From everyday conversation to political action: Talking austerity in online ‘third spaces’
Taking forward a new agenda for online political deliberation – the study of everyday political talk in non-political, online ‘third spaces’ – this article examines the dynamics of political talk across three general interest UK-based online forums. The quantitative analysis found that discussions about austerity were just as likely to emerge from non-political discussions as they were ones that began as ‘political’, demonstrating the links people made between everyday experiences and public policy. Our qualitative analysis represents the first real attempt to analyse political actions within third spaces, with some striking outcomes. Over half of all political discussions led to at least one political action (with significant variation between forums). The findings demonstrate that while such third spaces remain concerned with the preoccupations of everyday life, they can and do perform a role of mobilizing agent towards political participation
Disruption of a Proto-Planetary Disk by the Black Hole at the Milky Way Centre
Recently, an ionized cloud of gas was discovered plunging toward the
supermassive black hole, SgrA*, at the centre of the Milky Way. The cloud is
being tidally disrupted along its path to closest approach at ~3100
Schwarzschild radii from the black hole. Here, we show that the observed
properties of this cloud of gas can naturally be produced by a proto-planetary
disk surrounding a low-mass star, which was scattered from the observed ring of
young stars orbiting SgrA*. As the young star approaches the black hole, its
disk experiences both photo-evaporation and tidal disruption, producing a
cloud. Our model implies that planets form in the Galactic centre, and that
tidal debris from proto-planetary disks can flag low mass stars which are

Bioavailability and kidney responses to Diclofenac in the fathead minnow (Pimephales promelas)

The final version is available from the publisher via the DOI in this record.Diclofenac is one of the most widely prescribed nonsteroidal anti-inflammatory drugs worldwide. It is frequently detected in surface waters; however, whether this pharmaceutical poses a risk to aquatic organisms is debated. Here we quantified the uptake of diclofenac by the fathead minnow (Pimephales promelas) following aqueous exposure (0.2-25.0 μg L(-1)) for 21 days, and evaluated the tissue and biomolecular responses in the kidney. Diclofenac accumulated in a concentration- and time-dependent manner in the plasma of exposed fish. The highest plasma concentration observed (for fish exposed to 25 μg L(-1) diclofenac) was within the therapeutic range for humans. There was a strong positive correlation between exposure concentration and the number of developing nephrons observed in the posterior kidney. Diclofenac was not found to modulate the expression of genes in the kidney associated with its primary mode of action in mammals (prostaglandin-endoperoxide synthases) but modulated genes associated with kidney repair and regeneration. There were no significant adverse effects following 21 days exposure to concentrations typical of surface waters. The sign problem across the QCD phase transition
The average phase factor of the QCD fermion determinant signals the strength
of the QCD sign problem. We compute the average phase factor as a function of
temperature and baryon chemical potential using a two-flavor NJL model. This
allows us to study the strength of the sign problem at and above the chiral
transition. It is discussed how the anomaly affects the sign problem.
Finally, we study the interplay between the sign problem and the endpoint of

Endotoxin as a Marker for Water Quality
Background: Water quality testing is vital to protect human health. Current testing relies mainly on culture-based detection of faecal indicator organisms such as Escherichia coli (E.coli). However, bacterial cultures are a slow process, taking 24–48 h and requiring specialised laboratories and trained personnel. Access to such laboratories is often sparse in developing countries and there are many fatalities deriving from poor water quality. Endotoxin is a molecular component of Gram-negative bacterial cell walls and can be used to detect their presence in drinking water. Method: The current study used a novel assay (BacterisK) to rapidly detect endotoxin in various water samples and correlate the results with E. coli content measured by culture methods. The data generated by the BacterisK assay are presented as an ‘endotoxin risk’ (ER). Results: The ER values correlate with E. coli and thus endotoxin can be used as a marker of faecal contamination in water. Moreover, the BacterisK assay provides data in near real-time and can be used in situ allowing water quality testing at different spatial and temporal locations. Conclusion: We suggest that BacterisK can be used as a convenient risk assessment tool to assess water quality where results are required quickly or access to laboratories is lacking
Native American Children and Their Reports of Hope: Construct Validation of the Children's Hope Scale
Child reports of hope continue to be utilized as predictors of positive adjustment; however, the utilization of the hope construct has not been assessed within the culturally diverse Native American child group. The present study investigated the applicability of the Hope theory among 96 Native American children in the Midwest. Measures included the Children’s Hope Scale and a Hope Interview. Native American children in the current sample appear to conceptualize hope as a way to reach goals as did the children in the normative sample. Results from the factor analysis demonstrate that the factor structure found in the current study was similar to the factor structure found in the standardization sample. Because of the similar Hope theory conceptualization and factor structure, interventions focused on the positive psychology construct of hope may be applicable within a Native American child population
An Anti-Human ICAM-1 Antibody Inhibits Rhinovirus-Induced Exacerbations of Lung Inflammation
Human rhinoviruses (HRV) cause the majority of common colds and acute exacerbations of asthma and chronic obstructive pulmonary disease (COPD). Effective therapies are urgently needed, but no licensed treatments or vaccines currently exist. Of the 100 identified serotypes, ∼90% bind domain 1 of human intercellular adhesion molecule-1 (ICAM-1) as their cellular receptor, making this an attractive target for development of therapies; however, ICAM-1 domain 1 is also required for host defence and regulation of cell trafficking, principally via its major ligand LFA-1. Using a mouse anti-human ICAM-1 antibody (14C11) that specifically binds domain 1 of human ICAM-1, we show that 14C11 administered topically or systemically prevented entry of two major groups of rhinoviruses, HRV16 and HRV14, and reduced cellular inflammation, pro-inflammatory cytokine induction and virus load in vivo. 14C11 also reduced cellular inflammation and Th2 cytokine/chemokine production in a model of major group HRV-induced asthma exacerbation. Interestingly, 14C11 did not prevent cell adhesion via human ICAM-1/LFA-1 interactions in vitro, suggesting the epitope targeted by 14C11 was specific for viral entry. Thus a human ICAM-1 domain-1-specific antibody can prevent major group HRV entry and induction of airway inflammation in vivo

The association between pain-induced autonomic reactivity and descending pain control is mediated by the periaqueductal grey.
There is a strict interaction between the autonomic nervous system (ANS) and pain, which might involve descending pain modulatory mechanisms. The periaqueductal grey (PAG) is involved both in descending pain modulation and ANS, but its role in mediating this relationship has not yet been explored. Here, we sought to determine brain regions mediating ANS and descending pain control associations. Thirty participants underwent conditioned pain modulation (CPM) assessments, in which they rated painful pressure stimuli applied to their thumbnail, either alone or with a painful cold contralateral stimulation. Differences in pain ratings between ‘pressure-only’ and ‘pressure + cold’ stimuli provided a measure of descending pain control. In 18 of the 30 participants, structural scans and two functional MRI assessments, one pain-free and one during cold-pain were acquired. Heart rate variability (HRV) was simultaneously recorded. Normalised low-frequency HRV (LF-HRVnu) and the CPM score were negatively correlated; individuals with higher LF-HRVnu during pain reported reductions in pain during CPM. PAG-ventro-medial prefrontal cortex (vmPFC) and PAG-rostral ventromedial medulla (RVM) functional connectivity correlated negatively with the CPM. Importantly, PAG-vmPFC functional connectivity mediated the strength of the LF-HRVnu-CPM association. CPM response magnitude was also negatively correlated with vmPFC GM volume. Our multi-modal approach, using behavioural, physiological and MRI measures, provides important new evidence of interactions between ANS and descending pain mechanisms. ANS dysregulation and dysfunctional descending pain modulation are characteristics of chronic pain. We suggest that further investigation of body-brain interactions in chronic pain patients may catalyse the development of new treatments

This was done through a detailed evaluation of publicly available data and information. The investigation follows a receptor-pathway-source approach to back-track from designated shellfish waters in Lyme Bay and the Exe Estuary to potential upstream sources of contaminants in the Exe Estuary catchment. When reviewing information on potential sources and drivers of water quality issues, we highlight where there are significant data gaps. This report also provides a summary of activities and actions already underway within the wider catchment designed to achieve water quality improvements and discusses potential future catchment management interventions.Natural Environment Research Council (NERC

Sustained perturbation in functional connectivity induced by cold pain.
BACKGROUND: Functional connectivity (FC) perturbations have been reported in multiple chronic pain phenotypes, but the nature of reported changes varies between cohorts and may relate to the consequences of living with chronic-pain related comorbidities, such as anxiety and depression. Healthy volunteer studies provide opportunities to study the effects of tonic noxious stimulation independently of these sequelae. Connectivity changes in task negative and positive networks, for example, the default mode and salience networks (DMN/SN), respectively, have been described, but how these and other connectivity networks, for example, those governing descending pain control are affected by the presence of tonic, noxious stimulation in healthy, pain-free individuals, remains unknown. METHOD: In 20 healthy volunteers, we assessed FC prior to, during, and following tonic cold painful stimulation in the ventromedial prefrontal cortex (vmPFC), rostral anterior insula (rAI), subgenual anterior cingulate cortex (ACC) and periaqueductal grey (PAG). We also recorded subjectively reported pain using a computerised visual analogue scale. RESULTS: We saw DMN FC changes during painful stimulation and that inter-network connectivity between the rAI with the vmPFC increased during pain, whereas PAG-precuneus FC decreased. Pain-induced FC alterations persisted following noxious stimulation. FC changes related to the magnitude of individuals' subjectively reported pain. CONCLUSIONS: We demonstrate FC changes during and following tonic cold-pain in healthy participants. Similarities between our findings and reports of patients with chronic pain suggest that some FC changes observed in these patients may relate to the presence of an ongoing afferent nociceptive drive. SIGNIFICANCE: How pain-related resting state networks are affected by tonic cold-pain remains unknown. We investigated functional connectivity alterations during and following tonic cold pain in healthy volunteers. Cold pain perturbed the functional connectivity of the ventro-medial prefrontal cortex, anterior insula, and the periacquaductal grey area. These connectivity changes were associated with the magnitude of individuals' reported pain. We suggest that some connectivity changes described in chronic pain patients may be due to an ongoing afferent peripheral drive.This work was funded by a Medical
