Evaluation of the cost-benefit of the implementation of PAT systems for the production of solid forms in a pharmaceutical company

Tese de mestrado, Engenharia Farmacêutica, Universidade de Lisboa, Faculdade de Farmácia, 2016Process Analytical Technology (PAT) is defined by the FDA in the PAT initiative issued in 2004 as ”a system for designing, analyzing, and controlling manufacturing through timelymeasurements (i.e.,during processing) of critical quality and performance attributes of raw and inprocess materials and processes, with the goal of ensuring the final product quality”. Since then, the pharmaceutical companies have been adopting gradually this technology in their manufacturing lines. PAT tools are indeed well recognized as a critical tool for achieving better process and product control leading to the possibility of real time release testing. This thesis evaluated the technical and economic feasibility of implementation of PAT systems into a Portuguese pharmaceutical company manufacturing line of solid forms. To accomplish this task, two products were used as models. These products were selected since they cover most unit operations of the solid forms installed manufacturing line. The work was divided in two parts. The first part is concerned with the identification of critical quality attributes (CQAs) and critical process parameters (CPPs) of the two model products in order for the proposition of opportunities of implementation of PAT strategies. These opportunities are described in detail and evaluated according to a series of criteria. Nine opportunities were identified, mainly involving NIR and RAMAN spectroscopy as suitable technology. The implementation of the identified opportunities, is expected to permit the possibility of real time release for the two products. The second part focuses on the economical feasibility of the identified PAT implementation. The economic analysis indicated that the implementation of all opportunities will reduce manufacturing / quality control time and costs substantially. The expectation of return-of-investmentis achieved in a time frame between 5 to 8 years respectively considering optimistic and pessimist scenarios

Políticas de educação: as diferentes responsabilidades e opções dos poderes públicos

O presente trabalho procura analisar a evolução das teorias políticas e econômicas mais relevantes, bem como os tipos de Estado que lhes estão associados, que foram se sobrepondo no decorrer dos últimos três séculos nas sociedades democráticas ocidentais e que mais diretamente foram interferindo na elaboração e execução das políticas públicas de educação. Em seguida, reflete-se sobre as suas implicações na forma como o pensamento educativo foi sendo construído e sedimentado. Partindo dessa análise, termina-se por equacionar as mudanças fundamentais que se perspectivam no pensar de algumas dimensões das políticas educativas públicas, situando-as, sobretudo, na necessidade de associar as lógicas nacionais e transnacionais às políticas de base local, que, por meio de uma ação conjunta e concertada, mobilizem a comunidade no sentido de formar cidadãos capazes, sem perder de vista a escala global, de agir em âmbito local, a partir do conhecimento da sua própria realidade e induzir uma nova noção de desenvolvimento, o qual se espera que seja integrado e sustentado. Abstract This paper seeks to analyze the evolution of the most relevant political and economic theories, as well as the associated state types, during the last three centuries in western democratic societies and that most directly interfered in the elaboration and execution of public education policies, to reflect on their implications in the construction and consolidation of educational thinking. Starting from this analysis, it ends up by equating the fundamental changes that are perceived in the thinking of some dimensions of the public educational policies, placing them in the need to associate local policies to national and transnational logics, that can, through joint and concerted action, mobilize the community to train citizens who, without forgetting the global scale, are able to act at the local level, based on the knowledge of their own reality, and to induce a new notion of development, which is expected to be integrated and sustained

Perception of health status among the elderly population living in the historic center of Portalegre

In this communication we present the results of an research that aimed to assess the state of health, the feeling of loneliness and quality of life of Portalegre (Portugal) senior population. Our aim is to caracterize the perception of health status among the elderly population based on three indicators: self-assessment of health status; self-assessment of the existence of changes in health status and autonomy in performing everyday tasks

Molecular diagnosis of Mycoplasma bovis

Dissertação de Mestrado Integrado em Medicina VeterináriaMycoplasma bovis is a bacteria responsible for different disease presentations in cattle, such as pneumonia, mastitis, otitis, genital disorders, keratoconjuntivitis and arthritis, presently considered as one of the major emerging pathogens affecting cattle. Until this day, it is responsible for losses in animal production of over 150 million euros across Europe. The pathogenesis of Mycoplasma-associated diseases is multifactorial and the highly variable surface lipoproteins allows a fast and efficient dissemination of M. bovis within the host and the herd. Due to its high antigenic plasticity, its ability to survive within multiple host cells and the capacity to establish multiple synergistic interactions with other pathogens, makes M. bovis and associated infections are a major challenge in Veterinary Medicine, since the vaccine is not efficient and antibiotics are almost inefficient. This study aims at developing and validating a quantitative PCR protocol for the diagnosis of M. bovis. 93 milk samples, from 5 different Portuguese farms, were collected, processed and each one’s DNA extracted to be analyzed through a qPCR method targeting the uvrC and uvrC2024 genes. Given the percentage of positivity, which was high, the conclusion we can take from the study is that there is still work to do, in terms of establishing a uniformed practice to tackle the wide presence of M. bovis in farms.RESUMO - Diagnóstico Molecular de M. bovis - Mycoplasma bovis é uma bactéria cuja infecção pode ter diferentes apresentações tais como pneumonia, mastite, otite, afecções genitais, queratoconjuntivite e artrites. É um dos agentes considerados emergentes e que afecta a produção agropecuária, sendo responsável por perdas na ordem dos 150 milhões de euros na Europa. Tem uma patogénese multifactorial e as proteínas membranares à sua superfície conferem uma variabilidade que permite uma rápida e eficiente disseminação no hospedeiro, e no rebanho. Esta variabilidade e capacidade de resistir à imunidade do hospedeiro, assim como as suas interacções sinérgicas com outros agentes patogénicos, tornam as infecções por M. bovis um obstáculo difícil de conter e ultrapassar na Medicina Veterinária, isto porque quer a antibioterapia, quer a vacina, não são eficientes. Este estudo procura desenvolver e contribuir para o estabelecimento de um protocolo de diagnóstico para a detecção de M. bovis. Vindas de 5 produções portuguesas diferentes, 93 amostras foram processadas e analisadas através de um qPCR, com os genes uvrC e uvrC2024 como genes alvo. Dados os resultados, com uma positividade significativa pode-se considerar que ainda há trabalho pela frente em termos de estabelecer e uniformizar uma prática para combater a larga presença de M. bovis nas explorações.N/

A Gestão dos Militares na Situação de Reserva

O presente trabalho tem por objectivo caracterizar a situação de “Reserva”, como instrumento de gestão dos quadros do Exército e propor a implementação de medidas incentivadoras à sua adopção ao longo da carreira militar, enfatizando a reconversão profissional dos seus quadros, a valorização do capital humano da organização e as necessidades estruturais do ramo. Este assunto reveste-se de especial interesse pela heterogeneidade e dimensão dos quadros de pessoal do Exército, que associados a constrangimentos internos e externos se constituem como grandes desafios de gestão. No desenvolvimento do presente trabalho recorreu-se ao emprego de diferentes metodologias de investigação, salientando-se: o método indutivo na caracterização e análise do universo dos quadros permanentes na situação de “Reserva” e dos desvios identificados pelos órgãos de gestão de pessoal. Para tal contámos com a colaboração dos órgãos da Direcção de Administração e Mobilização de Pessoal do Exército, na recolha de informação crítica e na condução de entrevistas a entidades intervenientes no processo de gestão. . o método dedutivo na interpretação e análise do quadro legislativo e normativo orientador da função operativa de Gestão de Pessoal no Exército concorrentemente com a análise das orientações supranacionais e nacionais das políticas sectoriais de Emprego, Segurança Social e do Ensino e Formação Profissional. A recensão literária no âmbito dos recursos humanos orientou-nos para os actuais paradigmas de gestão e para a apreensão das orientações empresariais nesta área. . o método quantitativo foi desenvolvido pela aplicação de um questionário, que nos facilitou a interpretação do valor intrínseco que os quadros do Exército detêm relativamente à “Reserva” e simultaneamente alicerçou o levantamento de um modelo de aplicação da “Reserva”. O presente trabalho foi orientado para a maximização do instrumento de gestão de pessoal das Forças Armadas - “Reserva”, no pressuposto da realização técnico profissional dos seus quadros e da satisfação das necessidades estruturais do Exército em quadros permanentes. Abstract: The objective of this essay is to characterize the situation of the “Reserva”1 as an instrument of Army personnel management and to present the implementation of measures, which will encourage its adoption throughout the military career. These measures stress the development of professional capabilities rearrangements considering, simultaneously, their technical and professional anxieties, the valuation of the organization’s human assets and the Army’s needs. This issue presents special interest due to the heterogeneity and dimension of the Army personnel ranks, associated to internal and external constraints that forms the personnel management challenge. To develop the present essay, various research methods were employed, mainly: • the inductive method for the characterization and analysis of the “elements” in the “Reserva” and the evasion from normative framework, identified by the personnel management authorities. For this, we depended on the collaboration of the Administration and Mobilization of Army Personnel Department, for the compiling of critical information and interviews with entities involved in it • the deductive method for the interpretation and analysis of the legislative and normative framework which is the guideline for the activities of Personnel Management in the Army, simultaneously with the analysis of the supranational and national orientations of the Employment, Social Security and Educational/Professional Training policies. A literary survey in the human resources area guided us in the current paradigms of management and the apprehension of the entrepreneurial orientations in this area. • the quantitative method was developed through the application of a questionnaire which facilitated our interpretation of the intrinsic value which the Army ranks have in relation to the “Reserva” and simultaneously consolidated the improvement of a “Reserva” application model. This essay was oriented for the maximization of the “Reserva” as an Armed Forces personnel management tool in the assumption of the technical and professional accomplishment of their staff and the fulfilment of the personnel structure necessities of the Army
