The study of epigenetic regulation of gene HLA II. Clas within family relationships

Úvod: V současné postgenomické době představuje studium epigenetických regulací jeden z nástrojů pro porozumění funkci genů. Epigenetické regulace mohou přímo řídit časovou a prostorovou aktivitu genů či jejich umlčení. Molekulární podstata těchto regulací spočívá v modifikaci bází DNA, remodelaci chromatinu a RNA interferenci. Zároveň tyto mechanismy vykazují vlastní způsob přenosu dědičné informace do dalších generací nazývaný epigenetikcá dědičnost. V rámci mnoha chorob byla jako příčina prokázána deregulace epigenetické informace určitých genů. Z tohoto důvodu se jeví studium epigenomu HLA genů jako zvláště důležité, neboť tyto geny hrají zásadní roli v regulaci imunitního systému. Cíle: Cílem této práce je vytvořit popis epigenetických modifikací v rámci rodin. Jedná se o analýzu histonových modifikací a metylace DNA v promotorovém úseku genu HLA DQA1. Cílem bylo rovněž srovnat rozdíly v epigenetických modifikacích mezi alelami a také srovnání rozdílů v daných modifikacích mezi generacemi. Získané výsledky budou porovnány s analýzou míry exprese genu HLA DQA1. Metody: Z odebrané periferní krve dárců byly izolovány DNA, RNA a leukocyty. DNA byla použita ke genotypizaci. Ze získaných dat byla provedena analýza...Introduction: At our post-genomic era the studies of epigenetic regulation constitutes one of the tools for understanding the function of genes. Epigenetic regulation can directly control the temporal and spatial gene activity or silencing. The molecular basis of these regulations are DNA bases modifications, chromatin remodeling and RNA interference. At the same time, these mechanisms have a special way of transferring genetic information to subsequent generations called epigenetic inheritance. It has been proven epigenetic deregulation of certain genes as cause for many disease. For this reason, the study of epigenome HLA genes seems particularly important because these genes play a fundamental role in regulating the immune system. Aims: The aim of this work is to create a description of epigenetic modifications within families. It is an analysis of histone modifications and DNA methylation in the promoter region of the gene HLA DQA1. Legal and Extralegal Factors in Argumentation and Decision-making of the Constitutional Court of the Czech Republic Abstract The thesis deals with the influence of legal and extralegal factors on the decision-making of the Constitutional Court of the Czech Republic. Particularly, it focuses on the questions of which preconditions for decision-making of the Constitutional Court are created by the external political and social environment, how this decision-making reflects the different attitudes and approaches of individual judges and how it is influenced by the composition of the judicial panels. The author first summarizes a wide range of factors whose influence on court decisions has been observed. These include not only the content of legislation, but also judicial philosophy, including activism and self-restraint, and various extralegal factors observed by the attitudinal and strategic model of judicial decision-making, but also by psychological and economic studies. Subsequently, the thesis focuses on the Constitutional Court of the Czech Republic. Konkrétně se pak zaměřuje na otázky, jaké předpoklady pro rozhodování Ústavnímu soudu vytváří vnější politické a společenské prostředí, jak se v tomto rozhodování odráží odlišné postoje a přístupy jednotlivých soudů a jaký vliv na něj má složení soudních senátů. Autor nejprve shrnuje širokou škálu faktorů, u nichž byl pozorován vliv na rozhodování soudů. Mezi ně patří nejen obsah právní úpravy, ale také soudcovské filosofie včetně aktivismu a sebeomezení, a nejrůznější mimoprávní faktory, na něž poukazuje například tzv. postojový a strategický model soudního rozhodování, ale také psychologické a ekonomické studie. Scaling aspects of the sea-ice-drift dynamics and pack fracture

A study of the sea-ice dynamics in the periods of time prior to and during the cycles of basin-wide fragmentation of the ice cover in the Arctic Ocean is presented. The fractal geometry of the ice-sheets limited by leads and ridges was assessed using the satellite images, while the data on the correlated sea-ice motion were obtained in the research stations "North Pole 32" and "North Pole 33" established on the ice pack. The revealed decrease of the fractal dimension as a result of large-scale fragmentation is consistent with the localization of the fracture process (leads propagation). At the same time, the scaling properties of the distribution of amplitudes of ice-fields accelerations were insensitive to the event of sea-ice fragmentation. The temporal distribution of the accelerations was scale-invariant during "quiet" periods of sea-ice drift but disordered in the period of mechanical perturbation. The period of decorrelated (in time) ice-field motion during the important fracture event was interpreted as an inter-level transition in the hierarchic dynamical system. Import 03/08/2012Bakalářská práce v první části obsahuje popis dobývacích strojů spolu s jejich rozdělením. Dále se zabývá návrhem rozmístění ovládačů v kabině řidiče velkostroje. Nejdříve je uvedena rešerše ovládání velkostrojů, dále shrnuje požadavky na kabinu řidiče z hlediska norem a ergonomie. V závěru této části je samotný návrh rozmístění ovládačů v kabině. Druhá část se zabývá návrhem odpružení kabiny řidiče. Na začátku je zhodnocení dnešního stavu. Pokračuje návrh odpružení kabiny, nadefinování vstupních parametrů pro výpočet a samotný výpočet pružinového závěsu s jeho popisem.The first part of the bachelor thesis contains a description of mining machinery, together with its distribution. It also deals with the design layout of the controls in the cab giant machines. First, research of control of giant machines is presented, then it summarizes the requirements for cab drivers in terms of standards and ergonomics. At the end of this section is the actual design layout of the controls in the cab. The second part deals with cab suspension. At the beginning there is assessment of the current state. Information System for a Learning Center

Práce je zaměřena na použití jazyka PHP a MySQL při tvorbě informačního systému pro školící centrum. Před samotnou tvorbou aplikace je provedena důkladná analýza požadavků a následně navržena databázová struktura pro danou aplikaci tak, aby plně vyhovovala danému prostředí. Při analýze byl kladen zvýšený důraz na bezpečnost a rozšiřitelnost aplikace. Dále je popsána samotná implementace aplikace a problémy, které při tvorbě aplikace nastaly. V závěru jsou uvedena možná další rozšíření a zlepšení aplikace.This thesis is aimed at the usage of PHP language and MySQL database system towards the creation of information system for a learning center. A deep analysis is performed before the design of an application together with the database structure, which is appropriate for the environment. An increased emphasis is laid on security and easy expansibility of the application in this analysis. Further on is described the design phase of the application and all the problems that came up. In the end of this work are listed possible extensions and improvements of this application.

Import 05/08/2014Práce je rozdělena na několik částí. V úvodu je rešerše konstrukce a upevnění kabin řidiče velkostroje na rýpadlech používaných na obou lokalitách SD a.s. Následuje konstrukční návrh zdvihu kabiny řidiče, rozbor sil působících na kabinu řidiče za klidu i za provozu stroje a základní výpočet zdvihu kabiny. Dále je návrh kabiny řidiče velkostroje a jejího rozvržení. V závěru práce je návrh osy kabiny řidiče s její pevnostní kontrola.The work is divided into several parts. At the beginning of the research there are showned as the design as the mounting of the cabs on the excavators giant machines used at both locations SD a.s. The following part deals with the design of cab´s lift, analysis of the forces acting on the cab during the rest and during the machine operation and basic calculation of the lift´s cabin. The final part deals with the proposal of cab of giant machine and its patterns. Judge Rapporteur and the Panel: On the Influence of the Panel Composition on Decisions of the Constitutional Court of the Czech Republic about Constitutional Complaints

Článek se věnuje otázce, jaký vliv má složení senátu Ústavního soudu na rozhodnutí o ústavní stížnosti, konkrétně nakolik výsledek rozhodnutí závisí na míře vstřícnosti soudce zpravodaje a nakolik na míře vstřícnosti ostatních členů senátu. Tuto otázku nejprve zasazuje do kontextu obecné teorie senátního rozhodování a následně empiricky zkoumá v rozhodnutích Ústavního soudu. Na teoretické úrovni jsou formulovány tři základní účely senátního rozhodování: posílení legitimity rozhodnutí, eliminace chyb a dosažení promyšlenějšího rozhodnutí, a čtyři modely senátního rozhodování: konfliktní senát, strategický senát, kolegiální senát a formální senát. Objasněn je pojem a možné příčiny tzv. panelového efektu v senátním rozhodování. V rámci vlastního výzkumu je pak analyzováno, nakolik jsou uvedené funkce naplňovány v rozhodování Ústavního soudu, kterému z modelů se senáty Ústavního soudu nejvíce blíží a zda se v senátním rozhodování projevuje panelový efekt. Výzkum dospívá k závěru, že výsledek řízení o ústavních stížnostech závisí primárně na osobě soudce zpravodaje a jen minimálně na osobách dalších členů senátu. Senáty Ústavního soudu se proto nejvíce blíží formálnímu modelu fungování senátu a panelový efekt se v nich projevuje jen minimálně.The article deals with the influence of the composition of the panel of the Constitutional Court of the Czech Republic on the decision about a constitutional complaint. In particular a question is posed, to what extent does the result depend on the judge rapporteur and to what extent on other members of the panel? At first, the author analyses the theoretical context of the question and after that conducts an empirical research. On the theoretical level, three basic functions of panel decision-making are recognized: legitimacy-strengthening, mistake-correction and achieving deeper consideration of the decision. After that, four types of judicial panel are distinguished: adversarial, strategic, collegial and formal panel. The theoretical part also clarifies the concept and causes of the so-called panel effect. The empirical research analyses, which type of panels occurs at the Constitutional Court, how does the panel decision-making of the Constitutional Court fulfil its functions, and whether the panel effect can be observed. The author concludes, that the result of the proceedings about a constitutional complaint depends primarily on the judge rapporteur and the influence of other panel members is weak. The Constitutional court panels thus resemble the model of formal panel. The panel effect is insignificant in the panel decision-making of the Constitutional court
