A new UV and visible reflectance microspectroscopy technique has been applied to cometary samples from the Stardust mission. The results have been compared with terrestrial minerals and aerogel
A pesquisa efetuada permite demonstrar

Spectral studies of extra-terrestrial materials
Experiments were made using a state-of-the-art UV-Vis microspectrophotometer (MSP) in order to assess if the instrument is suitable for use on spectroscopy of terrestrial and extra-terrestrial materials. This new instrument brings advantages that cannot be found in instruments currently in use: it requires only extremely small samples (min of -2 um) and it is very quick to use (little sample preparation and spectra taken in less than 2 min).
If suitable, the instrument could help to show the relationship between meteorites and their parent bodies and in the study ' of very small fragile samples, such as cometary samples and Interplanetary Dust Particles (IDPs). Using samples of minerals commonly found in meteorites, it was concluded that the instrument is suitable for the study of these materials, however it has some limitations and certain conditions need to be met.
The method was then applied on two grains from comet 81P/Wild 2 returned by the space mission Stardust. Further limitations were found with these samples caused by the fact that they are covered in aerogel and embedded in gold foil. Results indicate however, that the samples seem to be composed of a mixture of different materials.
Results from the study of HED (howardites, eucrites and diogenites) type meteorites proved that if the conditions are met, the technique is suitable and comparable to other instruments and can be used to match the spectra of meteorites to that of their possible asteroidal parent bodies.
A complementary investigation studied the effects of impact by shock on the spectra of rocks using a Light Gas Gun and Near Infrared spectroscopy with the goal of investigating the effects of weathering on the spectra of asteroids. It was found that there is a change in the spectra of the samples and a relationship with a change in composition of the impacted area
Cyclic behavior of a two-span RC beam built with plain reinforcing bars
Reinforced concrete structural elements lacking appropriate seismic detailing and built with plain reinforcing bars, and subjected to cyclic loads like the ones induced by earthquakes, are particularly sensitive to the bond-slip mechanism. Though, existing studies on the cyclic behavior of RC structures generally refer to elements with deformed bars. As a result, the behavior of elements with plain bars is not yet fully understood. In this framework, the cyclic behavior of a two-span RC beam built with plain reinforcing bars, collected from an ancient building structure, was investigated. The support and loading conditions observed in-situ were simulated in the test setup. The beam displayed a flexural failure and the damage was concentrated in three short plastic hinges. The poor damage distribution evidences the effects of the bar slippage mechanism on the beam behaviorFCT - SFRH/BD/27406/2006FCT - SFRH/BD/62110/200
A partir do ouvir, nasce o perceber, e
A sacralidade
Structural and Functional Characterization of a New Bacterial Target Against Tuberculosis: The Phosphatase PtpA
Tuberculosis (TB) is one of the top causes of death remaining a major public health problem worldwide. Mycobacterium tuberculosis is the agent of TB, infecting the human respiratory tract. Its remarkable pathogenicity hinges upon the ability to challenge the immune system of the host by secreting phosphatases into macrophages. Among them, Protein Tyrosine Phosphatase A (PtpA) plays a key role on the infection process, preventing the phagosome-lysosome fusion and promoting the inhibition of phagosome acidification. Thus, PtpA becomes a promising target for the development of new anti-TB drugs.
The aim of this work is to contribute to find new structure-based drug design approaches against TB, studying the inhibitory properties of three different families of compounds towards PtpA – chalcones, thiosemicarbazones and azaindoles.
The protein was overexpressed in E. coli – final yield of 20 mg protein/ liter of culture – and successfully purified using affinity chromatography. To provide new insights into the binding mode of the studied compounds, molecular docking studies were performed suggesting thiosemicarbazones as non-competitive inhibitors and the chalcones and azaindoles with a preferential active site binding.
The protein was also biophysically characterized. The oligomeric state was confirmed by SEC, proving that PtpA is a monomer in solution. The protein stability was assessed through TSA revealing that, with 10% glycerol, PtpA resists to the effects of 10% DMSO. TSA was also used to find a suitable protein storage condition (-80°C) and to confirm PEG400 as an alternative solvent for the inhibitors. In addition, distinct biophysical approaches – TSA, MST and urea-gel electrophoresis – were implemented to detect protein-ligand interactions but definitive evidence were not obtained.
Ligand-free and co-crystallization assays were extensively explored and several crystals were tested at the ESRF, Diamond and MAX IV. Two crystal structures were obtained: a co-crystallization PtpA-Lap11 structure at 3.6 Å resolution and a soaking PtpA-C33 structure at 2.8 Å resolution. Despite the low/medium resolution obtained, both structures reveal the potential binding of the inhibitors with suspicious density blobs near His120B for Lap11 and at the active site for C33. The ligands were preliminarily modelled but further refinement cycles are required to elucidate the respective binding
mobilidade, utilizando escalas visuais 0-100 mm, e a capacidade funcional (HAQ).
moderada (rho 0.370 a 0.438, p<0.01).
capacidade funcional do doente (HAQ).
O impacto no DAS284vVS utilizando as
Background: Rheumatoid Arthritis´ activity´s evaluation according with the patient´s
perspective as assumed a big relevance, incorporating different rates of the disease’s activity.
In relation to how the patient is questioned (Patient Global Assessment – PGA), various
formulations of the question and the period of in which the evaluation is reported have been
published.
Objective: In this study, we intend to evaluate if the way the phrasing affects the answer given
by the patient and what its correlation with the other parameters of the DAS28 and other
measures to evaluate the disease´s impact.
Methods: One hundred and one patients, who were fulfilling the 1987 ACR or 2010
ACR/EULAR Diagnostic Criteria for the Rheumatoid Arthritis followed in Rheumatology
Department of the Hospital University of Coimbra, completed independently a questionnaire
which had seven versions of the PGA-VAS in a scale of 0-100mm and two versions assigning
a value from 0 to 100. They also evaluated fatigue, pain, deformity and mobility, using visual
scales 0-100mm, and the functional capacity (HAQ). Their medical assistant made an
evaluation, a determination of the activity of their disease (DAS28) and their individual components. All PGA-VAS were evaluated using Pearson's correlation coefficient. This has
also been used to correlate the PGA-VAS with other variables of the disease’s impact. In
allanalysis, p<0.05 was considered statistically significant.
Results: The PGA-VAS correlate weakly with the other variables of the DAS28, with the
exception of tender joints, score of 28, with which the correlation is moderate (rho 0.370 a
0.438, p<0.01). These correlate moderately/ strongly with other VAS (fatigue, pain, deformity
and mobility), but also with the patient's functional ability (HAQ). All PGA-VAS versions
correlate moderately/ strongly with each other (rho 0.452 to 0.786, p<0.01), with the
exception of the two scales developed by the researchers, where the issue is carried out in an
affirmative way, which correlation was weak or inexistent. The difference between the
DAS28 global scoring using all PGA-VAS versions was minimal (0.109±0.455).
Conclusion: The different PGA-VAS correlate each other moderately and with other scales in
which patients self-evaluates of disease impact. The impact in DAS284vVS was minimal. To
increase homogenization in the results, we recommend standardizing the question of PGAVAS
and its validation
Assembly and function of a protein cross-linking enzyme during bacterial spore morphogenesis
Sporulation in Bacillus subtilis culminates with the formation of a dormant
endospore. The endospore (or spore) is one of the most resilient cell types
known and can remain viable in the environment for extended periods of
time. Contributing to the spore’s resistance and its ability to interact with
and monitor its immediate environment is the coat, the outermost layer of
B. subtilis spores. The coat is composed by over 70 different proteins, which
are produced at different stages in sporulation and orderly assembled
around the developing spore.(...)Fundação para a Ciência e a Tecnologia (FCT
Backpacking culture: digital nomads in Porto - a Selina case study
This report incorporates the curricular internship of the Master’s degree in Intercultural Studies for Business, from Porto Accounting and Business School. The internship was performed at Selina Porto, an international company originally from Panama.
As globalization provides a more efficient path to mobility meanwhile facilitating the search for information, the urge to travel and seek new cultures became intense. People from around the world engaged in the act of traveling more deeply than what was known previously. The backpacker phenomenon evolved into a lifestyle, as being a tourist simply was not fulfilling for this new generation of travelers.
This type of culture also leads to the transformation of organizational culture. As digital transformation takes over due to a market in need of change and innovation, the work regulation adapts itself to flexible schedules and remote work. In this specific case, the hotel chain Selina faced the need of adapting their business in order to respond to the new demand, assuming a new role.
The purpose of this report is to present and analyze my internship at Selina, as an example of creativity and innovation in the hospitality business. This company targets backpackers and digital nomads, creating a market strategy suitable not only for them but locals as well, by enhancing national cultures. A study, based on qualitative research, was conducted by analyzing solo travelers’ reviews on Booking – a reservation platform –, in order to better perceive what attracts this particular cluster of travelers and the vision conveyed by the company. Features such as location, staff and environment are decidedly the most attractive.
It is also proposed to depict in full detail the tasks and functions carried out throughout the internship, alluding to the evolution experienced, as well as the challenges and difficulties encountered due to Covid-19.O presente relatório insere-se no âmbito do estágio curricular do Mestrado em Estudos Interculturais para Negócios, pelo Instituto Superior de Contabilidade e Administração do Porto.
Sudden Infant Death Syndrome: What healthcare professionals and parents know about how to prevent it
Ao alinhar o conhecimento e o comportamento com as mais recentes políticas de prevenção do SMSL, os profissionais de saúde poderiam efetivamente transmitir as mensagens corretas aos pais e, consequentemente, contribuir para diminuir o número de casos de SMSL.Background: Sudden Infant Death Syndrome (SIDS) is defined as the death of an infant of less than 1 year that is unexpected and unexplained after an extensive investigation including necropsy. In developed countries, SIDS is the first cause of mortality in the first year of life, excluding the neonatal period (first 28 days), and is the third leading cause of infant death in the world. Risk factors related to SIDS have already been identified, most of them related with the infant's sleep position and environment. The Task Force on Sudden Infant Death Syndrome of the American Academy of Pediatrics published in 2016 an update on the Recommendations for a Safe Infant Sleeping Environment in order to reduce the risk of all sleep-related infant deaths.
Objective: This study aims at assessing the knowledge about SIDS risk factors of parents of infants born in the Centro Hospitalar Universitário São João (CHUSJ) and of healthcare professionals taking care of newborns in the same hospital.
Materials and Methods: This study used two structured self-administered questionnaires aiming at evaluating the knowledge of parents and healthcare professionals about SIDS prevention and its risk factors. A statistical analysis of the responses was performed using STATA.
Results: Overall, 67.7% of parents and 100% of healthcare professionals knew what SIDS stands for. Besides, 82.3% of healthcare professionals and 47.5% of parents recognized the supine position as the safest sleep position to prevent SIDS. For each of the 13 questions in the questionnaire about SIDS risk factors, the majority of healthcare professionals identified the correct answer whereas among parents only 7 questions were correctly answered by the majority of respondents. Only 37.2% of healthcare professionals and 8.7% of parents responded correctly to 75% or more of the 13 questions about SIDS risk factors. Healthcare professionals with children aged 5 or less are more likely to have stronger knowledge about SIDS risk factors than the other respondents (p = 0.032) and doctors are more likely than nurses to have less correct knowledge about SIDS risk factors (p = 0.028 and 0.012, respectively).
Discussion and Conclusion: Healthcare professionals are not as well informed about SIDS as they should and, possibly due to their self-awareness of this, they have little confidence in discussing SIDS-related issues. This highlights how crucial the training of healthcare professionals about this topic is, and that their role of (partial) gatekeepers of this information for parents should be strengthened with appropriate support. By aligning their knowledge and behaviour with the latest SIDS prevention policies, they could effectively pass on to parents the correct messages, and consequently contribute to decrease the number of SIDS cases
