    Sudan Black B treatment reduces autofluorescence and improves resolution of in situ hybridization specific fluorescent signals of brain sections

    Interference by autofluorescence is one of the major concerns of immunofluorescence analysis of in situ hybridization-based diagnostic assays. We present a useful technique that reduces autofluorescent background without affecting the tissue integrity or direct immunofluorescence signals in brain sections. Using six different protocols, such as ammonia/ethanol, Sudan Black B (SBB) in 70% ethanol, photobleaching with UV light and different combinations of them in both formalin-fixed paraffin-embedded and frozen human brain tissue sections, we have found that tissue treatment of SBB in a concentration of 0.1% in 70% ethanol is the best approach to reduce/eliminate tissue autofluorescence and background, while preserving the specific fluorescence hybridization signals.     The contribution of a MOET nucleus scheme for the improvement of GuzerĂĄ ( Bos indicus) cattle for milk traits in Brazil.

    The Guzerå breed evolved from the introduction of breeds from India, mainly the Kankrej breed, into Brazilian livestock at the end of the 19th century. Guzerå adapted well to the climatic conditions of Brazil, where it is considered a dual-purpose breed and has been used for pasture-based beef, milk or dual-purpose production systems with the use of low-medium inputs. The importance of this genetic resource for milk production in tropical regions moved breeders to implement the National Breeding Program for the Improvement of Guzerå in 1994, based on both progeny testing and MOET nucleus schemes. We sought to evaluate the role of the MOET nucleus scheme in the phenotypic and genetic progress for milk traits in this breed. The initial database used in the present study consisted of 6,513 cows, daughters of 761 bulls. We performed genetic evaluations with different datasets using a linear mixed model in a single trait analysis, including the relationship matrix, in order to estimate breeding values. Inbreeding coefficients were also calculated using the relationship of descent between two parents. Annual phenotypic, genetic and inbreeding trends were obtained for each dataset, considering the genetic pathways of both the bull and the cow. The low genetic progress found for milk yield in the whole population (5.27 ± 0.30 kg/year) partially accounted for the dual-purpose selection goal, despite the higher genetic progress in the MOET nucleus (9.39 ± 0.79 kg/year). The inbreeding coefficient was minimized at the beginning of the breeding program based on the use of new lineages. Posteriorly, it started increasing again from 0.002 in 1991 to 0.008 in 2019. The results provided evidence of the significant contribution of the MOET nucleus scheme for the phenotypic and genetic progress of Guzerå breed for milk traits, as well as of the impact of the breeding program on the inbreeding coefficient rate in the early years. New strategies need to be designed for the Guzerå breed, to allow for greater improvement of milk traits and minimizing the rate of the inbreeding coefficient

    Gastric peroral endoscopic pyloromyotomy for refractory gastroparesis: a systematic review of early outcomes with pooled analysis

    Background and Aims: Gastroparesis (GP) is a chronic debilitating condition. Prior pyloric-targeted procedures are either invasive or have questionable efficacy. Gastric peroral pyloromyotomy (G-POEM) has been proposed as a minimally invasive approach. We performed a pooled analysis to evaluate the efficacy and safety of G-POEM for GP. Methods: Electronic databases (Medline, Scopus, EMBASE) were searched up to January 2019. Studies including patients who underwent G-POEM for GP were eligible. Procedural, clinical, and safety outcomes were assessed by pooling data with a random- or fixed-effect model according to the degree of heterogeneity to obtain a proportion with a 95% confidence interval. Results: Ten studies were eligible for inclusion (292 patients), and 2 of the 10 studies were prospective. Seven studies were performed in the United States, 2 in France, and 1 in China. Endoscopic pyloromyotomy was feasible in all patients. Significant symptomatic improvement was achieved after 83.9% of procedures (mean follow-up, 7.8 ± 5.5 months). When comparing the mean values of pre- and postprocedural scintigraphic evolution, there was a significant decrease of the residual percentage at 2 and 4 hours. The overall adverse events rate was 6.8%. Conclusions: G-POEM appears to be a promising approach for GP in terms of safety and efficacy outcomes in the short term

    The OPERA neutrino detector in the underground Gran Sasso Laboratory (LNGS) was designed to perform the first detection of neutrino oscillations in appearance mode through the study of ΜΌ→Μτ\nu_\mu\to\nu_\tau oscillations. The apparatus consists of an emulsion/lead target complemented by electronic detectors and it is placed in the high energy long-baseline CERN to LNGS beam (CNGS) 730 km away from the neutrino source. Runs with CNGS neutrinos were successfully carried out in 2007 and 2008 with the detector fully operational with its related facilities for the emulsion handling and analysis. After a brief description of the beam and of the experimental setup we report on the collection, reconstruction and analysis procedures of first samples of neutrino interaction events

    Emulsion sheet doublets as interface trackers for the OPERA experiment

    New methods for efficient and unambiguous interconnection between electronic counters and target units based on nuclear photographic emulsion films have been developed. The application to the OPERA experiment, that aims at detecting oscillations between mu neutrino and tau neutrino in the CNGS neutrino beam, is reported in this paper. In order to reduce background due to latent tracks collected before installation in the detector, on-site large-scale treatments of the emulsions ("refreshing") have been applied. Changeable Sheet (CSd) packages, each made of a doublet of emulsion films, have been designed, assembled and coupled to the OPERA target units ("ECC bricks"). A device has been built to print X-ray spots for accurate interconnection both within the CSd and between the CSd and the related ECC brick. Sample emulsion films have been extensively scanned with state-of-the-art automated optical microscopes. Efficient track-matching and powerful background rejection have been achieved in tests with electronically tagged penetrating muons. Further improvement of in-doublet film alignment was obtained by matching the pattern of low-energy electron tracks. The commissioning of the overall OPERA alignment procedure is in progress.Comment: 19 pages, 19 figure

    Relationship between the prevalence of subclinical tenosynovitis and treatment in patients with RA in clinical remission: STARTER study

    Objective: This study is a sub-analysis from the patient cohort of the STARTER (Sonographic Tenosynovitis Assessment in RheumaToid arthritis patiEnts in Remission) study. The aim was to evaluate differences in ultrasound-detected joint and/or tendon involvement between patients receiving therapies based on a combination of conventional synthetic DMARDs (csDMARDs) and biologic DMARDs (bDMARDs) and those who were treated with either csDMARDs or bDMARDs in monotherapy. Material and methods: Four hundred and twenty-seven consecutive patients with a diagnosis of RA were recruited between October 2013 and June 2014. They were divided into three subgroups based on their therapy at baseline: patients with bDMARD in monotherapy, patients with csDMARD in monotherapy and patients in combination therapy (csDMARD + bDMARD). At baseline, 6 months and 12 months, a clinical examination (28 joint count) and an ultrasound evaluation were performed in each patient. A score of grey-scale (GS) and power Doppler (PD) synovitis and tenosynovitis was calculated based on the OMERACT scoring systems. Results: Two hundred and fifty-six patients completed the observation period: 48 patients from the bDMARD group (18.75%), 152 patients from the csDMARD group (59.38%) and 56 patients from csDMARD + bDMARD group (21.88%). The analysis showed that GS tenosynovitis and PD tenosynovitis are better controlled in combination therapy than they are ith csDMARD alone (P=0.025 and P=0.047, respectively); for PD synovitis, there was a better response in those who were treated with the combination therapy when compared with the patients receiving csDMARD (P=0.01) or bDMARD (P=0.02) alone. Conclusions: The analysis showed a lower prevalence of subclinical inflammatory manifestations detected with ultrasound imaging in those patients treated with the combination therapy than in those in monotherapy

    Resumo A violĂȘncia de gĂȘnero, apesar de global, Ă© socialmente invisĂ­vel, sendo urgente o avanço de produção cientĂ­fica sobre esse tema. Um estudo bibliomĂ©trico majora a demanda de debates ao demonstrar que as pesquisas abrangeram poucos paĂ­ses, ignoraram as consequĂȘncias dos danos e ainda sĂŁo, apesar de sua ampliação, incipientes. Este artigo objetivou descrever o panorama da produção cientĂ­fica sobre violĂȘncia de gĂȘnero, procurando tecer uma anĂĄlise crĂ­tica diante da magnitude da demanda. Buscaram-se artigos em inglĂȘs, pelos descritores gender e violence, na base de dados Web of Science, de 1982 a 2012, excluindo-se os textos de ĂĄreas especĂ­ficas, que tratavam da violĂȘncia geral ou adotaram "gĂȘnero" em substituição a sexo, restando 450 artigos. A bibliometria ratificou que as mulheres encontram-se em mais situaçÔes de violĂȘncias, apesar da referĂȘncia a crianças e adolescentes, homossexuais e homens. A produtividade das pesquisas em diversas ĂĄreas do conhecimento Ă© crescente, revelando sua natureza multidisciplinar. Apenas 26 paĂ­ses sediaram estudos, nĂșmero reduzido diante da amplitude do problema. A maioria dos estudos nĂŁo abordou as consequĂȘncias da violĂȘncia de gĂȘnero, demonstrando uma importante lacuna na produção cientĂ­fica. Esta pesquisa evidenciou pontos pouco explorados pela produção cientĂ­fica, servindo de orientação para futuros estudos
