Biomarkers: a strategic tool in the assessment of environmental quality of coastal waters
Ecosystems are under the pressure of complex mixtures of contaminants whose effects are not always simple to assess. Biomarkers, acting as early warning signals of the presence of potentially toxic xenobiotics, are useful tools for assessing either exposure to, or the effects of these compounds providing information about the toxicant bioavailability. In fact, it has been argued that a full understanding of ecotoxicological processes must consider an integrated multi-level approach, in which molecular impact is related with higher-order biological consequences at the individual, population and community levels. Monitoring programs should make use of this tool to link contaminants and ecological responses fulfilling strategies like those launched by OSPAR (Commissions of Oslo and Paris) Convention on the protection of the marine environment of the North-East Atlantic and the International Council for the Exploration of the Sea (ICES). An overview of the work done in the past few years using biomarkers as in situ tools for pollution assessment in Portuguese coastal waters is presented as a contribution to the set up of a biomonitoring program for the Portuguese coastal zone. Considering the data set available the biomonitoring proposal should include the analysis of biomarkers and effects at individual levels. The aim of the program will include a spatial and temporal characterization of the biomarkers acetyl-cholinesterase, metallothioneins, DNA damage, adenylate energy charge and scope-for-growth levels. The investigation of the spatial variation of biomarkers is crucial to define sites for long term monitoring, which will be integrated with a chemical monitoring program. An SDRAM test education package that embeds the factory equipment into the e-learning server
SDRAM (Synchronous Dynamic Random Access Memory) demand has grown exponentially since the 1980s, as a result of technological factors and new areas of application, particularly concerning communication and consumer electronics. The SDRAM market represented in 2007 c. 20% of the total semiconductor business and is seen as a strategic area, justifying private and public investment in the western and far-eastern economic communities. SDRAM test education is therefore an important subject, but very high purchase and maintenance costs keep test equipment beyond reach of most university test courses. This paper presents a pilot project addressing an SDRAM test education course developed jointly by Qimonda and the University of Porto (FEUP), where the company offers remote access to one if its Advantest SDRAM automatic test equipments. Access to this remote tester was embedded into the Moodle e-learning server that supports a new course entitled Electronic Systems Testing (TSEL), which is part of the Integrated Masters degree on Electrical and Computer Engineering at FEUP. The excellent feedback received from students encouraged us to extend this cooperation into an educational network, which is also introduced in this paper
Motivating for Reading through Transmedia Storytelling: A Case Study with Students from a Middle School in the Médio Tejo Region
The evolution of information and communication technologies has changed the way we relate to each other and how we build our knowledge. This creates challenges for education systems, as school must provide all students with the educational experiences that will enable them to develop the skills reflected in the profile of the 21st-century student on completion of compulsory schooling. It is up to teachers to find new ways of teaching, making the most of the resources and digital tools made available by mobile technologies. Technology can make a significant contribution to increasing students' motivation because it is closer to what they like and use in their daily lives. And this introduction of technology into the classroom can promote student-oriented teaching, which contributes to the development of skills such as autonomy, critical thinking and self-esteem. One of the areas that can contribute to this paradigm shift is the creation of experiences in immersive learning environments such as Transmedia Storytelling. Immersive learning environments can favour the creation and implementation of projects that promote reading skills in schools. This is the focus of this article. The aim of this study is to evaluate the impact of transmedia storytelling on the level of motivation of students and on the improvement of pedagogical practices implemented by the teachers involved. This case study was carried out in the subject of Portuguese in three 7th-grade classes of a school from the Médio Tejo region. The results obtained suggest a high level of motivation of students and teachers. The latter recognise that pedagogical routes using Transmedia Storytelling contribute to the motivation, autonomy and improvement of students' learning.info:eu-repo/semantics/publishedVersio
On-line estimation of biomass in an E. coli fed-batch fermentation
In this work, an Extended Kalman Observer is applied to the on-line determination of biomass
concentration in a high-cell density fed-batch fermentation of Escherichia coli.
Although the importance of this fermentation process for the biopharmaceutical industry is widely
recognized, there are still several difficulties associated with the design of monitoring and control algorithms
that could improve the performance of the process by decreasing the production costs and increasing the
yield.
In this process, biomass concentration has an important role for model predictive control, estimation of
specific growth rates, prevention of acetate accumulation and optimization of the production of recombinant
proteins (regarding both productivity and moment of induction). However, nowadays it is still determined
using off-line laboratory analysis, making it of limited use for control purposes.
For the development of the Extended Kalman Observer, a dynamical mathematical model of the process was
used, which includes balance equations for the main state variables (biomass, glucose, acetate, dissolved
oxygen and carbon dioxide concentrations) together with a complex kinetic model describing the 3 main
metabolic pathways of Escherichia coli.
The observer applied in this work requires the on-line measurement of a subset of state variables (dissolved
oxygen and carbon dioxide concentrations) together with broth weight and gaseous mass transfer rates.
State-of-the-art sensors were used for measuring dissolved oxygen and carbon dioxide concentrations and
gaseous transfer rates were determined on-line using commercial gas analysers. The calculations were
performed on-line in a developed LabVIEW data acquisition and control system.
The extended Kalman observer exhibited a good convergence to the real values of biomass concentration,
with a very low quadratic difference between experimental and estimated data. Also, the sampling frequency

Identification of yield coefficients in an E. coli model : an optimal experimental design using genetic algorithms
An optimal experimental design for yield coefficients estimation in an
unstructured growth model of fed-batch fermentation of E. coli is presented. The feed
profile is designed by optimisation of a scalar function based on the Fischer Information
Matrix. A genetic algorithm is proposed as the optimisation method due to its efficiency

Design of on-line state estimators for a recombinant E. coli fed-batch fermentation
In recent years a remarkable effort has been made in the development of new
sensors and process analytical technology. However, it is still difficult to find reliable and
low cost commercial sensors for on-line measurements of important variables. Therefore,
considerable attention has been focused on the development of on-line software sensors.
Nevertheless, the application of those algorithms to complex biological processes is still
very incipient. In this work two different state estimators have been studied regarding
their applicability to the recombinant Escherichia coli fed-batch fermentation. Both
algorithms showed the ability to estimate on-line biomass and acetate concentrations.
However, the extended Kalman observer exhibited a better convergence in spite of being

Design of interval observers for an E. coli fed-batch fermentation with uncertain inputs
In bioreactors, the measurement of variables that play a key role in the quality and productivity of
fermentations, is of major importance. However, their direct measurement is often expensive or even
impossible considering the current sensor technology. Therefore, on-line estimation of unmeasured
variables in bioreactors can be an interesting approach.
The objective of this work is to introduce an alternative solution for the state observation of
bioprocesses in cases where the kinetic model is unclear and the concentration of the influent
substrates is badly known, a situation that is common in many practical applications.
The high-cell density fed-batch fermentation of Escherichia coli is studied in terms of applicability of
a simple interval observer for the estimation of relevant variables of the process, when uncertainties of
the process inputs exist.
The simple interval observer is designed on the basis of the cooperativity properties of the observer
error dynamics (Rapaport and Dochain, 2005). Further assumptions are the knowledge of the (lower
and upper) bounds of the influent substrate concentration. Furthermore, an appropriate state
transformation and conditions that guarantee system cooperativity have been introduced for that
purpose.

Monitoring of fed-batch E. coli fermentations with software sensors
Accurate monitoring and control of industrial bioprocess requires the knowledge of a great number of variables, being some of them not measurable with standard devices. To overcome this difficulty, software sensors can be used for on-line estimation of those variables and, therefore, its development is of paramount importance. An Asymptotic Observer was used for monitoring Escherichia coli fed-batch fermentations. Its performance was evaluated using simulated and experimental data. The results obtained showed that the observer was able to predict the biomass concentration profiles showing, however, less satisfactory results regarding the estimation of glucose and acetate concentrations. quite narrow, indicating that it is possible to accurately predict biomass concentration

Supplier selection and procurement in SMEs: Insights from the literature on key criteria and purchasing strategies
Effective strategic purchasing and supplier selection in companies provides businesses with leverage in acquiring goods and services. Thus, companies are in a better position to negotiate prices, discounts, delivery times and logistic channels. Also, strategic purchasing allows for performing a risk assessment to ensure the company's profitability. This research aimed to identify the key currently deliberated by SMEs for supplier selection, considering such purchasing strategies as (1) cost reduction, (2) risk management, (3) global sourcing, (4) total quality management, (5) sustainable management and (6) supplier management. Also, it aimed to identify the emerging issues related to purchasing strategies. This research work performed a content analysis following a literature review. The Scopus indexation database was selected to conduct the document search. After the refinement process, based on 59 analysed documents, bibliometric assessment tools were applied to identify the key criteria for supplier selection. The TOP6 highest ranked criteria, which corresponds to 80 % of the most referred criteria, include: (1) the quality of goods; (2) compliance with the delivery times; (3) price/cost; (4) supplier reputation and/or market positioning; (5) geographical location; and (6) supplier performance history. The goal of strategic purchasing is to support the companies in achieving long-term goals through its integration into the company's strategic planning process. It should be identified by the managers as an important resource. Several factors elevate the importance of strategic purchasing, namely, environmental protection, technology advances related to logistics 4.0, and risk assessment related to global sourcing and sustainability.
