Impact of volatile phenols and their precursors on wine quality and control measures of Brettanomyces/Dekkera yeasts
Volatile phenols are aromatic compounds and one of the key molecules responsible for olfactory defects in wine. The yeast genus Brettanomyces is the only major microorganism that has the ability to covert hydroxycinnamic acids into important levels of these compounds, especially 4-ethylphenol and 4-ethylguaiacol, in red wine. When 4-ethylphenols reach concentrations greater than the sensory threshold, all wineâs organoleptic characteristics might be influenced or damaged. The aim of this literature review is to provide a better understanding of the physicochemical, biochemical, and metabolic factors that are related to the levels of p-coumaric acid and volatile phenols in wine. Then, this work summarizes the different methods used for controlling the presence of Brettanomyces in wine and the production of ethylphenols
Latent tuberculosis infection, tuberculin skin test and vitamin D status in contacts of tuberculosis patients: a cross-sectional and case-control study
<p>Abstract</p> <p>Background</p> <p>Deficient serum vitamin D levels have been associated with incidence of tuberculosis (TB), and latent tuberculosis infection (LTBI). However, to our knowledge, no studies on vitamin D status and tuberculin skin test (TST) conversion have been published to date. The aim of this study was to estimate the associations of serum 25-hydroxyvitamin D<sub>3 </sub>(25[OH]D) status with LTBI prevalence and TST conversion in contacts of active TB in Castellon (Spain).</p> <p>Methods</p> <p>The study was designed in two phases: cross-sectional and case-control. From November 2009 to October 2010, contacts of 42 TB patients (36 pulmonary, and 6 extra-pulmonary) were studied in order to screen for TB. LTBI and TST conversion cases were defined following TST, clinical, analytic and radiographic examinations. Serum 25(OH)D levels were measured by electrochemiluminescence immunoassay (ECLIA) on a COBAS<sup>Ÿ </sup>410 ROCHE<sup>Ÿ </sup>analyzer. Logistic regression models were used in the statistical analysis.</p> <p>Results</p> <p>The study comprised 202 people with a participation rate of 60.1%. Only 20.3% of the participants had a sufficient serum 25(OH)D (℠30 ng/ml) level. In the cross-sectional phase, 50 participants had LTBI and no association between LTBI status and serum 25(OH)D was found. After 2 months, 11 out of 93 negative LTBI participants, without primary prophylaxis, presented TST conversion with initial serum 25(OH)D levels: a:19.4% (7/36): < 20 ng/ml, b:12.5% (4/32):20-29 ng/ml, and c:0%(0/25) ℠30 ng/ml. A sufficient serum 25(OH)D level was a protector against TST conversion a: Odds Ratio (OR) = 1.00; b: OR = 0.49 (95% confidence interval (CI) 0.07-2.66); and c: OR = 0.10 (95% CI 0.00-0.76), trends p = 0.019, adjusted for high exposure and sputum acid-fast bacilli positive index cases. The mean of serum level 25(OH)D in TST conversion cases was lower than controls,17.5 ± 5.6 ng/ml versus 25.9 ± 13.7 ng/ml (p = 0.041).</p> <p>Conclusions</p> <p>The results suggest that sufficient serum 25(OH)D levels protect against TST conversion.</p

GENCODE: reference annotation for the human and mouse genomes in 2023

GENCODE produces high quality gene and transcript annotation for the human and mouse genomes. All GENCODE annotation is supported by experimental data and serves as a reference for genome biology and clinical genomics. The GENCODE consortium generates targeted experimental data, develops bioinformatic tools and carries out analyses that, along with externally produced data and methods, support the identification and annotation of transcript structures and the determination of their function. Here, we present an update on the annotation of human and mouse genes, including developments in the tools, data, analyses and major collaborations which underpin this progress. For example, we report the creation of a set of non-canonical ORFs identified in GENCODE transcripts, the LRGASP collaboration to assess the use of long transcriptomic data to build transcript models, the progress in collaborations with RefSeq and UniProt to increase convergence in the annotation of human and mouse protein-coding genes, the propagation of GENCODE across the human pan-genome and the development of new tools to support annotation of regulatory features by GENCODE. Evaluation of appendicitis risk prediction models in adults with suspected appendicitis
Background
Appendicitis is the most common general surgical emergency worldwide, but its diagnosis remains challenging. The aim of this study was to determine whether existing risk prediction models can reliably identify patients presenting to hospital in the UK with acute right iliac fossa (RIF) pain who are at low risk of appendicitis.
Methods
A systematic search was completed to identify all existing appendicitis risk prediction models. Models were validated using UK data from an international prospective cohort study that captured consecutive patients aged 16â45âyears presenting to hospital with acute RIF in March to June 2017. The main outcome was best achievable model specificity (proportion of patients who did not have appendicitis correctly classified as low risk) whilst maintaining a failure rate below 5 per cent (proportion of patients identified as low risk who actually had appendicitis).
Results
Some 5345 patients across 154 UK hospitals were identified, of which twoâthirds (3613 of 5345, 67·6 per cent) were women. Women were more than twice as likely to undergo surgery with removal of a histologically normal appendix (272 of 964, 28·2 per cent) than men (120 of 993, 12·1 per cent) (relative risk 2·33, 95 per cent c.i. 1·92 to 2·84; Pâ<â0·001). Of 15 validated risk prediction models, the Adult Appendicitis Score performed best (cutâoff score 8 or less, specificity 63·1 per cent, failure rate 3·7 per cent). The Appendicitis Inflammatory Response Score performed best for men (cutâoff score 2 or less, specificity 24·7 per cent, failure rate 2·4 per cent).
Conclusion
Women in the UK had a disproportionate risk of admission without surgical intervention and had high rates of normal appendicectomy. Risk prediction models to support shared decisionâmaking by identifying adults in the UK at low risk of appendicitis were identified
Doença de Crohn isolada do apĂȘndice cecal como causa de enterorragia Crohnâs disease isolated of the appendix as a source of enterorrhagia
RACIONAL: A doença de Crohn isolada do apĂȘndice Ă© doença de paciente jovem, relativamente rara, com quadro histopatolĂłgico semelhante ao da doença de Crohn tĂpica. Seu quadro clĂnico, em geral, simula o da apendicite aguda, com manifestaçÔes protraĂdas e na palpação do abdome, alĂ©m dos sinais de irritação peritonial, nota-se com freqĂŒĂȘncia, um tumor palpĂĄvel. OBJETIVO: Apresentar um caso de doença de Crohn isolada do apĂȘndice que se manifestou por enterorragia e cuja origem foi identificada pela colonoscopia. PACIENTE: Masculino, com 16 anos de idade, sem outras manifestaçÔes digestivas apresentou dois episĂłdios de enterorragia com intervalo de cerca de 1 ano. No segundo episĂłdio, a colonoscopia identificou o sangramento como proveniente do Ăłstio apendicular. RESULTADO: Indicada a exploração cirĂșrgica, constatou-se que o ceco e o Ăleo eram normais e que o apĂȘndice cecal estava com sinais inflamatĂłrios e bloqueado pelo omento, realizando-se a apendicectomia. O exame anatomopatolĂłgico revelou tratar-se de processo inflamatĂłrio crĂŽnico com infiltrado linfoplasmocitĂĄrio e granulomas esparsos, sugestivo de doença de Crohn. Em acompanhamento clĂnico, nĂŁo apresentou manifestaçÔes de doença digestiva e a colonoscopia, realizada apĂłs 2 anos, nĂŁo mostrou evidĂȘncias de recidiva ou propagação da doença para o Ăleo. CONCLUSĂO: Neste caso, como nos demais relatados na literatura, a apendicectomia Ă© possivelmente curativa, devendo o paciente ser acompanhado, pelo menos, por 5 anos. No diagnĂłstico diferencial das enterorragias em pacientes jovens, a doença de Crohn isolada do apĂȘndice tambĂ©m pode ser considerada.<br>BACKGROUND: Crohnâs disease confined to the appendix is relatively rare as a sole primary manifestation of the disease. Young people are more affected. The medical history and the physical examination are similar to the findings in acute appendicitis, but the manifestations are protracted. On physical examination there are signs of peritoneal irritation and an abdominal mass is palpable in the right iliac fossa. AIMS: To report a case of Crohnâs disease confined to the appendix and presenting with enterorrhagia. The source of the bleeding was localized by colonoscopy. PATIENT: A 16-year old caucasian male without past history of gastrointestinal symptoms, presented with two episodes of enterorrhagia within a period of one year. In the second episode colonoscopy identified the appendicular ostium as the source of bleeding. RESULTS: At operation the cecum and terminal ileum were normal in thickness and texture, and an inflammatory appendix adherent to the omentum was removed. Microscopically there were non-caseating granulomas, intense infiltration of the wall with plasma cells, lymphocytes and macrophages. The patient has not suffered recurrence, and a colonoscopy realized 2 years after the operation did not show signs of Crohnâs disease. CONCLUSION: This case, like others in the literature, appendectomy is curative, but a 5-year follow-up is mandatory. When a young patient presents with enterorrhagia, this diagnosis has to be considered
Towards a more sustainable viticulture: Foliar application of N-doped calcium phosphate nanoparticles on Tempranillo grapes
The use of nanomaterials for the efficient delivery of active species in viticulture is still an unexplored opportunity. Nitrogen, an essential nutrient for grapevine development and wine quality, is commonly provided in the form of urea. However, the application of conventional fertilisers contributes to nitrate leaching and denitrification, thus polluting groundwater and causing a serious environmental impact. Nanotechnology is offering smart solutions towards more sustainable and efficient agriculture. In the present work, we assessed the efficiency of nontoxic amorphous calcium phosphate (ACP) nanoparticles as nanocarriers of urea (U-ACP) through field experiments on Tempranillo grapevines.
