    Fermi LAT AGN classification using supervised machine learning

    Classifying Active Galactic Nuclei (AGN) is a challenge, especially for BL Lac Objects (BLLs), which are identified by their weak emission line spectra. To address the problem of classification, we use data from the 4th Fermi Catalog, Data Release 3. Missing data hinders the use of machine learning to classify AGN. A previous paper found that Multiple Imputation by Chain Equations (MICE) imputation is useful for estimating missing values. Since many AGN have missing redshift and the highest energy, we use data imputation with MICE and K-nearest neighbor (kNN) algorithm to fill in these missing variables. Then, we classify AGN into the BLLs or the Flat Spectrum Radio Quasars (FSRQs) using the SuperLearner, an ensemble method that includes several classification algorithms like logistic regression, support vector classifiers, Random Forests, Ranger Random Forests, multivariate adaptive regression spline (MARS), Bayesian regression, Extreme Gradient Boosting. We find that a SuperLearner model using MARS regression and Random Forests algorithms is 91.1% accurate for kNN imputed data and 91.2% for MICE imputed data. Furthermore, the kNN-imputed SuperLearner model predicts that 892 of the 1519 unclassified blazars are BLLs and 627 are Flat Spectrum Radio Quasars (FSRQs), while the MICE-imputed SuperLearner model predicts 890 BLLs and 629 FSRQs in the unclassified set. Thus, we can conclude that both imputation methods work efficiently and with high accuracy and that our methodology ushers the way for using SuperLearner as a novel classification method in the AGN community and, in general, in the astrophysics community.Comment: 15 pages, 8 figures, to be published in Monthly Notices of the Royal Astronomical Societ

    Fibronectin as an adjuvant in the diagnosis of oral inflammatory myofibroblastic tumor

    Inflammatory myofibroblastic tumor is a distinctive lesion composed of myofibroblastic spindle shaped cells accompanied by inflammatory infiltrate that may arise in various organs. It is believed to be a noneoplastic inflammatory condition, although this is still controversial. The recognition of inflammatory myofibroblastic tumor as an entity is important especially to avoid unnecessary surgery. A few cases have been reported in the oral cavity. This report primarily presents a case of inflammatory myofibroblastic tumor that arose in the floor of mouth of a 23-year-old woman. The proliferating spindle cells were immunoreactive for vimentin, smooth muscle actin, and muscle specific actin and negative for desmin, AE1/AE3, S-100, CD68, MyoD1 and caldesmon. In an attempt to assess the potential neoplastic nature of this lesion, immunohistochemical expression of ALK protein was performed, although no immunoreactivity was detected. Also, the presence of well differentiated myofibroblasts identified by fibronectin is discussed, as well as the importance in establishing an immunoprofile to better consolidate the diagnosis. We conclude that the study of fibronectin in case series may aid the diagnosis as well as the prediction of the tumor course

    β-Microseminoprotein Endows Post Coital Seminal Plasma with Potent Candidacidal Activity by a Calcium- and pH-Dependent Mechanism

    The innate immune factors controlling Candida albicans are mostly unknown. Vulvovaginal candidiasis is common in women and affects approximately 70–75% of all women at least once. Despite the propensity of Candida to colonize the vagina, transmission of Candida albicans following sexual intercourse is very rare. This prompted us to investigate whether the post coital vaginal milieu contained factors active against C. albicans. By CFU assays, we found prominent candidacidal activity of post coital seminal plasma at both neutral and the acid vaginal pH. In contrast, normal seminal plasma did not display candidacidal activity prior to acidification. By antifungal gel overlay assay, one clearing zone corresponding to a protein band was found in both post coital and normal seminal plasma, which was subsequently identified as β-microseminoprotein. At neutral pH, the fungicidal activity of β-microseminoprotein and seminal plasma was inhibited by calcium. By NMR spectroscopy, amino acid residue E71 was shown to be critical for the calcium coordination. The acidic vaginal milieu unleashed the fungicidal activity by decreasing the inhibitory effect of calcium. The candidacidal activity of β-microseminoprotein was mapped to a fragment of the C-terminal domain with no structural similarity to other known proteins. A homologous fragment from porcine β-microseminoprotein demonstrated calcium-dependent fungicidal activity in a CFU assay, suggesting this may be a common feature for members of the β-microseminoprotein family. By electron microscopy, β-microseminoprotein was found to cause lysis of Candida. Liposome experiments demonstrated that β-microseminoprotein was active towards ergosterol-containing liposomes that mimic fungal membranes, offering an explanation for the selectivity against fungi. These data identify β-microseminoprotein as an important innate immune factor active against C. albicans and may help explain the low sexual transmission rate of Candida

GW useful in irrigation of a garden and aids to reduce cost as well as maintain the environmental prosperity. This paper discussed the effectiveness of Botryococcus sp. to clean GW in phycoremediation treatment. This process involves as growing the Botryococcus sp. in the GW which is contributing to utilize supplements in GW for its grow. The results indicated that Botryococcus sp. is effective to reduce COD (88%), BOD (82%), TIC (76%), TC (58%), TN (52%), TOC (39%), Phosphate (37.5%) and pH (7%) for 100% concentration of GW. Meanwhile, for the 50% of GW concentration Botryococcus sp. capable to remove such as COD (83%), TIC (82%), BOD (68%), TN (67%), Phosphate (36.8%), TC (34%), TOC (31%) and pH (1.2%). Then, the study concludes that Botryococcus sp. can grow effectively in GW and be able to reduce the rate of nutrient in GW

    Long-Range Activation of Systemic Immunity through Peptidoglycan Diffusion in Drosophila

    The systemic immune response of Drosophila is known to be induced both by septic injury and by oral infection with certain bacteria, and is characterized by the secretion of antimicrobial peptides (AMPs) into the haemolymph. To investigate other possible routes of bacterial infection, we deposited Erwinia carotovora (Ecc15) on various sites of the cuticle and monitored the immune response via expression of the AMP gene Diptericin. A strong response was observed to deposition on the genital plate of males (up to 20% of a septic injury response), but not females. We show that the principal response to genital infection is systemic, but that some AMPs, particularly Defensin, are induced locally in the genital tract. At late time points we detected bacteria in the haemolymph of immune deficient RelishE20 flies, indicating that the genital plate can be a route of entry for pathogens, and that the immune response protects flies against the progression of genital infection. The protective role of the immune response is further illustrated by our observation that RelishE20 flies exhibit significant lethality in response to genital Ecc15 infections. We next show that a systemic immune response can be induced by deposition of the bacterial elicitor peptidoglycan (PGN), or its terminal monomer tracheal cytotoxin (TCT), on the genital plate. This immune response is downregulated by PGRP-LB and Pirk, known regulators of the Imd pathway, and can be suppressed by the overexpression of PGRP-LB in the haemolymph compartment. Finally, we provide strong evidence that TCT can activate a systemic response by crossing epithelia, by showing that radiolabelled TCT deposited on the genital plate can subsequently be detected in the haemolymph. Genital infection is thus an intriguing new model for studying the systemic immune response to local epithelial infections and a potential route of entry for naturally occurring pathogens of Drosophila

    Accumulation capacity of ions in cabbage (Brassica oleracea L.) supplied with sea water

    Cabbage seedlings were grown hydroponically to study the effects of different concentrations of seawater on the seedling growth, ion content under one-fourth strength Hoagland's nutrient solution in the greenhouse. The biomass of various organs of cabbage seedlings as well as the whole plants was significantly higher in the treatments with 1 g and 2 g sea salt/L than the no-salt control, but the treatments with 4, 5 or 6 g sea salt/L caused a decrease in growth. Root/shoot ratio remained at the level of control regardless of the sea salt treatment. Na+ and Cl- concentration in different parts of cabbage seedlings increased significantly, whereas K+ and Ca2+ concentration generally increased at low concentrations of sea salt and then decreased with increasing seawater concentration. Sodium and K+ concentrations were significantly higher in the stems than roots and leaves regardless of the sea salt treatment. The sea salt treatment increased Mg2+ concentration in stems and leaves of cabbage seedlings. An increase in Na+ and Cl- concentration in roots, stems and leaves of cabbage seedlings was the main contributor to declining ratios of K+/Na+, Ca2+/Na+ and Mg2+/Na+. The obtained data suggest that cabbage seedlings have strong ability to sustain seawater stress by the regulation of transport and distribution of ions

    Immunogenicity, safety, and reactogenicity of heterologous COVID-19 primary vaccination incorporating mRNA, viral-vector, and protein-adjuvant vaccines in the UK (Com-COV2): a single-blind, randomised, phase 2, non-inferiority trial

    BACKGROUND: Given the importance of flexible use of different COVID-19 vaccines within the same schedule to facilitate rapid deployment, we studied mixed priming schedules incorporating an adenoviral-vectored vaccine (ChAdOx1 nCoV-19 [ChAd], AstraZeneca), two mRNA vaccines (BNT162b2 [BNT], Pfizer-BioNTech, and mRNA-1273 [m1273], Moderna) and a nanoparticle vaccine containing SARS-CoV-2 spike glycoprotein and Matrix-M adjuvant (NVX-CoV2373 [NVX], Novavax). METHODS: Com-COV2 is a single-blind, randomised, non-inferiority trial in which adults aged 50 years and older, previously immunised with a single dose of ChAd or BNT in the community, were randomly assigned (in random blocks of three and six) within these cohorts in a 1:1:1 ratio to receive a second dose intramuscularly (8-12 weeks after the first dose) with the homologous vaccine, m1273, or NVX. The primary endpoint was the geometric mean ratio (GMR) of serum SARS-CoV-2 anti-spike IgG concentrations measured by ELISA in heterologous versus homologous schedules at 28 days after the second dose, with a non-inferiority criterion of the GMR above 0·63 for the one-sided 98·75% CI. The primary analysis was on the per-protocol population, who were seronegative at baseline. Safety analyses were done for all participants who received a dose of study vaccine. The trial is registered with ISRCTN, number 27841311. FINDINGS: Between April 19 and May 14, 2021, 1072 participants were enrolled at a median of 9·4 weeks after receipt of a single dose of ChAd (n=540, 47% female) or BNT (n=532, 40% female). In ChAd-primed participants, geometric mean concentration (GMC) 28 days after a boost of SARS-CoV-2 anti-spike IgG in recipients of ChAd/m1273 (20 114 ELISA laboratory units [ELU]/mL [95% CI 18 160 to 22 279]) and ChAd/NVX (5597 ELU/mL [4756 to 6586]) was non-inferior to that of ChAd/ChAd recipients (1971 ELU/mL [1718 to 2262]) with a GMR of 10·2 (one-sided 98·75% CI 8·4 to ∞) for ChAd/m1273 and 2·8 (2·2 to ∞) for ChAd/NVX, compared with ChAd/ChAd. In BNT-primed participants, non-inferiority was shown for BNT/m1273 (GMC 22 978 ELU/mL [95% CI 20 597 to 25 636]) but not for BNT/NVX (8874 ELU/mL [7391 to 10 654]), compared with BNT/BNT (16 929 ELU/mL [15 025 to 19 075]) with a GMR of 1·3 (one-sided 98·75% CI 1·1 to ∞) for BNT/m1273 and 0·5 (0·4 to ∞) for BNT/NVX, compared with BNT/BNT; however, NVX still induced an 18-fold rise in GMC 28 days after vaccination. There were 15 serious adverse events, none considered related to immunisation. INTERPRETATION: Heterologous second dosing with m1273, but not NVX, increased transient systemic reactogenicity compared with homologous schedules. Multiple vaccines are appropriate to complete primary immunisation following priming with BNT or ChAd, facilitating rapid vaccine deployment globally and supporting recognition of such schedules for vaccine certification.
