Parâmetros oftálmicos de carnĂvoros do cerrado
Dissertação (mestrado)—Universidade de BrasĂlia, Faculdade de Agronomia e Medicina Veterinária, 2018.O presente estudo descreve os parâmetros oftálmicos morfolĂłgicos e fisiolĂłgicos de trĂŞs espĂ©cies de carnĂvoros do Cerrado, cachorro-do-mato (Cerdocyon thous), lobo-guará (Chrysocyon brachyurus) e quati (Nasua nasua). Todas os procedimentos foram realizados na Fundação Jardim ZoolĂłgico de BrasĂlia, contemplando seis cachorros-do-mato, oito lobos-guará e 15 quatis, submetidos a contenção quĂmica. Os animais foram submetidos a avaliação oftálmica, teste lacrimal de Schirmer (TLS), aferição de pressĂŁo intraocular (PIO) de rebote, PIO de aplanação (cachorro-do-mato e quati), aferição do comprimento da fenda palpebral (FP), gonioscopia (cachorro-do-mato e lobo-guará) e fundoscopia. Os dados obtidos foram submetidos Ă avaliação estatĂstica mediante testes ANOVA e de Tukey, de acordo com idade e sexo. Para cachorro-do-mato, os valores mĂ©dios ± desvio padrĂŁo foram de 4,3 ± 2,96 mm/min para TLS; 18,45 ± 1,5 mm para FP de machos; 16,45 ± 0,79 mm para FP de fĂŞmeas; tonometria de rebote: 15,50 ± 0,71 mmHg para animais jovens e 9,50 ± 2,62 mmHg para adultos com TonoVet®, calibragem D (TVD), 6,30 ± 1,37 mmHg para animais jovens e 5,00 ± 4,82 mmHg para adultos com TonoVet®, calibragem P (TVP), 25,00 ± 1,41 mmHg para animais jovens e 15,00 ± 2,14 para adultos com TonoVet® Plus, calibragem dog (TVPD); 19,50 ± 0,71 para tonometria de aplanação (TonoPen® XL) para animais jovens e 9,75 ± 4,46 para adultos. Para lobo-guará, os valores mĂ©dios ± desvio padrĂŁo foram de 12,30 ± 7,89 mm/min para TLS de animais adultos e de 4,33 ± 2,16 mm/min para animais idosos; 22, 79 ± 1,63 mm para FP; tonometria de rebote: 11,00 ± 2,77 mmHg (TVD), 6,78 ± 2,58 mmHg (TVP) e 18,29 ± 3,47 mmHg (TVPD). Para quati, TLS: 1 ± 0,89 mm/min para jovens e 4,21 ± 2,42 mm/min para adultos; FP: 9,66 ± 0,77 mm para jovens e 12,50 ± 0,95 mm para adultos; tonometria de rebote: 9,50 ± 2,43 mmHg (TVP), 13,06 ± 1,73 mmHg (TVD, machos), 16,35 ± 2,85 mmHg (TVD, fĂŞmeas), 16,69 ± 2,57 mmHg (TVPD, machos), 21,50 ± 3,84 mmHg (TVPD, fĂŞmeas); tonometria de aplanação: 18,66 ± 4,16 mmHg. Os ângulos iridocorneanos de cachorro-do-mato e de lobo-guará sĂŁo abertos, com malha trabecular fina e espaçada. Os fundos de olhos de todos os animais estudados sĂŁo similares aos dos demais mamĂferos, tapetais e holangiĂłticos. Os valores de referĂŞncia obtidos divergem dos estabelecidos para espĂ©cies domĂ©sticas e outras espĂ©cies de carnĂvoros selvagens, justificando a pesquisa e o estabelecimento de padrões espĂ©cie-especĂficos.Coordenação de Aperfeiçoamento de Pessoal de NĂvel Superior (CAPES).This study covers morphologic and physiologic ophthalmic parameters of three carnivore species that occur in brazilian’s Cerrado, the crab-eating fox (Cerdocyon thous), the maned wolf (Chrysocyon brachyurus) and the coati (Nasua nasua). All the data was collected at BrasĂlias’s Zoo (FJZB), contemplating six crab-eating foxes, eight maned wolves and 15 coatis, all under anesthesia. The animals were submitted to ophthalmic evaluation, Schirmer tear test (STT), intraocular pressure (IOP) with rebound or applanation (crab-eating fox and coati only) tonometers, measurement of palpebral fissure (PF), gonioscopy (crab-eating fox and maned wolf), and fundoscopy. The reference values were calculated and tested for significant statistical difference toward age and sex with ANOVA and Tukey tests. For the crab-eating fox, the median ± standard deviation values were 4,3 ± 2,96 mm/min for STT; 18,45 ± 1,5 mm for male PF; 16,45 ± 0,79 mm for female PF; rebound tonometry: 15,50 ± 0,71 mmHg for young animals and 9,50 ± 2,62 mmHg for adults with TonoVet®, D setting (TVD), 6,30 ± 1,37 mmHg for young and 5,00 ± 4,82 mmHg for adult animals with TonoVet®, P setting (TVP), 25,00 ± 1,41 mmHg for young and 15,00 ± 2,14 for adults with TonoVet® Plus, dog setting (TVPD); applanation tonometry: 19,50 ± 0,71 (TonoPen® XL) for young and 9,75 ± 4,46 for adult animals. For the maned wolf, the median ± standard deviation values were: STT: 12,30 ± 7,89 mm/min for adult and 4,33 ± 2,16 mm/min for old animals; 22, 79 ± 1,63 mm for PF; rebound tonometry: 11,00 ± 2,77 mmHg (TVD), 6,78 ± 2,58 mmHg (TVP), and 18,29 ± 3,47 mmHg (TVPD). For coati, TLS: 1 ± 0,89 mm/min for young and 4,21 ± 2,42 mm/min for adult animals; FP: 9,66 ± 0,77 mm for young and 12,50 ± 0,95 mm for adult animals; rebound tonometry: 14,6 ± 2,82 mmHg (TVP), 13,06 ± 1,73 mmHg (TVD, males), 16,35 ± 2,85 mmHg (TVD, females), 16,69 ± 2,57 mmHg (TVPD, males), 21,50 ± 3,84 mmHg (TVPD, females); applanation tonometry: 18,66 ± 4,16 mmHg. The iridocorneal angles for crab-eating foxes and maned wolves are open, with a thin, spaced, trabecular mesh. The fundus of the subjects of this study were very similar to the ones in other mammals, holangiotic and with tapetum. The reference values found in this study diverge from the ones stablished for domestic species, as well as from the ones for other wild carnivores, justifying the research and establishment of species-specific standards
Valores de referência para selecionados testes oftálmicos para a arara canindé (Ara araruna)
Objetivou-se determinar os valores normais para testes oftálmicos diagnósticos selecionados para a Arara Canindé. Trinta e cinco Ara ararauna (70 olhos), de sexo indeterminado, adultas, com peso médio de 1kg e provenientes de cativeiro no Distrito Federal, foram avaliadas. Aferiram-se a produção lacrimal pelo Teste lacrimal de Schirmer (TLS), a avaliação microbiológica da conjuntiva ocular, a pressão intra-ocular (PIO) utilizando a tonometria de rebote e o comprimento horizontal da rima palpebral Neste estudo 84.1% das amostras analisadas foram positivas para crescimento microbiológico. Bactérias, fungos e hifas foram isolados e Staphylococcus spp. (21.9%) e Bacillus spp. (26.8%) foram isolados mais frequentemente. Os valores médios do teste de Lacrimal de Schirmer (TLS) foram de 7.6±4.6 e 6.6±4.4mm/min para olhos direito (OD) e esquerdo (OE), respectivamente (média = 7,11±0,76mm/min). A pressão intraocular média foi de 11.4±2.5 (OD) e 11.6±1.8mmHg (OE) anteriormente à anestesia (média 11,49±0,22 mmHg) e 7.6 ± 2.4 mmHg (OD) e 7.8 ± 1.8 mm Hg (OE) (média 7,71±0,08mm Hg) após a anestesia, verificando-se que a PIO foi significativamente menor quando os animais se encontravam sob anestesia comparativamente ao momento em que não estavam anestesiados. O comprimento horizontal da rima horizontal palpebral foi de 11.7±0.1mm OD e de 11.8±0.1mm OE (média 11,72±0,07mm). Verificou-se correlação positiva do TLS com o comprimento da fissura palpebral para a espécie estudada. Estes valores de referencia serão úteis no diagnóstico de alterações oculares da Arara Canindé.The aim of this study was to establish reference values for selected ophthalmic diagnostic tests in healthy blue-and-yellow macaws. We investigated a total of 35 adult macaws (70 eyes) of undetermined sex and with an average weight of 1 kg, who were living in captivity in the Federal District, Brazil. Tear production using the Schirmer tear test (STT), normal conjunctival flora, intraocular pressure (IOP) using a rebound tonometer and horizontal palpebral fissure length (HPFL) were evaluated. In this study, 84.1% of samples were positive for microbial growth. Bacteria, fungi and yeasts were isolated, and Staphylococcus spp. (21.9%) and Bacillus spp. (26.8%) were the most frequently isolated microorganisms. The mean value for STT was 7.6±4.6mm/min in the right eye (OD) and 6.6±4.4mm/min in the left eye (OS) (median = 7,11±0,76mm/min). Mean IOP was 11.4±2.5mm Hg OD and 11.6±1.8mm Hg OS (median = 11.49±0.22mm Hg), prior to anesthesia, and 7.6±2.4mm Hg OD and 7.8±1.8mm Hg OS (median 7.71±0.08mmHg) after anesthesia. The IOP was significantly lower when the animals were under anesthesia as compared to when they were conscious (p≤0.05). Horizontal palpebral fissure length was 11.7±0.1mm OD and 11.8±0.1mm OS (median = 11.72±0.07mm). The STT showed a positive correlation with palpebral fissure measurement for this species. These selected ophthalmic reference values will be particularly useful in diagnosing pathological changes in the eyes of blue-and-yellow macaws
Angiotensin Converting Enzyme Regulates Cell Proliferation and Migration
Background The angiotensin-I converting enzyme (ACE) plays a central role in the renin-angiotensin system, acting by converting the hormone angiotensin-I to the active peptide angiotensin-II (Ang-II). More recently, ACE was shown to act as a receptor for Ang-II, and its expression level was demonstrated to be higher in melanoma cells compared to their normal counterparts. However, the function that ACE plays as an Ang-II receptor in melanoma cells has not been defined yet. Aim Therefore, our aim was to examine the role of ACE in tumor cell proliferation and migration. Results We found that upon binding to ACE, Ang-II internalizes with a faster onset compared to the binding of Ang-II to its classical AT1 receptor. We also found that the complex Ang-II/ACE translocates to the nucleus, through a clathrin-mediated process, triggering a transient nuclear Ca2+ signal. In silico studies revealed a possible interaction site between ACE and phospholipase C (PLC), and experimental results in CHO cells, demonstrated that the beta 3 isoform of PLC is the one involved in the Ca2+ signals induced by Ang-II/ACE interaction. Further studies in melanoma cells (TM-5) showed that Ang-II induced cell proliferation through ACE activation, an event that could be inhibited either by ACE inhibitor (Lisinopril) or by the silencing of ACE. In addition, we found that stimulation of ACE by Ang-II caused the melanoma cells to migrate, at least in part due to decreased vinculin expression, a focal adhesion structural protein. Carcinoma simples tubular mamário em Rattus Novergicus/ Mammary tubular simple carcinoma in Rattus Novergicus – case report
Tumores de origem na glândula mamária em ratas sĂŁo frequentes, sendo o carcinoma mais prevalente. O carcinoma mamário Ă© caracterizado pelo comportamento maligno, com relatos de metástase pulmonar em diversas espĂ©cies, e de prognĂłstico reservado. A excisĂŁo tumoral Ă© o mais indicado para fins terapĂŞuticos e diagnĂłsticos, por permitir o exame histopatolĂłgico. Relata-se um caso de carcinoma mamário em uma fĂŞmea twister (Rattus novergicus), com 30 meses de idade, atendida em uma Universidade de BrasĂlia, apresentando dois nĂłdulos em glândulas mamárias, removidos cirurgicamente. Em posterior exame histopatolĂłgico, constatou-se carcinoma simples tubular mamário. A intervenção cirĂşrgica lhe garantiu qualidade de vida e a paciente veio a Ăłbito apĂłs 6 meses, apresentando crescimento de nĂłdulo em regiĂŁo torácica direita. Ozonização de grãos de milheto pérola (Pennisetum glaucum) com foco na segurança do alimento
A ozonização gasosa aplicada Ă descontaminação microbiolĂłgica e quĂmica em grĂŁos de cereais tem grande potencial de aplicação na indĂşstria de armazenamento, transporte e processamento, aumentando assim a segurança no consumo dos produtos derivados dessas matĂ©rias-primas. O presente estudo teve como objetivo avaliar a influĂŞncia do gás ozĂ´nio nas caracterĂsticas fĂsico-quĂmicas e no potencial de germinação de grĂŁos de milheto pĂ©rola, quando estes sĂŁo submetidos a diferentes condições de exposição ao O3 de forma que nĂŁo afete a qualidade tecnolĂłgica dos grĂŁos. Os tratamentos aplicados no milheto nĂŁo causaram diferença significativa (p> 0,05) nos parâmetros fĂsico-quĂmicos (umidade, acidez e cor instrumental), somente uma amostra ozonizada diferiu da amostra controle (p< 0,05) quanto a coloração. No teste de germinação, verificou-se que o tratamento controle nĂŁo diferiu dos tratamentos P1, P2 e P3 (p> 0,05) e os tratamentos P4, P5 e P6, diferiram do tratamento controle (p <0,05). Assim, nas condições avaliadas neste estudo, Ă© recomendado a ozonização de milheto em concentrações de atĂ© 400 mg de O3/Kg, nĂŁo afetando a qualidade dos grĂŁos
Pervasive gaps in Amazonian ecological research
Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4
While the increasing availability of global databases on ecological communities has advanced our knowledge
of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In
the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of
Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus
crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced
environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian
Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by
2050. This means that unless we take immediate action, we will not be able to establish their current status,
