    Energetics and development modes of Asteroidea (Echinodermata) from the Southwestern Atlantic Ocean including Burdwood Bank/MPA Namuncurá

    Reproduction is a highly expensive process that during gonadal development requires an important supply of nutrients. The allocation of energy can vary throughout the reproductive cycle, between sexes and development modes. During research cruise aboard the RV Puerto Deseado in April 2016, we collected fifteen species of sea stars from the southernmost region of Argentina. The main purposes of the present study were threefold: first, to revise and report novel information on the reproductive strategies and energetic density (ED) of body components from the asteroids of the southwestern Atlantic Ocean including Burdwood Bank/ MPA Namuncurá; second, to compare the ED of these body components across species with contrasting reproductive strategies; third, to provide a tool to estimate the ED from dry mass of organs of the sea star species examined. Ovaries of Diplopteraster verrucosus (a brooder with a nidamental chamber) revealed a significantly greater ED than did the testes (29.81 ± 1.38 and 17.76 ± 1.59, respectively). In Glabraster antarctica (a broadcaster with yolky eggs and facultative planktotrophic larvae), the gonads had EDs of 25.78 ± 3.16 and 19.21 ± 0.52 (females and males, respectively). While in Peribolaster folliculatus (a broadcaster with eggs with low yolk content and inferred planktotrophic larvae) there was no significant difference in the ED values between sexes (females: 22.79 ± 1.10 and males: 20.46 ± 1.05). For the pyloric caeca, ED values did not reveal any difference between sexes, although in P. folliculatus, the ED was significantly higher than that for D. verrucosus and G. antarctica (25.90 ± 1.00, 23.03 ± 0.34, and 22.66 ± 0.65, respectively). The body wall had higher ED values in D. verrucosus and P. folliculatus than that for all the sea star species analyzed (46.48 ± 1.63, 51.17 ± 1.34, respectively). Higher ED values in the ovaries could be related to the nutrition of offspring, while differences found in the body wall may reflect the skeletal structure of this body component. This research provides basic information for understanding the differences on energetic allocation when contrasting development modes are considered.Fil: Fraysse, Cintia Pamela.     Enhancement of cortical extracellular 5-HT by 5-HT1A and 5-HT2C receptor blockade restores the antidepressant-like effect of citalopram in non-responder mice.

    We recently found that the response of DBA/2 mice to SSRIs in the forced swim test (FST) was impaired and they also had a smaller basal and citalopram-stimulated increase in brain extracellular serotonin (5-HT) than 'responder' strains. We employed intracerebral microdialysis, FST and selective antagonists of 5-HT1A and 5-HT2C receptors to investigate whether enhancing the increase in extracellular 5-HT reinstated the anti-immobility effect of citalopram in the FST. WAY 100635 (0.3 mg/kg s.c.) or SB 242084 (1 mg/kg s.c.), respectively a selective 5-HT1A and 5-HT2C receptor antagonist, raised the effect of citalopram (5 mg/kg) on extracellular 5-HT in the medial prefrontal cortex of DBA/2N mice (citalopram alone 5.2+/-0.3 fmol/20 microl, WAY 100635+citalopram 9.9+/-2.1 fmol/20 microl, SB 242084+ citalopram 7.6+/-1.0 fmol/20 microl) to the level reached in 'responder' mice given citalopram alone. The 5-HT receptor antagonists had no effect on the citalopram-induced increase in extracellular 5-HT in the dorsal hippocampus. The combination of citalopram with WAY 100635 or SB 242084 significantly reduced immobility time in DBA/2N mice that otherwise did not respond to either drug singly. Brain levels of citalopram in mice given citalopram alone or with 5-HT antagonists did not significantly differ. The results confirm that impaired 5-HT transmission accounts for the lack of effect of citalopram in the FST and suggest that enhancing the effect of SSRIs on extracellular 5-HT, through selective blockade of 5-HT1A and 5-HT2C receptors, could be a useful strategy to restore the response in treatment-resistant depression

    Computer simulations of heterologous immunity: Highlights of an interdisciplinary cooperation

    The relationship between biological research and mathematical modeling is complex, critical, and vital. In this review, we summarize the results of the collaboration between two laboratories, exploring the interaction between mathematical modeling and wet-lab immunology. During this collaboration several aspects of the immune defence against viral infections were investigated, focusing primarily on the subject of heterologous immunity. In this manuscript, we emphasize the topics where computational simulations were applied in conjunction with experiments, such as immune attrition, the growing and shrinking of cross-reactive T cell repertoires following repeated infections, the short and long-term effects of cross-reactive immunological memory, and the factors influencing the appearance of new clonal specificities. For each topic, we describe how the mathematical model used was adapted to answer specific biological questions, and we discuss the hypotheses that were generated by simulations. Finally, we propose rules for testing hypotheses that emerge from model experimentation in the wet lab, and vice-versa

    Advances in Therapies and Imaging for Systemic Vasculitis

    Vasculitis is a systemic disease characterized by immune-mediated injury of blood vessels. Current treatments for vasculitis, such as glucocorticoids and alkylating agents, are associated with significant side effects. Furthermore, the management of both small and large vessel vasculitis is challenging because of a lack of robust markers of disease activity. Recent research has advanced our understanding of the pathogenesis of both small and large vessel vasculitis, and this has led to the development of novel biologic therapies capable of targeting key cytokine and cellular effectors of the inflammatory cascade. In parallel, a diverse range of imaging modalities with the potential to monitor vessel inflammation are emerging. Continued expansion of combined structural and molecular imaging using positron emission tomography with computed tomography or magnetic resonance imaging may soon provide reliable longitudinal tracking of vascular inflammation. In addition, the emergence of radiotracers able to assess macrophage activation and immune checkpoint activity represents an exciting new frontier in imaging vascular inflammation. In the near future, these advances will allow more precise imaging of disease activity enabling clinicians to offer more targeted and individualized patient management

    Mapping pathological phenotypes in a mouse model of CDKL5 disorder.

Here we describe the physiological, molecular, and behavioral phenotyping of a Cdkl5 conditional knockout mouse model of CDKL5 disorder. Behavioral analysis of constitutive Cdkl5 knockout mice revealed key features of the human disorder, including limb clasping, hypoactivity, and abnormal eye tracking. Anatomical, physiological, and molecular analysis of the knockout uncovered potential pathological substrates of the disorder, including reduced dendritic arborization of cortical neurons, abnormal electroencephalograph (EEG) responses to convulsant treatment, decreased visual evoked responses (VEPs), and alterations in the Akt/rpS6 signaling pathway. Selective knockout of Cdkl5 in excitatory and inhibitory forebrain neurons allowed us to map the behavioral features of the disorder to separable cell-types.     Memory-enhancing effects of GEBR-32a, a new PDE4D inhibitor holding promise for the treatment of Alzheimer\u2019s disease.

    Memory loss characterizes several neurodegenerative disorders, including Alzheimer’s disease (AD). Inhibition of type 4 phosphodiesterase (PDE4) and elevation of cyclic adenosine monophosphate (cAMP) has emerged as a promising therapeutic approach to treat cognitive deficits. However, PDE4 exists in several isoforms and pan inhibitors cannot be used in humans due to severe emesis. Here, we present GEBR-32a, a new PDE4D full inhibitor that has been characterized both in vitro and in vivo using biochemical, electrophysiological and behavioural analyses. GEBR-32a efficiently enhances cAMP in neuronal cultures and hippocampal slices. In vivo pharmacokinetic analysis shows that GEBR-32a is rapidly distributed within the central nervous system with a very favourable brain/blood ratio. Specific behavioural tests (object location and Y-maze continuous alternation tasks) demonstrate that this PDE4D inhibitor is able to enhance memory in AD transgenic mice and concomitantly rescues their hippocampal long-term potentiation deficit. Of great relevance, our preliminary toxicological analysis indicates that GEBR-32a is not cytotoxic and genotoxic, and does not seem to possess emetic-like side effects. In conclusion, GEBR-32a could represent a very promising cognitive-enhancing drug with a great potential for the treatment of Alzheimer’s disease

    Effects of resistance training on jumping performance in pre-adolescent rhythmic gymnasts: a randomized controlled study

    The aim of this study was to determine the effectiveness of two different resistance training programs on lower limb explosive and reactive strength in young female athletes. Fifty seven rhythmic gymnasts were randomly assigned to unspecific resistance training with dumbbells (12 repetition maximum squats) (n=19; age=12.0±1.8 years) or to specific resistance training with weighted belts (6% of body mass; n=18; age=11.9 ± 1.0 years). Squat jump test, counter movement jump test, hopping test, flexibility of the hip, and anthropometric measures were assessed before and after six weeks training. The main result was that both unspecific resistance training and specific resistance training protocols positively affected the jumping performance, with an increase of the lower limb explosive strength of 6-7%, with no side effects. Counter movement jump flight time increased significantly (p<0.01) while hopping test ground contact time significantly decreased (p<0.01). No significant differences were detected among groups for flexibility, body mass, calf and thigh circumferences. Therefore, six weeks of resistance training that integrates different elements of rhythmic gymnastics training enhance jumping ability in young female athletes
