    Stress-inducible phosphoprotein 1 (HOP/STI1/STIP1) regulates the accumulation and toxicity of α-synuclein in vivo

    The predominantly pre-synaptic intrinsically disordered protein α-synuclein is prone to misfolding and aggregation in synucleinopathies, such as Parkinson’s disease (PD) and Dementia with Lewy bodies (DLB). Molecular chaperones play important roles in protein misfolding diseases and members of the chaperone machinery are often deposited in Lewy bodies. Here, we show that the Hsp90 co-chaperone STI1 co-immunoprecipitated α-synuclein, and co-deposited with Hsp90 and Hsp70 in insoluble protein fractions in two mouse models of α-synuclein misfolding. STI1 and Hsp90 also co-localized extensively with filamentous S129 phosphorylated α-synuclein in ubiquitin-positive inclusions. In PD human brains, STI1 transcripts were increased, and in neurologically healthy brains, STI1 and α-synuclein transcripts correlated. Nuclear Magnetic Resonance (NMR) analyses revealed direct interaction of α-synuclein with STI1 and indicated that the STI1 TPR2A, but not TPR1 or TPR2B domains, interacted with the C-terminal domain of α-synuclein. In vitro, the STI1 TPR2A domain facilitated S129 phosphorylation by Polo-like kinase 3. Moreover, mice over-expressing STI1 and Hsp90ß presented elevated α-synuclein S129 phosphorylation accompanied by inclusions when injected with α-synuclein pre-formed fibrils. In contrast, reduced STI1 function decreased protein inclusion formation, S129 α-synuclein phosphorylation, while mitigating motor and cognitive deficits as well as mesoscopic brain atrophy in α-synuclein-over-expressing mice. Our findings reveal a vicious cycle in which STI1 facilitates the generation and accumulation of toxic α-synuclein conformers, while α-synuclein-induced proteostatic stress increased insoluble STI1 and Hsp90

    Arrhythmogenic mechanisms in the isolated perfused hypokalaemic murine heart

    AIM: Hypokalaemia is associated with a lethal form of ventricular tachycardia (VT), torsade de pointes, through pathophysiological mechanisms requiring clarification. METHODS: Left ventricular endocardial and epicardial monophasic action potentials were compared in isolated mouse hearts paced from the right ventricular epicardium perfused with hypokalaemic (3 and 4 mm [K(+)](o)) solutions. Corresponding K(+) currents were compared in whole-cell patch-clamped epicardial and endocardial myocytes. RESULTS: Hypokalaemia prolonged epicardial action potential durations (APD) from mean APD(90)s of 37.2 ± 1.7 ms (n = 7) to 58.4 ± 4.1 ms (n =7) and 66.7 ± 2.1 ms (n = 11) at 5.2, 4 and 3 mm [K(+)](o) respectively. Endocardial APD(90)s correspondingly increased from 51.6 ± 1.9 ms (n = 7) to 62.8 ± 2.8 ms (n = 7) and 62.9 ± 5.9 ms (n = 11) giving reductions in endocardial–epicardial differences, ΔAPD(90), from 14.4 ± 2.6 to 4.4 ± 5.0 and −3.4 ± 6.0 ms respectively. Early afterdepolarizations (EADs) occurred in epicardia in three of seven spontaneously beating hearts at 4 mm [K(+)](o) with triggered beats followed by episodes of non-sustained VT in nine of 11 preparations at 3 mm. Programmed electrical stimulation never induced arrhythmic events in preparations perfused with normokalemic solutions yet induced VT in two of seven and nine of 11 preparations at 4 and 3 mm [K(+)](o) respectively. Early outward K(+) current correspondingly fell from 73.46 ± 8.45 to 61.16±6.14 pA/pF in isolated epicardial but not endocardial myocytes (n = 9) (3 mm [K(+)](o)). CONCLUSIONS: Hypokalaemic mouse hearts recapitulate the clinical arrhythmogenic phenotype, demonstrating EADs and triggered beats that might initiate VT on the one hand and reduced transmural dispersion of repolarization reflected in ΔAPD(90) suggesting arrhythmogenic substrate on the other

    Observation of excited states in the near-drip-line nucleus 125Pr

    High-spin states have been observed in the near-drip-line nucleus 125Pr following the reaction 64Zn(64Zn, p2n). The detection of charged particles and neutrons evaporated from the compound system, along with the M/q of the recoiling nucleus, have allowed the identification of excited states in 125Pr and the unambiguous assignment of five rotational structures to this nucleus. This is the most neutron-deficient Pr isotope in which excited states have been observed. The strongest band is identified as the h11/2 single-quasiproton configuration, and is observed to a maximum spin of I = (67/2hℏ). Another structure is interpreted as the g9/2 proton hole state, which is associated with bands of enhanced deformation observed in several nuclei in this mass region. These two bands are compared with analogous bands in the heavier odd-A Pr isotopes and changes in deformation are discussed

    Irbesartan in Marfan syndrome (AIMS): a double-blind, placebo-controlled randomised trial

    BACKGROUND: Irbesartan, a long acting selective angiotensin-1 receptor inhibitor, in Marfan syndrome might reduce aortic dilatation, which is associated with dissection and rupture. We aimed to determine the effects of irbesartan on the rate of aortic dilatation in children and adults with Marfan syndrome. METHODS: We did a placebo-controlled, double-blind randomised trial at 22 centres in the UK. Individuals aged 6-40 years with clinically confirmed Marfan syndrome were eligible for inclusion. Study participants were all given 75 mg open label irbesartan once daily, then randomly assigned to 150 mg of irbesartan (increased to 300 mg as tolerated) or matching placebo. Aortic diameter was measured by echocardiography at baseline and then annually. All images were analysed by a core laboratory blinded to treatment allocation. The primary endpoint was the rate of aortic root dilatation. This trial is registered with ISRCTN, number ISRCTN90011794. FINDINGS: Between March 14, 2012, and May 1, 2015, 192 participants were recruited and randomly assigned to irbesartan (n=104) or placebo (n=88), and all were followed for up to 5 years. Median age at recruitment was 18 years (IQR 12-28), 99 (52%) were female, mean blood pressure was 110/65 mm Hg (SDs 16 and 12), and 108 (56%) were taking ÎČ blockers. Mean baseline aortic root diameter was 34·4 mm in the irbesartan group (SD 5·8) and placebo group (5·5). The mean rate of aortic root dilatation was 0·53 mm per year (95% CI 0·39 to 0·67) in the irbesartan group compared with 0·74 mm per year (0·60 to 0·89) in the placebo group, with a difference in means of -0·22 mm per year (-0·41 to -0·02, p=0·030). The rate of change in aortic Z score was also reduced by irbesartan (difference in means -0·10 per year, 95% CI -0·19 to -0·01, p=0·035). Irbesartan was well tolerated with no observed differences in rates of serious adverse events. INTERPRETATION: Irbesartan is associated with a reduction in the rate of aortic dilatation in children and young adults with Marfan syndrome and could reduce the incidence of aortic complications

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE Δ4 allele

    The Cholecystectomy As A Day Case (CAAD) Score: A Validated Score of Preoperative Predictors of Successful Day-Case Cholecystectomy Using the CholeS Data Set

    Background Day-case surgery is associated with significant patient and cost benefits. However, only 43% of cholecystectomy patients are discharged home the same day. One hypothesis is day-case cholecystectomy rates, defined as patients discharged the same day as their operation, may be improved by better assessment of patients using standard preoperative variables. Methods Data were extracted from a prospectively collected data set of cholecystectomy patients from 166 UK and Irish hospitals (CholeS). Cholecystectomies performed as elective procedures were divided into main (75%) and validation (25%) data sets. Preoperative predictors were identified, and a risk score of failed day case was devised using multivariate logistic regression. Receiver operating curve analysis was used to validate the score in the validation data set. Results Of the 7426 elective cholecystectomies performed, 49% of these were discharged home the same day. Same-day discharge following cholecystectomy was less likely with older patients (OR 0.18, 95% CI 0.15–0.23), higher ASA scores (OR 0.19, 95% CI 0.15–0.23), complicated cholelithiasis (OR 0.38, 95% CI 0.31 to 0.48), male gender (OR 0.66, 95% CI 0.58–0.74), previous acute gallstone-related admissions (OR 0.54, 95% CI 0.48–0.60) and preoperative endoscopic intervention (OR 0.40, 95% CI 0.34–0.47). The CAAD score was developed using these variables. When applied to the validation subgroup, a CAAD score of ≀5 was associated with 80.8% successful day-case cholecystectomy compared with 19.2% associated with a CAAD score >5 (p < 0.001). Conclusions The CAAD score which utilises data readily available from clinic letters and electronic sources can predict same-day discharges following cholecystectomy

    Mendepani kekangan inovasi dan teknologi dalam kalangan pekebun kecil sawit di Sabah

    Pelbagai usaha melibatkan inovasi dan teknologi berkaitan teknik penanaman dan pengeluaran hasil telah diberi penekanan oleh organisasi yang terlibat dalam penanaman kelapa sawit. Usaha ini bertujuan untuk menjamin kesejahteraan pekebun kecil sawit antaranya melalui interaksi dan pendedahan terhadap kaedah penanaman serta aktiviti hiliran. Oleh itu, kajian dilakukan untuk mengenalpasti tindakbalas pekebun kecil sawit terhadap pelbagai inovasi dan teknologi yang diperkenalkan oleh agensi terbabit. Hal ini perlu diberi penekanan kerana maklumat daripada pihak agensi berkaitan menjelaskan bahawa produktiviti per hektar di kawasan kajian tidak mencapai sasaran pengeluaran yang ditetapkan oleh pihak agensi. Seramai 300 orang pekebun kecil sawit di daerah Lahad Datu, Semporna dan Tawau di Sabah telah ditemubual dengan menggunakan soal selidik. Hasil kajian dianalisis secara diskriptif bagi memahami kekangan pekebun kecil sawit terhadap penggunaan inovasi dan teknologi dalam penanaman kelapa sawit di kawasan kajian. Dapatan kajian mendapati majoriti pekebun kecil menjelaskan bahawa kekangan penggunaan inovasi dan teknologi berpunca daripada faktor pemilikan tanah, masalah akses kepada kredit, akses kepada pasaran output dan input serta kekangan terhadap sumber dan bukan sumber. Kekangan tersebut telah mewujudkan jurang interaksi antara organisasi dengan pekebun kecil sawit. Jurang berkenaan telah mendorong mereka untuk terus bergantung kepada orang tengah. Oleh itu, pihak agensi dan pekebun kecil sawit perlu bekerjasama untuk menambahbaik jurang dengan memberi penekanan terhadap program inovasi dan teknologi dalam institusi pekebun kecil

    Artikel ini bertujuan untuk meneliti keterlibatan kominiti dalam pembangunan pertanian mampan. Kesedaran dan keperluan untuk memahami “apa” dan “apa yang seharusnya” memerlukan keterlibatan kedua-dua belah pihak sama ada komuniti dan agensi. Metod kajian dilakukan dikawasan pertanian Kuala Pilah telah melibatkan 60 orang responden. Pengaruh keterlibatan diteliti dengan menggunakan kaedah kolerasi. Hasil analisis mendapati taraf pendidikan yang rendah telah menyebabkan pengaruh keterlibatan berada pada tahap yang rendah. Sehubungan itu, hasil kajian mencadangkan agar pihak agensi memainkan peranan yang lebih proaktif bagi mewujudkan kefahaman petani dikawasan kajian berhubung dengan amalan pembangunan pertanian mampan
