    Detection of androgenic anabolic steroids (AAS) in urine samples by gas chromatography-mass spectrometry

    EsterĂłides androgĂȘnicos anabĂłlicos (EAA) sĂŁo substĂąncias naturais, sintĂ©ticas ou semi-sintĂ©ticas derivadas da testosterona, utilizadas em atividades esportivas com o objetivo de melhorar o desempenho fĂ­sico pelo aumento de massa e força muscular. Apesar de causarem efeitos tĂłxicos graves, principalmente sobre os sistemas cardiovascular, hepĂĄtico e neuro-endĂłcrino, os EAA tĂȘm sido amplamente utilizados no Ăąmbito esportivo. Desta forma, o presente trabalho teve como objetivo a validação de um mĂ©todo para a determinação de esterĂłides e/ou seus produtos de biotransformação em amostras de urina por cromatografia gasosa acoplada Ă  espectrometria de massas (CG-EM). O mĂ©todo baseou-se na hidrĂłlise enzimĂĄtica de esterĂłides conjugados, no emprego da extração lĂ­quido-lĂ­quido e na derivação do extrato para posterior anĂĄlise por CG-EM, no modo de monitoramento seletivo de Ă­ons (SIM). O mĂ©todo validado cumpriu com os princĂ­pios de confiança necessĂĄrios para a determinação de EAA em amostras de urina com a finalidade de controle de dopagem no esporte. Os limites de detecção dos esterĂłides analizados variaram de 0,5 a 15 ng/mL. Bons valores de precisĂŁo (intra e interensaio) tambĂ©m foram obtidos (CV< 11%). Amostras de voluntĂĄrios (n=10) que relataram uso de EAA foram submetidas ao mĂ©todo. Pela anĂĄlise das amostras, foi verificado o uso dos principais esterĂłides utilizados no Ăąmbito esportivo no Brasil.Androgenic anabolic steroids (AAS) are defined as natural, synthetic or semi-synthetic drugs chemically derived from testosterone, used with the aim to improve physical performance by increasing both muscle strength and mass. Despite their reported toxicological effects on the cardiovascular, hepatic and neuro-endocrine systems, the AAS have been extensively used in sports activities. The objective of this study was to validate an analytical method for the detection of AAS and/or their metabolites in urine samples. The method was based on enzymatic hydrolysis, liquid-liquid extraction, derivatization and further analysis by gas chromatography-mass spectrometry (GC-MS) operated in the selected ion monitoring mode (SIM). The validated method agrees with the confidence concepts for AAS detection in urine samples for anti-doping control programs. The limits of detection of AAS varied from 0,5 to 15ng/mL. Good inter and intra-assay precisions were also observed (CV< 11%). Urine samples collected from volunteers (n=10) who reported AAS use were submitted to the method and the most used AAS in Brazil were successfully detected

    Advances in Recombinant Lipases: Production, Engineering, Immobilization and Application in the Pharmaceutical Industry

    Lipases are one of the most used enzymes in the pharmaceutical industry due to their efficiency in organic syntheses, mainly in the production of enantiopure drugs. From an industrial viewpoint, the selection of an efficient expression system and host for recombinant lipase production is highly important. The most used hosts are Escherichia coli and Komagataella phaffii (previously known as Pichia pastoris) and less often reported Bacillus and Aspergillus strains. The use of efficient expression systems to overproduce homologous or heterologous lipases often require the use of strong promoters and the co-expression of chaperones. Protein engineering techniques, including rational design and directed evolution, are the most reported strategies for improving lipase characteristics. Additionally, lipases can be immobilized in different supports that enable improved properties and enzyme reuse. Here, we review approaches for strain and protein engineering, immobilization and the application of lipases in the pharmaceutical industry

    Estudo do comportamento do chumbo em latossolos brasileiros tratados com fosfatos: contribuiçÔes para a remediação de sítios contaminados

    Phosphates have been used for lead immobilization in soils but the influence of soil type is not fully understood. In this work, lead chemical behaviour in two Brazilian latosoils (LA and LV) was studied via treatment with phosphates. The Pb concentration in Toxicity Characteristic Leaching Procedure (TCLP) solutions was decreased in all treatments. After treatment with H3PO4 the Pb concentration in the LA remained within the regulatory limit established by EPA. The ecotoxicological results with Daphnia pulex showed that this treatment reduced the lead bioavailability. Sequential extraction analyses showed that the lead was transferred from the most available to the residual fraction. Relevant decrease of soluble lead was observed in all phosphate treatments

    A single dose of ChAdOx1 Chik vaccine induces neutralising antibodies against four chikungunya virus lineages in a phase 1 clinical trial

    Chikungunya virus (CHIKV) is a reemerging mosquito-borne virus that causes swift outbreaks. Major concerns are the persistent and disabling polyarthralgia in infected individuals. Here we present the results from a first-in-human trial of the candidate simian adenovirus vectored vaccine ChAdOx1 Chik, expressing the CHIKV full-length structural polyprotein (Capsid, E3, E2, 6k and E1). 24 adult healthy volunteers aged 18–50 years, were recruited in a dose escalation, open-label, nonrandomized and uncontrolled phase 1 trial (registry NCT03590392). Participants received a single intramuscular injection of ChAdOx1 Chik at one of the three preestablished dosages and were followed-up for 6 months. The primary objective was to assess safety and tolerability of ChAdOx1 Chik. The secondary objective was to assess the humoral and cellular immunogenicity. ChAdOx1 Chik was safe at all doses tested with no serious adverse reactions reported. The vast majority of solicited adverse events were mild or moderate, and self-limiting in nature. A single dose induced IgG and Tcell responses against the CHIKV structural antigens. Broadly neutralizing antibodies against the four CHIKV lineages were found in all participants and as early as 2 weeks after vaccination. In summary, ChAdOx1 Chik showed excellent safety, tolerability and 100% PRNT50 seroconversion after a single dose

    Fungal pathogens are able to produce inoculum (spores) on plant debris present on the soil surface of vineyards. These spores can then reach plant surfaces and cause severe grapevine infections when environmental conditions are favourable. The capacity of plant diversity to increase the resistance of crops towards pests and invasive species is very well-known. For instance, Brassica spp. have been already investigated for their capacity to effectively suppress soil-borne inoculum of some causal agents of Black-foot disease in grapevines in vineyard soils. It may also have positive effect on the some dagger nematodes. Cover crops also stimulate the development of microbial communities such as arbuscular mycorrhizal fungi. Many management strategies have been developed against these important grapevine pathogens, but the effects of soil cover vegetation or organic mulching against spore dispersal, acting as a barrier, have been scarcely explored. Thus, in the BIOVINE project (www.biovine.eu) specific experiments were planned in order to verify the possibility of using cover crops: i) to control some relevant pathogens producing inoculum (spores) on plant debris present on the soil surface of vineyards; ii) to determine the presence of causal agents of Petri disease of grapevines on the roots of cover crops; iii) to promote mykorrhizal communities associated with grapevine roots; iv) to control arthropod pests (repellent of arthropods or attracting beneficials); v) to investigate Brassica plants effect on the soil-borne pest nematode Xiphinema index

    Imaginal Discs – A New Source of Chromosomes for Genome Mapping of the Yellow Fever Mosquito Aedes aegypti

    Dengue fever is an emerging health threat to as much as half of the human population around the world. No vaccines or drug treatments are currently available. Thus, disease prevention is largely based on efforts to control its major mosquito vector Ae. aegypti. Novel vector control strategies, such as population replacement with pathogen-incompetent transgenic mosquitoes, rely on detailed knowledge of the genome organization for the mosquito. However, the current genome assembly of Ae. aegypti is highly fragmented and requires additional physical mapping onto chromosomes. The absence of readable polytene chromosomes makes genome mapping for this mosquito extremely challenging. In this study, we discovered and investigated a new source of chromosomes useful for the cytogenetic analysis in Ae. aegypti – mitotic chromosomes from imaginal discs of 4th instar larvae. Using natural banding patterns of these chromosomes, we developed a new band-based approach for physical mapping of DNA probes to the precise chromosomal positions. Further application of this approach for genome mapping will greatly enhance the utility of the existing draft genome sequence assembly for Ae. aegypti and thereby facilitate application of advanced genome technologies for investigating and developing novel genetic control strategies for dengue transmission

    Anxiety, concerns and COVID-19: Cross-country perspectives from families and individuals with neurodevelopmental conditions

    BACKGROUND: The COVID-19 pandemic had a major impact on the mental health and well-being of children with neurodevelopmental conditions (NDCs) and of their families worldwide. However, there is insufficient evidence to understand how different factors (e.g., individual, family, country, children) have impacted on anxiety levels of families and their children with NDCs developed over time. METHODS: We used data from a global survey assessing the experience of 8043 families and their children with NDCs (mean of age (m) = 13.18 years, 37% female) and their typically developing siblings (m = 12.9 years, 45% female) in combination with data from the European Centre for Disease Prevention and Control, the University of Oxford, and the Central Intelligence Agency (CIA) World Factbook, to create a multilevel data set. Using stepwise multilevel modelling, we generated child-, family- and country-related factors that may have contributed to the anxiety levels of children with NDCs, their siblings if they had any, and their parents. All data were reported by parents. RESULTS: Our results suggest that parental anxiety was best explained by family-related factors such as concerns about COVID-19 and illness. Children’s anxiety was best explained by child-related factors such as children’s concerns about loss of routine, family conflict, and safety in general, as well as concerns about COVID-19. In addition, anxiety levels were linked to the presence of pre-existing anxiety conditions for both children with NDCs and their parents. CONCLUSIONS: The present study shows that across the globe there was a raise in anxiety levels for both parents and their children with NDCs because of COVID-19 and that country-level factors had little or no impact on explaining differences in this increase, once family and child factors were considered. Our findings also highlight that certain groups of children with NDCs were at higher risk for anxiety than others and had specific concerns. Together, these results show that anxiety of families and their children with NDCs during the COVID-19 pandemic were predicted by very specific concerns and worries which inform the development of future toolkits and policy. Future studies should investigate how country factors can play a protective role during future crises

    TRY plant trait database – enhanced coverage and open access

    Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait‐based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for ‘plant growth form’. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait–environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE Δ4 allele
