidases (MFO) system and enzyme activities. We further tested whether metabolic rates or relative liver size were associated with the variation in detoxification capacity. We found no association between EROD activity and relative (per mass unit) basal metabolic rate (BMR). Instead, EROD activity and relative liver mass (% of body mass) correlated positively, suggesting that a proportionally large liver also functions efficiently. Our results suggest that granivores and non-migratory birds may be more vulnerable to environmental contaminants than insectivores and migratory birds. The diet and migration status, however, are phylogenetically strongly connected to each other, and their roles cannot be fully separated in our analysis with only ten passerine species

    Standardized voluntary force measurement in a lower extremity rehabilitation robot

    BACKGROUND: Isometric force measurements in the lower extremity are widely used in rehabilitation of subjects with neurological movement disorders (NMD) because walking ability has been shown to be related to muscle strength. Therefore muscle strength measurements can be used to monitor and control the effects of training programs. A new method to assess isometric muscle force was implemented in the driven gait orthosis (DGO) Lokomat. To evaluate the capabilities of this new measurement method, inter- and intra-rater reliability were assessed. METHODS: Reliability was assessed in subjects with and without NMD. Subjects were tested twice on the same day by two different therapists to test inter-rater reliability and on two separate days by the same therapist to test intra-rater reliability. RESULTS: Results showed fair to good reliability for the new measurement method to assess isometric muscle force of lower extremities. In subjects without NMD, intraclass correlation coefficients (ICC) for inter-rater reliability ranged from 0.72 to 0.97 and intra-rater reliability from 0.71 to 0.90. In subjects with NMD, ICC ranged from 0.66 to 0.97 for inter-rater and from 0.50 to 0.96 for intra-rater reliability. CONCLUSION: Inter- and intra- rater reliability of an assessment method for measuring maximal voluntary isometric muscle force of lower extremities was demonstrated. We suggest that this method is a valuable tool for documentation and controlling of the rehabilitation process in patients using a DGO

    The clinical features of the piriformis syndrome: a systematic review

    Piriformis syndrome, sciatica caused by compression of the sciatic nerve by the piriformis muscle, has been described for over 70 years; yet, it remains controversial. The literature consists mainly of case series and narrative reviews. The objectives of the study were: first, to make the best use of existing evidence to estimate the frequencies of clinical features in patients reported to have PS; second, to identify future research questions. A systematic review was conducted of any study type that reported extractable data relevant to diagnosis. The search included all studies up to 1 March 2008 in four databases: AMED, CINAHL, Embase and Medline. Screening, data extraction and analysis were all performed independently by two reviewers. A total of 55 studies were included: 51 individual and 3 aggregated data studies, and 1 combined study. The most common features found were: buttock pain, external tenderness over the greater sciatic notch, aggravation of the pain through sitting and augmentation of the pain with manoeuvres that increase piriformis muscle tension. Future research could start with comparing the frequencies of these features in sciatica patients with and without disc herniation or spinal stenosis

    Estimating and Modelling Bias of the Hierarchical Partitioning Public-Domain Software: Implications in Environmental Management and Conservation

    BACKGROUND: Hierarchical partitioning (HP) is an analytical method of multiple regression that identifies the most likely causal factors while alleviating multicollinearity problems. Its use is increasing in ecology and conservation by its usefulness for complementing multiple regression analysis. A public-domain software "hier.part package" has been developed for running HP in R software. Its authors highlight a "minor rounding error" for hierarchies constructed from >9 variables, however potential bias by using this module has not yet been examined. Knowing this bias is pivotal because, for example, the ranking obtained in HP is being used as a criterion for establishing priorities of conservation. METHODOLOGY/PRINCIPAL FINDINGS: Using numerical simulations and two real examples, we assessed the robustness of this HP module in relation to the order the variables have in the analysis. Results indicated a considerable effect of the variable order on the amount of independent variance explained by predictors for models with >9 explanatory variables. For these models the nominal ranking of importance of the predictors changed with variable order, i.e. predictors declared important by its contribution in explaining the response variable frequently changed to be either most or less important with other variable orders. The probability of changing position of a variable was best explained by the difference in independent explanatory power between that variable and the previous one in the nominal ranking of importance. The lesser is this difference, the more likely is the change of position. CONCLUSIONS/SIGNIFICANCE: HP should be applied with caution when more than 9 explanatory variables are used to know ranking of covariate importance. The explained variance is not a useful parameter to use in models with more than 9 independent variables. The inconsistency in the results obtained by HP should be considered in future studies as well as in those already published. Some recommendations to improve the analysis with this HP module are given

    Birds use structural properties when selecting materials for different parts of their nests

    Bird nests can have various roles but all act as the location for incubation, so at least have to serve to hold and support the incubating bird and its clutch of eggs. Nest construction is species specific and the use of materials varies between different parts of the nest. At present we know very little about the role that these materials play in the structural characteristics of the nest. This study examined materials from deconstructed nests from four species of thrush (Turdidae) and two species of finch (Fringillidae) that all constructed nests made of woody stems. It was hypothesised that structural properties would vary within the different regions of a nest, with thicker and stronger materials being found in parts of the nest needing the most support. Secondly, it was predicted that structural properties would vary little between nests of members of the same family, but would be quite different between nests of different families. Nests were deconstructed to quantify the materials used in the cup lining, and the upper and lower parts of the outer nest. The 20 thickest pieces of material were selected from each nest part and for each piece, and their diameter and mass quantified. Each piece was then subjected to a three-point bending test using an Instron universal testing machine to determine its rigidity and bending strength. Placement of materials in the nest was non-random in all species. The materials used in the outer part of the nest were thicker, stronger and stiffer than those materials found in the cup lining. The extent to which these structural properties varied between families depended on where the material was taken from the nest. Both strength and rigidity strongly positively correlated with the diameter of the piece of material. We hypothesise that birds are not directly aware of the structural properties of the material per se but rather assess diameter and mass of the material when they pick it up by the bill. Using this information they decide on whether the piece is suitable for that appropriate stage of nest construction

    Levels of DNA methylation vary at CpG sites across the BRCA1 promoter, and differ according to triple negative and "BRCA-like" status, in both blood and tumour DNA

    Triple negative breast cancer is typically an aggressive and difficult to treat subtype. It is often associated with loss of function of the BRCA1 gene, either through mutation, loss of heterozygosity or methylation. This study aimed to measure methylation of the BRCA1 gene promoter at individual CpG sites in blood, tumour and normal breast tissue, to assess whether levels were correlated between different tissues, and with triple negative receptor status, histopathological scoring for BRCA-like features and BRCA1 protein expression. Blood DNA methylation levels were significantly correlated with tumour methylation at 9 of 11 CpG sites examined (p<0.0007). The levels of tumour DNA methylation were significantly higher in triple negative tumours, and in tumours with high BRCA-like histopathological scores (10 of 11 CpG sites; p<0.01 and p<0.007 respectively). Similar results were observed in blood DNA (6 of 11 CpG sites; p<0.03 and 7 of 11 CpG sites; p<0.02 respectively). This study provides insight into the pattern of CpG methylation across the BRCA1 promoter, and supports previous studies suggesting that tumours with BRCA1 promoter methylation have similar features to those with BRCA1 mutations, and therefore may be suitable for the same targeted therapies

    Low-cost, deep-sea imaging and analysis tools for deep-sea exploration: a collaborative design study

    A minuscule fraction of the deep sea has been scientifically explored and characterized due to several constraints, including expense, inefficiency, exclusion, and the resulting inequitable access to tools and resources around the world. To meet the demand for understanding the largest biosphere on our planet, we must accelerate the pace and broaden the scope of exploration by adding low-cost, scalable tools to the traditional suite of research assets. Exploration strategies should increasingly employ collaborative, inclusive, and innovative research methods to promote inclusion, accessibility, and equity to ocean discovery globally. Here, we present an important step toward this new paradigm: a collaborative design study on technical capacity needs for equitable deep-sea exploration. The study focuses on opportunities and challenges related to low-cost, scalable tools for deep-sea data collection and artificial intelligence-driven data analysis. It was conducted in partnership with twenty marine professionals worldwide, covering a broad representation of geography, demographics, and domain knowledge within the ocean space. The results of the study include a set of technical requirements for low-cost deep-sea imaging and sensing systems and automated image and data analysis systems. As a result of the study, a camera system called Maka Niu was prototyped and is being field-tested by thirteen interviewees and an online AI-driven video analysis platform is in development. We also identified six categories of open design and implementation questions highlighting participant concerns and potential trade-offs that have not yet been addressed within the scope of the current projects but are identified as important considerations for future work. Finally, we offer recommendations for collaborative design projects related to the deep sea and outline our future work in this space

    Outcomes of Brood Parasite–Host Interactions Mediated by Egg Matching: Common Cuckoos Cuculus canorus versus Fringilla Finches

    Antagonistic species often interact via matching of phenotypes, and interactions between brood parasitic common cuckoos (Cuculus canorus) and their hosts constitute classic examples. The outcome of a parasitic event is often determined by the match between host and cuckoo eggs, giving rise to potentially strong associations between fitness and egg phenotype. Yet, empirical efforts aiming to document and understand the resulting evolutionary outcomes are in short supply.We used avian color space models to analyze patterns of egg color variation within and between the cuckoo and two closely related hosts, the nomadic brambling (Fringilla montifringilla) and the site fidelic chaffinch (F. coelebs). We found that there is pronounced opportunity for disruptive selection on brambling egg coloration. The corresponding cuckoo host race has evolved egg colors that maximize fitness in both sympatric and allopatric brambling populations. By contrast, the chaffinch has a more bimodal egg color distribution consistent with the evolutionary direction predicted for the brambling. Whereas the brambling and its cuckoo host race show little geographical variation in their egg color distributions, the chaffinch's distribution becomes increasingly dissimilar to the brambling's distribution towards the core area of the brambling cuckoo host race.High rates of brambling gene flow is likely to cool down coevolutionary hot spots by cancelling out the selection imposed by a patchily distributed cuckoo host race, thereby promoting a matching equilibrium. By contrast, the site fidelic chaffinch is more likely to respond to selection from adapting cuckoos, resulting in a markedly more bimodal egg color distribution. The geographic variation in the chaffinch's egg color distribution could reflect a historical gradient in parasitism pressure. Finally, marked cuckoo egg polymorphisms are unlikely to evolve in these systems unless the hosts evolve even more exquisite egg recognition capabilities than currently possessed
