    Structure of a SLC26 Anion Transporter STAS Domain in Complex with Acyl Carrier Protein: Implications for E. coli YchM in Fatty Acid Metabolism

    SummaryEscherichia coli YchM is a member of the SLC26 (SulP) family of anion transporters with an N-terminal membrane domain and a C-terminal cytoplasmic STAS domain. Mutations in human members of the SLC26 family, including their STAS domain, are linked to a number of inherited diseases. Herein, we describe the high-resolution crystal structure of the STAS domain from E. coli YchM isolated in complex with acyl-carrier protein (ACP), an essential component of the fatty acid biosynthesis (FAB) pathway. A genome-wide genetic interaction screen showed that a ychM null mutation is synthetically lethal with mutant alleles of genes (fabBDHGAI) involved in FAB. Endogenous YchM also copurified with proteins involved in fatty acid metabolism. Furthermore, a deletion strain lacking ychM showed altered cellular bicarbonate incorporation in the presence of NaCl and impaired growth at alkaline pH. Thus, identification of the STAS-ACP complex suggests that YchM sequesters ACP to the bacterial membrane linking bicarbonate transport with fatty acid metabolism

    Quantifying farmers’ preferences for cropping systems intensification: A choice experiment approach applied in coastal Bangladesh’s risk prone farming systems

    Sustainable intensification (SI) is envisioned as an effective strategy for developing countries to increase farm productivity while reducing negative environmental and social externalities. The development of regionally appropriate SI options however requires accounting for the knowledge and preferences of key stakeholders. In Bangladesh, the Government has requested international donors to support the development of dry season rice expansion in the coastal region. Policies however tend to be made without adequate study of farmers’ preferences and ambitions; this can render crop intensification efforts ineffective. Understanding farmers’ preferences for alternative crops and crop management practices are therefore crucial for success where agricultural development investments aim at incorporating the principles of SI. OBJECTIVE(S): Using coastal Bangladesh as a case study– we aim to (1) quantify farmers’ preferences for alternative irrigated crop and crop management options in comparison to the status quo (land fallowing), (2) analyze whether farmers’ preferences are conditioned by concerns regarding the cost and availability of irrigation and fertilizer inputs in comparison to expected net revenues, (3) understand how the heterogeneity in preferences can be attributed to farmer and/or farm characteristics, institutional, and biophysical factors, (4) determine how much farmers’ are willing to invest in different crops and crop management options – including those reliant and not reliant on irrigation. METHODS: Taking 300 farmers in two diverse coastal environments, a choice experiment (CE) was employed to explore the heterogeneity in farmers’ preferences for different dry “rabi” season intensification options (‘boro’ rice, maize, wheat and mungbean) against the status quo (dry season land fallowing after harvest of the monsoon season rice crop). Analyses included random parameter logit modeling followed by willingness-to-invest and profit simulations. RESULTS AND CONCLUSIONS: Analyses revealed strong farmer preferences against rice and in favor of irrigated maize, and also in favor of rainfed or partially irrigated mungbean as an alternative to land fallowing. Irrespective of their location and environmental conditions, respondents had largely a negative preference for irrigation and fertilizer use due to high investment costs and associated production risks in the dry season. Nonetheless, a significant positive effect on their willingness-to-intensify cropping was observed where farmers felt it feasible to provide in-field drainage to limit waterlogging risks.Estación Experimental Agropecuaria BarilocheFil: Aravindakshan, Sreejith.     Revisiting Bimaximal Neutrino Mixing in a Model with S4 Discrete Symmetry

    In view of the fact that the data on neutrino mixing are still compatible with a situation where Bimaximal mixing is valid in first approximation and it is then corrected by terms of order of the Cabibbo angle, arising from the diagonalization of the charged lepton masses, we construct a model based on the discrete group S4 where those properties are naturally realized. The model is supersymmetric in 4-dimensions and the complete flavour group is S4 x Z4 x U(1)_FN, which also allows to reproduce the hierarchy of the charged lepton spectrum. The only fine tuning needed in the model is to reproduce the small observed value of r, the ratio between the neutrino mass squared differences. Once the relevant parameters are set to accommodate r then the spectrum of light neutrinos shows a moderate normal hierarchy and is compatible, within large ambiguities, with the constraints from leptogenesis as an explanation of the baryon asymmetry in the Universe.Comment: 30 pages, 5 figures; added reference

    Identification of 181Hg and shape coexistence in odd-A Hg isotopes

    In-beam γ-ray transitions in 181Hg, the lightest odd-A Hg isotope known thus far, have been identified from fragment mass-γ and γ-γ coincidence measurements. Five prolate deformed rotational bands were placed in the level scheme. A decoupled band built on the strongly prolate deformed 1/2-[521] ground state was observed up to 29/2-. A 5/2-[512] configuration is suggested for a pair of strongly coupled bands displaying no signature splitting. The other two bands are also signature partner bands. They are populated with the largest intensity and exhibit splitting. They have been associated with the mixed neutron i13/2 orbitals and are proposed to decay to an i13/2 isomeric state associated with an oblate state

    Burden and risk factors for Pseudomonas aeruginosa community-acquired pneumonia:a Multinational Point Prevalence Study of Hospitalised Patients

    Pseudornonas aeruginosa is a challenging bacterium to treat due to its intrinsic resistance to the antibiotics used most frequently in patients with community-acquired pneumonia (CAP). Data about the global burden and risk factors associated with P. aeruginosa-CAP are limited. We assessed the multinational burden and specific risk factors associated with P. aeruginosa-CAP. We enrolled 3193 patients in 54 countries with confirmed diagnosis of CAP who underwent microbiological testing at admission. Prevalence was calculated according to the identification of P. aeruginosa. Logistic regression analysis was used to identify risk factors for antibiotic-susceptible and antibiotic-resistant P. aeruginosa-CAP. The prevalence of P. aeruginosa and antibiotic-resistant P. aeruginosa-CAP was 4.2% and 2.0%, respectively. The rate of P. aeruginosa CAP in patients with prior infection/colonisation due to P. aeruginosa and at least one of the three independently associated chronic lung diseases (i.e. tracheostomy, bronchiectasis and/or very severe chronic obstructive pulmonary disease) was 67%. In contrast, the rate of P. aeruginosa-CAP was 2% in patients without prior P. aeruginosa infection/colonisation and none of the selected chronic lung diseases. The multinational prevalence of P. aeruginosa-CAP is low. The risk factors identified in this study may guide healthcare professionals in deciding empirical antibiotic coverage for CAP patients

    The Cholecystectomy As A Day Case (CAAD) Score: A Validated Score of Preoperative Predictors of Successful Day-Case Cholecystectomy Using the CholeS Data Set

    Background Day-case surgery is associated with significant patient and cost benefits. However, only 43% of cholecystectomy patients are discharged home the same day. One hypothesis is day-case cholecystectomy rates, defined as patients discharged the same day as their operation, may be improved by better assessment of patients using standard preoperative variables. Methods Data were extracted from a prospectively collected data set of cholecystectomy patients from 166 UK and Irish hospitals (CholeS). Cholecystectomies performed as elective procedures were divided into main (75%) and validation (25%) data sets. Preoperative predictors were identified, and a risk score of failed day case was devised using multivariate logistic regression. Receiver operating curve analysis was used to validate the score in the validation data set. Results Of the 7426 elective cholecystectomies performed, 49% of these were discharged home the same day. Same-day discharge following cholecystectomy was less likely with older patients (OR 0.18, 95% CI 0.15–0.23), higher ASA scores (OR 0.19, 95% CI 0.15–0.23), complicated cholelithiasis (OR 0.38, 95% CI 0.31 to 0.48), male gender (OR 0.66, 95% CI 0.58–0.74), previous acute gallstone-related admissions (OR 0.54, 95% CI 0.48–0.60) and preoperative endoscopic intervention (OR 0.40, 95% CI 0.34–0.47). The CAAD score was developed using these variables. When applied to the validation subgroup, a CAAD score of ≤5 was associated with 80.8% successful day-case cholecystectomy compared with 19.2% associated with a CAAD score >5 (p < 0.001). Conclusions The CAAD score which utilises data readily available from clinic letters and electronic sources can predict same-day discharges following cholecystectomy

    A century of trends in adult human height

    Being taller is associated with enhanced longevity, and higher education and earnings. We reanalysed 1472 population-based studies, with measurement of height on more than 18.6 million participants to estimate mean height for people born between 1896 and 1996 in 200 countries. The largest gain in adult height over the past century has occurred in South Korean women and Iranian men, who became 20.2 cm (95% credible interval 17.5-22.7) and 16.5 cm (13.3-19.7) taller, respectively. In contrast, there was little change in adult height in some sub-Saharan African countries and in South Asia over the century of analysis. The tallest people over these 100 years are men born in the Netherlands in the last quarter of 20th century, whose average heights surpassed 182.5 cm, and the shortest were women born in Guatemala in 1896 (140.3 cm; 135.8-144.8). The height differential between the tallest and shortest populations was 19-20 cm a century ago, and has remained the same for women and increased for men a century later despite substantial changes in the ranking of countries

    Rising rural body-mass index is the main driver of the global obesity epidemic in adults

    Body-mass index (BMI) has increased steadily in most countries in parallel with a rise in the proportion of the population who live in cities 1,2 . This has led to a widely reported view that urbanization is one of the most important drivers of the global rise in obesity 3�6 . Here we use 2,009 population-based studies, with measurements of height and weight in more than 112 million adults, to report national, regional and global trends in mean BMI segregated by place of residence (a rural or urban area) from 1985 to 2017. We show that, contrary to the dominant paradigm, more than 55 of the global rise in mean BMI from 1985 to 2017�and more than 80 in some low- and middle-income regions�was due to increases in BMI in rural areas. This large contribution stems from the fact that, with the exception of women in sub-Saharan Africa, BMI is increasing at the same rate or faster in rural areas than in cities in low- and middle-income regions. These trends have in turn resulted in a closing�and in some countries reversal�of the gap in BMI between urban and rural areas in low- and middle-income countries, especially for women. In high-income and industrialized countries, we noted a persistently higher rural BMI, especially for women. There is an urgent need for an integrated approach to rural nutrition that enhances financial and physical access to healthy foods, to avoid replacing the rural undernutrition disadvantage in poor countries with a more general malnutrition disadvantage that entails excessive consumption of low-quality calories. © 2019, The Author(s)
