    Spatial distribution of the most abundant sea urchin populations on the southeast coast of SĂŁo Paulo (Brazil)

    Se investigó la variación espacial en la abundancia de Echinometra lucunter, Lytechinus variegatus y Arbacia lixula con respecto a cuatro escalas espaciales (105, 104, 103 y 102 m) y dos profundidades (0–3 y 3–8 m), a lo largo de 100 km del litoral de São Paulo (agosto a octubre de 1996). Echinometra lucunter fue la especie más abundante, prefiriendo los fondos más someros bajo el efecto del oleaje. Se identificó un gradiente en su abundancia, aumentando en dirección del suroeste al noreste. Arbacia lixula mostró el mismo gradiente, pero con una mayor abundancia en el sector más profundo. Ambas especies mostraron diferencias significativas a una escala espacial de centenares de metros y E. lucunter también a una escala de kilómetros. Lytechinus variegatus mostró un patrón en mosaico, siendo abundante en ciertos sitios. Las variaciones en la calidad del agua y en la heterogeneidad natural del hábitat podrían explicar de forma conjunta la distribución espacial de estas poblaciones.The spatial variability of Echinometra lucunter, Lytechinus variegatus and Arbacia lixula was studied in relation to four spatial scales (105, 104, 103 and 102 m) and two depths (0–3 and 3–8 m), along 100 km of the São Paulo coastline (August to October 1996). Echinometra lucunter was the most abundant species, with preference for shallow substrates subject to wave action. An abundance gradient was identified, increasing from southwest to northeast. Arbacia lixula showed the same pattern of spatial variation, but was more abundant in the deeper areas. Both species showed significant differences on a spatial scale of hundreds of meters, and E. lucunter also on a scale of kilometers. Lytechinus variegates presented a patchy distribution, being more abundant at certain sites.     Safflower oil: an integrated assessment of phytochemistry, antiulcerogenic activity, and rodent and environmental toxicity

    Gastric ulcers are a significant medical problem and the development of complications lead to significant mortality rates worldwide. In Brazil, Carthamus tinctorius L., Asteraceae, seeds essential oil, the safflower oil, is currently used as a thermogenic compound and as treatment for problems related to the cardiovascular system. In this study, by Raman spectroscopy, it was shown that oleic and linoleic acids are the compounds present in higher concentrations in the safflower oil. We demonstrated that safflower oil (750 mg/kg, p.o.) decrease the ulcerogenic lesions in mice after the administration of hydrochloric acid-ethanol. The gastric ulcers induced by non-steroidal anti-inflammatory drug (NSAID) in mice treated with cholinomimetics were treated with four different doses of safflower oil, of which, the dose of 187.5 mg/kg (p.o.) showed significant antiulcerogenic properties (**p < 0.01). Moreover, the safflower oil at doses of 187.5 mg/kg (i.d.) increased the pH levels, gastric volume (**p < 0.01) and gastric mucus production (***p < 0.001), and decreased the total gastric acid secretion (***p < 0.001). The acute toxicity tests showed that safflower oil (5.000 mg/kg, p.o.) had no effect on mortality or any other physiological parameter. Ecotoxicological tests performed using Daphnia similis showed an EC50 at 223.17 mg/l, and therefore safflower oil can be considered “non-toxic” based on the directive 93/67/EEC on risk assessment for new notified substances by European legislation.     Fluoxetine can make marine organisms unhappy: a study on the sub-lethal effects on marine invertebrates

    The environmental effects caused by selective serotonin reuptake inhibitor drugs have been investigated for marine organisms and coastal ecosystems but are scarce in neotropical organisms. This investigation aimed to evaluate the sublethal effects of fluoxetine on the embryonic development of the sea urchin Echinometra lucunter and the survival and swimming behavior of the brine shrimp Artemia sp. The organisms were exposed to four different concentrations of fluoxetine (30, 300, 3000 and 30000 ng L-1) and to a negative control (filtered seawater), following the respective standard testing protocols. We verified a significant reduction of the embryos development to pluteus larvae, starting from 3000 ng L-1 (54.0±10.9% normal larvae), in comparison with the controls (83.5±3.1%). The non-observed effect concentration (NOEC) was estimated at 300 ng L-1, and the lowest observed effect concentration (LOEC) was 3000 ng L-1. In the behavior tests with Artemia sp, no significant adverse effects were reported for mobility, swimming speed and inactivity time. These results show that Fluoxetine can interfere on the development of species like the sea urchin E. lucunter, but short term exposure did not affected the swimming behavior of the brine shrimp Artemia sp. Fluoxetine presents thus a potential to affect marine biota and disrupt the equilibrium of the coastal ecosystems

    Evolutionary and Experimental Assessment of Novel Markers for Detection of Xanthomonas euvesicatoria in Plant Samples

    BACKGROUND: Bacterial spot-causing xanthomonads (BSX) are quarantine phytopathogenic bacteria responsible for heavy losses in tomato and pepper production. Despite the research on improved plant spraying methods and resistant cultivars, the use of healthy plant material is still considered as the most effective bacterial spot control measure. Therefore, rapid and efficient detection methods are crucial for an early detection of these phytopathogens. METHODOLOGY: In this work, we selected and validated novel DNA markers for reliable detection of the BSX Xanthomonas euvesicatoria (Xeu). Xeu-specific DNA regions were selected using two online applications, CUPID and Insignia. Furthermore, to facilitate the selection of putative DNA markers, a customized C program was designed to retrieve the regions outputted by both databases. The in silico validation was further extended in order to provide an insight on the origin of these Xeu-specific regions by assessing chromosomal location, GC content, codon usage and synteny analyses. Primer-pairs were designed for amplification of those regions and the PCR validation assays showed that most primers allowed for positive amplification with different Xeu strains. The obtained amplicons were labeled and used as probes in dot blot assays, which allowed testing the probes against a collection of 12 non-BSX Xanthomonas and 23 other phytopathogenic bacteria. These assays confirmed the specificity of the selected DNA markers. Finally, we designed and tested a duplex PCR assay and an inverted dot blot platform for culture-independent detection of Xeu in infected plants. SIGNIFICANCE: This study details a selection strategy able to provide a large number of Xeu-specific DNA markers. As demonstrated, the selected markers can detect Xeu in infected plants both by PCR and by hybridization-based assays coupled with automatic data analysis. Furthermore, this work is a contribution to implement more efficient DNA-based methods of bacterial diagnostics

    Single domain antibodies: promising experimental and therapeutic tools in infection and immunity

    Antibodies are important tools for experimental research and medical applications. Most antibodies are composed of two heavy and two light chains. Both chains contribute to the antigen-binding site which is usually flat or concave. In addition to these conventional antibodies, llamas, other camelids, and sharks also produce antibodies composed only of heavy chains. The antigen-binding site of these unusual heavy chain antibodies (hcAbs) is formed only by a single domain, designated VHH in camelid hcAbs and VNAR in shark hcAbs. VHH and VNAR are easily produced as recombinant proteins, designated single domain antibodies (sdAbs) or nanobodies. The CDR3 region of these sdAbs possesses the extraordinary capacity to form long fingerlike extensions that can extend into cavities on antigens, e.g., the active site crevice of enzymes. Other advantageous features of nanobodies include their small size, high solubility, thermal stability, refolding capacity, and good tissue penetration in vivo.     Geoeconomic variations in epidemiology, ventilation management, and outcomes in invasively ventilated intensive care unit patients without acute respiratory distress syndrome: a pooled analysis of four observational studies

    Background: Geoeconomic variations in epidemiology, the practice of ventilation, and outcome in invasively ventilated intensive care unit (ICU) patients without acute respiratory distress syndrome (ARDS) remain unexplored. In this analysis we aim to address these gaps using individual patient data of four large observational studies. Methods: In this pooled analysis we harmonised individual patient data from the ERICC, LUNG SAFE, PRoVENT, and PRoVENT-iMiC prospective observational studies, which were conducted from June, 2011, to December, 2018, in 534 ICUs in 54 countries. We used the 2016 World Bank classification to define two geoeconomic regions: middle-income countries (MICs) and high-income countries (HICs). ARDS was defined according to the Berlin criteria. Descriptive statistics were used to compare patients in MICs versus HICs. The primary outcome was the use of low tidal volume ventilation (LTVV) for the first 3 days of mechanical ventilation. Secondary outcomes were key ventilation parameters (tidal volume size, positive end-expiratory pressure, fraction of inspired oxygen, peak pressure, plateau pressure, driving pressure, and respiratory rate), patient characteristics, the risk for and actual development of acute respiratory distress syndrome after the first day of ventilation, duration of ventilation, ICU length of stay, and ICU mortality. Findings: Of the 7608 patients included in the original studies, this analysis included 3852 patients without ARDS, of whom 2345 were from MICs and 1507 were from HICs. Patients in MICs were younger, shorter and with a slightly lower body-mass index, more often had diabetes and active cancer, but less often chronic obstructive pulmonary disease and heart failure than patients from HICs. Sequential organ failure assessment scores were similar in MICs and HICs. Use of LTVV in MICs and HICs was comparable (42\ub74% vs 44\ub72%; absolute difference \u20131\ub769 [\u20139\ub758 to 6\ub711] p=0\ub767; data available in 3174 [82%] of 3852 patients). The median applied positive end expiratory pressure was lower in MICs than in HICs (5 [IQR 5\u20138] vs 6 [5\u20138] cm H2O; p=0\ub70011). ICU mortality was higher in MICs than in HICs (30\ub75% vs 19\ub79%; p=0\ub70004; adjusted effect 16\ub741% [95% CI 9\ub752\u201323\ub752]; p&lt;0\ub70001) and was inversely associated with gross domestic product (adjusted odds ratio for a US$10 000 increase per capita 0\ub780 [95% CI 0\ub775\u20130\ub786]; p&lt;0\ub70001). Interpretation: Despite similar disease severity and ventilation management, ICU mortality in patients without ARDS is higher in MICs than in HICs, with a strong association with country-level economic status.
