    Formation and removal of alkylthiolate self-assembled monolayers on gold in aqueous solutions

    We report the development of novel reagents and approaches for generating recyclable biosensors. The use of aqueous media for the formation of protein binding alkylthiolate monolayers on Au surfaces results in accelerated alkylthiolate monolayer formation and improvement in monolayer integrity as visualized by fluorescence microscopy and CV techniques. We have also developed an electrocleaning protocol that is compatible with microfluidics devices, and this technique serves as an on-chip method for cleaning Au substrates both before and after monolayer formation. The techniques for the formation and dissociation of biotinylated SAMs from aqueous solvents reported here may be applied towards the development of Au-based sensor devices and microfluidics chips in the future. A potential use of these devices includes the specific capture and triggered release of target cells, proteins, or small molecules from liquid samples

    Prevalence of feline immunodeficiency virus and Toxoplasma gondii in Feral Cats on St. Kitts, West Indies

    Toxoplasma gondii (T. gondii) is a cosmopolitan protozoan parasite that infects all warm-blooded species including humans. The definitive hosts of T. gondii are felid vertebrates including the domestic cat. Domestic cats shed oocysts for approximately two weeks in their feces after the primary infection. It has been shown that feline immunodeficiency virus (FIV) positive cats have a higher prevalence of and a higher titer of antibodies to T. gondii than those of FIV-negative cats. The main purposes of this study were to determine FIV prevalence and to investigate the oocysts shedding in FIV-positive and FIV-negative feral cats on St. Kitts. Fecal samples were collected from feral cats while their FIV statues were determined using a commercial SNAP kit. Total fecal DNA of each cat was tested for the presence of T. gondii DNA using a polymerase chain reaction (PCR) consistently detecting one genome equivalent. A FIV-positive status was detected in 18 of 105 (17.1%, 95% confidence interval (CI): 9.9%−24.3%) feral cats sampled. Furthermore, males were three times more likely to be FIV positive than females (p = 0.017) with an odds ratio of 3.93 (95% CI: 1.20–12.89). Adults were found to have at least twice the prevalence of FIV compared to cats younger than one year of age (p = 0.056) with an odds ratio of 3.07 (95% CI: 0.94–10.00). Toxoplasma gondii DNA was not detected in the feces of any of the 18 FIV-positive (95% CI: 0%−0.18%) and 87 FIV-negative cats (95% CI: 0%−0.04%). A follow-up study with a much bigger sample size is needed to prove or disprove the hypothesis that FIV-positive cats have a higher prevalence of shedding T. gondii oocysts than FIV-negative cats

    Occurrence of Aspergillus species and aflatoxin contamination in raw and roasted peanuts from formal and informal markets in Eldoret and Kericho towns, Kenya

    Published Online: August 2013.The population and diversity of fungal species and levels of aflatoxin contamination were investigated in 228 marketed peanut samples; 140 from formal and 88 from informal markets, in Kericho and Eldoret towns of Kenya. Ground pea- nut samples were cultured on Modified Dichloran Rose Bengal (MDRB) agar while aflatoxin level was quantified based on indirect competitive ELISA. Correlation between the incidence of major aflatoxin-producing fungal species and aflatoxin levels was also established. Fungal species commonly isolated from the peanut samples included Asper-gillus flavus L strain, A. flavus S strain, A. parasiticus, A. tamarii, A. caelatus, A. alliaceus (all of Aspergillus section Flavi) and A. niger. Fungi isolated in low frequency included Fusarium spp., Penicillium spp., Mucor spp. and Rhi- zopus spp. Aflatoxin levels in peanut products ranged from 0 to 2345 ÎŒg/kg in raw peanuts, 0 to 382 ÎŒg/kg in roasted coated peanuts, and 0 to 201 ÎŒg/kg in roasted de-coated peanuts. Overall, levels of total aflatoxin were higher in sam- ples from informal (mean = 97.1 ÎŒg/kg) than formal (mean = 55.5 ÎŒg/kg) market outlets. There was a positive and sig- nificant correlation (R2 = 0.63; p ≀ 0.05) between aflatoxin levels and the major aflatoxin producing fungi in raw pea- nuts from formal markets in Eldoret town. Additionally, total aflatoxin in raw peanut samples from informal markets in Kericho was positively and significantly correlated (R2 = 0.81; p ≀ 0.05) to the population of A. flavus (L and S strains). In roasted coated peanuts sampled from formal market outlets in Eldoret, aflatoxin levels correlated positively and sig- nificantly (R2 = 0.37; p ≀ 0.05) with A. flavus S strain. There is need to create awareness among peanut traders and con- sumers on proper handling of peanuts and health risks associated with consumption of unsafe peanut products

    Strategic support for a range of data management needs has emerged as a campus-level priority at UVM, as at many other colleges and universities. In Spring 2017, the UVM Libraries reconfigured the vacant position of “Science Librarian” as “Science and Data Librarian” with new responsibilities for supporting data management. This presentation will describe a case study of how the UVM Libraries aligned these responsibilities with campus needs and initiatives; what we learned about data librarianship from the recruitment process; and the opportunities for an incoming librarian shaping the course of new library services

    Used cooking oil can be used as raw material for biodiesel, but the levels of free fatty acids (Free Fatty Acid, FFA) is quite high. It is necessary for pretreatment in the form of the adsorption process to reduce levels of FFA. This study aims to determine the optimal conditions of adsorption process and determine the quality of biodiesel produced from adsorption processes and transesterification. Natural zeolites are used as adsorbents activated beforehand using ammonium chloride, calcined and heated to obtain H-zeolite. Furthermore, the adsorption process optimization includes the time, the adsorbent concentration, temperature and particle size. The oil that is already in the adsorption catalyst is reacted with methanol and KOH to obtain biodiesel. The optimum adsorption conditions obtained at the time of 90 minutes, the concentration of H-zeolite 12%, temperature 90 ° C, and a particle size of 0.2 mm that can lower FFA levels from 3.2% to 1.1%. Biodiesel produced meets the quality requirements of SNI 04-7182-2006 with a water content of 0.02%, a density of 857.60 kg / m3, the acid value of 0.29 mg-KOH / g, iodine number 15.71, saponification 168 , 02 and cetane index of 75.62. Compounds contained in biodiesel are methyl 9-octadecanoic (49.45%), methyl heksadekanoat (20.79%), and methyl 9,12oktaekanoat 9.12 (18.87%)

    Stressful Life Events and Behavior Change: A Qualitative Examination of African American Women\u27s Participation in a Weight Loss Program

    We qualitatively assessed how life stressors affected African American women\u27s participation in a weight reduction program. A sample of 9 women, who completed a behavioral lifestyle intervention, participated in individual, structured, in-depth interviews. Life stressors, ranging from personal illness to changes in employment status, had varied effects on participation. Some women coped with life stressors by using them as a motivational tool to improve their own health, while others reported limited ability to devote time to attend meetings or engage in the prescribed lifestyle changes due to life stressors. A critical key to improving weight loss outcomes for African American women may be using intervention strategies that teach positive coping skills to alter maladaptive responses to life stressors
