    Rising water flow as a factor of organic material importation into caves

    The Carlúcio Cave is located on the left bank of the Peruaçu River canyon, in a National Park located in the semiarid zone of Minas Gerais, Southeastern Brazil. Although the Carlúcio Cave is in an unfavorable relief position in terms of material importation into its interior, it harbors an important fossil deposit. This paper sought to investigate such fossil assemblage and describe material importation and deposition events. For that purpose, a survey of the existing fossils was conducted, including a description of the deposition sites and taphonomic signatures. A topographic survey was carried out of representative samples and the deposit chronology was established by means of carbon-14 and uranium-series dating. The fossil assemblage comprises mainly terrestrial epigeal mollusk shells, totaling 1,399 individuals, 70% of which are Drymaeus sp. (Bulimuloidea, Bulimulidae). In addition, 20 bones, 57 vegetal deposition sites, four charcoals, one palm fruit and one corncob were recorded. The cave location and morphology and the deposit’s taphonomic signature indicate that the material was carried upward by the rise in water level (WL) of the Peruaçu River due to episodic barriers to the natural river flow, followed by floating and retention of the material once the WL went gradually down. At least four flood events that reached the Carlúcio Cave were identified in the Peruaçu River canyon during the Holocene. This study provides evidence of a mechanism of material transport and accumulation in the cave caused by the rising water flow in connection with flood events in the river canyon.Key words: Cave deposits, Holocene chronology, fluvial geomorphology, Peruaçu River canyon.Naraščanje nivoja vode kot faktor vnosa organskega materiala v jameJama Carlúcio je na levem bregu kanjona reke Peruaçu v nacionalnem parku na polpuščavskem območju province Minas Gerais v jugovzhodni Braziliji. Čeprav je jama v neugodnem reliefnem položaju glede vnosa materiala, v njej ležijo pomembne fosilne najdbe. Ta članek je poskušal raziskati fosile in opisati dogodke vnosa in odlaganja materiala. Zato smo pregledali obstoječe fosile ter med drugim opisali odlagališča in tafonomske podpise. Izvedli smo topografsko raziskavo reprezentativnih vzorcev ter z določanjem starosti z ogljikom C14 in uranovo serijo vzpostavili depozitno kronologijo. Fosilni ostanki obsegajo predvsem kopenske epigealne mehkužce s 1399 posamezniki, od tega 70 % Drymaeus sp. (Bulimuloidea, Bulimulidae). Poleg tega smo opazili 20 kosti, 57 rastlinskih tališč, štiri oglja, eno palmovo sadje in eno koruzo. Lokacija jame, njena morfologija in tafonomske raziskave sedimentov nakazujejo, da je material odneslo navzgor s porastom nivoja vode reke Peruaçu zaradi epizodnih ovir pri naravnem toku reke. Temu sledi lebdenje in zadrževanje snovi, ko je nivo postopoma upadel. V holocenu smo v kanjonu reke Peruaçu identificirali vsaj štiri poplavne dogodke, ki so segli v jamo Carlúcio. Ta študija dokazuje mehanizem materialnega transporta in akumulacije v jami, ki jo povzroča naraščajoči nivo vode v povezavi s poplavnimi dogodki v rečnem kanjonu.Ključne besede: jamski sedimenti, holocenska kronologija, rečna geomorfologija, kanjon reke Peruçu

    Periodical reactivation under the effect of caffeine attenuates fear memory expression in rats

    In the last decade, several studies have shown that fear memories can be attenuated by interfering with reconsolidation. However, most of the pharmacological agents used in preclinical studies cannot be administered to humans. Caffeine is one of the world’s most popular psychoactive drugs and its effects on cognitive and mood states are well documented. Nevertheless, the influence of caffeine administration on fear memory processing is not as clear. We employed contextual fear conditioning in rats and acute caffeine administration under a standard memory reconsolidation protocol or periodical memory reactivation. Additionally, potential rewarding/aversion and anxiety effects induced by caffeine were evaluated by conditioning place preference or open field, respectively. Caffeine administration was able to attenuate weak fear memories in a standard memory reconsolidation protocol; however, periodical memory reactivation under caffeine effect was necessary to attenuate strong and remote memories. Moreover, caffeine promoted conditioned place preference and anxiolytic-like behavior, suggesting that caffeine weakens the initial learning during reactivation through counterconditioning mechanisms. Thus, our study shows that rewarding and anxiolytic effects of caffeine during fear reactivation can change the emotional valence of fear memory. It brings a new promising pharmacological approach based on drugs widely used such as caffeine to treat fear-related disorders

    Human helminth co-infection: no evidence of common genetic control of hookworm and Schistosoma mansoni infection intensity in a Brazilian community.

    Strong statistical associations between soil-transmitted helminths and schistosomes are frequently observed in co-endemic human populations, although the underlying explanations remain poorly understood. This study investigates the contribution of host genetics and domestic environment to hookworm and Schistosoma mansoni infection intensity and evaluates the role of genetic and non-genetic factors in co-variation of infection intensity. Detailed genealogical information allowed assignment of 1303 individuals living in the Brazilian community of Americaninhas, Minas Gerais state, to 25 pedigrees (containing between two and 1159 members) residing in 303 households. The prevalence of co-infection with both hookworms and schistosomes was high (38.5%), with significant correlation between Necator americanus and S. mansoni faecal egg counts. Bivariate variance component analysis demonstrated a modest but significant species-specific heritability for intensity of N. americanus (h(2)=0.196) and S. mansoni infection (h(2)=0.230). However, after accounting for demographic, socio-economic and household risk factors, no evidence for common genetic control of intensity of hookworm and schistosome infection was observed. There was some evidence for residual clustering within households but the majority (63%) of the covariance between N. americanus and S. mansoni infection intensity remained specific to the individual and could not be explained by shared genes, shared environment or other shared demographic, socio-economic or environmental risk factors. Our results emphasize the importance of exposure to hookworm and schistosome infection in driving the association between levels of infection with these species in hosts resident in areas of high transmission and suggest that much of this common exposure occurs outside the home

    Influence of environmental factors on tenuazonic acid production by Epicoccum sorghinum: An integrative approach of field and laboratory conditions

    Sorghum is the fifth most cultivated and consumed grain in the world. However, this grain is frequently contaminated with toxins from fungi. The present study evaluated the effects of environmental factors on tenuazonic acid (TeA) production by Epicoccum sorghinum in the field and in controlled laboratory conditions. In this study, 50 sorghum grain samples were collected from summer and autumn growing seasons and analyzed for TeA contamination using LC-MS/MS. To further understand the ecophysiology of this fungus, an isolated strain of E. sorghinum from the field was investigated for its development and TeA production under controlled environmental conditions in the laboratory. In the ecophysiological investigation, the effects of water activity (0.90, 0.95, 0.99) and temperature (18, 22, 26 and 30?°C) were evaluated on the radial growth, enzymatic production and expression of TAS1, which is the gene involved in TeA production. Results showed that in the field, the summer season presented the highest TeA average level in the grains (587.8??g/kg) compared to level found in the autumn (440.5??g/kg). The ecophysiological investigation confirmed that E. sorghinum produces more actively TeA under environmental conditions simulating the summer season. Optimum growth, maximum TAS1 gene expression, and higher extracellular enzymatic production were observed at 26?°C with a water activity of 0.99. Pearson correlation analyses showed that the production of TeA highly correlates with fungal growth. The present study demonstrates that abiotic factors in a combined approach of field and laboratory conditions will assist in predicting the driving environmental factors that could affect growth of E. sorghinum and TeA production in sorghum grains

    Schistosoma mansoni: experimental infection of mice through the ear and estimation of skin parasitism

    No presente trabalho, desenvolveu-se método de infecção de camundongos através da orelha e de recuperação de esquistossômulos resultantes dessas infecções. Cerca de 80% das cercarías postas em contacto com orelhas de camundongos penetraram. Destas, 30% foram recuperadas. como vermes adultos, do sistema porta. Da pele (das orelhas) as maiores recuperações de esquistossômulos ocorreram nos dois primeiros dias após a infecção. Os parasitas permaneceram nesse sítio por dois dias. No terceiro dia, os parasitas foram recuperados tanto da pele como dos pulmões. A partir do 4.° dia, foi predominante a recuperação de esquistossômulos ao nível dos pulmões. Do total de parasitas que potencialmente atingiriam o sistema porta, proporção elevada (73-80%) pode ser recuperada da pele, no segundo dia após a infecção, como esquistossômulos. Revelando-se apropriadas ao acesso, à migração no hospedeiro e às técnicas de recuperação de parasitas, sugere-se que orelhas de camundongos podem ser utilizadas como sítio de infecção para estudos que visem a análise parasitológica dos eventos iniciais da infecção em animais normais ou imunes.A method is presented to recover schistosomula from the skin of ear infected mice. About 80% of the cercariae applied to the ear were able to penetrate the mouse skin and 30% of them were recovered from the portal system as adult worms. The best recovery of larvae from the ear occurred in the two days that follow the penetration. From the 3rd day on, parasites were recovered both from skin and lungs and on the 4th day they were already found mainly in the lungs. From 73 to 80% of the parasites eventually found in the portal system can be recovered as schistosomula from the skin on tjhe 2nd day of infection. The use of the mouse ears is suggested for the parasitolqgical analysis of the initial events of the S. mansoni infection both in normal and in immune animals, for they are very convenient to the cercarial access and to the use of methods of study of migration and recuperation of the larvae

    Schistosoma Mansoni Reinfection: Analysis of Risk Factors By Classification and Regression Tree (CART) Modeling.

    Praziquantel (PZQ) is an effective chemotherapy for schistosomiasis mansoni and a mainstay for its control and potential elimination. However, it does not prevent against reinfection, which can occur rapidly in areas with active transmission. A guide to ranking the risk factors for Schistosoma mansoni reinfection would greatly contribute to prioritizing resources and focusing prevention and control measures to prevent rapid reinfection. The objective of the current study was to explore the relationship among the socioeconomic, demographic, and epidemiological factors that can influence reinfection by S. mansoni one year after successful treatment with PZQ in school-aged children in Northeastern Minas Gerais state Brazil. Parasitological, socioeconomic, demographic, and water contact information were surveyed in 506 S. mansoni-infected individuals, aged 6 to 15 years, resident in these endemic areas. Eligible individuals were treated with PZQ until they were determined to be negative by the absence of S. mansonieggs in the feces on two consecutive days of Kato-Katz fecal thick smear. These individuals were surveyed again 12 months from the date of successful treatment with PZQ. A classification and regression tree modeling (CART) was then used to explore the relationship between socioeconomic, demographic, and epidemiological variables and their reinfection status. The most important risk factor identified for S. mansoni reinfection was their “heavy” infection at baseline. Additional analyses, excluding heavy infection status, showed that lower socioeconomic status and a lower level of education of the household head were also most important risk factors for S. mansoni reinfection. Our results provide an important contribution toward the control and possible elimination of schistosomiasis by identifying three major risk factors that can be used for targeted treatment and monitoring of reinfection. We suggest that control measures that target heavily infected children in the most economically disadvantaged households would be most beneficial to maintain the success of mass chemotherapy campaigns

    The Right Tool for the Job: Detection of Soil-Transmitted Helminths in Areas Co-endemic for Other Helminths

    Due to the recent increased use of the McMaster (MM) fecal egg counting method for assessing benzimidazole drug efficacy for treating soil-transmitted helminth (STH) infections, the aim of the current study was to determine the operational value of including the MM method alongside the Kato-Katz (KK) fecal thick smear to increase the diagnostic sensitivity when STHs are co-endemic with trematode helminths (e.g., Schistosoma mansoni). Methods A cross-sectional study was conducted in school-aged children aged 4-18 years in the northeastern region of the State of Minas Gerais (Brazil), where Necator americanus, Ascaris lumbricoides, Trichuris trichiura, and S. mansoni are co-endemic. One fecal sample from each participant was collected and transported to the field laboratory for analysis. Coprological diagnosis was performed on each fecal sample by three different methods: Formalin-Ether Sedimentation (FES), KK and the MM technique. The diagnostic sensitivity and negative predictive value (NPV) of each technique was calculated using the combination of all three techniques as the composite standard. In order to determine the agreement between the three techniques Fleiss´ kappa was used. Both the Cure Rate (CR) and the Fecal Egg Count Reduction (FECR) were calculated using the two quantification techniques (i.e., the MM and KK). Results Fecal samples from 1260 children were analyzed. The KK had higher diagnostic sensitivity than the MM for the detection of both A. lumbricoides (KK 97.3%, MM 69.5%) and hookworm (KK 95.1%, MM 80.8%). The CR of a single dose of mebendazole varied significantly between the KK and MM for both A. lumbricoides (p = 0.016) and hookworm (p = 0.000), with lower rates obtained with the KK. On the other hand, the FECR was very similar between both techniques for both A. lumbricoides and hookworm. Conclusion The MM did not add any diagnostic value over the KK in areas where both STHs and trematodes were co-endemic. The lower sensitivity of the MM would have an important impact on the administration of selective school-based treatment in this area since if only the MM were used, 36 (13.9%) children diagnosed with A. lumbricoides would have gone untreated. Author Summary Diagnosis of intestinal helminths and Schistosoma mansoni infections is based on the detection of eggs in feces. There are many techniques available for both detection and quantification of infection. For the quantification of helminth infections, the methods traditionally used are the Kato-Katz (KK) fecal think smear in humans, and the McMaster (MM) counting method in animals. Recently, the MM has been used for assessing the efficacy of benzimidazole drugs for treating soil-transmitted helminth (STH) infections in humans. In most parts of the world, however, STHs occur simultaneously with other helminth species, and the MM does not detect other helminth eggs. Therefore, in this study we sought to determine if the use of the MM in an area of Brazil were both STHs and S. mansoni are co-endemic, added any value to the current standard of diagnosis using the KK

    TitDrop II, a practice low-cost titrator based on drop counting is proposed. A microcomputer counts drops in an opto-switch and it receives pH values from a commercial pHmeter with RS-232 serial port. The volume of drops must kept constant and the amount of base in each drop is obtained by standardization in the titrator. A linear behavior between height of reservoir of titrant and drop frequency was observed, but there is no influence of low height on drop volume. Acetic acidity in samples of vinager was compared with volumetric titration, and deviation between -2.6 to 3.9% was observed

    EVIDENCE OF LONG TERM BENEFIT OF MORBIDITY REDUCTION DUE TO PRAZIQUANTEL TREATMENT AGAINST SCHISTOSOMA MANSONI IN KIGUNGU FISHING VILLAGE IN ENTEBBE, UGANDA

    Praziquantel (PZQ) is efficacious against Schistosoma mansoni. This was prospective cohort study. This study was carried out at Kigungu fishing village, Entebbe, Uganda. The goal of the study was to establish cost effective regiment for mass drug administration (MDA) of Praziquentel in the morbidity reduction of S.mansoni infection. In January 2004, nine hundred and forty five (945) participants were registered in this study. Our analysis was based on examining microscopically three slides prepared from each of 945 stool specimens delivered by each of the participant using modified Kato/Katz method. These included male and female, children and adults living in Kigungu fishing village in Entebbe Uganda. In total 901, cohorts were re-examined for infections clearance six months later in July 2004 and 18 months later in June 2005, 625 cohorts were again re-evaluated for S.mansoni infections after the baseline study. At baseline, (448) of 945 (47.5%) cohorts were S. mansoni positive. All these participants were treatment with a single oral dose of praziquantel at 40mg/kg. At the same time, 495 (52.5%) were S. mansoni negative. Of the 625 (66.3%) cohorts who came back for final review, 80 (12.8%) were still positive for S. mansoni while 210 (33.6%) remained negative after the base line treatment with praziquantel. On the other hand 103 (16.3%) of cohorts who were initially negative at the base line became S.mansoni positive after 18 months and 213(34.1%) remained negative for S.mansoni. The force of re-infection after six months was significant {(P=0.0001), (OR 0.47) CI at 95% (0.31-0.71)}. Nevertheless the force of reinfection was not significant after 18 months {(P=0.766), (OR 0.95) CI at 95% (0.68-1.34)}.The geometric mean eggs excretion of the 80 cohorts who were S.mansoni positive at 18 months was 151.967.This did not reach the geometric mean egg excreted by the same cohorts at baseline which was 285.05. The egg excretion was reduced by 46.8%. Similarly there was marked decrease in clinical symptoms amongst the cohorts. Our study suggests evidence of long-term benefit of praziquantel in Kigungu and that a yearly administration of praziquantel to the community could be a regiment for mass drug administration (MAD) for this community to control schistosomiasis morbidity
