    A silicon carbide reinforced aluminum (SiC/Al) material fabricated by Dural Aluminum Composites Corporation was tested for various components of rotary engines. Properties investigated included hardness, high temperature strength, wear resistance, fatigue resistance, thermal conductivity, and expansion. SiC/Al appears to be a viable candidate for cast rotors, and may be applicable to other components, primarily housings

    After Treatment Decrease of Bone Marrow Tregs and Outcome in Younger Patients with Newly Diagnosed Acute Myeloid Leukemia

    An emerging body of evidence demonstrates that defects in antileukemic effector cells in patients with acute myeloid leukemia (AML) can contribute to the development and/or persistence of the disease. In particular, immune suppressive regulatory T cells (Tregs) may contribute to this defective antileukemic immune response, being recruited by bone marrow leukemic cells to evade immune surveillance. We evaluated Tregs (CD4+/CD45RA-/CD25high/CD127low), performing multiparametric flow cytometry on freshly collected bone marrow aspirate (BMA), in addition to the usual molecular and cytogenetic work-up in newly diagnosed AML patients to look for any correlation between Tregs and the overall response rate (ORR). We studied 39 AML younger patients (<65 years), all treated with standard induction chemotherapy. ORR (complete remission (CR)+CR with incomplete hematologic recovery (CRi)) was documented in 21 out of 39 patients (54%); two partial responder patients were also recorded. Apart from the expected impact of the molecular-cytogenetic group (p=0.03) and the NPM mutation (p=0.05), diagnostic BMA Tregs did not show any correlation with ORR. However, although BMA Tregs did not differ in the study population after treatment, their counts significantly decreased in responder patients (p=0.039), while no difference was documented in nonresponder ones. This suggested that the removal of Treg cells is able to evoke and enhance anti-AML immune response. However, the role of BMA Tregs in mediating immune system-AML interactions in the diagnostic and posttreatment phase should be confirmed in a greater number of patients

    Immunohistochemical and molecular detection of equine herpesvirus 1 in Uruguay

    Equine herpesvirus 1 (EHV-1) is a major cause of epidemic abortion, neonatal mortality, respiratory disease and neurological disorders in horses. In South America, the virus has been isolated in Brazil, Argentina and Colombia. In Chile pathological findings from one aborted foetus have been reported, and in Uruguay only serological data about EHV-1 activity have been found. Some pathological findings were reported in Uruguay several years ago, but these data have never been officially confirmed. The present work describes the relevant findings of a study of EHV-1 infections in the Uruguayan equine population using polymerase chain reaction (PCR) and histological and immunohistochemical analysis techniques. The sequence analysis of a portion of the glycoprotein C gene amplified by PCR confirmed EHV-1 activity. The real-time PCR revealed the association of the virus with the non-neuropathogenic genotype. This study describes for the first time the immunohistochemical and molecular detection of EHV-1 in Uruguay.Facultad de Ciencias Veterinaria

    Revealing the coral habitat effect on benthopelagic fauna diversity in the Santa Maria di Leuca cold-water coral province using different devices and Bayesian hierarchical modelling

    Data from two experimental longline surveys and two video inspections carried out in Santa Maria di Leuca cold-water coral province (Mediterranean Sea) during spring-autumn 2010 were used in order to compare the benthopelagic abundance and diversity between coral and non-coral habitats and between different devices. The sampling was carried out in two types of habitat: a coral habitat with carbonate mounds and a non-coral habitat characterized by intermound sea floor. A Bayesian hierarchical modelling approach to accommodate factors influencing community assemblages was used considering the number of species, the Shannon-Wiener diversity index and the two most abundant species represented by the European conger (Conger conger) and blackbelly rosefish (Helicolenus dactylopterus). A relevant effect of the habitat factor was observed for both the number of species and the diversity index, showing a higher species number and diversity index in the coral habitat than in the non-coral habitat. Concerning the relevance of fixed effects from the model on the probability of observing non-zero (positive) abundances, the devices considered, longline and baited lander, did not show different influence for either C. conger or H. dactylopterus. In the case of positive abundance, a relevant device effect was only observed for H. dactylopterus, showing higher abundances for longline than for baited lander. A habitat effect was detected, with positive abundances for both species in the coral habitat. This study proves that structurally complex habitats generated by cold-water corals influence the distribution and diversity of the benthopelagic fauna, and that the use of different devices can provide complementary useful results. Increased knowledge about the role of cold-water corals in the associated benthopelagic fauna could lead to better conservation of one of the most important hot spots of biodiversity in the Mediterranean Sea

    ADAMTS2 gene dysregulation in T/myeloid mixed phenotype acute leukemia.

    Background: Mixed phenotype acute leukemias (MPAL) include acute leukemias with blasts that express antigens of more than one lineage, with no clear evidence of myeloid or lymphoid lineage differentiation. T/myeloid (T/My) MPAL not otherwise specified (NOS) is a rare leukemia that expresses both T and myeloid antigens, accounting for less than 1% of all leukemias but 89% of T/My MPAL. From a molecular point of view, very limited data are available on T/My MPAL NOS. Case presentation: In this report we describe a T/My MPAL NOS case with a complex rearrangement involving chromosomes 5 and 14, resulting in overexpression of the ADAM metallopeptidase with thrombospondin type 1 motif, 2 (ADAMTS2) gene due to its juxtaposition to the T cell receptor delta (TRD) gene segment. Conclusion: Detailed molecular cytogenetic characterization of the complex rearrangement in the reported T/My MPAL case allowed us to observe ADAMTS2 gene overexpression, identifying a molecular marker that may be useful for monitoring minimal residual disease. To our knowledge, this is the first evidence of gene dysregulation due to a chromosomal rearrangement in T/My MPAL NOS.     Imaging the structural style of an active normal fault through multidisciplinary geophysical investigation: a case study from the Mw 6.1, 2009 L'Aquila earthquake region (central Italy)

    The normal fault-system responsible of the 2009 Mw 6.1 L'Aquila earthquake (Paganica-San Demetrio fault-system) comprises several narrow, fault-parallel valleys of controversial origin. We investigated a key section of the southeastern portion of this fault network along the small Verupola Valley. In order to characterize its nature and possible tectonic activity, we applied multiple-geosciences techniques able to image at depth the structure associated to this peculiar landform. We integrated magnetometry, 2-D P wave and resistivity tomography, surface waves and seismic noise analysis coupled with field mapping, shallow boreholes and trenching. According to our results, the Verupola Valley is a ∼30–40-m-deep graben controlled by a SW-dipping master fault and synthetic splays paired with an antithetic NE-dipping fault. The SW-dipping splays are active and cut very shallow (<2 m deep) Late Pleistocene sediments. The small amount of cumulated vertical offset (∼15 m) across the conjugated system may indicate a young fault inception or very low Quaternary slip-rates. Due to its structural continuity with the adjacent mapped strands of the Paganica–San Demetrio fault network, we relate the Verupola Valley to the recent activity of the southeastern segment of this fault system. We also suggest that other fault-parallel valleys can have the same tectonic origin and setting of the Verupola Valley. This latter represents a scale-independent analogue from metric scale (exposed in the palaeoseismological trenches) to the Middle Aterno Basin scale (seen from seismic profiles and fault mapping). Overall, the imaged structural style is coherent with the regional tectonic setting due to Quaternary crustal extension

    Macro- and megafauna recorded in the submarine Bari Canyon (southern Adriatic, Mediterranean Sea) using different tools

    Macro- and megafauna were recorded in the submarine Bari Canyon (southern Adriatic Sea, Mediterranean Sea) during an oceanographic cruise carried out in May-June 2012 and an experimental fishing survey conducted in November 2013. During the former, a total of 20 benthic samples were taken using a Van Veen grab at depths between 268 and 770 m and 4 deployments of a baited lander, for about 43 hours of video records, were carried out at depths between 443 and 788 m. During the latter, 8 longline fishing operations were conducted from 338 down to 612 m. Eighty-five living benthic and benthopelagic species were recorded: 29 Porifera, 1 Cnidaria, 2 Mollusca, 11 Annelida, 1 Arthropoda, 19 Bryozoa, 3 Echinodermata and 19 Chordata. A total of 51 species are new records for the Bari Canyon, 29 new records for the Adriatic Sea. Among the Porifera Cerbaris curvispiculifer is a new addition for the Italian Sponge Fauna. The first certain record of living specimens for the bryozoan Crisia tenella longinodata is reported. A total of 6 Mediterranean endemic species have been identified: 4 Porifera and 2 Annelida. The bathymetric range of some species has been extended. New information acquired for deep sea species confirms their importance in the structure of cold-water coral communities. This study has updated the knowledge on the biodiversity of the Adriatic Sea, as well as of the Bari Canyon in particular, one of the sites designated as “jewels of the Mediterranean” for which urgent conservation measures are needed

    Pulsed Photoconductive Connected Slot Array Operating at the Sub-mm Wavelength Band

    A novel pulsed photoconductive THz source is presented that is able to radiate mW-level average powers, over a large bandwidth by exploiting both the optical and electrical properties of photoconductive sources and the ultrawideband properties of connected antenna arrays. An optical system composed of a micro-lenses array splits the laser beam into N x N spots that host the active excitation of the antenna arrays. An “ad hoc” network has been adopted to bias the array active spots in order to implement a connected antenna array configuration. The array feeds a silicon lens to increase the directivity of the radiated THz beam. A slot array prototype has been designed, fabricated, and measured. The proposed solutions achieve excellent power radiation levels by making use of an accurate electromagnetic design. This solution can offer enhancements to any active system relying on pulsed photoconductive antennas

    The Impact of Graft CD3 Cell/Regulatory T Cell Ratio on Acute Graft-versus-Host Disease and Post-Transplantation Outcome: A Prospective Multicenter Study of Patients with Acute Leukemia Undergoing Allogeneic Peripheral Blood Stem Cell Transplantation

    Although it is well known that tumor site- or bone marrow-infiltrating regulatory T cells (Tregs) might be correlated with worse outcomes in solid tumors and acute leukemias by promoting immune surveillance escape, their contribution to the immediate post-allogeneic transplantation phase by peripheral blood (PB) allografts remains unclear. Moreover, the Treg content in stem cells harvested from PB has been suggested to be correlated with acute graft versus-host-disease (aGVHD) and immunologic recovery after allogeneic PB stem cell transplantation (allo-PBSCT). This study aimed to investigate the impact of the graft content of Tregs, as graft CD3+/Tregs ratio (gCD3/TregsR), on acute GVHD and post-allo-PBSCT outcomes. We prospectively enrolled 94 consecutive patients at 9 Italian centers of the Gruppo Italiano Trapianto di Midollo Osseo (GITMO) with acute myelogenous (n = 71; 75%) or lymphoblastic (n = 23; 25%) leukemia in complete remission who underwent matched related donor (n = 35; 37%) or unrelated donor (n = 59; 63%) allo-PBSCT. The median graft CD3+ cell, Treg, and gCD3/TregsR values were 196 × 106/kg body weight (range, 17 to 666 × 106/kg), 3 × 106/kg (range, 0.1 to 35 × 106/kg), and 71 (range, 1 to 1883), respectively. The discriminatory power of the gCD3/TregsR value to predict grade ≥II aGVHD was assessed by estimating the area under the receiver operating characteristic (ROC) curve (AUC). Any grade and grade ≥II aGVHD occurred in 24 (26%) and 17 (18%) allo-PBSCT recipients, respectively. By ROC analysis, AUC (0.74; 95% confidence interval [CI], 0.608 to 0.866; P =.002) identified 70 as the optimal gCD3/TregsR cutoff value predicting the appearance of grade ≥II aGVHD with 76% sensitivity and 71% specificity. Patients were subdivided into a high (ROC curve value ≥70) gCD3/TregsR group (HR; n = 48) and a low (ROC curve value &lt;70) gCD3/TregsR group (LR; n = 46). The incidence of grade II-IV aGVHD was lower in the LR group compared with the HR group (9% [4 of 46] versus 27% [13 of 48]) in both univariate analysis (odds ratio [OR], 4.8; 95% CI, 1.44 to 16.17; P =.015) and multivariate analysis (OR, 5.0; 95% CI, 1.34 to 18.93; P =.017), whereas no differences were documented taking into account aGVHD of any grade. The overall survival, disease-free survival, nonrelapse mortality, and relapse rates at 2 and 3 years were 61% and 54%, 62% and 55%, 15% and 23%, and 27% and 30%, respectively. Of note, gCD3/TregsR did not significantly correlate with relapse (P =.135). Taken together, our data from this prospective multicenter study confirm the value of Tregs in preventing aGVHD while maintaining the graft-versus-leukemia effect. © 2021 American Society for Transplantation and Cellular Therapy.     The secretome derived from mesenchymal stromal cells cultured in a xeno-free medium promotes human cartilage recovery in vitro

    Osteoarthritis (OA) is a disabling joint disorder causing articular cartilage degeneration. Currently, the treatments are mainly aimed to pain and symptoms relief, rather than disease amelioration. Human bone marrow stromal cells (hBMSCs) have emerged as a promising paracrine mechanism-based tool for OA treatment. Here, we investigate the therapeutic potential of conditioned media (CM) and extracellular vesicles (EVs) isolated from hBMSC and grown in a xeno-free culture system (XFS) compared to the conventional fetal bovine serum-culture system (FBS) in an in vitro model of OA. First, we observed that XFS promoted growth and viability of hBMSCs compared to FBS-containing medium while preserving their typical phenotype. The biological effects of the CM derived from hBMSC cultivated in XFS-and FBS-based medium were tested on IL-1α treated human chondrocytes, to mimic the OA enviroment. Treatment with CM derived from XFS-cultured hBMSC inhibited IL-1α-induced expression of IL-6, IL-8, and COX-2 by hACs compared to FBS-based condition. Furthermore, we observed that hBMSCs grown in XFS produced a higher amount of EVs compared to FBS-culture. The hBMSC-EVs not only inhibit the adverse effects of IL-1α-induced inflammation, but play a significant in vitro chondroprotective effect. In conclusion, the XFS medium was found to be suitable for isolation and expansion of hBMSCs with increased safety profile and intended for ready-to-use clinical therapies
