Towards Skeleton based Reconstruction : From Projective Skeletonization to Canal Surface Estimation
International audienceWe present a novel approach to reconstruct a 3D object from images corresponding to two different viewpoints: we estimate the skeleton of the object instead of its surface. The originality of the method is to be able to reconstruct a tubular object with a limited number of input images. Unlike classical reconstruction methods, like multi-view stereo or more recently structure-from-motion, this approach does not rely on interest points but estimates the topology of the object and derives its surface. Our contribution are twofold. First, given two perspective images of the 3D shape, the projection of the skeleton is computed in 2D. Secondly the 3D skeleton is reconstructed from the two projections using triangulation and matching. A mesh is finally derived for each skeleton branch
Caractérisation de la projection du squelette d'une surface canal 3D : Application à la reconstruction 3D à partir de deux images
International audienceNous présentons dans cet article une nouvelle approche pour reconstruire un objet 3D à partir de deux images de celui-ci. L'originalité de notre approche vient du fait que nous n'estimons pas directement la surface de l'objet 3D mais son squelette. Ce travail s'appuie sur les deux contributions suivantes. PremiÚrement, nous décrivons la relation existant entre un squelette 3D et sa projection, orthographique ou perspective, sur un plan image. Ensuite, nous montrons comment retrouver le squelette 3D à partir de deux de ses projections. Contrairement aux méthodes de reconstruction 3D classiques qui génÚrent un nuage de points sans maillage, cette approche a pour avantage de reconstruire aussi la topologie de l'objet, c'est-à -dire d'en retrouver un maillage sans traitement annexe. Nous traitons ici des objets représentables par un squelette 3D curviligne et nous supposons aussi que les points de vue sont calibrés
beDNA : un projet visant Ă la collection systĂ©matique dâĂ©chantillons humains archĂ©ologiques Ă vocation palĂ©ogĂ©nĂ©tique â une premiĂšre expĂ©rimentation
La palĂ©ogĂ©nĂ©tique occupe dĂ©sormais une place importante dans les problĂ©matiques archĂ©ologiques. Toutefois, les analyses dâADN ancien peuvent ĂȘtre desservies, voire empĂȘchĂ©es, par lâĂ©tat de prĂ©servation des Ă©chantillons en raison de contamination par de lâADN moderne ou de mauvaises conditions de stockage. Le projet beDNA, "banque dâĂ©chantillons et de DonnĂ©es Nationale ArchĂ©ogĂ©nĂ©tique", souhaite donner les moyens dâanalyses palĂ©ogĂ©nĂ©tiques futures, en proposant le stockage systĂ©matique dâĂ©chantillons des squelettes humains tenant compte des contraintes inhĂ©rentes Ă la prĂ©servation de lâADN ancien. Le projet implique (1) un protocole systĂ©matique dâĂ©chantillonnage "propre" des restes humains sur le terrain commun Ă toutes les opĂ©rations archĂ©ologiques, (2) un espace de stockage dĂ©diĂ© Ă ces Ă©chantillons adaptĂ© Ă la conservation de lâADN ancien, (3) une base de donnĂ©es faisant le lien entre les sites et les Ă©chantillons conservĂ©s dans la banque, (4) lâapprobation par lâĂtat des demandes dâanalyse dâĂ©chantillons aprĂšs expertise. La phase de test du projet, initiĂ©e en septembre 2020 sur la rĂ©gion Ăle-de-France, nous a permis dâĂ©valuer et dâajuster le protocole dâĂ©chantillonnage sur le terrain et les dispositifs de transfert vers la banque. Cette note prĂ©sente les Ă©tapes envisagĂ©es pour chaque Ă©chantillon, depuis les terrains jusquâaux laboratoires dâanalyse gĂ©nĂ©tique, ainsi que le dĂ©roulement de sa phase test, en cours, et les premiers retours dâexpĂ©rience.Palaeogenetics is becoming increasingly important in tackling archaeological issues. However, analyses of ancient DNA can be hampered or even prevented by the state of preservation of samples due to poor storage conditions, and because of contamination by modern DNA. The beDNA project for a national archaeological genetic data and sample bank (banque dâĂ©chantillons et de DonnĂ©es Nationale ArchĂ©ogĂ©nĂ©tique) is developing the means to enable future palaeogenetic analyses by systematically storing human skeletal samples, with the constraints inherent to the preservation of ancient DNA taken into account. This project comprises (1) a systematic protocol for "clean" sampling of human remains to be common to all archaeological operations, (2) a dedicated storage space for samples, suited to aDNA preservation, (3) a database linking sites with the samples stored in the bank, (4) approval of sample analysis requests by authorities, after expert review. The test phase of the project, which began in September 2020 in the Ăle-de-France region, enabled us to evaluate and adjust both the sampling protocol in the field and the transfer process to the beDNA bank. This note describes the different stages envisaged for each sample, from the archaeological field to the genetics laboratory, as well as the development of the experimental phase and initial feedback from it
The PP2A inhibitor I2PP2A is essential for sister chromatid segregation in oocyte meiosis II.
Haploid gametes are generated through two consecutive meiotic divisions, with the segregation of chromosome pairs in meiosis I and sister chromatids in meiosis II. Separase-mediated stepwise removal of cohesion, first from chromosome arms and later from the centromere region, is a prerequisite for maintaining sister chromatids together until their separation in meiosis II [1]. In all model organisms, centromeric cohesin is protected from separase-dependent removal in meiosis I through the activity of PP2A-B56 phosphatase, which is recruited to centromeres by shugoshin/MEI-S332 (Sgo) [2-5]. How this protection of centromeric cohesin is removed in meiosis II is not entirely clear; we find that all the PP2A subunits remain colocalized with the cohesin subunit Rec8 at the centromere of metaphase II chromosomes. Here, we show that sister chromatid separation in oocytes depends on a PP2A inhibitor, namely I2PP2A. I2PP2A colocalizes with the PP2A enzyme at centromeres at metaphase II, independently of bipolar attachment. When I2PP2A is depleted, sister chromatids fail to segregate during meiosis II. Our findings demonstrate that in oocytes I2PP2A is essential for faithful sister chromatid segregation by mediating deprotection of centromeric cohesin in meiosis II
Synthesis and stability evaluation of novel peptidomimetic Caspase-1 inhibitors for topical application
International audienceDuring our search for topically-active Caspase-1 inhibitors, we identified a novel class of potent in-hibitors based on a 1,3,5-trisubstituted uracil motif equipped with an L-aspartate semi-aldehyde derived warhead. In the literature, the majority of Caspase-1 inhibitors possessing the same warhead have been designed and evaluated for oral administration as the ethyl acetal pro-drug form. For our topical program , the pro-drug acetal form was not fully hydrolysed in the skin and was unstable in many of our standard topical excipients, therefore, we were obliged to focus on the actual hemiacetal drug form of the molecule during our drug discovery program. Our work focuses on both the synthesis and achiral and chiral stability of the final drug molecules in topical excipients
The p300/CBP-associated factor (PCAF) is a cofactor of ATF4 for amino acid-regulated transcription of CHOP
When an essential amino acid is limited, a signaling cascade is triggered that leads to increased translation of the âmaster regulatorâ, activating transcription factor 4 (ATF4), and resulting in the induction of specific target genes. Binding of ATF4 to the amino acid response element (AARE) is an essential step in the transcriptional activation of CHOP (a CCAAT/enhancer-binding protein-related gene) by amino acid deprivation. We set out to identify proteins that interact with ATF4 and that play a role in the transcriptional activation of CHOP. Using a tandem affinity purification (TAP) tag approach, we identified p300/CBP-associated factor (PCAF) as a novel interaction partner of ATF4 in leucine-starved cells. We show that the N-terminal region of ATF4 is required for a direct interaction with PCAF and demonstrate that PCAF is involved in the full transcriptional response of CHOP by amino acid starvation. Chromatin immunoprecipitation analysis revealed that PCAF is engaged on the CHOP AARE in response to amino acid starvation and that ATF4 is essential for its recruitment. We also show that PCAF stimulates ATF4-driven transcription via its histone acetyltransferase domain. Thus PCAF acts as a coactivator of ATF4 and is involved in the enhancement of CHOP transcription following amino acid starvation
RÎle du récepteur des androgÚnes (RA) et du récepteur activé par les proliférateurs des peroxysomes b (PPARb) dans le muscle squelettique chez la souris
Chez les MammifÚres, les muscles squelettiques représentent 55% de la masse corporelle. Leurs propriétés d'excitabilité, de contractilité et d'élasticité leur permettent de générer force et mouvements. Ils possÚdent également une fonction métabolique essentielle. Mon travail de thÚse se propose de caractériser le rÎle de deux récepteurs nucléaires, le récepteur des androgÚnes (RA) et le récepteur activé par les proliférateurs des peroxysomes b (PPARb) dans ces fonctions contractiles et métaboliques. Pour ce faire, nous avons invalidé sélectivement dans les myocytes du muscle squelettique chez la souris, soit le RA, soit PPARb, à l'aide du systÚme Cre-LoxP. Dans la premiÚre partie de ce travail, nous avons démontré que le RA des myocytes relaye les effets anaboliques des androgÚnes dans les muscles du périnée, mais pas dans ceux des membres. En revanche, nous avons établi que, dans les muscles des membres, il joue un rÎle essentiel dans la régulation des capacités contractiles. Dans la deuxiÚme partie de la thÚse, nous avons démontré que l'invalidation de PPARb sélectivement dans les myocytes, aussi bien de maniÚre constitutive que spécifiquement à l'ùge adulte, induit une conversion de fibres lentes et oxydatives en fibres rapides et glycolytiques. De plus, cette conversion de fibres est associée à une diminution des capacités d'oxydation des acides gras par les muscles et conduit au développement d'une obésité et d'un diabÚte de type II avec la sédentarité. En conclusion, ces résultats apportent une meilleure compréhension des fonctions musculaires et permettront à long terme le développement de nouvelles thérapies dans le traitement de la perte de masse et de force musculaire, ainsi que dans le traitement des désordres métaboliques.In mammals, skeletal muscles account for up to 55% of total body mass. Thanks to their excitability, contractility and elasticity properties, they generate strength and movements. They also play a major role in metabolic functions. The aim of this work is to characterize the role of the two following nuclear receptors, the androgen receptor (AR), and the peroxisome proliferator-activated receptor b (PPARb), in skeletal muscle contractile and metabolic functions. To this end, we selectively invalidated either AR or PPARb, using the Cre-LoxP system in mice myocytes. In the first part of this work, we demonstrate that myocytic AR mediates androgen-induced perineal muscle fiber hypertrophy. In contrast, in hindlimb muscles, myocytic AR do not control their mass, but is required for optimal strength of fast-twitch and intermediary muscles. In the second part of this work, we demonstrate that both constitutive and selective ablation at adulthood of PPARb in skeletal muscles leads to a oxidative-to-glycolytic fiber type switching and decreases fatty acid oxidative capacities in muscles, thus resulting In conclusion, our work provides a better understanding of muscle functions and will allow the development of new therapies against muscle wasting and metabolic disorders
