Membrane interactions of S100A12 (Calgranulin C)
S100A12 (Calgranulin C) is a small acidic calcium-binding peripheral membrane protein with two EF-hand structural motifs. It is expressed in macrophages and lymphocytes and highly up-regulated in several human inflammatory diseases. In pigs, S100A12 is abundant in the cytosol of granulocytes, where it is believed to be involved in signal modulation of inflammatory process. In this study, we investigated the interaction of the porcine S100A12 with phospholipid bilayers and the effect that ions (Ca2+, Zn2+ or both together) have in modifying protein-lipid interactions. More specifically, we intended to address issues such as: (1) is the protein-membrane interaction modulated by the presence of ions? (2) is the protein overall structure affected by the presence of the ions and membrane models simultaneously? (3) what are the specific conformational changes taking place when ions and membranes are both present? (4) does the protein have any kind of molecular preferences for a specific lipid component? To provide insight into membrane interactions and answer those questions, synchrotron radiation circular dichroism spectroscopy, fluorescence spectroscopy, and surface plasmon resonance were used. The use of these combined techniques demonstrated that this protein was capable of interacting both with lipids and with ions in solution, and enabled examination of changes that occur at different levels of structure organization. The presence of both Ca2+ and Zn2+ ions modify the binding, conformation and thermal stability of the protein in the presence of lipids. Hence, these studies examining molecular interactions of porcine S100A12 in solution complement the previously determined crystal structure information on this family of proteins, enhancing our understanding of its dynamics of interaction with membranes

Constructions of algebraic lattices
In this work we present constructions of algebraic lattices in Euclidean space with optimal center density in dimensions 2, 3, 4, 6, 8 and 12, which are rotated versions of the lattices Λn, for n = 2,3,4,6,8 and K12. Characterizing Width Uniformity by Wave Propagation
This work describes a novel image analysis approach to characterize the
uniformity of objects in agglomerates by using the propagation of normal
wavefronts. The problem of width uniformity is discussed and its importance for
the characterization of composite structures normally found in physics and
biology highlighted. The methodology involves identifying each cluster (i.e.
connected component) of interest, which can correspond to objects or voids, and
estimating the respective medial axes by using a recently proposed wavefront
propagation approach, which is briefly reviewed. The distance values along such
axes are identified and their mean and standard deviation values obtained. As
illustrated with respect to synthetic and real objects (in vitro cultures of
neuronal cells), the combined use of these two features provide a powerful
description of the uniformity of the separation between the objects, presenting

Alterações músculo-esqueléticas relacionadas com o envelhecimento: causas e consequências
O aumento da população idosa e a crescente procura de tratamentos mais adequados para
aliviar a dor causada pelos fatores que caracterizam o envelhecimento, particularmente a
instalação da sarcopenia, condição associada ao sedentarismo e em alguns casos a
enfermidades como artrites e artrose, são responsáveis pelo desenvolvimento de dores
articulares que impossibilitam a prática de exercÃcio fÃsico, despoletando outros riscos como
a osteoporose e perda da estabilidade postural que em conjunto aumentam o risco de
quedas, lesões e fraturas associadas (Powers & Howley, 1997). Porém, a sua origem
deverá ser igualmente interpretada muitas vezes como consequência das crescentes
limitações do foro ortopédico.
A inatividade conduz a um desuso do sistema músculo-esquelético e naturalmente a uma
hipo-estimulação do tecido muscular conduzindo à atrofia do mesmo com diminuição da
sua função (Tavares, C. 2003). Um exemplo categórico é o caso da osteoartrose do joelho,
uma patologia articular crónica degenerativa, frequente em adultos e idosos que interfere na
mobilidade e autonomia. A osteoartrose é caracterizada pela presença de dor e limitação
funcional, consequência da formação de osteófitos e/ou alteração na integridade da
cartilagem articular, que conduzem a uma perda gradual da força, diminuição da mobilidade
e instabilidade articular. Frequentemente assistimos nos indivÃduos com esta patologia a
uma diminuição gradual nos seus nÃveis de atividade fÃsica, já que a imobilização da
articulação contribui para diminuir o processo inflamatório na mesma, diminuindo também a
dor. No entanto, a imobilização da articulação com artrose vai potenciar: a atrofia, o
encurtamento de músculos e tendões, o surgimento de contracturas, a perda de massa
óssea, o aumento do risco de fratura, a redução da mobilidade articular e a degeneração da
cartilagem (Gordon, N., 1992). Assim, a inatividade fÃsica torna-se contraproducente, sendo
a prescrição de exercÃcio fÃsico adequado uma forma de combater o avanço progressivo da
doença. O fortalecimento da musculatura peri articular é fundamental para a obtenção do
equilÃbrio da articulação, controlando o impacto do pé sobre o solo durante a marcha,
lubrificando a articulação e reduzindo os movimentos anormais entre as superfÃcies
articulares o que ajuda a diminuir a degeneração da articulação (Roddy, E., W. Zhang, and
M. Doherty, 2005).
A manutenção do equilÃbrio é um aspeto determinante na funcionalidade e autonomia
estando relacionado como o sistema visual, somatosensorial e vestibular, com a força dos
membros inferiores, nomeadamente os grupos musculares flexores e extensores das
articulações do joelho e tibiotársica, que tendem a diminuir com a idade. Além disso, as
alterações degenerativas dos discos intervertebrais, somados à diminuição da flexibilidade
e da força das estruturas musculares e das posturas incorretas frequentes, conduzem a
desalinhamentos nas curvaturas da coluna que por sua vez contribuem para a deterioração
do equilÃbrio, não só estático, como também dinâmico, e consequentemente do padrão de
marcha. Neste contexto, a realização de atividade fÃsica é fundamental na prevenção de
quedas e de fraturas associadas, tendo sido demonstrado por vários estudos que a
participação em programas de exercÃcio e o treino de tarefas especificamente orientadas
para o sistema sensorial e a manutenção da estabilidade postural reduzem
significativamente o número de quedas quando comparado a grupos de controlo, tanto em
homens como em mulheres (Izquierdo et al., 2005; 2004). Em suma, o treino da força com
intensidade moderada a elevada pode ser efetuado com elevada tolerância por parte de
adultos e idosos, com resultados bastante satisfatórios em termos de adaptação
morfológica e funcional, assim como também em termos de propriedades elétricas e
contrácteis.
Tentativa preliminar para desenvolver um modelo de análise (path-flow) da performance em jovens nadadoras
O objectivo do presente trabalho foi desenvolver um modelo de análise da
performance (path flow) em jovens nadadoras, com base em parâmetros antropométricos,
hidrodinâmicos, biomecânicos e bioenergéticos
Stability of elite freestyle performance from childhood to adulthood
Stability of athletic performance is important for practitioners and coaches, since it allows the selection of appropriate training
methods and prediction of ages for best results. We performed a longitudinal study of 1694 season-best performances of 242
elite-standard swimmers throughout their careers, from 12 to 18 years of age. Mean stability (descriptive statistics and oneway
repeated-measures ANOVA, followed by a Bonferroni post-hoc test) and normative stability (Cohen’s kappa tracking
index and the Pearson correlation coefficient) were determined for seven consecutive seasons. Performance improvements in
all events were observed (14.36–18.97%). Bonferroni post-hoc tests verified changes in almost all events assessed. Cohen’s
kappa demonstrated low stability (0.17–0.27) in relative performance. Pearson correlations only became high from 15 to 16
years in the 50-m and 100-m events, and from 16 to 17 years in the 200-m, 400-m, and 1500-m events. Our results show that:
(a) swimmers should display a substantial improvement (14–19%) to become elite standard as adults, such as at 18 years; (b)
16 is the age at which the ability to predict adult performance increases markedly
Longitudinal interventions in elite swimming: a systematic review based on energetics, biomechanics and performance
Longitudinal information requires the notion of repeated measurements throughout time. Such data is important because it allows the determination of the effectiveness of an intervention program. Research in competitive swimming has given special emphasis to energetics and biomechanics as determinant domains to improve performance. The purpose of this systematic review was to summarize longitudinal evidences on the energetic, biomechanical, and performance status of elite swimmers. A computerized search was made in 6 databases, conference proceedings, and department files. The 28 studies that satisfied the inclusion criteria were selected for analysis. Studies' qualitative evaluation was made by 2 independent reviewers using the Quality Index. These studies were then gathered into 3 main categories according to their reported data: energetics (n = 18), biomechanics (n = 9), and performance (n = 8). The conclusions were as follows: (a) elite swimmers are able to demonstrate from slight to substantial changes in their performance and energetic and biomechanical profiles within and between seasons; (b) the magnitude of change is dependent on the characteristics of the training programs, the duration of the intervention, and subject's gender; and (c) future research should emphasize the use of more complex procedures to improve the quality of the interventions
