    Beliefs About Psychological Problems Inventory (BAPPI) : development and psychometric properties

Thus, it is desirable to include clients’ beliefs about their psychological problems on systematic assessment protocols underlying the process of systematic treatment selection and of tailoring the treatment to the person. However, assessment instruments which specifically capture clients’ beliefs about their psychological problems are scarce. The objective of the studies presented was to evaluate the psychometric properties of the Beliefs About Psychological Problems Inventory (BAPPI), an assessment instrument of the clients’ beliefs about their psychological problems. Study 1 (Exploratory Factor Analysis) involved 200 participants, and Study 2 (Confirmatory Factor Analysis and other validity studies), involved 545 participants. Results revealed that the BAPPI presents a stable factorial structure of six dimensions (Psychodynamic, Humanistic, Biomedical, Cognitive-Behavioral, Systemic, and Eclectic/Integrative). Altogether, analyses of items, internal consistency, reliability, and external validity revealed that the BAPPI is a valid assessment instrument for use in mental health research and practice, especially in the process of systematic treatment selection and, therefore, of matching/tailoring the treatment to the client’s characteristics

    Mapping atmospheric pollutants emissions in European countries

    In this paper we present a methodology which enables the graphical representation, in a bi-dimensional Euclidean space, of atmospheric pollutants emissions in European countries. This approach relies on the use of Multidimensional Unfolding (MDU), an exploratory multivariate data analysis technique. This technique illustrates both the relationships between the emitted gases and the gases and their geographical origins. The main contribution of this work concerns the evaluation of MDU solutions. We use simulated data to define thresholds for the model fitting measures, allowing the MDU output quality evaluation. The quality assessment of the model adjustment is thus carried out as a step before interpretation of the gas types and geographical origins results. The MDU maps analysis generates useful insights, with an immediate substantive result and enables the formulation of hypotheses for further analysis and modeling

    Trust and fake news: Exploratory analysis of the impact of news literacy on the relationship with news content in Portugal

    In order to understand the role of contemporary journalism and the media system it is vital to consider consumers’ relationship with news content in terms of trust and perception of dubious content. This analysis is particularly relevant in a context where intense flows of information raise serious questions about individual ability to interpret, validate, and reproduce content. This analysis explores a news literacy scale used by Maskl et al. (2015) and Fletcher (in Newman et al., 2018) to investigate the links between news literacy profiles and their relationship with content, with particular focus on illegitimate/doubtful news pieces. Results suggest individuals with higher news literacy tend to trust news in general but not when content originates in social media. Higher literacy profiles are also associated with increased concern regarding online content legitimacy. These conclusions are particularly relevant in the currently volatile media sphere, highly dependent on a substantially informed public to ensure the legitimacy and importance of journalistic content and to distinguish it from other kinds of content flooding communication networks.     Activation of the endoplasmic reticulum stress response by the amyloid-beta 1–40 peptide in brain endothelial cells

    Neurovascular dysfunction arising from endothelial cell damage is an early pathogenic event that contributes to the neurodegenerative process occurring in Alzheimer's disease (AD). Since the mechanisms underlying endothelial dysfunction are not fully elucidated, this study was aimed to explore the hypothesis that brain endothelial cell death is induced upon the sustained activation of the endoplasmic reticulum (ER) stress response by amyloid-beta (Aβ) peptide, which deposits in the cerebral vessels in many AD patients and transgenic mice. Incubation of rat brain endothelial cells (RBE4 cell line) with Aβ1–40 increased the levels of several markers of ER stress-induced unfolded protein response (UPR), in a time-dependent manner, and affected the Ca2 + homeostasis due to the release of Ca2 + from this intracellular store. Finally, Aβ1–40 was shown to activate both mitochondria-dependent and -independent apoptotic cell death pathways. Enhanced release of cytochrome c from mitochondria and activation of the downstream caspase-9 were observed in cells treated with Aβ1–40 concomitantly with caspase-12 activation. Furthermore, Aβ1–40 activated the apoptosis effectors' caspase-3 and promoted the translocation of apoptosis-inducing factor (AIF) to the nucleus demonstrating the involvement of caspase-dependent and -independent mechanisms during Aβ-induced endothelial cell death. In conclusion, our data demonstrate that ER stress plays a significant role in Aβ1–40-induced apoptotic cell death in brain endothelial cells suggesting that ER stress-targeted therapeutic strategies might be useful in AD to counteract vascular defects and ultimately neurodegeneration

    Activation of PI3K Is Indispensable for Interleukin 7–mediated Viability, Proliferation, Glucose Use, and Growth of T Cell Acute Lymphoblastic Leukemia Cells

    Interleukin (IL)-7 is essential for normal T cell development. Previously, we have shown that IL-7 increases viability and proliferation of T cell acute lymphoblastic leukemia (T-ALL) cells by up-regulating Bcl-2 and down-regulating the cyclin-dependent kinase inhibitor p27kip1. Here, we examined the signaling pathways via which IL-7 mediates these effects. We investigated mitogen-activated protein kinase (MEK)–extracellular signal-regulated kinase (Erk) and phosphatidylinositol-3-kinase (PI3K)–Akt (protein kinase B) pathways, which have active roles in T cell expansion and have been implicated in tumorigenesis. IL-7 induced activation of the MEK–Erk pathway in T-ALL cells; however, inhibition of the MEK–Erk pathway by the use of the cell-permeable inhibitor PD98059, did not affect IL-7–mediated viability or cell cycle progression of leukemic cells. IL-7 induced PI3K-dependent phosphorylation of Akt and its downstream targets GSK-3, FOXO1, and FOXO3a. PI3K activation was mandatory for IL-7–mediated Bcl-2 up-regulation, p27kip1 down-regulation, Rb hyperphosphorylation, and consequent viability and cell cycle progression of T-ALL cells. PI3K signaling was also required for cell size increase, up-regulation of CD71, expression of the glucose transporter Glut1, uptake of glucose, and maintenance of mitochondrial integrity. Our results implicate PI3K as a major effector of IL-7–induced viability, metabolic activation, growth and proliferation of T-ALL cells, and suggest that PI3K and its downstream effectors may represent molecular targets for therapeutic intervention in T-ALL

    A study of the antitumour potential of three Portuguese wild mushrooms

    Natural matrixes such as mushrooms represent a rich source of biologically active compounds with recognized potential in drug discovery and development [1,2]. Indeed, many pre-clinical studies have been conducted in human tumour cell lines and in some cases, a number of compounds extracted from mushrooms have entered clinical trials [3]. Our previous results showed that extracts from Agaricus arvensis, Suillus collinitus and Clytocibe alexandri are promising sources of low molecular weight bioactive compounds [4]. The aim of the present work was to study the antitumour potential of the extracts and isolated compounds from three Portuguese wild mushrooms by verifying their effect on various human tumour cell lines in what concerns effect on cell growth, cell cycle profile and programmed cell death. Wild mushrooms were collected from the Northeast of Portugal and classified as Agaricus arvensis, Suillus collinitus and Clitocybe alexandri. Phenolic (methanolic and ethanolic) and polissacharidic extracts were prepared. The effect of the extracts on tumour cell growth inhibition was verified with the SRB assay and the GI50 of each extract was determined for each of the cell lines studied (NCI-H460, MCF-7, AGS and HCT-15). Our preliminary results revealed that all the extracts from Clitocybe alexandri are capable of causing cell growth inhibition and provided GI50 concentrations bellow 60µg/ml in all the cell lines tested [4]. Regarding the effect of the Agaricus arvensis extracts, they all caused an inhibition of cell growth in all cell lines, particularly the methanolic extract which revealed to be a very potent inhibitor of cell growth, especially in the MCF-7 cell line. The evaluation of the effect of the Suillus collinitus extracts will be carried out as well as cell cycle and apoptosis analyses, by flow cytometry. Finally, the isolation and characterization of compounds from these extracts will also be carried out, using HPLC-DAD or HPLC-RI.     Stealth magnetoliposomes based on calcium-substituted magnesium ferrite nanoparticles for curcumin transport and release

    Despite the promising pharmacological properties of curcumin, the transport and effective release of curcumin is still a challenge. The advances in functionalized nanocarriers for curcumin have also been motivated by the anticancer activity of this natural compound, aiming at targeted therapies. Here, stealth (aqueous and solid) magnetoliposomes containing calcium-substituted magnesium ferrite nanoparticles, CaxMg1−xFe2O4 (with x = 0.25, 0.50, 0.75) were developed as nanocarriers for curcumin. The magnetic nanoparticles exhibit superparamagnetic properties and crystalline structure, with sizes below 10 nm. The magnetoliposomes based on these nanoparticles have hydrodynamic diameters around or below 150 nm and a low polydispersity. The influence of an alternating magnetic field (AMF) on drug release over time was evaluated and compared with curcumin release by diffusion.
