    Impacts of in vivo and in vitro exposures to tamoxifen: comparative effects on human cells and marine organisms

    Tamoxifen (TAM) is a first generation-SERM administered for hormone receptor-positive (HER+) breast cancer in both pre- and post-menopausal patients and may undergo metabolic activation in organisms that share similar receptors and thus face comparable mechanisms of response. The present study aimed to assess whether environmental trace concentrations of TAM are bioavailable to the filter feeder M. galloprovincialis (100 ng L-1) and to the deposit feeder N. diversicolor (0.5, 10, 25 and 100 ng L-1) after 14 days of exposure. Behavioural impairment (burrowing kinetic), neurotoxicity (AChE activity), endocrine disruption by alkali-labile phosphate (ALP) content, oxidative stress (SOD, CAT, GPXs activities), biotransformation (GST activity), oxidative damage (LPO) and genotoxicity (DNA damage) were assessed. Moreover, this study also pertained to compare TAM cytotoxicity effects to mussels and targeted human (i.e. immortalized retinal pigment epithelium - RPE; and human transformed endothelial cells - HeLa) cell lines, in a range of concentrations from 0.5 ng L-1 to 50 μg L-1. In polychaetes N. diversicolor, TAM exerted remarkable oxidative stress and damage at the lowest concentration (0.5 ng L-1), whereas significant genotoxicity was reported at the highest exposure level (100 ng L-1). In mussels M. galloprovincialis, 100 ng L-1 TAM caused endocrine disruption in males, neurotoxicity, and an induction in GST activity and LPO byproducts in gills, corroborating in genotoxicity over the exposure days. Although cytotoxicity assays conducted with mussel haemocytes following in vivo exposure was not effective, in vitro exposure showed to be a feasible alternative, with comparable sensitivity to human cell line (HeLa).info:eu-repo/semantics/publishedVersio

    EURISWEB – Web-based epidemiological surveillance of antibiotic-resistant pneumococci in Day Care Centers

    BACKGROUND: EURIS (European Resistance Intervention Study) was launched as a multinational study in September of 2000 to identify the multitude of complex risk factors that contribute to the high carriage rate of drug resistant Streptococcus pneumoniae strains in children attending Day Care Centers in several European countries. Access to the very large number of data required the development of a web-based infrastructure – EURISWEB – that includes a relational online database, coupled with a query system for data retrieval, and allows integrative storage of demographic, clinical and molecular biology data generated in EURIS. METHODS: All components of the system were developed using open source programming tools: data storage management was supported by PostgreSQL, and the hypertext preprocessor to generate the web pages was implemented using PHP. The query system is based on a software agent running in the background specifically developed for EURIS. RESULTS: The website currently contains data related to 13,500 nasopharyngeal samples and over one million measures taken from 5,250 individual children, as well as over one thousand pre-made and user-made queries aggregated into several reports, approximately. It is presently in use by participating researchers from three countries (Iceland, Portugal and Sweden). CONCLUSION: An operational model centered on a PHP engine builds the interface between the user and the database automatically, allowing an easy maintenance of the system. The query system is also sufficiently adaptable to allow the integration of several advanced data analysis procedures far more demanding than simple queries, eventually including artificial intelligence predictive models

    Evaluation of Jackknife and Bootstrap for Defining Confidence Intervals for Pairwise Agreement Measures

    Several research fields frequently deal with the analysis of diverse classification results of the same entities. This should imply an objective detection of overlaps and divergences between the formed clusters. The congruence between classifications can be quantified by clustering agreement measures, including pairwise agreement measures. Several measures have been proposed and the importance of obtaining confidence intervals for the point estimate in the comparison of these measures has been highlighted. A broad range of methods can be used for the estimation of confidence intervals. However, evidence is lacking about what are the appropriate methods for the calculation of confidence intervals for most clustering agreement measures. Here we evaluate the resampling techniques of bootstrap and jackknife for the calculation of the confidence intervals for clustering agreement measures. Contrary to what has been shown for some statistics, simulations showed that the jackknife performs better than the bootstrap at accurately estimating confidence intervals for pairwise agreement measures, especially when the agreement between partitions is low. The coverage of the jackknife confidence interval is robust to changes in cluster number and cluster size distribution

    No período de agosto/86 a fevereiro/87, três pacientes foram tratados com VP-16-1800mg/m2, em infusão única em 26 horas no dia -6, seguida por Ciclo fosfamida (EDX) 50mg/kg/d nos dias -4 e -3. Irradiação corpórea total (ICT) 1000 cGy em 5 frações de 200cG y nos dias -2, -1 e 0. O transplante de medula óssea (TMO) foi realizado no dia O utilizando doadores HLA compatíveis. O estudo incluiu três pacientes: um paciente portador de Leucemia Linfoblástica Aguda (LLA) em 1.ª recaída medular; um paciente portador de Leucemia Mielóide Crônica (LMC) em 2.ª fase crônica e um paciente portador de LLA em 2.ª remissão. A idade variou entre quatro e 29 anos. A principal toxicidade apresentada por este regime foi mucosite severa que ocorreu nos três pacientes. Todos os três pacientes apresentaram ulcerações orais e um deles apresentou diarréia hemorrágica com volume superiora dois litros. Em todos os pacientes o quadro se resolveu nas três semanas que se seguiram ao TMO. Um paciente apresentou alterações eletrocardiográficas significativas sem, entretanto, ocorrerem manifestações clínicas de cardiotoxicidade. A eficácia do regime pode ser testada no paciente em recaída que permanece em remissão 210 dias pós-TMO. No paciente portador de LMC houve eliminação do cromossomo Ph1. Concluímos que VP-16/EDX/ICT é um regime eficaz e bem tolerado no tratamento de neoplasias hematológicas

The magnetization reversal mechanism depends upon the orientation of the cooling field with respect to the twinned microstructure of the AF, and whether the applied field is increased to (or decreased from) a positive saturating field; i.e., the magnetization reversal is asymmetric. The reversal of the sample magnetization from one saturated state to the other occurs via either domain wall motion or magnetization rotation on opposite sides of the same hysteresis loop

    Clear-PEM: A PET imaging system dedicated to breast cancer diagnostics

    The Clear-PEM scanner for positron emission mammography under development is described. The detector is based on pixelized LYSO crystals optically coupled to avalanche photodiodes and readout by a fast low-noise electronic system. A dedicated digital trigger (TGR) and data acquisition (DAQ) system is used for on-line selection of coincidence events with high efficiency, large bandwidth and small dead-time. A specialized gantry allows to perform exams of the breast and of the axilla.     Cross-referencing unidentified fish sound data sets to unravel sound sources: a case study from the Temperate Northern Atlantic

    There is growing evidence that studying aquatic acoustic communities can provide ecologically relevant information. Understanding these communities may offer unique insights into species behaviour and ecology, while consolidating passive acoustic monitoring as a tool for mapping the presence of target species or estimating changes in aquatic biodiversity. Fish can be significant soundscape contributors, but most soniferous fish species are yet to be identified. Here, we crossed information of three key fish acoustic communities in the Lusitanian Province of the Temperate Northern Atlantic (the Madeira archipelago, the Azores archipelago and Arrábida in mainland Portugal) to unveil potential sources of unidentified fish sounds. We found that the three communities shared various sound types and we were able to narrow down the list of possible fish sound sources. Several sound types were suggested to be produced by species of the Pomacentridae, Scorpaenidae and Serranidae families. We also observed that the sound type /kwa/, associated with Scorpaena spp., exhibited more variations in the geographic area where more species of this genus are known to be present. This study showcases that, as databases of unidentified fish sounds continue to grow, future comparisons of multiple acoustic communities may provide insights into unknown fish sound sources and sound types

    Is the meiofauna a good indicator for climate change and anthropogenic impacts?

    Our planet is changing, and one of the most pressing challenges facing the scientific community revolves around understanding how ecological communities respond to global changes. From coastal to deep-sea ecosystems, ecologists are exploring new areas of research to find model organisms that help predict the future of life on our planet. Among the different categories of organisms, meiofauna offer several advantages for the study of marine benthic ecosystems. This paper reviews the advances in the study of meiofauna with regard to climate change and anthropogenic impacts. Four taxonomic groups are valuable for predicting global changes: foraminifers (especially calcareous forms), nematodes, copepods and ostracods. Environmental variables are fundamental in the interpretation of meiofaunal patterns and multistressor experiments are more informative than single stressor ones, revealing complex ecological and biological interactions. Global change has a general negative effect on meiofauna, with important consequences on benthic food webs. However, some meiofaunal species can be favoured by the extreme conditions induced by global change, as they can exhibit remarkable physiological adaptations. This review highlights the need to incorporate studies on taxonomy, genetics and function of meiofaunal taxa into global change impact research
