High-Throughput System for the Early Quantification of Major Architectural Traits in Olive Breeding Trials Using UAV Images and OBIA Techniques
The need for the olive farm modernization have encouraged the research of more efficient crop management strategies through cross-breeding programs to release new olive cultivars more suitable for mechanization and use in intensive orchards, with high quality production and resistance to biotic and abiotic stresses. The advancement of breeding programs are hampered by the lack of efficient phenotyping methods to quickly and accurately acquire crop traits such as morphological attributes (tree vigor and vegetative growth habits), which are key to identify desirable genotypes as early as possible. In this context, an UAV-based high-throughput system for olive breeding program applications was developed to extract tree traits in large-scale phenotyping studies under field conditions. The system consisted of UAV-flight configurations, in terms of flight altitude and image overlaps, and a novel, automatic, and accurate object-based image analysis (OBIA) algorithm based on point clouds, which was evaluated in two experimental trials in the framework of a table olive breeding program, with the aim to determine the earliest date for suitable quantifying of tree architectural traits. Two training systems (intensive and hedgerow) were evaluated at two very early stages of tree growth: 15 and 27 months after planting. Digital Terrain Models (DTMs) were automatically and accurately generated by the algorithm as well as every olive tree identified, independently of the training system and tree age. The architectural traits, specially tree height and crown area, were estimated with high accuracy in the second flight campaign, i.e. 27 months after planting. Differences in the quality of 3D crown reconstruction were found for the growth patterns derived from each training system. These key phenotyping traits could be used in several olive breeding programs, as well as to address some agronomical goals. In addition, this system is cost and time optimized, so that requested architectural traits could be provided in the same day as UAV flights. This high-throughput system may solve the actual bottleneck of plant phenotyping of "linking genotype and phenotype," considered a major challenge for crop research in the 21st century, and bring forward the crucial time of decision making for breeders
Eficacia de la insuflación con dióxido de carbono en enteroscopia de un solo balón para determinar profundidad de intubación y dolor posprocedimiento en pacientes referidos para evaluación del intestino delgado
ResumenIntroducciónLa intubación profunda del intestino delgado es limitada por la distensión debido a la insuflación con aire durante el procedimiento. El dióxido de carbono (CO2) es rápidamente absorbido de la luz intestinal previniendo la distensión abdominal.ObjetivoComparar el efecto de la insuflación con CO2 y aire en la intubación profunda durante enteroscopia de un solo balón (ESB) y dolor posprocedimiento.DiseñoEstudio descriptivo de las series de casos y comparación de los resultados de sus variables, en un hospital terciario.Pacientes y métodosCiento cuarenta y siete ESB fueron realizadas en 130 pacientes con sospecha de enfermedad del intestino delgado usando insuflación con CO2 en 65 procedimientos y 82 con aire. Enteroscopio Olympus SIF 180-Q y bomba CO2 UCR Olympus fueron usados. Intubación profunda fue medida usando una escala validada. El dolor fue evaluado post-ESB y a las 12 h.ResultadosCiento cuarenta y siete ESB fueron realizadas en 130 pacientes con una media de intubación en el grupo CO2 significativamente mayor que en el grupo aire (p = 0.008). Noventa y cinco ESB anterógrada y 52 ESB retrógrada fueron realizadas con una media de intubación significativamente mayor en el grupo CO2 (p = 0.035 y p= 0.004, respectivamente). Carbon dioxide (CO2) is rapidly absorbed from the bowel lumen and prevents distention of abdomen.ObjectiveA comparison was made between the effects of CO2 and air insufflations on the intubation depth during single balloon enteroscopy (SBE), as well as post-procedure pain.DesignDescriptive case series study and comparison of the results in a tertiary care hospital.Patients and methodsA total of 147 SBE procedures were performed on 130 patients with suspected small bowel disease using CO2 insufflation in 65 procedures, and air insufflation in 82. Enteroscopy was performed using an Olympus SIF 180-Q and a UCR CO2 pump. Intubation depth was recorded using a validated form, and pos-procedure pain immediately after SBE and 12 hours later were evaluated.ResultsA total of 147 SBE procedures were performed on 130 patients with a mean small bowel intubation depth in the CO2 group significantly larger than the air group (p=.008). An oral approach was used in 95 procedures, and anal in 52 procedureswith a mean intubation depth significantly larger in the CO2 group (p=.035 and p=.004, respectively). Histological characterization of table olive fruit bruising cv. Manzanilla de Sevilla
Sin paginarActualmente el sector del olivar en España incluye dos subsectores claramente diferenciados por su aprovechamiento: el olivar de almazara y el de mesa o verdeo, cada uno con unas particularidades específicas que los diferencian, tales como variedades empleada s y técnicas de cultivo como la poda o la recolección. En los últimos años, la necesidad de optimizar los costes de producción ha generalizado la recolección mecanizada mediante brazos vibradores o máquinas cabalgadoras en explotaciones de olivar de aceite. Sin embargo, no se ha extendido de igual manera en plantaciones de aceituna de mesa, debido a los altos porcentajes de frutos dañados por los golpes que se producen durante la recolección mecanizada. El molestado, manchas oscuras en la parte externa del exocarpo de los frutos, hace referencia a los daños provocados en los frutos por los golpes durante la recolección, más notables en la recolección mecánica. Estas manchas deprecian el fruto a nivel comercial con el consecuente rechazo del consumidor. No todos los cultivares presentan la misma susceptibilidad al molestado. ‘Manzanilla de Sevilla’, variedad por excelencia de aceituna de mesa, es considerada sensible al molestado lo que dificulta su recolección por medios mecánicos. El Departamento de Ciencias Agroforestales de la Un iversidad de Sevilla empezó a trabajar en el año 2003 en un programa de mejora genética de olivo para mesa. Una de las líneas de trabajo dentro de este proyecto ha consistido en estudiar diferentes parámetros de calidad, entre los que se encuentra el molestado. En colaboración con el Instituto de Agricultura Sostenible-CSIC, se ha desarrollado una metodología para evaluar el mismo en aceituna de mesa. En este trabajo se describe anatómicamente el molestado en frutos frescos de ‘Manzanilla de Sevilla’, después de un golpe inducido, a los 30 minutos y 24 horas del impacto y fijados posteriormente con FAE (formalina, ácido acético, etanol del 95% y agua destilada (10:5:50:3 5 v/v/v/v)). Se han evaluado histológicamente los cambios (rupturas celulares y pérdida de grosor en paredes celulares) que se producen en el mesocarpo de las zonas dañadas. Las observaciones histológicas han revelad o diferencias en los daños para los dos momentos evaluados. La presencia de rupturas celulares y la pérdida de grosor de las paredes celulares, en las zonas dañadas, han sido más evidentes a las 24 horas tras el golpe.Currently olive growing in Spain includes two main industries: olive oil and table olives, each of which has its
own peculiarities, such as different varieties and specific performance of cultivation techniques like pruning or
harvesting. In recent years, due to the need of reducing production costs, mechanical harvesting using trunk
shakers or straddle harvesters has become widespread in olive oil orchards. However, that is not the case in table
olive plantations, due to the high percentage of fruit damaged by the impacts that take place during mechanical
harvesting.
The term fruit bruising, dark spots on the outside of the fruit exocarp, refers to the fruit damage caused by
impacts during collection, most notably in mechanical harvesting. These spots decreases commercial fruit value,
being rejected by the consumer. Not all cultivars have the same fruit bruise susceptibility. Manzanilla de Sevilla,
Spanish leading table olive cultivar, is considered to be sensitive to bruising being therefore difficult to harvest
by mechanical devices
Researchers from the Departamento de Ciencias Agroforestales at the University of Seville, Spain, are working
since 2003 on a table olive breeding program. One of the research lines within the program has been to study. different table olive quality parameters, including fruit bruising. An evaluation methodology is being developed
to asses this phenomenon in collaboration with the Instituto de Agricultura Sostenible-CSIC.
This paper describes anatomical changes related to fruit bruising in 'Manzanilla de Sevilla' fresh fruits after an
induced impact. Bruised fruits were chemically fixed in FAE (formalin, acetic acid, 95% ethanol and distilled
water (10:5:50:35 v/v/v/v)) 30 minutes and 24 hours after the impact. Histological changes (cell breakage, loss
of cell wall thickness and higher colour intensity) produced in the mesocarp of the damaged areas have been
evaluated. Histological observations revealed differences between the evaluated times. The presence of cell
ruptures and the loss of thickness in the cell walls, in the damaged areas, has been most evident at 24 hours after

Plasmodium species differentiation by non-expert on-line volunteers for remote malaria field diagnosis
BACKGROUND: Routine field diagnosis of malaria is a considerable challenge in rural and low resources endemic areas mainly due to lack of personnel, training and sample processing capacity. In addition, differential diagnosis of Plasmodium species has a high level of misdiagnosis. Real time remote microscopical diagnosis through on-line crowdsourcing platforms could be converted into an agile network to support diagnosis-based treatment and malaria control in low resources areas. This study explores whether accurate Plasmodium species identification-a critical step during the diagnosis protocol in order to choose the appropriate medication-is possible through the information provided by non-trained on-line volunteers. METHODS: 88 volunteers have performed a series of questionnaires over 110 images to differentiate species (Plasmodium falciparum, Plasmodium ovale, Plasmodium vivax, Plasmodium malariae, Plasmodium knowlesi) and parasite staging from thin blood smear images digitalized with a smartphone camera adapted to the ocular of a conventional light microscope. Visual cues evaluated in the surveys include texture and colour, parasite shape and red blood size. RESULTS: On-line volunteers are able to discriminate Plasmodium species (P. falciparum, P. malariae, P. vivax, P. ovale, P. knowlesi) and stages in thin-blood smears according to visual cues observed on digitalized images of parasitized red blood cells. Friendly textual descriptions of the visual cues and specialized malaria terminology is key for volunteers learning and efficiency. CONCLUSIONS: On-line volunteers with short-training are able to differentiate malaria parasite species and parasite stages from digitalized thin smears based on simple visual cues (shape, size, texture and colour). While the accuracy of a single on-line expert is far from perfect, a single parasite classification obtained by combining the opinions of multiple on-line volunteers over the same smear, could improve accuracy and reliability of Plasmodium species identification in remote malaria diagnosis.M.L. holds a postdoctoral Fellowship of the Spanish Ministry of Economy and Competitiveness (FPDI-2013-16409). CYLD regulates keratinocyte differentiation and skin cancer progression in humans
CYLD is a gene mutated in familial cylindromatosis and related diseases, leading to the development of skin appendages tumors. Although the deubiquitinase CYLD is a skin tumor suppressor, its role in skin physiology is unknown. Using skin organotypic cultures as experimental model to mimic human skin, we have found that CYLD acts as a regulator of epidermal differentiation in humans through the JNK signaling pathway. We have determined the requirement of CYLD for the maintenance of epidermal polarity, keratinocyte differentiation and apoptosis. We show that CYLD overexpression increases keratinocyte differentiation while CYLD loss of function impairs epidermal differentiation. In addition, we describe the important role of CYLD in the control of human non-melanoma skin cancer progression. Our results show the reversion of the malignancy of human squamous cell carcinomas that express increased levels of CYLD, while its functional inhibition enhances the aggressiveness of these tumors which progress toward spindle cell carcinomas. We have found that the mechanisms through which CYLD regulates skin cancer progression include the control of tumor differentiation, angiogenesis and cell survival. Longitudinally extensive transverse myelitis with AQP4 antibodies revealing ovarian teratoma.
Paraneoplastic myelitis is a rare inflammatory disorder most frequently associated with solid tumors or lymphoproliferative disorders. Patients often harbor onconeuronal antibodies and their prognosis is usually poor. Here we report a 42-year old woman with longitudinally extensive transverse myelitis and aquaporin-4 (AQP4) antibodies that led to the diagnosis of ovarian teratoma. After tumor removal and immune therapy (including corticosteroids, plasma exchange, intravenous immunoglobulins and rituximab) the patient progressively improved achieving complete recovery. Histological study of the teratoma demonstrated neural tissue containing AQP4 expressing cells and intense inflammatory infiltrates, providing evidence for a possible paraneoplastic link between both disorders
Origin of inverse Rashba-Edelstein effect detected at the Cu/Bi interface using lateral spin valves
Under the terms of the Creative Commons Attribution License 3.0 (CC-BY).The spin transport and spin-to-charge current conversion properties of bismuth are investigated using permalloy/copper/bismuth (Py/Cu/Bi) lateral spin valve structures. The spin current is strongly absorbed at the surface of Bi, leading to ultrashort spin-diffusion lengths. A spin-to-charge current conversion is measured, which is attributed to the inverse Rashba-Edelstein effect at the Cu/Bi interface. The spin-current-induced charge current is found to change direction with increasing temperature. A theoretical analysis relates this behavior to the complex spin structure and dispersion of the surface states at the Fermi energy. Ischemic aetiology, self-reported frailty, and gender with respect to cognitive impairment in chronic heart failure patients
Background: decisive information on the parameters involved in cognitive impairment in patients with chronic heart failure is as yet lacking. Our aim was to determine the functional and psychosocial variables related with cognitive impairment using the mini-mental-state examination (MMSE) with age-and education-corrected scores. Methods: a cohort study of chronic heart failure patients included in an integrated multidisciplinary hospital/primary care program. The MMSE (corrected for age and education in the Spanish population) was administered at enrolment in the program. Analyses were performed in 525 patients. Demographic and clinical variables were collected. Comprehensive assessment included depression (Yesavage), family function (family APGAR), social network (Duke), dependence (Barthel Index), frailty (Barber), and comorbidities. Univariate and multivariate logistic regression were performed to determine the predictors of cognitive impairment. Results: cognitive impairment affected 145 patients (27.6 %). Explanatory factors were gender (OR: 2.77 (1.75-4.39) p 3.5 (OR: 0.59 (0.35-0.99) p = 0.048), and beta-blocker treatment (OR: 0.36 (0.17 to 0.76, p = 0.007)). No association was found between cognitive impairment and social support or family function. Conclusion: the observed prevalence of cognitive impairment using MMSE corrected scores was 27.6 %. A global approach in the management of these patients is needed, especially focusing on women and patients with frailty, low albumin levels, and ischemic aetiology heart failure
