    Knoop Hardness Of Composites Cured With Halogen And Led Lightcuring Units In Class I Restorations

    Aim: To evaluate the effect of light-curing units (LCUs) on the microhardness of class I composite restorations at different depths. Methods: Two light emitting diodes (LED) (Freelight 2, Radii) and one halogen (Optilux 501) LCUs were evaluated. Thirty class I cavities prepared in human third molars were restored with a microhybrid (Charisma) and a microfilled (Renamel) resin composite. After seven days of water storage, the teeth were decoronated and the crowns were bisected mesiodistally and tested for microhardness under a 25 g load for 20 seconds. Fifteen indentations were performed at three depths for each half-crown. Results: Charisma presented significantly higher Knoop hardness number (KHN) values than Renamel. At the superficial depth, there were no statistically significant differences (p > 0.05) when Charisma was cured with both LED curing units. However, statistically significant difference (p < 0.05) was found when Charisma was light-cured with the halogen LCU. The lowest KHN value was obtained by Renamel light-cured with both LED LCUs, regardless of the composite. Nevertheless, when the microfilled composite was light-cured with the halogen LCU, hardness was significantly higher compared to those cured with LED units at all evaluated depths. Conclusions: The effectiveness of polymerization is related not only to the light-curing source, but also to the type of composite and the curing depth.8130-3

    Fios de uma hashtag: entextualização de resistência face a desafios políticos e sanitários no Brasil

    The scenario for this paper is the combined sanitary and political crisis that Brazil has been facing in these unprecedented times of the COVID-19 pandemic. The country is ideologically polarized and this polarization has been enhanced in the last year and a half in Bolsonaro’s government. A bleak period characterized by a way of governing that Silva (2020) names “pragmatics of chaos” due to denialism (even of the very pandemic) and to the neoliberal values cherished by a radical right-wing that supports the President and backs the discontinuation of important public policies, mainly those directed towards minoritized groups and environmental protection. Having in mind such a scenario, and amidst the societal bewilderment and inaction regarding the President’s contradictory actions towards democracy, we chose to focus on a rare event of direct resistance featured by a migrant of Haitian origin, who told in the face of the president what maybe many Brazilians had not dared to say. What is of interest here is that this performance, recorded on a mobile phone, and posted as a tweet, went viral, and gave rise to a hashtag - #acaboubolsonaro [ItsOverBolsonaro] - that is still alive seven months later. With a focus on the man’s performance of resistance and its entextualization on social media, we analyze his discourse and the impacts of his positioning. In the analysis, we try to retrace some of the threads traversed by the hashtag in two Twitter accounts and some of the reverberations on Instagram. We also discuss how the discursive thread of the postings weaves a metareflexive fabric about the man’s performance. Um período sombrio caracterizado por uma forma de governar que Silva (2020) denomina “pragmática do caos” devido ao negacionismo (até mesmo da própria pandemia) e aos valores neoliberais caros a uma direita radical, que apoia o Presidente e defende a descontinuação de políticas públicas importantes, principalmente as voltadas a grupos minoritários e de proteção ambiental. Tendo em vista tal cenário, e em meio ao espanto e à inação da sociedade em relação às ações contraditórias do Presidente no que concerne à democracia, optamos por nos concentrar em um raro evento de resistência direta protagonizado por um migrante de origem haitiana, que disse na cara do Presidente o que talvez muitos brasileiros não tenham ousado dizer. Essa performance, gravada em um celular e postada como um tweet, viralizou, dando origem a uma hashtag - #acaboubolsonaro [ItsOverBolsonaro] - que continua viva sete meses depois. Focalizando a performance de resistência e sua entextualização nas redes sociais, tentamos reconstituir alguns dos tópicos percorridos pela hashtag em duas contas do Twitter e algumas das reverberações no Instagram. Também discutimos como o fio discursive das postagens delineia um tecido metarreflexivo sobre a performance do Homem.     Controle glicêmico em pacientes diabéticos atendidos em centros de atenção primária à saúde

    OBJECTIVE: To identify factors associated to poor glycemic control among diabetic patients seen at primary health care centers. METHODS: A cross-sectional study was carried out in a sample of 372 diabetic patients attending 32 primary health care centers in southern Brazil. Data on three hierarchical levels of health unit infrastructure, medical care and patient characteristics were collected. RESULTS: The frequency of poor glycemic control was 50.5%. Multivariate analysis (multilevel method) showed that patients with body mass indexes below 27 kg/m², patients on oral hypoglycemic agents or insulin, and patients diagnosed as diabetic over five years prior to the interview were more likely to present poor glycemic control when compared to their counterparts. CONCLUSIONS: Given the hierarchical data structuring, all associations found suggest that factors associated to hyperglycemia are related to patient-level characteristics.OBJETIVO: Identificar fatores associados à falta de controle glicêmico em pacientes diabéticos atendidos em centros de atenção primária à saúde. MÉTODOS: Estudo transversal em amostra de 372 pacientes diabéticos atendidos nos 32 centros de atenção primária de uma cidade do sul do Brasil. Foram coletados dados ordenados em três níveis hierárquicos: estrutura das unidades de saúde, características do processo do cuidado médico e pacientes diabéticos. RESULTADOS: A freqüência de falta de controle glicêmico foi de 50,5%. A análise multivariada (método multinível) mostrou que pacientes com Índice de Massa Corporal abaixo de 27 kg/m², em tratamento medicamentoso e com mais de cinco anos de diagnóstico de diabetes, tiveram maior probabilidade de apresentar hiperglicemia quando comparados a seus pares. CONCLUSÕES: Considerando a estrutura hierárquica dos dados, todas as associações encontradas sugerem que os fatores associados à hiperglicemia são relacionados a características dos pacientes

    Near-infrared spectroscopy for the detection and quantification of bacterial contaminations in pharmaceutical products

    Accurate detection and quantification of microbiological contaminations remains an issue mainly due the lack of rapid and precise analytical techniques. Standard methods are expensive and time-consuming being associated to high economic losses and public health threats. In the context of pharmaceutical industry, the development of fast analytical techniques able to overcome these limitations is crucial and spectroscopic techniques might constitute a reliable alternative. In this work we proved the ability of Fourier transform near infrared spectroscopy (FT-NIRS) to detect and quantify bacteria (Bacillus subtilis, Escherichia coli, Pseudomonas fluorescens, Salmonella enterica, Staphylococcus epidermidis) from 10-108 CFUs/mL in sterile saline solutions (NaCl 0.9%). Partial least squares discriminant analysis (PLSDA) models showed that FT-NIRS was able to discriminate between sterile and contaminated solutions for all bacteria as well as to identify contaminant bacteria. Partial least squares (PLS) models allowed bacterial quantification with limits of detection ranging from 5.1 to 9CFU/mL for E. coli and B. subtilis, respectively. This methodology was successfully validated in three pharmaceutical preparations (contact lens solution, cough syrup and topic anti-inflammatory solution) proving that this technique possess a high potential to be routinely used for the detection and quantification of bacterial contaminations.This work was funded by Fundacao para a Ciencia e a Tecnologia (FCT) under the project UID/QUI/50006/2013.     Blood molecular profile to predict genotoxicity from exposure to antineoplastic drugs

However, the usually applied cytogenetic assays are laborious and time-consuming, the reason why it is critical to developing rapid and economic new methods. The aim of this study was to evaluate if the molecular profile of frozen whole blood, acquired by Fourier Transform Infrared (FTIR) spectroscopy, allows to assess genotoxicity in occupational exposure to antineoplastic drugs, as obtained by the cytokinesis-block micronucleus assay. For that purpose, 92 samples of peripheral blood were studied: 46 samples from hospital professionals occupationally exposed to antineoplastic drugs and 46 samples from workers in academia without exposure (controls). It was first evaluated the metabolome from frozen whole blood by methanol precipitation of macromolecules as haemoglobin, followed by centrifugation. The metabolome molecular profile resulted in 3 ratios of spectral bands, significantly different between the exposed and non-exposed group (p<0.01), and a spectral principal component-linear discriminant analysis (PCA-LDA) model enabling to predict genotoxicity from exposure with 73 % accuracy. After optimization of the dilution degree and solution used, it was possible to obtain a higher number of significant ratios of spectral bands, i.e., 10 ratios significantly different (p<0.001), highlighting the high sensitivity and specificity of the method. Indeed, the PCA-LDA model, based on the molecular profile of whole blood, enabled to predict genotoxicity from exposure with an accuracy, sensitivity, and specificity of 92 %, 93 %, and 91 %, respectively. All these parameters were achieved based on 1 μL of frozen whole blood, in a high-throughput mode, i.e., based on the simultaneous analysis of 92 samples, in a simple and economic mode.     Interactive Digital Television (iDTV) is a technology that has potential to mitigate economic and social difficulties related to knowledge access and digital inclusion in Brazil. Nevertheless, iDTV solutions did not yet bring anything new and disappointed most users. Thinking about social and cultural issues in iDTV applications might result in design solutions that make sense to people affected by this new communication medium. In this paper, we propose a set of questions to support designers of iDTV applications to address cultural aspects during requirement analysis in a structured way. As practical use example, we apply these questions to prospect requirements from a cultural perspec-tive for an overlaid iDTV application in the real context of a Brazilian Company’s TV show

    A simple, label-free, and high-throughput method to evaluate the epigallocatechin-3-gallate impact in plasma molecular profile

    Epigallocatechin-3-gallate (EGCG), the major catechin present in green tea, presents diverse appealing biological activities, such as antioxidative, anti-inflammatory, antimicrobial, and antiviral activities, among others. The present work evaluated the impact in the molecular profile of human plasma from daily consumption of 225 mg of EGCG for 90 days. Plasma from peripheral blood was collected from 30 healthy human volunteers and analyzed by high-throughput Fourier transform infrared spectroscopy. To capture the biochemical information while minimizing the interference of physical phenomena, several combinations of spectra pre-processing methods were evaluated by principal component analysis. The pre-processing method that led to the best class separation, that is, between the plasma spectral data collected at the beginning and after the 90 days, was a combination of atmospheric correction with a second derivative spectra. A hierarchical cluster analysis of second derivative spectra also highlighted the fact that plasma acquired before EGCG consumption presented a distinct molecular profile after the 90 days of EGCG consumption. It was also possible by partial least squares regression discriminant analysis to correctly predict all unlabeled plasma samples (not used for model construction) at both timeframes. We observed that the similarity in composition among the plasma samples was higher in samples collected after EGCG consumption when compared with the samples taken prior to EGCG consumption. Diverse negative peaks of the normalized second derivative spectra, associated with lipid and protein regions, were significantly affected (p < 0.001) by EGCG consumption, according to the impact of EGCG consumption on the patients' blood, low density and high density lipoproteins ratio. In conclusion, a single bolus dose of 225 mg of EGCG, ingested throughout a period of 90 days, drastically affected plasma molecular composition in all participants, which raises awareness regarding prolonged human exposure to EGCG. Because the analysis was conducted in a high-throughput, label-free, and economic analysis, it could be applied to high-dimension molecular epidemiological studies to further promote the understanding of the effect of bio-compound consumption mode and frequency.info:eu-repo/semantics/publishedVersio
