    General Relativistic 1+3 Orthonormal Frame Approach Revisited

    The equations of the 1+3 orthonormal frame approach are explicitly presented and discussed. Natural choices of local coordinates are mentioned. A dimensionless formulation is subsequently given. It is demonstrated how one can obtain a number of interesting problems by specializing the general equations. In particular, equation systems for ``silent'' dust cosmological models also containing magnetic Maxwell fields, locally rotationally symmetric spacetime geometries and spatially homogeneous cosmological models are presented. We show that while the 3-Cotton--York tensor is zero for Szekeres dust models, it is nonzero for a generic representative within the ``silent'' class.Comment: 41 pages, uufiles encoded postscript file, submitted to Phys.     HSD3B1 genotype identifies glucocorticoid responsiveness in severe asthma

    Asthma resistance to glucocorticoid treatment is a major health problem with unclear etiology. Glucocorticoids inhibit adrenal androgen production. However, androgens have potential benefits in asthma. HSD3B1 encodes for 3β-hydroxysteroid dehydrogenase-1 (3β-HSD1), which catalyzes peripheral conversion from adrenal dehydroepiandrosterone (DHEA) to potent androgens and has a germline missense-encoding polymorphism. The adrenal restrictive HSD3B1(1245A) allele limits conversion, whereas the adrenal permissive HSD3B1(1245C) allele increases DHEA metabolism to potent androgens. In the Severe Asthma Research Program (SARP) III cohort, we determined the association between DHEA-sulfate and percentage predicted forced expiratory volume in 1 s (FEV1PP). HSD3B1(1245) genotypes were assessed, and association between adrenal restrictive and adrenal permissive alleles and FEV1PP in patients with (GC) and without (noGC) daily oral glucocorticoid treatment was determined (n = 318). Validation was performed in a second cohort (SARP I&II; n = 184). DHEA-sulfate is associated with FEV1PP and is suppressed with GC treatment. GC patients homozygous for the adrenal restrictive genotype have lower FEV1PP compared with noGC patients (54.3% vs. 75.1%; P < 0.001). In patients with the homozygous adrenal permissive genotype, there was no FEV1PP difference in GC vs. noGC patients (73.4% vs. 78.9%; P = 0.39). Results were independently confirmed: FEV1PP for homozygous adrenal restrictive genotype in GC vs. noGC is 49.8 vs. 63.4 (P < 0.001), and for homozygous adrenal permissive genotype, it is 66.7 vs. 67.7 (P = 0.92). The adrenal restrictive HSD3B1(1245) genotype is associated with GC resistance. This effect appears to be driven by GC suppression of 3β-HSD1 substrate. Our results suggest opportunities for prediction of GC resistance and pharmacologic intervention

    Solubility and Permeation of Hydrogen Sulfide in Lipid Membranes

    Hydrogen sulfide (H2S) is mainly known for its toxicity but has recently been shown to be produced endogenously in mammalian tissues and to be associated with physiological regulatory functions. To better understand the role of biomembranes in modulating its biological distribution and effects; we measured the partition coefficient of H2S in models of biological membranes. The partition coefficients were found to be 2.1±0.2, 1.9±0.5 and 2.0±0.6 in n-octanol, hexane and dilauroylphosphatidylcholine liposome membranes relative to water, respectively (25°C). This two-fold higher concentration of H2S in the membrane translates into a rapid membrane permeability, Pm = 3 cm s−1. We used a mathematical model in three dimensions to gain insight into the diffusion of total sulfide in tissues. This model shows that the sphere of action of sulfide produced by a single cell expands to involve more than 200 neighboring cells, and that the resistance imposed by lipid membranes has a significant effect on the diffusional spread of sulfide at pH 7.4, increasing local concentrations. These results support the role of hydrogen sulfide as a paracrine signaling molecule and reveal advantageous pharmacokinetic properties for its therapeutic applications

    Critical Exponents, Hyperscaling and Universal Amplitude Ratios for Two- and Three-Dimensional Self-Avoiding Walks

    We make a high-precision Monte Carlo study of two- and three-dimensional self-avoiding walks (SAWs) of length up to 80000 steps, using the pivot algorithm and the Karp-Luby algorithm. We study the critical exponents ν\nu and 2Δ4γ2\Delta_4 -\gamma as well as several universal amplitude ratios; in particular, we make an extremely sensitive test of the hyperscaling relation dν=2Δ4γd\nu = 2\Delta_4 -\gamma. In two dimensions, we confirm the predicted exponent ν=3/4\nu = 3/4 and the hyperscaling relation; we estimate the universal ratios  / =0.14026±0.00007\ / \ = 0.14026 \pm 0.00007,  / =0.43961±0.00034\ / \ = 0.43961 \pm 0.00034 and Ψ=0.66296±0.00043\Psi^* = 0.66296 \pm 0.00043 (68\% confidence limits). In three dimensions, we estimate ν=0.5877±0.0006\nu = 0.5877 \pm 0.0006 with a correction-to-scaling exponent Δ1=0.56±0.03\Delta_1 = 0.56 \pm 0.03 (subjective 68\% confidence limits). This value for ν\nu agrees excellently with the field-theoretic renormalization-group prediction, but there is some discrepancy for Δ1\Delta_1. Earlier Monte Carlo estimates of ν\nu, which were  ⁣0.592\approx\! 0.592, are now seen to be biased by corrections to scaling. We estimate the universal ratios  / =0.1599±0.0002\ / \ = 0.1599 \pm 0.0002 and Ψ=0.2471±0.0003\Psi^* = 0.2471 \pm 0.0003; since Ψ>0\Psi^* > 0, hyperscaling holds. The approach to Ψ\Psi^* is from above, contrary to the prediction of the two-parameter renormalization-group theory.     Delegation and coordination with multiple threshold public goods: experimental evidence

    When multiple charities, social programs and community projects simultaneously vie for funding, donors risk mis-coordinating their contributions leading to an inefficient distribution of funding across projects. Community chests and other intermediary organizations facilitate coordination among donors and reduce such risks. To study this, we extend a threshold public goods framework to allow donors to contribute through an intermediary rather than directly to the public goods. Through a series of experiments, we show that the presence of an intermediary increases public good success and subjects’ earnings only when the intermediary is formally committed to direct donations to socially beneficial goods.
