Adaptações fisiológicas e estratégias de eliminação de toxinas em bivalves face a blooms de algas tóxicas
Bivalves play a marked role in coastal marine ecosystems, impacting primary
productivity with a strong top-down control on phytoplankton communities. In
addition, they are an important food source for higher trophic levels, including
humans. However, bivalves can be impaired by several external factors, either
biotic or abiotic, natural or anthropogenic. Exposure to these stressors, coupled
with intensive farming, raised the need for adequate management of the wild
populations and farmed production. To improve the difficult balance between
ecological and socio-economic interests, it must be investigated how bivalves
cope with external stressors. For this reason, the effects caused by the
following stressors were studied in the present thesis: (i) harmful algal blooms
(HABs) and (ii) climate change drivers in the seawater, such as warming (W)
and acidification (A). HABs are increasing in frequency, intensity, persistence
and geographic distribution, but are ambiguous regarding their ecological
impacts and physiological effects on bivalves. A strong relationship is emerging
between HABs and climate change drivers, what may affect both shellfisheries
and wild populations. Hence, the objectives of this work are: 1) To determine
whether native and invasive species cope with exposure to HABs differently,
providing new insights on species-specificities and ecosystem functioning
fragilities in the presence of invasive species and HABs; 2) To evaluate how
commercially valuable bivalve species cope with simultaneous exposure to
several climate change drives and HABs, providing new insights on how
environmental changes affects toxicokinetics, physiological and genotoxic
bivalves responses, under HABs; 3) To assess, marine biotoxins
bioaccessibility, contributing for new information relevant for risk assessment.
Different approaches were used to achieve these goals. Complementary field
and laboratory works were performed to assess toxicokinetics patterns of
marine biotoxins in native vs. invasive bivalve species. Bivalves were exposed
to toxic blooming algae species under climate change scenarios in laboratory controlled conditions to assess toxicokinetics as well as physiological and
genotoxic responses. Naturally contaminated bivalves were used to investigate
their role as vectors of marine biotoxins to humans through in vitro digestion
methodology.
Relevant new data were obtained regarding the effects of several marine
biotoxins in bivalves. Assessing accumulation of regulated and non-regulated
biotoxins in native (Ruditapes decussatus) and invasive (R. philippinarum) clam
species from Aveiro Lagoon revealed that higher toxin content, particularly
regarding OA-group, the most abundant and frequent toxins in the Portuguese
coast, is reached by native species. Accumulation of lower toxin levels by
invasive clam may then favour farmers interest for their production. The kinetics
and genotoxicity study in native and invasive clams reinforced the existence of
species-specific behaviours. Exposure to the dinoflagellate Prorocentrum lima, an OA and dinophysistoxin-1 producer, increases the pressure over native
clams, with higher toxin accumulation and genetic damage, as well as early
and increased induction of DNA repair activity. Invasive clams, on the other
hand, are better adapted to cope with these challenges.
Investigation of the effects of combined exposure to multiple stressors revealed
that W, A and HABs alter the accumulation/elimination dynamics of Paralytic
Shellfish Poisoning (PSP) toxins in mussels (Mytilus galloprovincialis). Lower
accumulation levels and slower elimination rates were observed. The predicted
climate change scenarios and exposure to HABs may then lead to lower
contamination levels but to longer harvesting closures. Simultaneous exposure
to altered environmental conditions and HABs also had significant impacts in
the antioxidant system and DNA integrity, resulting in an organ-specific
modulation of the antioxidant response, increasing genetic damage and
preventing/retarding DNA damage repair. However, the DNA damage observed
seems to be non-oxidative. While the investigation of factors enhancing the
elimination of toxins was not a main objective of this study, it was shown that
acidification might promote PSP toxins elimination in mussels.
Finally, the bioaccessibility studies revealed a significant reduction in the OA group content available after in vitro digestion, suggesting an overestimation of
exposure to these biotoxins and an over-conservative approach in safety levels
definition and risk assessment studies. This was the first study assessing
bioaccessibility of OA-group toxins, pointing to in vitro digestion as a promising
tool to obtain accurate data regarding toxin ingestion from bivalves to the
consumer. Overall, the complexity of the relation between HABs, bivalves and
the environmental factors to consider in marine biotoxins management is
increasing, and only continuous and extensive monitoring of environmental,
biological and anthropogenic conditions may allow for a healthier balance
between environmental and socio-economic interests.Os bivalves desempenham um papel fundamental nos ecossistemas marinhos
costeiros, limitando a produtividade primária através de um forte mecanismo
de controlo descendente (top-down) sobre as comunidades fitoplanctónicas e
constituindo uma importante fonte de alimento para níveis tróficos superiores,
incluindo o Homem. No entanto, os bivalves são afetados negativamente por
diversos fatores externos, sejam estes bióticos ou abióticos, naturais ou
antropogénicos. A exposição a estes fatores, juntamente com a exploração
intensiva, leva a que seja necessária a implementação de uma gestão
adequada, quer das populações selvagens, quer da produção em viveiros.
Para melhorar esta gestão e alcançar o difícil equilíbro entre os interesses
ecológicos e socioeconómicos é então necessário entender como os bivalves
lidam com estes fatores. Na presente tese foram estudados os efeitos
provocados por (i) blooms de algas tóxicas (HABs do inglês harmful algal
blooms) e (ii) alterações das condições climáticas da água do mar,
nomeadamente o aumento da temperatura (W) e acidificação (A). Os HABs
são fenómenos que estão a aumentar em frequência, intensidade, persistência
e abrangência geográfica, mas cujos impactos ecológicos e efeitos fisiológicos
em bivalves são ainda ambíguos. Concomitantemente com a pressão causada
pelo aumento da procura de bivalves como alimento para as populações
humanas, fortes indícios apontam para uma relação entre a ocorrência de
HABs e os parâmetros de alterações climáticas, o que pode afectar quer a
indústria, quer as populações de bivalves selvagens. Assim, os objetivos deste
trabalho são: 1) Determinar se as espécies nativas e invasoras lidam com a
exposição a HABs de maneira diferente, dando particular atenção às respostas
específicas de cada espécie e às fragilidades do ecossistema em relação à
presença de espécies invasoras e ocorrência dos referidos blooms; 2) Avaliar
como espécies de bivalves de elevado valor comercial lidam com a exposição
simultânea a alterações climáticas e HABs, focando o efeito destas
modificações ambientais na toxicocinética, respostas fisiológicas e
genotoxicidade exibida pelos bivalves; 3) Avaliar a bioacessibilidade das
biotoxinas marinhas, focando a obtenção de informação relevante para
estudos de avaliação de risco.
Foram realizados trabalhos complementares de campo e laboratório, de modo
a avaliar os padrões de toxicocinética de biotoxinas marinhas em espécies
nativas e invasoras. Em laboratório, foram expostos bivalves a algas tóxicas
em condições alteradas, simulando a ocorrência de HABs sob cenários
previstos de alterações climáticas, de modo a avaliar toxicocinética, respostas
fisiológicas e genotoxicidade. Exemplares naturalmente contaminados foram
utilizados para investigar o papel dos bivalves como vectores de biotoxinas
marinhas para humanos, através da simulação da digestão in vitro. O estudo da acumulação de biotoxinas marinhas regulamentadas e não
regulamentadas em espécies de amêijoa nativa (Ruditapes decussatus) e
invasora (R. philippinarum) na Ria de Aveiro revelou que a espécie nativa
acumula, recorrentemente, concentrações de toxinas mais elevadas,
particularmente no que toca a toxinas grupo do ácido ocadaico (OA do inglês
Okadaic acid), sendo estas as toxinas mais abundantes e frequentes na costa
Portuguesa. A acumulação de menores concentrações por parte da amêijoa
invasora pode promover o interesse dos produtores na sua exploração. A
avaliação da cinética e da genotoxicidade em amêijoas nativas e invasoras
reforçou a existência de comportamentos específicos para cada espécie. A
exposição a Prorocentrum lima, dinoflagelado produtor de OA e dinofisistoxina 1, aumenta a pressão sobre a espécie nativa, uma vez que esta apresenta as
concentrações de toxina mais elevadas, os maiores níveis de dano genético e
ainda uma indução mais precoce e intensa dos mecanismos de reparação de
dano no ADN. A espécie invasora, por outro lado, está melhor adaptada para
lidar com estes desafios.
O estudo dos efeitos de fatores múltiplos combinados revelou que a exposição
a W, A e HABs altera a dinâmica de acumulação/eliminação de toxinas
paralisantes (PSP) em mexilhões (Mytilus galloprovincialis). Os cenários
previstos de alterações climáticas e a exposição a HABs sugerem níveis de
contaminação mais baixos, mas períodos de interdição de apanha mais
prolongados. A exposição simultânea a condições ambientais alteradas e
HABs também teve um impacto significativo no sistema antioxidante e na
integridade do ADN, resultando numa modulação da resposta antioxidante
especifica para cada órgão, aumentando o dano genético e
prevenindo/atrasando a sua reparação. O dano encontrado no ADN parece, no
entanto, não ser oxidativo. Embora a investigação de fatores que favoreçam a
rápida eliminação de toxinas não constasse dos objetivos deste estudo, foi
demonstrado que a acidificação pode promover a eliminação de toxinas PSP
em mexilhões.
Por último, os estudos de bioacessibilidade revelaram uma redução na
quantidade de toxinas do grupo-OA disponível após a digestão in vitro,
sugerindo uma sobrestimação da exposição a estas biotoxinas e
recomendando uma abordagem conservadora na definição dos níveis de
segurança e na subsequente avaliação do risco. Este foi o primeiro estudo a
determinar a bioacessibilidade das toxinas do grupo-OA e sugere a técnica de
digestão in vitro como uma ferramenta promissora na obtenção de dados
rigorosos no que diz respeito à ingestão de toxinas através de bivalves e a
acessibilidade destes compostos no organismo humano. De um modo geral, a
complexidade da relação entre HABs, bivalves e os fatores ambientais a serem
considerados na gestão de biotoxinas marinhas está a aumentar, e apenas
uma contínua e extensiva monitorização das condições ambientais, biológicas
e antropogénicas pode permitir um melhor equilíbrio entre os interesses

Relatório de estágio de mestrado, Ecologia e Gestão Ambiental, Universidade de Lisboa, Faculdade de Ciências, 2020A procura e utilização de Fontes de Energia Renovável (FER) tem tido um crescimento elevado nos últimos anos, concentrado nos centros eletroprodutores do país, como os parques eólicos. Este crescimento levou a que Portugal atingisse o limite de capacidade para esta energia onshore e que assim fossem procuradas estratégias que permitissem ultrapassar esta saturação. Uma das soluções comuns passa pela repotenciação (repowering) dos parques, processo que permite a manutenção ou aumento da potência instalada sem aumentar o número de aerogeradores presentes. Este facto é particularmente importante para a redução do impacte habitualmente gerado por estas infraestruturas para com a biodiversidade local, nomeadamente as mais afectadas: as comunidades de aves e quirópteros. Entre os impactes diretos, como a colisão dos indivíduos com as pás dos aerogeradores, e indiretos, como a alteração de comportamentos, existe sempre um potencial de perturbação que deve ser estudado à luz do quadro legal em vigor, que enquadram a Avaliação de Impacte Ambiental (AIA), e das metodologias de monitorização mais adequadas. Os Estudos de Impacte Ambiental (EIA) surgem assim como uma ferramenta indispensável à análise da convivência das comunidades faunísticas com empreendimentos eólicos e avaliação de possíveis efeitos decorrentes da sua atividade. O presente trabalho visou estudar o efeito da remodelação do Parque Eólico (PE) de Vila Lobos (Viseu, Portugal) nas comunidades de aves em geral, aves de rapina e outras planadoras, e quirópteros no decorrer do seu 2º ano de exploração (2018-2019). O projeto de remodelação passou pela repotenciação do parque e consequente redução do número de aerogeradores de 20 para 3, com aumento da potência unitária das 3 novas máquinas. O projeto inicial de Vila Lobos não foi sujeito ao processo de AIA e o único indício do impacte potencial da sua operação é considerado como o ponto de partida do projeto de remodelação, correspondendo à sua fase de pré-construção (2014-2015). Assim, este estudo visou identificar um eventual efeito de exclusão sobre as comunidades consideradas, alterações no uso da área do PE pelas aves de rapina e outras planadoras, a influência na utilização de abrigos de quirópteros, e determinar a mortalidade de indivíduos associada ao PE. Os resultados para o 2º ano de exploração indicam que a remodelação do PE de Vila Lobos não se encontra a exercer um efeito de exclusão nas comunidades consideradas e que, de um modo geral, a área aparenta estar a ser progressivamente colonizada. Tal poderá ser consequência da redução do número de aerogeradores existentes e respetiva substituição por novas máquinas, com características diferentes das originais, que contrariam um potencial risco de colisão. O historial de incêndios surge como um importante factor de perturbação local que poderá explicar as variações encontradas na abundância relativa e riqueza específica das comunidades em anos anteriores. Verificou-se ainda uma intensificação da utilização da área PE pelas aves de rapina e outras planadoras, associada a um natural aumento do risco de colisão. No que diz respeito à comunidade de quirópteros, denotou-se a tendência esperada de aumento da sua atividade de acordo com o aumento da temperatura e com menores velocidades de vento, bem como a ausência de perturbação quer pela análise acústica, quer pela amostragem de abrigos na área de influência do PE. Não foi registada qualquer carcaça de ave ou quiróptero no 2º ano de exploração do PE de Vila Lobos, pelo que se considerou a mortalidade como sendo eventualmente residual por motivos de eficácia técnica. O estudo permitiu assim corroborar o potencial do processo de repotenciação de parques eólicos na redução global do seu impacte paisagístico e na biodiversidade local, aliada à sua sustentabilidade financeira e energética.The demand and use of Renewable Energy Sources (FER) has been growing strongly in recent years, concentrated in the country's power plants such as wind farms. This growth has led Portugal to reach the capacity limit for this onshore energy and thus strategies were sought to overcome this saturation. One of the common solutions is the repowering of wind farms, a process that allows the maintenance or increase of installed power without increasing the number of wind turbines present. This is particularly important for reducing the impact usually generated by these infrastructures on local biodiversity, particularly those most affected: the communities of birds and chiropters. Among the direct impacts, such as the collision of individuals with the blades of the wind turbines, and indirect impacts, such as changes in behaviour, there is always a potential for disturbance that should be studied in the light of the legal framework in force, which frames the Environmental Impact Assessment (AIA), and the most appropriate monitoring methodologies. Environmental Impact Studies (EIA) emerge as an indispensable tool for the analysis of the coexistence of faunal communities with wind farms and the evaluation of possible effects arising from their activity. This work aimed to study the effect of the remodelling of the Vila Lobos Wind Farm (PE) (Viseu, Portugal) on the communities of birds in general, birds of prey and other gliders, and chiropters during its 2nd year of operation (2018-2019). The refurbishment project involved repowering the park and consequently reducing the number of wind turbines from 20 to 3, with an increase in the unit power of the 3 new machines. The initial project at Vila Lobos was not subject to the EIA process and the only indication of the potential impact of its operation is considered to be the starting point of the refurbishment project, corresponding to its pre-construction phase (2014-2015). Thus, this study aimed to identify a possible exclusionary effect on the communities considered, changes in the use of the PE area by birds of prey and other gliders, the influence on the use of chiroptera shelters, and to determine the mortality of individuals associated with the PE. The results for the 2nd year of exploration indicate that the remodelling of the PE of Vila Lobos is not having an exclusionary effect on the communities considered and that, in general, the area appears to be progressively colonised. This may be a consequence of the reduction in the number of existing wind turbines and their replacement by new machines, with different characteristics from the original ones, which counteract a potential collision risk. The history of fires appears to be an important local disturbance factor which may explain the variations found in the relative abundance and specific wealth of communities in previous years. There has also been an intensification of the use of the PE area by birds of prey and other gliders, associated with a natural increase in the risk of collision. With regard to the chiropter community, there was an expected tendency to increase its activity according to the increase in temperature and lower wind speeds, as well as the absence of disturbance either by acoustic analysis or by sampling of shelters in the area of influence of PE. No bird or chiropter carcass was registered in the 2nd year of operation of Vila Lobos PE, so the mortality was considered as possibly residual for reasons of technical efficiency. The study thus made it possible to corroborate the potential of the process of repowering wind farms in the overall reduction of their impact on the landscape and local biodiversity, together with their financial and energy sustainability
Autoria, tradição e inovação no folclore do Baixo Minho: Um estudo de caso da Rusga de São Vicente de Braga
Esta dissertação de mestrado tem o propósito de esclarecer de que forma é que os
conceitos de autoria e pertença da música popular (a que se canta e toca associada à
ruralidade) influenciam os processos de categorização de autenticidade e tradição.
Questiona a identidade cultural e o pensamento de autenticidade ligados ao folclore
português.
O principal objetivo deste estudo é perceber como é que as noções de autoria e
pertença no repertório musical e coreográfico no âmbito do “universo” do folclore se
relacionam com os conceitos de “autenticidade” e “tradição”. Para cumprir este objetivo,
é realizado um estudo de caso da Rusga de São Vicente de Braga – Grupo Etnográfico do
Baixo Minho.
A partir de todos os dados recolhidos e analisados, foi possível concluir que a
autoria da música popular, enquanto conceito se situa no cerne de uma problemática
complexa e controversa, que consiste na mitificação da música popular como fruto
intemporal da alma do povo e elemento importante da definição da nacionalidade e
identidade de um povo. O conceito de autenticidade é aqui posto em causa através da
análise do processo de criação da música folclórica.
Conclui-se que o processo de criação é visto, cada vez mais, como um fenómeno
individual, que consiste no processo em que o autor ou compositor compõe segundo os
parâmetros que a tradição ou escola anterior lhe transmite. Na chamada tradição popular,
esses parâmetros são fornecidos via oral. Com o passar do tempo, a identificação desses
autores ficou perdida. A partir daí, a composição musical foi apropriada pelos grupos
folclóricos como sua e transmitida para a geração seguinte. Os grupos recebem,
assimilam, adaptam e recriam as composições deixadas pelas gerações anteriores. A
“tradição” consiste na transmissão; e a autoria é a receção dessa transmissão, dessa
“tradição”. Desta forma, constato que estes dois conceitos tão díspares estão
completamente interligados e articulados.This master dissertation aims to clarify how the concepts of authorship and
belonging in popular music (the one that is sung and played in a rural context) influence
the process of categorization of authenticity and tradition. It questions the cultural identity
and the thought of authenticity related to Portuguese folklore.
The main object of study in this work, it is to understand how the notions of
authorship and belonging in musical and choreographic repertoire within the “universe”
of folklore are related to the concepts of “authenticity” and “tradition”. To fulfill this
objective, it is realized a case of study of Rusga de São Vicente de Braga – Grupo
Etnográfico do Baixo-Minho.
From all the collected and analyzed data, it was possible to conclude that the
popular music authorship as a concept relies on the heart of a complex and controversial
problematic, which consists in the mythification of popular music as a timeless fruit of a
certain community’s soul and an important element of nationality definition and a
population’s identity. The concept of authenticity is here questioned through the analysis
of the folk music creation process.
It is concluded that the creative process is increasingly seen as an individual
phenomenon, which consists in the process where the author or composer creates
according to the parameters that the previous tradition or school transmits. In popular
tradition, these parameters are provided orally and over time, the identification of this
authors was lost. From there, the musical composition was appropriated by the folkloric
groups and transmitted to the next generation. The groups receive, assimilate, adapt and
recreate the compositions left by previous generations. “Tradition” consists on
transmission; and the authorship is the reception of that transmission, of that “tradition”.
In this way, I found that these two concepts, which are so distinct, are completely
interconnected and articulated
Does Ocean Sunfish Mola spp. (Tetraodontiformes: Molidae) represent a risk for Tetrodotoxin Poisoning in the Portuguese Coast?
Tetrodotoxin (TTX) is a potent neurotoxin naturally occurring in terrestrial and marine
organisms such as pufferfish. Due to the risk of TTX poisoning, fish of Tetraodontidae family and
other puffer-related species must not be placed in the EU markets. This restriction applies to fish of
the family Molidae even though no data on toxins’ occurrence is available. In this study, the presence
of TTX and its analogues was investigated in the main edible tissue (the white muscle) and the main
xenobiotics storage organ (the liver) of ocean sunfish Mola spp. (n = 13) from the South Portuguese
coast. HILIC-MS/MS analyses did not reveal TTX in the analyzed samples, suggesting an inexistent

Estilos parentais dos pais de crianças com perturbação de hiperactividade e défice de atenção
A presente investigação teve como objectivo estudar a relação entre o diagnóstico de
Perturbação de Hiperactividade e Défice de Atenção nas crianças e o Estilo Parental
adoptado pelos respectivos pais para a educação dos seus filhos. Foi recolhida uma
amostra de conveniência constituída por 50 crianças (36 do género masculino e 14 do
género feminino), com idades compreendidas entre os 5 e os 14 anos de idade, com
diagnóstico de Perturbação de Hiperactividade e Défice de Atenção e um representante educacional de cada criança (pai/mãe). Os instrumentos utilizados foram o Questionário de Conners para pais e professores e o Inventário de Estilos Parentais. De uma forma geral, os resultados demonstram que não existem diferenças estatisticamente significativas nos Estilos Parentais adoptados pelos pais, quer para rapazes quer para raparigas, assim como a idade dos pais não exerce influência sobre os Estilos Parentais postos em prática na educação dos filhos. Na avaliação de diagnóstico de hiperactividade, os pais são mais severos em relação aos professores e ao mesmo tempo não se verificam diferenças estatisticamente significativas na avaliação de pais e professores no que se refere às diferenças de género. Quanto à avaliação feita por pais e professores, as crianças com idades entre os 5 e os 8 anos de idade, apresentam maior grau de hiperactividade e são a faixa etária que representa diferenças significativas na prática educativa "abuso físico", evidenciando valores mais altos quando comparada com a faixa etária dos 9 aos 14 anos de idade. Apesar de os pais serem severos na avaliação do diagnóstico dos seus filhos, a prática educativa mais utilizada é o “Comportamento Moral”. Espera-se que estes resultados possam contribuir para a investigação nesta área e para a criação de intervenções eficazes que permitam aos pais adaptar um estilo parental que vá de encontro às necessidades que a patologia requer.This investigation aims to assess the relationship between the diagnosis of Attention
Deficit Hyperactivity Disorder in children and the Parental style adopted in these children’s up-bringing. A consecutive sample of 50 children (36 male and 14 female) between the ages of 5 to 14 years old, with Attention Deficit Hyperactivity Disorder and an educational representative of each child (parent) was collected. The assessing
instruments for this investigation were: the Conner’s Questionnaire for parents and
teachers and the Parenting Styles Inventory. The overall results revealed no statistically
significant differences between the Parenting Styles adopted by these parents, either for
the boys or the girls, as well as the parents´ age presented no influence on Parenting
Styles put into practice in the up-bringing of their children. In the diagnostic evaluation
of hyperactivity, the parents are more severe than teachers and at the same time there
are no statistically significant differences in the assessment of parents and teachers in
regards to gender differences. Regarding the evaluation made by parents and teachers,
children with ages between 5 to 8 years old present a higher degree of hyperactivity and
it is this age group that accounts for significant differences in the educational practice of “physical abuse”, showing higher values in comparison to the age group of 9 to 14 years old. Despite the parents being strict in the diagnostic assessment of their children, the
most widely used educational practice is the “Moral Behaviour”. It is hoped that these
findings can contribute in this area of research and create effective interventions that
enable parents to tailor a parenting style that goes according to the needs that the pathology requires
Bivalve shellfish safety in Portugal: variability of faecal levels, metal contaminants and marine biotoxins during the last decade (2011–2020)
Bivalves are a high-value product whose production has markedly increased, reaching 9863 tonnes in Portugal in 2021. Bivalves' habitats-lagoons, estuaries and coastal waters-are exposed to biological and anthropogenic contaminants, which can bioaccumulate in these organisms and pose a significant public health risk. The need to obtain a safe product for human consumption led to the implementation of standardised hygiene regulations for harvesting and marketing bivalve molluscs, resulting in routine monitoring of bivalve production areas for microbial quality, metal contaminants, and marine biotoxins. While excessive levels of biotoxins and metal contamination lead to temporary harvesting bans, high faecal contamination leads to area reclassification and impose post-harvest treatments. In this study, the seasonal and temporal variability of these parameters were analysed using historical data generated by the monitoring programme during the last decade. Moreover, the impact of the monitoring program on bivalve harvesting from 2011 to 2020 was assessed.
