    Variable readthrough responsiveness of nonsense mutations in hemophilia A

    Readthrough therapy relies on the use of small molecules that enable premature termination codons in mRNA open reading frames to be misinterpreted by the translation machinery, thus allowing the generation of full-length, potentially functional proteins from mRNA carrying nonsense mutations. In patients with hemophilia A, nonsense mutations potentially sensitive to readthrough agents represent approximately 16% of the point mutations. The aim of this study was to measure the readthrough effect of different compounds and to analyze the influence of premature termination codon context in selected nonsense mutations causing hemophilia A. To this end, primary fibroblasts from three patients with hemophilia A caused by nonsense mutations (p.W1586X, p.Q1636X and p.R1960X) and Chinese hamster ovary (CHO) cells transfected with 12 different plasmids encoding mutated F8 (p.Q462X, p.Q1705X, p.Q1764X, p.W274X, p.W1726X, p.W2015X, p.W2131X, p.R1715X, p.R1822X, p.R1960X, p.R2071X and p.R2228X) were treated with gentamicin, geneticin, PTC124, RTC13 or RTC14. Responses were assessed by analyzing not only F8 mRNA expression and FVIII biosynthesis (FVIII antigen by ELISA, western blot and immunofluorescence) but also the FVIII activity (by chromogenic assay). In the patients' fibroblasts, readthrough agents neither stabilized F8 mRNA nor increased FVIII protein or activity to detectable levels. In CHO cells, only in five of the 12 F8 variants, readthrough treatment increased both FVIII antigen and activity levels, which was associated with a reduction in intracellular accumulation of truncated forms and an increase in full-length proteins. These results provide experimental evidence of genetic context dependence of nonsense suppression by readthrough agents and of factors predicting responsiveness

    Estudio del comportamiento de pinturas acrílicas sobre tablero contrachapado tras envejecimiento artificial

    [ES] Entre los soportes más empleados en arte contemporáneo se encuentran los tableros derivados de la madera. Hasta el momento, las investigaciones para determinar la influencia de un soporte de tablero contrachapado en capas pictóricas acrílicas así como las condiciones óptimas para su conservación, resultan insuficientes o completamente ausentes. En la elaboración de estos tableros se utilizan adhesivos para darles consistencia pero la influencia de estos adhesivos en las capas pictóricas artísticas estará en función del tipo de tablero, del tipo de adhesivo empleado, de las condiciones ambientales y de las características de las capas pictóricas. Es evidente pues, la necesidad de modelos comparativos. Por todos estos motivos, el objetivo principal de esta investigación ha sido comprender la transformación que experimentan estos materiales durante su envejecimiento y las causas que pueden desestabilizar las obras, con el fin de adoptar las medidas necesarias para evitar un envejecimiento prematuro. Mediante la Espectroscopía Infrarroja por Transformada de Fourier FTIR se han analizado los cambios sufridos por modelos experimentales realizados con pinturas acrílicas imprimidas sobre tablero contrachapado tras ser sometidos a ciclos de envejecimiento por choque térmico (Cold-check test) y envejecimiento en condiciones de alta humedad.[EN] Among the most used stands in contemporary art are wood-derived boards. To date, research to determine the influence of a plywood backing on acrylic pictorial layers as well as optimal conditions for its preservation is insufficient or completely absent. In the production of these boards adhesives are used to give them consistency, but the influence of these adhesives on artistic pictorial layers will depend on the type of panel, the type of adhesive used, the environmental conditions and the characteristics of the pictorial layers. Obviously, the need for comparative models. For all these reasons, the main objective of this research has been to understand the transformation experienced by these materials during their aging and the causes that can destabilize the works, in order to take the necessary measures to avoid premature aging.     The recent neophyte Opuntia aurantiaca (Cactaceae): distribution and potential invasion in the Iberian Peninsula

    The Cactaceae, and especially its most emblematic genus, Opuntia, is one of the groups of plants with greater invasion potential in the Iberian Peninsula. One of the most recently detected species is Opuntia aurantiaca, a small cactus with an enormous capacity of dispersion. Probably native to Argentina and Uruguay, it behaves as a very aggressive invader in Australia and South Africa. In Europe, it only occurs on the Mediterranean coast of the Iberian Peninsula (Catalonia and Valencian Community). In this study, the geographic range of the species is accurately delineated at the peninsular level. Detected firstly at the beginning of the last decade in Navajas (Castelló Province), it has been subsequently observed in other places of Castelló, but also in Valencia, Tarragona and Barcelona, and since 2017 in Girona. With all gathered occurrence data, the potential distribution of O. aurantiaca is estimated (for the current climatic conditions as well as for different scenarios of global warming).     Los superhéroes como herramienta para el aprendizaje: la ciencia ficción llega a las aulas

    [EN] The innovation and results obtained from designing and applying a survey based on science fiction superheroes are presented as a leveling instrument regarding the association of legal concepts and rights with the chosen characters. The innovation is applied to different degrees of the Polytechnic University of Valencia. Characters well known to the students are chosen and the results obtained show us the ability to associate fundamental rights, as well as branches of the right to the characters, showing that the students perfectly differentiate and correctly associate the concepts. It is a gamification tool in the classroom based on science fiction that is very attractive for students and that can be extrapolated to any legal discipline and at any University for the assessment of knowledge by students.[ES] Se presenta la innovación y resultados obtenidos de diseñar y aplicar una encuesta basada en superhéroes de ciencia ficción como instrumento de nivelación referente a la asociación de conceptos jurídicos y derechos con los personajes elegidos. La innovación se aplica a distintas titulaciones de la Universitat Politècnica de València. Se eligen personajes muy conocidos por el alumnado. Los resultados obtenidos nos demuestran la capacidad de asociar derechos fundamentales, así como ramas del derecho a los personajes, evidenciando que el alumnado diferencia perfectamente y asocia de forma correcta los conceptos.     The application of gamified dynamics related to science fiction in legal learning in the Degree in Food Science and Technology of the Polytechnic University of Valencia

    [EN] The present work presents the strategy and the results obtained in a teaching innovation developed within the Educational Innovation and Improvement Project (PIMEs) «Gamification and ICTs: design of audiovisual activities based on science fiction for the dynamization of teachers in a face-to-face environment, blended and virtual» in which various activities are designed for legal learning in the Degree in Food Science and Technology, at the Universitat Politècnica de València. Case study approach through films, as well as the design of a card game and the use of futuristic food paintings allow the students to apply the corresponding legislation. The results obtained are very positive, due to the ability to associate images with the applicable legislation, in addition to being attractive activities for students and increasing their participation in the classroom, developing transversal competences.[ES] En el presente trabajo se presenta la estrategia y los resultados obtenidos en una innovación docente desarrollada dentro del Proyecto de Innovación y Mejora Educativa (PIMEs) «Gamificación y TICs: diseño de actividades audiovisuales basadas en la ciencia ficción para la dinamización docente en un entorno presencial, semipresencial y virtual» en el que se diseñan diversas actividades para el aprendizaje jurídico en la titulación del Grado en Ciencia y Tecnología de Alimentos, en la Universitat Politècnica de València.     Endoscopic treatment (endoscopic balloon dilation/self-expandable metal stent) vs surgical resection for the treatment of de novo stenosis in Crohn's disease (ENDOCIR study): an open-label, multicentre, randomized trial.

    Background: Stenosis is one of the most common complications in patients with Crohn's disease (CD). Endoscopic balloon dilation (EBD) is the treatment of choice for a short stenosis adjacent to the anastomosis from previous surgery. Self-expandable metal stents (SEMS) may be a suitable treatment option for longer stenoses. To date, however, there is no scientific evidence as to whether endoscopic (EBD/SEMS) or surgical treatment is the best approach for de novo or primary stenoses that are less than 10 cm in length. Methods/design: Exploratory study as "proof-of-concept", multicentre, open-label, randomized trial of the treatment of de novo stenosis in the CD; endoscopic treatment (EBD/SEMS) vs surgical resection (SR). The type of endoscopic treatment will initially be with EDB; if a therapeutic failure occurs, then a SEMS will be placed. We estimate 2 years of recruitment and 1 year of follow-up for the assessment of quality of life, costs, complications, and clinical recurrence. After the end of the study, patients will be followed up for 3 years to re-evaluate the variables over the long term. Forty patients with de novo stenosis in CD will be recruited from 15 hospitals in Spain and will be randomly assigned to the endoscopic or surgical treatment groups. The primary aim will be the evaluation of the patient quality of life at 1 year follow-up (% of patients with an increase of 30 points in the 32-item Inflammatory Bowel Disease Questionnaire (IBDQ-32). The secondary aim will be evaluation of the clinical recurrence rate, complications, and costs of both treatments at 1-year follow-up.     Study of behaviour on simulated daylight ageing of artists¿ acrylic and poly(vinyl acetate) paint films

    [EN] This work proposes a multi-method approach that combines advanced microscopy (SEM/EDX, AFM) and spectroscopy (UV-vis and FTIR) techniques. This approach not only characterises the behaviour of the additives of two commercial poly(vinyl acetate) (PVAc) and acrylic emulsion paints but also simultaneously characterises the changes in chemical composition and morphology observed in the paint films as a result of ageing due to the paints being exposed to an intense source of simulated daylight. In parallel, a series of mechanical tests were performed that correlate the chemical changes in composition and the changes observed in the films' mechanical properties. This work was a comparative study between both types of acrylic and PVAc paints.     On-line database of voltammetric data of immobilized particles for identifying pigments and minerals in archaeometry, conservation and restoration (ELCHER database)

    [EN] A web-based database of voltammograms is presented for characterizing artists' pigments and corrosion products of ceramic, stone and metal objects by means of the voltammetry of immobilized particles methodology. Description of the website and the database is provided. Voltammograms are, in most cases, accompanied by scanning electron microphotographs, X-ray spectra, infrared spectra acquired in attenuated total reflectance Fourier transform infrared spectroscopy mode (ATR-FTIR) and diffuse reflectance spectra in the UV-Vis-region. For illustrating the usefulness of the database two case studies involving identification of pigments and a case study describing deterioration of an archaeological metallic object are presented.
