The 19th International Electronic Conference on Synthetic Organic Chemistry session Ionic LiquidsIonic liquids (ILs) are considered one of the most promising green alternatives to molecular organic solvents. Nevertheless, before a widespread use of these compounds in a determined application, the knowledge of their ecotoxicity and biodegradability must be defined. In this work the effect of addition of different doses of 1-butil-2,3-dimethylimidazolium trifluoromethanesulfonate, [C4C1C1Im][OTf], on microbial activity of a soil under Pinus pinaster Aiton and on the seed germination of species of P. pinaster , Pinus sylvestris L., Pinus radiata D. Don and Eucalyptus globulus Labill were analyzed. Additionally, seed germination test were also applied to this IL after being subjected to heat treatment. A microcalorimeter Thermal Activity Monitor (TAM-III) TA-Instruments was used to determine the influence of the addition of four doses (10 %, 25 %, 50 % and 75 %) on the microbial activity of a Galician soil, under P. pinaster. Methodology was widely exposed in previous works [1]. From the data obtained in these experiments, the calculation of microbial growth was performed [2]. Seed germination test of selected species were carried out for several doses of ILs (10 %, 1 %, 0.1 % and 0.01 %). The degradation treatment of the IL was carried out during 24 h in an oven at the temperature of 200 °C. Five replies with 25 seeds per Petri dish were incubated in a Phytotron (Climas AGP890) for every species and treatments. Seeds were maintained for 16 h under light at 24 °C and in the dark for 8 h at 16 °C during 45 days, when the germination was completed in all the species [3, 4]. Results of soil microbial activity and germination obtained for this IL were compared with the corresponding to other ILs, previously studied in our laboratory. Dose of 10 % inhibited totally the germination of the four species and dose of 1 % provoked an important reduction with regard to the control, both, for degraded and no-degraded IL. Nevertheless, in spite of all the doses showed differences on calorimetric parameters with regard to control; only the dose of 75 % does not show soil microbial response

Optical and electrochemical properties of spirobifluorene iridanaphthalene complexes

Three new spirobifluorene iridaaromatic compounds bearing electron-withdrawing or electron-donor substituents or another iridanaphthalene moiety have been synthesized and structurally characterized. Thorough experimental and theoretical evaluation revealed that these novel systems present a high thermal, air and electrochemical stability as well as low optical and electronic energy gap values with a significant redshift of the absorption maximum in the UV-Vis spectra and predicted remarkably higher first hyperpolarizabilities compared to their organic counterparts. Therefore, the combination of a metallaaromatic system with a spirobifluorene moiety leads to the design and development of new spirobifluorene derivatives. These new systems have shown interesting optical and electronic properties making them of interest for future applications in optoelectronics.Xunta de Galicia | Ref. GRC2019/2

Silver nanoparticles produced by laser ablation and re-irradiation are effective preventing peri-implantitis multispecies biofilm formation

Implant-associated infection due to biofilm formation is a growing problem. Given that silver nanoparticles (Ag-NPs) have shown antibacterial effects, our goal is to study their effect against multispecies biofilm involved in the development of peri-implantitis. To this purpose, Ag-NPs were synthesized by laser ablation in de-ionized water using two different lasers, leading to the production of colloidal suspensions. Subsequently, part of each suspension was subjected to irradiation one and three times with the same laser source with which it was obtained. Ag-NPs were immobilized on the surface of titanium discs and the resultant materials were compared with unmodified titanium coupons. Nanoparticles were physico-chemically analysed to determine their shape, crystallinity, chemical composition, and mean diameter. The materials were incubated for 90 min or 48 h, to evaluate bacterial adhesion or biofilm formation respectively with Staphylococcus aureus or oral mixed bacterial flora composed of Streptococcus oralis, Actinomyces naeslundii, Veionella dispar, and Porphyromonas gingivalis. Ag-NPs help prevent the formation of biofilms both by S. aureus and by mixed oral bacterial flora. Nanoparticles re-irradiated three times showed the biggest antimicrobial effects. Modifying dental implants in this way could prevent the development of peri-implantitis.

ISSN: 0893-133X. eISSN: 1740-634X. DOI:10.1038/npp.2013.280α-Synuclein (α-Syn) appears to play a crucial role in the pathogenesis of several neurodegenerative disorders including Parkinson's disease (PD). The brains of Parkinson patients typically contain insoluble intracellular protein inclusions called Lewy bodies. Increased neuronal α-Syn levels represent a major component of Lewy bodies and therefore, the suppression of α-Syn expression provides a valid therapeutic target for PD. The goal of this study was to assess the ability of various small interfering RNA (siRNA) and antisense oligonucleotide (ASO) sequences directed against α-Syn to downregulate endogenous or overexpressed α-Syn mRNA levels in BE-M17 neuroblastoma cells. Moreover, we evaluated the feasibility of reducing α-Syn expression selectively in PD-vulnerable brain areas including substantia nigra pars compacta (SNc), ventral tegmental area (VTA), locus coeruleus (LC) and dorsal raphe nucleus (DR) of mice after the internalization of conjugated siRNA/ASO molecules into monoamine neurons following intranasal administration. Conclusions: These results set the stage for the testing of these molecules as potential disease-modifying agents in neurotoxin-based and genetic models of PD linked to pathogenic increases in α-Syn. In this study we have characterized conjugated siRNA and ASO molecules that actively reduce endogenous α-Syn expression in vivo using the intranasal route to deliver directly siRNA/ASO into the brainPeer Reviewe

Inherited retinal diseases (IRDs), defined by dysfunction or progressive loss of photoreceptors, are disorders characterized by elevated heterogeneity, both at the clinical and genetic levels. Our main goal was to address the genetic landscape of IRD in the largest cohort of Spanish patients reported to date. A retrospective hospital-based cross-sectional study was carried out on 6089 IRD affected individuals (from 4403 unrelated families), referred for genetic testing from all the Spanish autonomous communities. Clinical, demographic and familiar data were collected from each patient, including family pedigree, age of appearance of visual symptoms, presence of any systemic findings and geographical origin. Genetic studies were performed to the 3951 families with available DNA using different molecular techniques. Overall, 53.2% (2100/3951) of the studied families were genetically characterized, and 1549 different likely causative variants in 142 genes were identified. The most common phenotype encountered is retinitis pigmentosa (RP) (55.6% of families, 2447/4403). The most recurrently mutated genes were PRPH2, ABCA4 and RS1 in autosomal dominant (AD), autosomal recessive (AR) and X-linked (XL) NON-RP cases, respectively; RHO, USH2A and RPGR in AD, AR and XL for non-syndromic RP; and USH2A and MYO7A in syndromic IRD. Pathogenic variants c.3386G > T (p.Arg1129Leu) in ABCA4 and c.2276G > T (p.Cys759Phe) in USH2A were the most frequent variants identified. Our study provides the general landscape for IRD in Spain, reporting the largest cohort ever presented. Our results have important implications for genetic diagnosis, counselling and new therapeutic strategies to both the Spanish population and other related populations. This work was supported by the Instituto de Salud Carlos III (ISCIII) of the Spanish Ministry of Health (FIS; PI16/00425 and PI19/00321), Centro de Investigación Biomédica en Red Enfermedades Raras (CIBERER, 06/07/0036), IIS-FJD BioBank (PT13/0010/0012), Comunidad de Madrid (CAM, RAREGenomics Project, B2017/BMD-3721), European Regional Development Fund (FEDER), the Organización Nacional de Ciegos Españoles (ONCE), Fundación Ramón Areces, Fundación Conchita Rábago and the University Chair UAM-IIS-FJD of Genomic Medicine.
