    Functional MR Imaging Correlates of Neuropsychological Impairment in Primary-Progressive Multiple Sclerosis

    BACKGROUND AND PURPOSE: Cognitive deficits affect ≀30% of patients with PPMS. We investigated the functional correlates of cognitive network dysfunction in patients with PPMS and their correlation with the extent of structural MR imaging damage. MATERIALS AND METHODS: From 16 right-handed patients with PPMS and 17 matched controls, structural and fMRIs (during the performance of the 2-back task) were acquired. Neuropsychological tests exploring memory, attention, and frontal lobe cognitive domains were administered. T2 LL, NBV, and CC areas were measured. RESULTS: Six patients with PPMS were CI. Structural MR imaging measures did not differ between patients who were CI and those who were CP. Compared with patients who were CI, patients who were CP had increased activations of the left caudate nucleus, PFC, and inferior parietal lobule. Compared with controls and patients who were CP, patients who were CI had increased activations of the SII, cerebellum, and insula. Compared with controls, they also had increased activations of the right precentral gyrus and a reduced recruitment of the left PFC. In patients with PPMS, a decreased composite cognitive score correlated with increased activity of the cerebellum, insula, and SII, as well as decreased PFC activity. T2 LL correlated with decreased PFC recruitment and increased SII recruitment. CONCLUSIONS: In PPMS, an increased recruitment of cognitive-related networks might represent a functional reserve with the potential to limit the severity of cognitive impairment. The accumulation of T2 lesions and the consequent exhaustion of frontal lobe plasticity might contribute to cognitive impairment in PPMS

    Early life supply of competitive exclusion products reduces colonization of Extended Spectrum Beta-Lactamase producing Escherichia coli in broilers

    Broilers are an important reservoir of ESBL/pAmpC-producing bacteria. In previous studies a single supply of a competitive exclusion (CE) product before challenge with a high dose of ESBL/pAmpC-producing E. coli led to reduced colonization, excretion and transmission, but could not prevent colonization. The hypothesized mechanism is competition; therefore in this study the effect of a prolonged supply of CE products on colonization, excretion and transmission of ESBL-producing E. coli after challenge with a low dose at day 0 or day 5 was investigated. Day-old broilers (Ross 308) (n=220) were housed in isolators. Two CE products, containing unselected fermented intestinal bacteria (CEP) or a selection of pre- and probiotics (SYN), were supplied in drinking water from day 0 to 14. At day 0 or day 5 broilers were challenged with 0.5 mL with 101 or 102 cfu/mL E. coli encoding the beta–lactamase gene blaCTX-M-1 on an IncI plasmid (CTX-M-1-E. coli). Presence and concentration of CTX-M-1-E. coli was determined using cloacal swabs (day 0-14, 16, 19, 21) and cecal content (day 21). Cox proportional hazard model and a mixed linear regression model were used to determine the effect of the intervention on colonization and excretion (log10 cfu/g). When challenged at day of hatch, no effect of CEP was observed. When challenged at day 5, both CEP and SYN led to a prevention of colonization with CTX-M-1-E. coli in some isolators. In the remaining isolators, we observed reduced time until colonization (HR between 3.71 ×10-3 and 3.11), excretion (up to -1.60 log10 cfu/g) and cecal content (up to -2.80 log10 cfu/g) and a 1.5 to 3-fold reduction in transmission rate. Colonization after a low dose challenge with ESBL-producing E. coli can be prevented by CE products. However, if at least one bird is colonized it spreads through the whole flock. Prolonged supply of CE products provided shortly after hatch, may be applicable as an intervention to reduce prevalence of ESBL/pAmpC-producing bacteria in the broiler production chain

DOI: 10.1086/511161Stars form in the densest, coldest, most quiescent regions of molecular clouds. Molecules provide the only probes that can reveal the dynamics, physics, chemistry, and evolution of these regions, but our understanding of the molecular inventory of sources and how this is related to their physical state and evolution is rudimentary and incomplete. The Spectral Legacy Survey (SLS) is one of seven surveys recently approved by the James Clerk Maxwell Telescope (JCMT) Board of Directors. Beginning in 2007, the SLS will produce a spectral imaging survey of the content and distribution of all the molecules detected in the 345 GHz atmospheric window (between 332 and 373 GHz) toward a sample of five sources. Our intended targets are a low-mass core (NGC 1333 IRAS 4), three high-mass cores spanning a range of star-forming environments and evolutionary states (W49, AFGL 2591, and IRAS 20126), and a photodissociation region (the Orion Bar). The SLS will use the unique spectral imaging capabilities of HARP-B/ACSIS (Heterodyne Array Receiver Programme B/Auto- Correlation Spectrometer and Imaging System) to study the molecular inventory and the physical structure of these objects, which span different evolutionary stages and physical environments and to probe their evolution during the star formation process. As its name suggests, the SLS will provide a lasting data legacy from the JCMT that is intended to benefit the entire astronomical community. As such, the entire data set (including calibrated spectral data cubes, maps of molecular emission, line identifications, and calculations of the gas temperature and column density) will be publicly available.Peer reviewe

    The composition of the protosolar disk and the formation conditions for comets

    Conditions in the protosolar nebula have left their mark in the composition of cometary volatiles, thought to be some of the most pristine material in the solar system. Cometary compositions represent the end point of processing that began in the parent molecular cloud core and continued through the collapse of that core to form the protosun and the solar nebula, and finally during the evolution of the solar nebula itself as the cometary bodies were accreting. Disentangling the effects of the various epochs on the final composition of a comet is complicated. But comets are not the only source of information about the solar nebula. Protostellar disks around young stars similar to the protosun provide a way of investigating the evolution of disks similar to the solar nebula while they are in the process of evolving to form their own solar systems. In this way we can learn about the physical and chemical conditions under which comets formed, and about the types of dynamical processing that shaped the solar system we see today. This paper summarizes some recent contributions to our understanding of both cometary volatiles and the composition, structure and evolution of protostellar disks.Comment: To appear in Space Science Reviews.     Gain of FGF4 is a frequent event in KIT/PDGFRA/SDH/RAS-P WT GIST

    Gastrointestinal stromal tumors (GIST) lacking mutations in KIT/PDGFRA or RAS pathways and retaining an intact SDH complex are usually referred to as KIT/PDGFRA/SDH/RAS-P WT GIST or more simply quadruple WT GIST (~5% of all GIST). Despite efforts made, no recurrent genetic event in quadruple WT GIST has been identified so far. To further investigate this disease, we performed high throughput copy number analysis on quadruple WT GIST specimens identifying a recurrent focal gain in band 11q13.3 (involving FGF3/FGF4) in 6/8 cases. This event was not found in the other molecular GIST subgroups. FGF3/FGF4 duplication was associated with high expression of FGF4, both at mRNA and protein level, a growth factor normally not expressed in adult tissues or in KIT/PDGFRA-mutated GIST. FGFR1 was found to be the predominant FGF receptor expressed and phosphorylation of AKT was detected, suggesting that a FGF4-FGFR1 autocrine loop could stimulate downstream signaling in quadruple WT GIST. Together with the recent reports of quadruple WT cases carrying FGFR1 activating alterations, these findings strengthen the hypothesis of a potential involvement of FGFR pathway deregulation in quadruple WT GIST, which may represent a rationale for novel therapeutic approaches

    On a coupled PDE model for image restoration

    In this paper, we consider a new coupled PDE model for image restoration. Both the image and the edge variables are incorporated by coupling them into two different PDEs. It is shown that the initial-boundary value problem has global in time dissipative solutions (in a sense going back to P.-L. Lions), and several properties of these solutions are established. This is a rough draft, and the final version of the paper will contain a modelling part and numerical experiments

    Measurement of the azimuthal anisotropy of Y(1S) and Y(2S) mesons in PbPb collisions at √S^{S}NN = 5.02 TeV

    The second-order Fourier coefficients (υ2_{2}) characterizing the azimuthal distributions of ΄(1S) and ΄(2S) mesons produced in PbPb collisions at sNN\sqrt{s_{NN}} = 5.02 TeV are studied. The ΄mesons are reconstructed in their dimuon decay channel, as measured by the CMS detector. The collected data set corresponds to an integrated luminosity of 1.7 nb−1^{-1}. The scalar product method is used to extract the υ2_{2} coefficients of the azimuthal distributions. Results are reported for the rapidity range |y| < 2.4, in the transverse momentum interval 0 < pT_{T} < 50 GeV/c, and in three centrality ranges of 10–30%, 30–50% and 50–90%. In contrast to the J/ψ mesons, the measured υ2_{2} values for the ΄ mesons are found to be consistent with zero
