    Effects of various inulin levels on in vitro digestibility of corn silage, perennial ryegrass (Lolium perenne L.) and common vetch (Vicia sativa L.)/oat (Avena sativa L.) hay

    The aim of this study was to investigate the effects of various inulin levels on in vitro true dry matter digestibility (IVTDDM) and in vitro neutral detergent fibre digestibility (IVTDNDF) of corn silage (CS), perennial ryegrass (PR), and common vetch/oat hay (VO). Inulin was added to the fermenter at concentrations of 0 (CSC, PRC, VOC), 100 (CS100, PR100, VO100), 200 (CS200, PR200, VO200), and 300 (CS300, PR300, VO300) mg/litre of total culture fluid using an in vitro DaisyII incubator. Each fermenter contained 1600 ml buffer solution and 400 ml rumen fluid. The IVTDDM and IVTDNDF were determined with a DaisyII incubator and rumen fluid obtained from three cannulated Karayaka rams. The IVTDDM values (%) for PRC, PR100, PR200, and PR300 were 70.06 ± 1.133, 73.21 ± 4.153, 70.36 ± 0.506, and 66.69 ± 1.317, respectively. The effects of various inulin levels on IVTDDM and IVTDNDF values of PR were significant (P <0.05). The IVTDDM and IVTDNDF values for CS and VO were not significantly (P >0.05) different. Among the treatments, supplementation of inulin to CS and VO did not have a significant (P >0.05) effect on IVTDDM and IVTDNDF values. The high dose of inulin (PR300) reduced IVTDDM and IVTDNDF, whereas PR100 showed a statistically significant (P <0.05) increase on IVTDDM and IVTDNDF. However, in vivo studies with PR may be required to show the effects of various levels of inulin supplementation to support the IVTD findings of the current study.Keywords: Inulin, in vitro true digestibility, hay, silag

    The effects of seasonal variation on the microbial-n flow to the small intestine and prediction of feed intake in grazing karayaka sheep

    The objectives of the present study were to estimate the microbial-N flow to the small intestine and to predict the digestible organic matter intake (DOMI) in grazing Karayaka sheep based on urinary excretion of purine derivatives (xanthine, hypoxanthine, uric acid and allantoin) by the use of spot urine sampling under field conditions. In the trial, 10 Karayaka sheep from 2 to 3 years of age were used. The animals were grazed in a pasture for ten months and fed with concentrate and vetch plus oat hay for the other two months (January and February) indoors. Highly significant linear and cubic relationships (P<0.001) were found among months for purine derivatives index, purine derivatives excretion, purine derivatives absorption, microbial-N and DOMI. Through urine sampling and the determination of levels of excreted urinary PD and the Purine Derivatives:Creatinine ratio (PDC index), microbial-N values were estimated and they indicated that the protein nutrition of the sheep was insufficient. In conclusion, the prediction of protein nutrition of sheep under the field conditions may be possible with the use of spot urine sampling, urinary excreted PD and PDC index. The mean purine derivative levels in spot urine samples from sheep were highest in June, July and October. Protein nutrition of pastured sheep may be affected by weather changes, including rainfall. Spot urine sampling may useful in modeling the feed consumption of pasturing sheep. However, further studies are required under different field conditions with different breeds of sheep to develop spot urine sampling as a model

Investigation of this entity have increased our insight into the important role of the leptin-melanocortin pathway in energy balance. Here, we present a patient with POMC deficiency due to a homozygous c.206delC mutation in the POMC gene. We discuss the pathogenesis of this condition with emphasis on the crosstalk between hypothalamic and peripheral signals in the development of obesity and the POMC-melanocortin 4 receptors system as a target for therapeutic intervention

    A service-oriented admission control strategy for class-based IP networks

    The clear trend toward the integration of current and emerging applications and services in the Internet launches new demands on service deployment and management. Distributed service-oriented traffic control mechanisms, operating with minimum impact on network performance, assume a crucial role as regards controlling services quality and network resources transparently and efficiently. In this paper, we describe and specify a lightweight distributed admission control (AC) model based on per-class monitoring feedback for ensuring the quality of distinct service levels in multiclass and multidomain environments. The model design, covering explicit and implicit AC, exhibits relevant properties that allow managing quality of service (QoS) and service-level specifications (SLSs) in multiservice IP networks in a flexible and scalable manner. These properties, stemming from the way service-dependent AC and on-line service performance monitoring are proposed and articulated in the model’s architecture and operation, allow a self-adaptive service and resource management, while abstracting from network core complexity and heterogeneity. A proof of concept is provided to illustrate the AC criteria ability in satisfying multiple service class commitments efficiently. The obtained results show that the self-adaptive behavior inherent to on-line measurement-based service management, combined with the established AC rules, is effective in controlling each class QoS and SLS commitments consistently

    Proerythroblast Cells of Diamond-Blackfan Anemia Patients With RPS19 and CECR1 Mutations Have Similar Transcriptomic Signature

    Diamond Blackfan Anemia (DBA) is an inherited bone marrow (BM) failure syndrome, characterized by a paucity of erythroid differentiation. DBA is mainly caused by the mutations in ribosomal protein genes, hence classified as ribosomopathy. However, in approximately 30% of patients, the molecular etiology cannot be discovered. RPS19 germline mutations caused 25% of the cases. On the other hand, CECR1 mutations also cause phenotypes similar to DBA but not being a ribosomopathy. Due to the blockade of erythropoiesis in the BM, we investigated the transcriptomic profile of three different cell types of BM resident cells of DBA patients and compared them with healthy donors. From BM aspirates BM mononuclear cells (MNCs) were isolated and hematopoietic stem cells (HSC) [CD71–CD34+ CD38mo/lo], megakaryocyte–erythroid progenitor cells (MEP) [CD71–CD34+ CD38hi] and Proerythroblasts [CD71+ CD117+ CD38+] were sorted and analyzed with a transcriptomic approach. Among all these cells, proerythroblasts had the most different transcriptomic profile. The genes associated with cellular stress/immune responses were increased and some of the transcription factors that play a role in erythroid differentiation had altered expression in DBA proerythroblasts. We also showed that gene expression levels of ribosomal proteins were decreased in DBA proerythroblasts. In addition to these, colony formation assay (CFU-E) provided functional evidence of the failure of erythroid differentiation in DBA patients. According to our findings that all patients resembling both RPS19 and CECR1 mutations have common transcriptomic signatures, it may be possible that inflammatory BM niche may have a role in DBA pathogenesis

    Global burden of human brucellosis : a systematic review of disease frequency

    BACKGROUND: This report presents a systematic review of scientific literature published between 1990-2010 relating to the frequency of human brucellosis, commissioned by WHO. The objectives were to identify high quality disease incidence data to complement existing knowledge of the global disease burden and, ultimately, to contribute towards the calculation of a Disability-Adjusted Life Years (DALY) estimate for brucellosis.METHODS/PRINCIPAL FINDINGS: Thirty three databases were searched, identifying 2,385 articles relating to human brucellosis. Based on strict screening criteria, 60 studies were selected for quality assessment, of which only 29 were of sufficient quality for data analysis. Data were only available from 15 countries in the regions of Northern Africa and Middle East, Western Europe, Central and South America, Sub-Saharan Africa, and Central Asia. Half of the studies presented incidence data, six of which were longitudinal prospective studies, and half presented seroprevalence data which were converted to incidence rates. Brucellosis incidence varied widely between, and within, countries. Although study biases cannot be ruled out, demographic, occupational, and socioeconomic factors likely play a role. Aggregated data at national or regional levels do not capture these complexities of disease dynamics and, consequently, at-risk populations or areas may be overlooked. In many brucellosis-endemic countries, health systems are weak and passively-acquired official data underestimate the true disease burden.CONCLUSIONS: High quality research is essential for an accurate assessment of disease burden, particularly in Eastern Europe, the Asia-Pacific, Central and South America and Africa where data are lacking. Providing formal epidemiological and statistical training to researchers is essential for improving study quality. An integrated approach to disease surveillance involving both human health and veterinary services would allow a better understand of disease dynamics at the animal-human interface, as well as a more cost-effective utilisation of resources

    Predator Cat Odors Activate Sexual Arousal Pathways in Brains of Toxoplasma gondii Infected Rats

    Cat odors induce rapid, innate and stereotyped defensive behaviors in rats at first exposure, a presumed response to the evolutionary pressures of predation. Bizarrely, rats infected with the brain parasite Toxoplasma gondii approach the cat odors they typically avoid. Since the protozoan Toxoplasma requires the cat to sexually reproduce, this change in host behavior is thought to be a remarkable example of a parasite manipulating a mammalian host for its own benefit. Toxoplasma does not influence host response to non-feline predator odor nor does it alter behavior on olfactory, social, fear or anxiety tests, arguing for specific manipulation in the processing of cat odor. We report that Toxoplasma infection alters neural activity in limbic brain areas necessary for innate defensive behavior in response to cat odor. Moreover, Toxoplasma increases activity in nearby limbic regions of sexual attraction when the rat is exposed to cat urine, compelling evidence that Toxoplasma overwhelms the innate fear response by causing, in its stead, a type of sexual attraction to the normally aversive cat odor

    A Large Hadron Electron Collider at CERN

    This document provides a brief overview of the recently published report on the design of the Large Hadron Electron Collider (LHeC), which comprises its physics programme, accelerator physics, technology and main detector concepts. The LHeC exploits and develops challenging, though principally existing, accelerator and detector technologies. This summary is complemented by brief illustrations of some of the highlights of the physics programme, which relies on a vastly extended kinematic range, luminosity and unprecedented precision in deep inelastic scattering. Illustrations are provided regarding high precision QCD, new physics (Higgs, SUSY) and electron-ion physics. The LHeC is designed to run synchronously with the LHC in the twenties and to achieve an integrated luminosity of O(100) fb1^{-1}. It will become the cleanest high resolution microscope of mankind and will substantially extend as well as complement the investigation of the physics of the TeV energy scale, which has been enabled by the LHC
