Expression of Microbial Enzymes in Mammalian Astrocytes to Modulate Lactate Release
Astrocytes support and modulate neuronal activity through the release of L-lactate. The
suggested roles of astrocytic lactate in the brain encompass an expanding range of vital functions,
including central control of respiration and cardiovascular performance, learning, memory, executive
behaviour and regulation of mood. Studying the effects of astrocytic lactate requires tools that limit
the release of lactate selectively from astrocytes. Here, we report the validation in vitro of novel
molecular constructs derived from enzymes originally found in bacteria, that when expressed in
astrocytes, interfere with lactate handling. When lactate 2-monooxygenase derived from M. smegmatis
was specifically expressed in astrocytes, it reduced intracellular lactate pools as well as lactate release
upon stimulation. D-lactate dehydrogenase derived from L. bulgaricus diverts pyruvate towards
D-lactate production and release by astrocytes, which may affect signalling properties of lactate in
the brain. Together with lactate oxidase, which we have previously described, this set of transgenic
tools can be employed to better understand astrocytic lactate release and its role in the regulation of
neuronal activity in different behavioural contexts
Linearized stability analysis of gravastars in noncommutative geometry
In this work, we find exact gravastar solutions in the context of
noncommutative geometry, and explore their physical properties and
characteristics. The energy density of these geometries is a smeared and
particle-like gravitational source, where the mass is diffused throughout a
region of linear dimension due to the intrinsic uncertainty
encoded in the coordinate commutator. These solutions are then matched to an
exterior Schwarzschild spacetime. We further explore the dynamical stability of
the transition layer of these gravastars, for the specific case of
, where M is the black hole mass, to linearized
spherically symmetric radial perturbations about static equilibrium solutions.
It is found that large stability regions exist and, in particular, located
sufficiently close to where the event horizon is expected to form.Comment: 6 pages, 3 figure
Social representations of needlestick injuries
OBJECTIVES: understand the Social Representations about needlestick injuries elaborated by Nursing Technicians and analyze how these representations influence their conducts. METHOD: the data, obtained by interviews, were processed using ALCESTE software and their analysis was based on Serge Moscovici's Social Representations Theory. RESULTS: it was evidenced that, after the accident, these professionals take care of the affected area. Then, they report the accident, motivated by the fear of catching HIV and hepatitis. The different feelings experienced are due to this fear and the way they were forwarded by the institution, reflecting in the cause they attribute to their accident. CONCLUSIONS: it was verified that knowledge about the accident as a whole is very incipient in this professional group, demanding continuing education and greater emphasis on this subject in professional training. It is expected that this study draws public authorities and health institutions' attention to the problem and that it modifies Nursing Technicians' Social Representations about percutaneous exposure.OBJETIVOS: Comprender las Representaciones Sociales de los accidentes con materiales corto-punzantes, elaboradas por Técnicos de Enfermería, así como analizar de que modo esas representaciones influencian las conductas de eses profesionales. MÉTODO: Los datos, logrados por medio de entrevistas, fueron procesados por el software ALCESTE y el análisis de los mismos fue basado en la Teoría de las Representaciones Sociales de Serge Moscovici. RESULTADOS: se evidenció que, después de accidentarse, los Técnicos de Enfermería cuidan del área afectada. Enseguida, notifican el accidente, motivados por el miedo de que contraigan HIV y hepatitis. Los diversos sentimientos vividos son consecuentes de ese miedo y de la manera como fueron encaminados por la institución, reflejando en la causa que ellos atribuyen a su accidente. CONCLUSIONES: se constató que hay un conocimiento bastante incipiente por parte de esa clase profesional sobre el accidente como un todo, habiendo necesidad de una educación continuada y mayor énfasis en cuanto a ese asunto durante la formación profesional. Se espera que ese estudio llame la atención de los órganos gubernamentales e instituciones de salud sobre el problema y modifique las Representaciones Sociales de la exposición percutánea de los Técnicos de Enfermería.OBJETIVOS: compreender as representações sociais dos acidentes com materiais perfurocortantes, elaboradas por técnicos de enfermagem, bem como analisar de que modo essas representações influenciam as condutas desses profissionais. RESULTADOS: evidenciou-se que, após se acidentarem, os técnicos de enfermagem cuidam da área afetada. Em seguida, notificam o acidente, motivados pelo medo de contraírem HIV e hepatite. Os diversos sentimentos vivenciados são decorrentes desse medo e da forma como foram encaminhados pela instituição, refletindo na causa que eles atribuem ao seu acidente. CONCLUSÕES: constatou-se que há conhecimento bastante incipiente por parte dessa classe profissional sobre o acidente, como um todo, havendo necessidade de educação continuada e maior ênfase quanto a esse assunto, durante a formação profissional. Espera-se que este estudo chame a atenção dos órgãos governamentais e instituições de saúde sobre o problema e modifique as representações sociais da exposição percutânea dos técnicos de enfermagem
Transcriptome Analysis of the Octopus vulgaris Central Nervous System
Background: Cephalopoda are a class of Mollusca species found in all the world's oceans. They are an important model organism in neurobiology. Unfortunately, the lack of neuronal molecular sequences, such as ESTs, transcriptomic or genomic information, has limited the development of molecular neurobiology research in this unique model organism. Results: With high-throughput Illumina Solexa sequencing technology, we have generated 59,859 high quality sequences from 12,918,391 paired-end reads. Using BLASTx/BLASTn, 12,227 contigs have blast hits in the Swissprot, NR protein database and NT nucleotide database with E-value cutoff 1e(-5). The comparison between the Octopus vulgaris central nervous system (CNS) library and the Aplysia californica/Lymnaea stagnalis CNS ESTs library yielded 5.93%/13.45% of O. vulgaris sequences with significant matches (1e(-5)) using BLASTn/tBLASTx. Meanwhile the hit percentage of the recently published Schistocerca gregaria, Tilapia or Hirudo medicinalis CNS library to the O. vulgaris CNS library is 21.03%-46.19%. We constructed the Phylogenetic tree using two genes related to CNS function, Synaptotagmin-7 and Synaptophysin. Lastly, we demonstrated that O. vulgaris may have a vertebrate-like Blood-Brain Barrier based on bioinformatic analysis. Conclusion: This study provides a mass of molecular information that will contribute to further molecular biology research on O. vulgaris.
