Hypoxia up-regulates SERPINB3 through HIF-2\u3b1 in human liver cancer cells.
SERPINB3 is a cysteine-proteases inhibitor up-regulated in a significant number of cirrhotic patients carrying hepatocellular carcinoma (HCC) and recently proposed as a prognostic marker for HCC early recurrence. SERPINB3 has been reported to stimulate proliferation, inhibit apoptosis and, similar to what reported for hypoxia, to trigger epithelial-to-mesenchymal transition (EMT) and increased invasiveness in liver cancer cells. This study has investigated whether SERPINB3 expression is regulated by hypoxia-related mechanisms in liver cancer cells. Exposure of HepG2 and Huh7 cells to hypoxia up-regulated SERPINB3 transcription, protein synthesis and release in the extracellular medium. Hypoxia-dependent SERPINB3 up-regulation was selective (no change detected for SERPINB4) and operated through hypoxia inducible factor (HIF)-2\u3b1 (not HIF-1\u3b1) binding to SERPINB3 promoter, as confirmed by chromatin immuno-precipitation assay and silencing experiments employing specific siRNAs. HIF-2\u3b1-mediated SERPINB3 up-regulation under hypoxic conditions required intracellular generation of ROS. Immuno-histochemistry (IHC) and transcript analysis, performed in human HCC specimens, revealed co-localization of the two proteins in liver cancer cells and the existence of a positive correlation between HIF-2\u3b1 and SERPINB3 transcript levels, respectively. Hypoxia, through HIF-2\u3b1-dependent and redox-sensitive mechanisms, up-regulates the transcription, synthesis and release of SERPINB3, a molecule with a high oncogenic potential
Viscerocutaneous form of loxoscelism and erythrocyte glucose-6-phosphate deficiency
Em um perĂodo de cinco anos, todos os pacientes que exibiram a forma viscerocutânea do loxoscelismo foram investigados para a deficiĂŞncia de glicose-6 fosfato desidrogenase eritrocitária, e em dois pacientes em um total de sete, esta deficiĂŞncia foi encontrada. Este achado sugere que esta deficiĂŞncia enzimática genĂ©tica poderia ser uma das causas da hemĂłlise que ocorre em decorrĂŞncia da picada por Loxosceles.In a period of time of five years, all patients who exhibited viscerocutaneous form of loxoscelism were investigated for erythrocyte glucose-6-phosphate deficiency, and in two patients out of seven it was found this deficiency. This finding suggests that this genetical enzyme deficiency could account for the hemolysis after Loxosceles bite, at least in some of the cases
First record of Ozognathus cornutus (LeConte, 1859) (Coleoptera Ptinidae) from Sardinia, Italy.
Ozognathus cornutus (LeConte, 1859) is recorded on the invasive alien tree Robinia pseudocacia L. (Fabaceae) in an urban area in Italy. The species has already been reported in two other Italian regions, but this is the first record for the Sardinian fauna. Due to the great adaptability of this alien species and the increasingly cosmopolitan trend, we suggest monitoring it with caution for early detection and to plan an appropriate rapid management response

Changing Style in Style-Changing Industries: The Role of Critics as Gatekeepers in High-End Fashion
Mastering aesthetics is a precious source of competitive advantage in creative industries. In fashion, innovation is reflected by how and how much styles change. Elite designers claim to be the only endogenous force shaping fashion innovation season by season. Yet, each season, fashion critics vet the new collections these designers introduce, assessing what is original as opposed toreworked and uninspired, in this way playing a fundamental role as gatekeepers in setting taste within the industry. In this research, we document how stylistic innovation, vis-`a-vis the styles premier design houses introduced each season, is impacted, among the others, by the specific exogenous force of critics’ assessments of designers’ past work. Our data, which include 61 measures detailing the styles introduced by 38 prestigious Italian and French design houses over a nine-year period, suggest designers move further away from styles reviewed less favourably while adhering more closely to styles reviewed more positively. Additionally, the styles a designer introduces are shown to depend on critical assessments of competing designers’ styles, revealing how design houses attend to each other’s work. This work documents the strong correlation between style dynamics and critics’ feedback. It also has important implications for any company trying to find a balance between independence and conformity in setting its own unique positioning into the market
Focusing on the role of abiotic and biotic drivers on cross-taxon congruence
Diversity patterns can show congruence across taxonomic groups. Consistent diversity patterns allow the identification of indicator surrogates potentially representative of unobserved taxa or the broader biodiversity patterns. However, the effective use of biodiversity surrogates depends on underlying mechanisms driving the strength of the relationship among taxonomic groups. Here, we explored congruence patterns in community composition among taxa occupying different trophic levels, accounting for abiotic and biotic factors: vascular plants and six groups of ground-dwelling arthropods (pseudoscorpions, spiders, darkling beetles, rove beetles, ground beetles and ants) were chosen as potential indicator surrogates. We evaluated the cross-taxon relationships using Mantel test; subsequently, we investigated if these relationships could partially depend on abiotic drivers, using partial Mantel tests; then, we evaluated the partial contributions of abiotic and biotic drivers in explaining these relationships through a series of variation partitioning analyses. Our results showed that a consistent cross-taxon congruence pattern was evident across almost all group pairs: pseudoscorpions, spiders, ground beetles and vascular plants showed the largest number of significant correlations with other taxa. Environmental gradients resulted as drivers of cross-taxon congruence, shaping composition patterns. However, they were not the only ones. Biotic drivers account for part of cross-taxon congruence among vascular plants and arthropod predators (i.e., pseudoscorpions and spiders, but also ground beetles), as well as among taxa at high trophic levels. Almost all strictly predatory taxa, known as biological control agents, emerged as the best predictors of plant community composition even when the role of environmental factors was considered. Spiders/ants and spiders/ground beetles showed close relationships and congruent composition patterns, irrespective of environmental parameters. Relationships among taxa might be driven by several complex biotic interactions (e.g., non-trophic and trophic interactions, direct and indirect interactions). Bottom-up and top-down forces, consumptive and non-consumptive interactions may play a role in influencing the community composition of taxa and driving the observed relationships. Future studies should broaden knowledge about the role of these forces and interactions in determining the congruence across taxa. The multi-trophic perspective in cross-taxon studies can be promising for identifying biodiversity surrogates and their application in conservation planning
