    Continuous dependence estimates for nonlinear fractional convection-diffusion equations

    We develop a general framework for finding error estimates for convection-diffusion equations with nonlocal, nonlinear, and possibly degenerate diffusion terms. The equations are nonlocal because they involve fractional diffusion operators that are generators of pure jump Levy processes (e.g. the fractional Laplacian). As an application, we derive continuous dependence estimates on the nonlinearities and on the Levy measure of the diffusion term. Estimates of the rates of convergence for general nonlinear nonlocal vanishing viscosity approximations of scalar conservation laws then follow as a corollary. Our results both cover, and extend to new equations, a large part of the known error estimates in the literature.Comment: In this version we have corrected Example 3.4 explaining the link with the results in [51,59

    Flow and bubble statistics of turbulent bubble-laden downflow channel

    Direct numerical simulations of fully developed turbulent channel downflow at bulk Re equal to 6300, loaded with monodisperse bubbles at gas volume fractions α=0.5% , α=2.5% and α=10 have been carried out. Bubble deformability, surface tension, as well as discontinuity in the material properties across the bubble interfaces are explicitly accounted for. A full-scale channel of size 4πH × 2H × 4πH/3 in terms of the channel half-width H containing a number of bubbles up to O(103) is considered. The statistical structure of the bubbles, the probability density function describing the bubble velocity and the liquid kinetic energy spectra have been determined. A close range preferential clustering of the bubbles was found with a maximum density independent of the gas volume fraction at a separation distance of about 2.2R, with R the bubble radius. Preferential horizontal alignment and a general tendency to repulsion is shown for separation distances smaller than 3R. At larger separation distances a close to random distribution is observed for α=2.5% and α=10%, while tendency to vertical alignment is observed for α=0.5% . The pdf of the bubble velocity fluctuations was found to be well approximated by a Gaussian distribution. The liquid kinetic energy spectra in the channel core do not show a marked -3  scaling, which was previously reported for homogeneous isotropic turbulence and pseudo-turbulence

    Mixed modeling for large-eddy simulation: The single-layer and two-layer minimum-dissipation-Bardina models

    Predicting the behavior of turbulent flows using large-eddy simulation requires modeling of the subgrid-scale stress tensor. This tensor can be approximated using mixed models, which combine the dissipative nature of functional models with the capability of structural models to approximate out-of-equilibrium effects. We propose a mathematical basis to mix (functional) eddy-viscosity models with the (structural) Bardina model. By taking an anisotropic minimum-dissipation (AMD) model for the eddy viscosity, we obtain the (single-layer) AMD-Bardina model. In order to also obtain a physics-conforming model for wall-bounded flows, we further develop this mixed model into a two-layer approach: the near-wall region is parameterized with the AMD-Bardina model, whereas the outer region is computed with the Bardina model. The single-layer and two-layer AMD-Bardina models are tested in turbulent channel flows at various Reynolds numbers, and improved predictions are obtained when the mixed models are applied in comparison to the computations with the AMD and Bardina models alone. The results obtained with the two-layer AMD-Bardina model are particularly remarkable: both first- and second-order statistics are extremely well predicted and even the inflection of the mean velocity in the channel center is captured. Hence, a very promising model is obtained for large-eddy simulations of wall-bounded turbulent flows at moderate and high Reynolds numbers.Comment: 29 pages, 14 figures, 3 tables; revised, accepted manuscrip

    Towards LES of bubble-laden channel flows:Sub-grid scale closures for momentum advection

    This paper presents an a-posteriori assessment of different LES sub-grid scale closures for momentum advection in the context of bubble-laden channel flows. The numerical approach is based on the Volume-of-Fluid method in combination with the onefluid formulation of the incompressible Navier-Stokes equations. To study the behavior of different subgrid scale models, a turbulent bubble-laden downflow channel is simulated at a friction Reynolds number of ReΤ = 590. The setup is chosen such that the bubbles are nearly spherical, but mildly wobbling. Both functional models of eddy viscosity type and scale similarity type models are used to close the sub-grid scale stresses. The results are compared to a direct numerical simulation of the same setup. It is found that the stream-wise volumetric flow rate depends strongly on the closure model as well as the grid resolution. While some models lead to an improvement compared to the LES without an explicit model, the comparably dissipative nature of the QUICK scheme prevents a clear assessment of some more advanced modeling strategies

    Possible common central pathway for resistin and insulin in regulating food intake.

    Aim: Adipose tissue has been the object of intense research in the field of obesity and diabetes diseases in the last decade. Examination of adipocyte-secreted peptides led to the identification of a unique polypeptide, resistin (RSTN), which has been suggested as a link between obesity and diabetes. RSTN plays a clearly documented role in blocking insulin (INS)-induced hypoglycaemia. As brain injection of INS affects feeding behaviour, we studied the possible interaction between INS and RSTN in food-deprived rats, measuring effects on food intake. In addition, we examined how RSTN might affect neuropeptide Y (NPY)-induced feeding, as studies have shown that rat RSTN can interfere with the NPY system. Methods: Overnight food-deprived rats were injected into the third brain ventricle (3V) with either INS (10 or 20 mUI), RSTN (0.1–0.4 nmol/rat), or saline before access to food. Another group of rats was injected into the 3V with RSTN alone, NPY alone or RSTN plus NPY. Their food intake and body weight were measured. Results: Our results confirm the hypophagic effect of RSTN on food deprivation-induced food intake, and more importantly, show that RSTN neither potentiates nor blocks the effects of INS on food intake, but does reduce the hyperphagic effect of NPY. Conclusion:  The observation that RSTN does not modify feeding INS-induced hypophagia, but does influence NPY-induced feeding, points to the possibility that RSTN may be involved in control of food intake through an NPY-ergic mechanism as INS

    Epidemiological significance of the domestic Black Pig ( Sus scrofa) in the maintenance of Bovine Tuberculosis in Sicily

    Bovine tuberculosis (bTB) is an emerging disease among wild animals in many parts of the world. Wildlife reservoir hosts may thus represent a potential source of infection for livestock and humans. We investigated the role played by the Sicilian black pig, an autochthonous free- or semi-free-ranging domestic pig breed, as a potential source of bTB infection in an area where bTB prevalence in cattle is high. We initially performed a preliminary field study to assess the occurrence of bTB in such animals. We sampled 119 pigs at abattoir and found 6.7% and 3.4% of them to be affected by gross tuberculous-like lesions (TBL) and Mycobacterium bovis culture positive, respectively. We then proceeded to investigate the dissemination and characteristics of lesions in a second field study performed on 100 animals sampled from infected herds. Here, tissues collected at the abattoir were examined macroscopically, microscopically, and by culture tests. Most pigs with TBL showed generalized lesions in both gross and histological examinations (53% and 65.5%, respectively). Head lymph nodes were the most frequently affected in both localized and generalized TB cases observed macroscopically and microscopically. M. bovis was the most frequently isolated etiologic agent. The molecular characterization of isolates from both field studies by spoligotyping and analysis of 12 mycobacterial interspersed repetitive-unit–variable number tandem repeat (MIRU-VNTR) loci, followed by their comparison to isolates of cattle origin, suggested a potential transmission of mycobacteria from domestic animals to black pigs and vice versa. Our findings, along with ethological, ecological, and management considerations, suggest that the black pig might act as a bTB reservoir in the ecosystem under study. However, additional studies will be necessary to establish the true epidemiological significance of the Sicilian black pig

    Novel highly potent and selective sigma1 receptor antagonists effectively block the binge eating episode in female rats

    In this paper, the benzo-cracking approach was applied to the potent sigma1 (σ1) receptor antagonist 1 to afford the less conformationally constrained 1,3-dioxane derivatives 2 and 3. To evaluate the effect of the increase in the distance between the two hydrophobic structural elements that flank the basic function, the cis and trans diastereomers of 4 and 5 were also prepared and studied. Compounds 2 and 3 showed affinity values at the σ1 receptor significantly higher than that of the lead compound 1. In particular, 3 displayed unprecedented selectivity over the σ2 receptor, the phencyclidine site of the NMDA receptor, and opioid receptor subtypes, as well as over the dopamine transporter. Docking results supported the structure-activity relationship studies. Due to its interesting biological profile, derivative 3, selected for an in vivo study in a validated preclinical model of binge eating, was able to counteract the overeating of palatable food only in binging rats, without affecting palatable food intake in the control group and anxiety-like and depression-related behaviors in female rats. This result strengthened the involvement of the σ1 receptor in the compulsive-like eating behavior and supported the σ1 receptor as a promising target for the management of eating disorders

    Stress-Induced Reinstatement of Drug Seeking: 20 Years of Progress

    In human addicts, drug relapse and craving are often provoked by stress. Since 1995, this clinical scenario has been studied using a rat model of stress-induced reinstatement of drug seeking. Here, we first discuss the generality of stress-induced reinstatement to different drugs of abuse, different stressors, and different behavioral procedures. We also discuss neuropharmacological mechanisms, and brain areas and circuits controlling stress-induced reinstatement of drug seeking. We conclude by discussing results from translational human laboratory studies and clinical trials that were inspired by results from rat studies on stress-induced reinstatement. Our main conclusions are (1) The phenomenon of stress-induced reinstatement, first shown with an intermittent footshock stressor in rats trained to self-administer heroin, generalizes to other abused drugs, including cocaine, methamphetamine, nicotine, and alcohol, and is also observed in the conditioned place preference model in rats and mice. This phenomenon, however, is stressor specific and not all stressors induce reinstatement of drug seeking. (2) Neuropharmacological studies indicate the involvement of corticotropin-releasing factor (CRF), noradrenaline, dopamine, glutamate, kappa/dynorphin, and several other peptide and neurotransmitter systems in stress-induced reinstatement. Neuropharmacology and circuitry studies indicate the involvement of CRF and noradrenaline transmission in bed nucleus of stria terminalis and central amygdala, and dopamine, CRF, kappa/dynorphin, and glutamate transmission in other components of the mesocorticolimbic dopamine system (ventral tegmental area, medial prefrontal cortex, orbitofrontal cortex, and nucleus accumbens). (3) Translational human laboratory studies and a recent clinical trial study show the efficacy of alpha-2 adrenoceptor agonists in decreasing stress-induced drug craving and stress-induced initial heroin lapse

    Memantine reduces consumption of highly palatable food in a rat model of binge eating

    Excessive consumption of highly palatable food has been linked to the development of eating disorders and obesity, and can be modeled in non-food-deprived rats by offering them a limited (2-h daily) access to an optional dietary fat. Since the glutamatergic system has recently emerged as a viable target for binge-eating medication development, we compared the effects of subchronic treatment with glutamatergic receptor antagonists to the effects of a reference appetite-suppressing agent sibutramine on highly palatable food (lard) and normal chow intake. In three separate experiments, the consumption of a standard laboratory chow and lard were measured during 12 days of medication treatment and for 6 days afterwards. Generalized estimating equations analysis demonstrated that sibutramine (7.5 mg/kg, PO) significantly decreased lard consumption, with a concurrent increase in chow consumption. Sibutramine effects disappeared after treatment discontinuation. The NMDA receptor antagonist memantine (5 mg/kg, IP) significantly decreased lard consumption and increased chow consumption, comparable to effects of sibutramine; however, memantine’s effects persisted after treatment discontinuation. The effects of the mGluR5 antagonist MTEP (7.5 mg/kg, IP) on food consumption were in the same direction as seen with memantine, but the observed differences were not significant. In an additional control experiment, sibutramine and memantine reduced unlimited (24 h) chow intake during the treatment phase. Present results provide evidence that glutamatergic neurotransmission might be involved in the regulation of excessive consumption of highly palatable foods, and suggest that NMDA receptor may be an attractive target for developing obesity and disordered eating pharmacotherapies
