    Actinobacteria from Greenwich Island and Dee Island: isolation, diversity and distribution

    Actinobacteria from underexplored and unusual environments have gained significant attention for their capability in producing novel bioactive molecules of diverse chemical entities. Streptomyces is the most prolific Actinobacteria in producing useful molecules. Rapid decline effectiveness of existing antibiotics in the treatment of infections are caused by the emergence of multidrug-resistant pathogens. Intensive efforts are urgently required in isolating non-Streptomyces or rare Actinobacteria and understanding of their distribution in the harsh environment for new drug discovery. In this study, pretreatment of soil samples with 1.5% phenol was used for the selective isolation of Actinobacteria from Dee Island and Greenwich Island. A high number of non-Streptomyces (69.4%) or rare Actinobacteria was significantly recovered despite the Streptomyces (30.6%), including the genera Micromonospora, Micrococcus, Kocuria, Dermacoccus, Brachybacterium, Brevibacterium, Rhodococcus, Microbacterium and Rothia. Reduced diversity and shift of distribution were observed at the elevated level of soil pH. The members of genera Streptomyces, Micromonospora and Micrococcus were found to distribute and tolerate to a relatively high pH level of soil (pH 9.4-9.5), and could potentially be alkaliphilic Actinobacteria. The phylogenetic analysis had revealed some potentially new taxa members of the genera Micromonospora, Micrococcus and Rhodococcus. Principal Component Analysis of soil samples was used to uncover the factors that underlie the diversity of culturable Actinobacteria. Water availability in soil was examined as the principal factor that shaped the diversity of the Actinobacteria, by providing a dynamic source for microbial interactions and elevated diversity of Actinobacteria

    Prevalence of Listeria monocytogenes in frozen burger patties in Malaysia

    A total of 112 burger patties (35 beef burger patties, 39 chicken burger patties and 38 fish burger patties) which are commercially available at retail level were investigated for the presence and number of Listeria monocytogenes. These samples were analyzed using MPN-PCR method and conventional culturing methods. L. monocytogenes was detected in 33.3% of chicken burger patties, 22.9%of beef patties, and 10.5% of fish patty samples. From all contaminated raw burger patties, the estimated count of L. monocytogenes was ranged from 3 to 75 MPN/g. The results suggest that burger act as a potential source of listeriosis if the contaminated burger patty is consumed without adequate cooking. The risk associated with consumption of these samples was found to be high particularly for processed food at retail level in Malaysia. Therefore, food manufacturers play an importantrole in monitoring the manufacturing process and conduct a periodical surveillance on microbiological quality assessment on the processing plants. Besides, there is a need to increase awareness ofconsumers and food handlers to practice proper cooking of the burger patties before the point of consumption, to reduce the risk of listeria infection

    The perspective of students on drivers and benefits of building information modelling incorporation into quantity surveying profession in Klang Valley Malaysia

    Building Information Modelling (BIM) is a very useful tool that facilitates architecture, engineering and construction (AEC) professionals and stakeholders in planning, designing and constructing the buildings through 3D models. BIM can be widened to building operations and data storage which can be accessible by owners and others. Such data help owners and stakeholders to generate results according to the information gained through BIM models. The objectives of this study were to identify the perspective of students on drivers of BIM incorporation into the quantity surveying profession and to identify the perspective of students on benefits of BIM incorporation into the quantity surveying profession. A questionnaire survey was carried out to gain the students’ perspective on drivers and benefits of BIM incorporation into the quantity surveying profession in Klang Valley, Malaysia. Specifically, this study investigated twelve drivers and fourteen benefits of BIM incorporation into the quantity surveying profession. The top three drivers were improving the capacity to provide whole-life value to the client, desire for innovation to remain competitive and strong support from university management and industry. The top three benefits were BIM provides fast, effective and efficient quantity take-off and cost estimation, time savings in the preparation of estimating cost and improved visualization for better understanding of designs for measurement and minimise omissions. For future research, it is recommended that the study be replicated at other regions so that a clearer view of this topic can be obtained. Besides, qualitative research methods could be used in identifying other drivers and benefits not covered in this study. By answering the questions in the survey form, the students were able to gain some knowledge on BIM and its importance to the quantity surveying profession. Also, it would be interesting to include industrial practitioners in this kind of study, allowing comparisons of the results between academia and industry at a later stage. Nonetheless, this study benefited the undergraduate students pursuing the Bachelor of Science (Hons) Quantity Surveying programme, universities, colleges and other institutions that offered the quantity surveying programmes at various levels and quantity surveyors working in the construction industry by exposing them to a comprehensive list of drivers and benefits of BIM incorporation into quantity surveying profession. In a way, this study helped promoted BIM and its implementation in the field of quantity surveying in Klang Valley, Malaysia

    Biofilm formation by Salmonella Typhi and Salmonella Typhimurium on plastic cutting board and its transfer to dragon fruit

    Adhesion of microorganism to food contact surface can become a source of microbial contamination.Enhanced resistances to environmental stresses are exhibited by biofilm producers. In this study, biofilm formation by Salmonella Typhi and Salmonella Typhimurium on plastic cutting board was accessed before the evaluation of the transfer of these two pathogens from plastic cutting board to dragon fruit. By using crystal violet assay, it was found that the adhesion on plastic cutting board by these two pathogens was the greatest at time 12 h. Results showed that Salmonella adhesion is strain-dependent and varied with time. The mean transfer rate from contaminated plastic cutting board to dragon fruit was examined to be 0.79 and 0.72 for Salmonella Typhi and Salmonella Typhimurium, respectively. This indicated that there is a risk of cross-contamination which should be concerned

    Occurrance and antibiotic resistance of Campylobacter jejuni and Campylobacter coli in retail broiler chicken

    Broiler part samples (80 fresh and 80 chilled) were examined for the prevalence and numbers of C. jejuni and C. coli by employing most-probable-number (MPN) and polymerase chain reaction (PCR) techniques. The prevalence of the bacteria was high where C. jejuni was detected in 92.5% fresh and 53.8% chilled samples while C. coli in 80.0% fresh and 56.3% chilled. The number of these bacteria in the positive fresh and chilled samples was from 3 to more than 2400 MPN/g and from 3 to 290 MPN/g, respectively. Antibiotic resistance test (using Kirby-Bauer disc diffusion method) on 10 C. jejuni and 13 C. coli isolates toward ampicillin, tobramycin, enrofloxacin, ciprofloxacin, tetracycline, cephalothin, gentamicin and norfloxacin revealed high resistance toward all antibiotics (20.0% - 100.0%). All isolates were resistant to at least two antibiotics. This study highlights the potential of multidrug-resistant C. jejuni and C. coli transmission to humans through fresh and chilled broiler parts. Consecutive studies with bigger sample sizes and covering all over Malaysia are warranted in future

    Characterization of multiple-antimicrobial-resistant Salmonella enterica Subsp. enterica isolated from indigenous vegetables and poultry in Malaysia.

    Aims: The aims of this communication were to study characterization of serogroups among Salmonella isolates and the relationship of antimicrobial resistance to serogroups. Multiple antimicrobial resistance (MAR) was performed on 189 Salmonella enterica isolates associated with 38 different serovars that were recovered from poultry and four types of indigenous vegetables. Methods and Results: Disc diffusion analysis was performed with a selection of 10 different antimicrobial agents. Isolates recovered from indigenous vegetables showed 100% (134/134) resistant to erythromycin and followed by 42%, 34%, 19% for tetracycline, streptomycin and trimethroprim-sulfamethoxazole respectively. In general, 90·1% (50/55) and 56·7% (76/134) of Salmonella isolated from poultry and indigenous vegetables, respectively, exhibited MAR index more than 0·2. Conclusions: Characterization of Salmonella isolates based on the MAR results indicated that poultry still remains as the main reservoir for multi-drug-resistant Salmonella. Four isolates from the indigenous vegetables showed the highest MAR index in this study. Further investigations need to be conducted to determine if Salmonella isolates recovered from indigenous vegetables were gaining more antimicrobial resistance.
