    Financing obstacles and growth: an analysis for euro area non-financial corporations

    This paper investigates whether financial obstacles, and, more generally, financial pressure faced by firms, significantly affect firm growth. For this purpose, we use an unbalanced panel of about 1,000,000 observations for around 155,000 non-financial corporations in five euro area countries. In addition to the balance sheet information in this panel, we also rely on firm-level survey data. In this way we are able to work out a direct measure of the firms’ probability of facing financing obstacles. Our results indicate that, though based on few variables, this measure appears to be relevant in explaining firm growth in four out of the five countries considered. Other firm-level variables related to the financial pressure faced by firms, such as cash flow (debt burden) are found to exert a positive (negative) impact on firm growth, while the results for leverage are less clear-cut.     Multimodal arguments in the mainstream press: Illustrating portrayals of migration

    This paper sketches a methodological integration of tools from multimodal discourse analysis and argumentation in order to unveil opaque argumentative inferences emerging in multimodal configurations (i.e., headlines and press photos) of seemingly non-argumentative genres such as news articles. We offer illustrative examples from the Italian mainstream press in the context of the so-called European ‘refugee crisis.’ Overall, our methodologically oriented proposal aims to deepen the debate in the area of multimodal argumentation. To that end, we sketch a dialogue with other perspectives that specifically study argumentative inference in multimodal configurations. We contend that this approach enables a better examination of the argumentative potential that is implicitly sustained in multimodal configurations of news articles

    Global Stability Analysis of a Compressible Turbulent Flow around a High-Lift Configuration

    The purpose of this work is to analyze a complex high lift configuration for which significant regions of separated flow are present. Current state of the art methods have some diffculty to predict the origin and the progression of this separated flow when increasing the angle of attack. The mechanisms responsible for the maximum lift limit on multi-element wing con?gurations are not clear; this stability analysis could help to understand the physics behind the phenomenon and to find a relation between the flow separation and the instability onset. The methodology presented herein consists in the computation of a steady base flow solution based on a finite volume discretization and a proposal of the solution for a generalized eigenvalue problem corresponding to the perturbed and linearized problem. The eigenvalue problem has been solved with the Arnoldi iterative method, one of the Krylov subspace projection methods. The described methodology was applied to the NACA0012 test case in subsonic and in transonic conditions and, finally, for the first time to the authors knowledge, on an industrial multi-component geometry, such as the A310 airfoil, in order to identify low frequency instabilities related to the separation. One important conclusion is that for all the analyzed geometries, one unstable mode related to flow separation appears for an angle of attack greater than the one correspondent to the maximum lift coe?cient condition. Finally, an adjoint study was carried out in order to evaluate the receptivity and the structural sensitivity of the geometries, giving an indication of the domain region that could be modified resulting in the biggest change of the flowfield

    Bridging the gap: returning genetic results to indigenous communities in Latin America

    In response to inequality in access to genomics research, efforts are underway to include underrepresented minorities, but explicit (and enforcing) guidelines are mostly targeted toward the Global North. In this work, we elaborate on the need to return scientific results to indigenous communities, reporting the actions we have taken in a recent genomic study with Mapuche communities in Chile. Our approach acknowledged the social dynamics perpetuating colonial hierarchies. We framed genetic results to empower indigenous knowledge and communities’ history and identities. A fundamental step in our strategy has been sharing the results with the communities before publishing the scientific paper, which allowed us to incorporate community perspectives. We faced the challenge of translating genetic concepts like admixture, emphasizing the distinction between identity and biology. To reach a broad and diverse audience, we disseminated the study results to single community members, cultural representatives, and high schools, highlighting the importance of the history of the region before the European contact. To facilitate results dissemination, we prepared didactic material and a report in Spanish written in non-specialized language, targeting a wider Latin American readership. This work illustrates the benefits of discussing scientific findings with indigenous communities, demonstrating that a collaborative and culturally sensitive approach fosters knowledge sharing and community empowerment and challenges power dynamics in genetic research.     Progressive Seizure Aggravation in the Repeated 6-Hz Corneal Stimulation Model Is Accompanied by Marked Increase in Hippocampal p-ERK1/2 Immunoreactivity in Neurons

    The 6-Hz corneal stimulation test is used to screen novel antiepileptic molecules to overcome the problem of drug refractoriness. Although recognized as a standard test, it has been evaluated only recently in the attempt to characterize the putative neuronal networks involved in seizures caused by corneal stimulation. In particular, by recording from the CA1 region we previously established that the hippocampus participates to propagation of seizure activity. However, these findings were not corroborated by using markers of neuronal activation such as FosB/ΔFosB antigens. In view of this discrepancy, we performed new experiments to characterize the changes in levels of phosphorylated extracellular signal-regulated kinases1/2 (p-ERK1/2), which are also used as markers of neuronal activation. To this aim, mice underwent corneal stimulation up to three different times, in three sessions separated by an interval of 3 days. To characterize a group in which seizures could be prevented by pharmacological treatment, we also considered pretreatment with the ghrelin receptor antagonist EP-80317 (330 ÎŒg/kg). Control mice were sham-treated. Video electrocorticographic (ECoG) recordings were obtained from mice belonging to each group of treatment. Animals were finally used to characterize the immunoreactivity for FosB/ΔFosB and p-ERK1/2 in the hippocampus. As previously shown, FosB/ΔFosB levels were highly increased throughout the hippocampus by the first induced seizure but, in spite of the progressively increased seizure severity, they were restored to control levels after the third stimulation. At variance, corneal stimulation caused a progressive increase in p-ERK1/2 immunoreactivity all over the hippocampus, especially in CA1, peaking in the third session. Predictably, EP-80317 administration reduced both duration and severity of seizures, prevented the increase in FosB/ΔFosB levels in the first session, and partially counteracted the increase in p-ERK1/2 levels in the third session. The vast majority of p-ERK1/2 immunopositive cells were co-labeled with FosB/ΔFosB antibodies, suggesting the existence of a relationship between the investigated markers in a subpopulation of neurons activated by seizures. These findings suggest that p-ERK1/2 are useful markers to define the aggravation of seizures and the response to anticonvulsant treatments. In particular, p-ERK1/2 expression clearly identified the involvement of hippocampal regions during seizure aggravation in the 6-Hz model

    Development and Validation of MPS-Based System for Human Appearance Prediction in Challenging Forensic Samples

    Forensic DNA phenotyping (FDP) provides the ability to predict the human external traits from unknown sample donors, directly from minute amounts of DNA found at the crime scene. We developed a MPS multiplex assay, with the aim of genotyping all 41 DNA markers included in the HIrisPlex-S system for simultaneous prediction of eye, hair and skin colours. Forensic samples such as blood, skeletal remains, touch DNA, saliva swab, artificially degraded samples together with individuals with known phenotypes and a set of 2800 M control DNA were sequenced on the Ion Torrent platform in order to evaluate the concordance testing results and the forensic suitability of the 41-plex MPS assay. The panel was evaluated by testing a different number of PCR cycles and the volume of reagents for library preparation. The study demonstrated that full and reliable profiles were obtained with 0.1–5 ng, even with high degraded DNA. The increment of the number of PCR cycles results in an improvement of correctly genotyping and phenotyping for samples with low amounts of degraded DNA but higher frequencies of artefacts were found. The high DNA degradation level did not influence the correct genotyping and phenotyping and the critical parameter affecting the result is the quantity of input DNA. Eye and hair colour was predicted in 92.60% of individuals and skin colour in 85.15% of individuals. The results suggest that this MPS assay is robust, highly sensitive and useful for human pigmentation prediction in the forensic genetic field

    Involvement of PPAR\u3b3 in the anticonvulsant activity of EP-80317, a ghrelin receptor antagonist

    Ghrelin, des-acyl ghrelin and other related peptides possess anticonvulsant activities. Although ghrelin and cognate peptides were shown to physiologically regulate only the ghrelin receptor, some of them were pharmacologically proved to activate the peroxisome proliferator-activated receptor gamma (PPAR\u3b3) through stimulation of the scavenger receptor CD36 in macrophages. In our study, we challenged the hypothesis that PPAR\u3b3 could be involved in the anticonvulsant effects of EP-80317, a ghrelin receptor antagonist. For this purpose, we used the PPAR\u3b3 antagonist GW9662 to evaluate the modulation of EP-80317 anticonvulsant properties in two different models. Firstly, the anticonvulsant effects of EP-80317 were studied in rats treated with pilocarpine to induce status epilepticus (SE). Secondly, the anticonvulsant activity of EP-80317 was ascertained in the repeated 6-Hz corneal stimulation model in mice. Behavioral and video electrocorticographic (ECoG) analyses were performed in both models. We also characterized levels of immunoreactivity for PPAR\u3b3 in the hippocampus of 6-Hz corneally stimulated mice. EP-80317 predictably antagonized seizures in both models. Pre-treatment with GW9662 counteracted almost all EP-80317 effects both in mice and rats. Only the effects of EP-80317 on power spectra of ECoGs recorded during repeated 6-Hz corneal stimulation were practically unaffected by GW9662 administration. Moreover, GW9662 alone produced a decrease in the latency of tonic-clonic seizures and accelerated the onset of SE in rats. Finally, in the hippocampus of mice treated with EP-80317 we found increased levels of PPAR\u3b3 immunoreactivity. Overall, these results support the hypothesis that PPAR\u3b3 is able to modulate seizures and mediates the anticonvulsant effects of EP-80317

    Urban food waste for soil amendment? Analysis and characterisation of waste-based compost for soil fertility management in agroecological horticultural production systems in the city of Rosario, Argentina

    Urban and peri-urban lands can be an important source of food production for localised and sustainable food systems, however, their soils can be of poor quality, degraded or damaged by anthropic activities, and little is known about their suitability or safety. This paper aims to contribute to this knowledge gap by assessing the soil remediation capacity and qualities of different types of compost made from urban and peri-urban organic wastes for agroecological food production. Prepared over the course of 2021, and used in 2022 for food growing, five different composts were observed and analysed, in two different farms in the city of Rosario, Argentina. Four raw materials generated largely by local industries were used to make the composts: chicken manure, rumen (cow’s stomachs), brewer’s bagasse (byproducts of the beer industry) and urban leaves collected from the municipality waste collection. These were mixed in different proportions (all reaching the 20–30 C/N ratio, typical of quality compost) to produce viable growing substrates where radishes and lettuces were grown. The aim of the study was to assess the possibility, quality and limitations to use locally available organic inputs for soil fertility management in agroecological farming, in the context of urbanisation and to assess pathways to develop closed-cycle agroecological agriculture at metropolitan level. Natural manure substrates (raw and composted) were analysed, as well as crops grown and fertilised with each of the substrates. The attributes and limiting factors of each substrate and their response to local soil conditions were compared and physicochemical, biochemical, and microbiological analyses were performed, including among others, the study of microbial biomass, biological activity, biophytotoxicity, pH, aerobic heterotrophs, nitrogen fixation, and the presence of antibiotics, agrochemicals and heavy metals. The results of the analyses show that all the composted materials improved the physical, chemical and biological properties. However, in some cases, pollutants were present even after composting. Analysis carried out on the vegetables generally indicate undetectable levels or levels below the admissible limits, demonstrating the filtering capacity of the different composts and the soil
