    Interactions between individual plant species and soil nutrient status in Shortgrass Steppe

    Includes bibliographical references (pages 1131-1133).The effect of plant community structure on nutrient cycling is fundamental to our understanding of ecosystem function. We examined the importance of plant species and plant cover (i.e., plant covered microsites vs. bare soil) on nutrient cycling in shortgrass steppe of northeastern Colorado. We tested the effects of both plant species and cover on soils in an area of undisturbed shortgrass steppe and an area that had undergone nitrogen and water additions from 1971 to 1974, resulting in significant shifts in plant species composition. Soils under plants had consistently higher C and N mineralization rates and, in some cases, higher total and microbial C and N levels than soils without plant cover. Four native grasses, one sedge, and one shrub differed from one another in the quantity and quality of above- and belowground biomass but differences among the six species in soil nutrient cycling under their canopies were slight. However, soils under bunchgrasses tended to have higher C mineralization and microbial biomass C than soil under the rhizomatous grass, Agropyron smithii. Also, the one introduced annual in the study, Kochia scoparia, had soils with less plant-induced heterogeneity and higher rates of C and N mineralization as well as higher levels of microbial biomass C than soils associated with the other species. This species was abundant only on plots that had received water and nitrogen for a 4-yr period that ended 20 year ago, where it has persisted in the absence of resource additions for 20 yr, suggesting a positive feedback between plant persistence and soil nutrient status. Plant cover patterns had larger effects on ecosystem scale estimates of soil properties than the attributes of a particular plant species. This result may be due to the semiarid nature of this grassland in which plant cover is discontinuous and decomposition and nutrient availability are primarily limited by water, not by plant species-mediated characteristics such as litter quality. That local plant-induced patterns in soil properties significantly affected ecosystem scale estimates of these properties indicates that consideration of structural attributes, particularly plant cover patterns, is critical to estimates of ecosystem function in shortgrass steppe

    SIV-specific CD8+ T cells are clonotypically distinct across lymphoid and mucosal tissues

    CD8+ T cell responses are necessary for immune control of simian immunodeficiency virus (SIV). However, the key parameters that dictate antiviral potency remain elusive, conceivably because most studies to date have been restricted to analyses of circulating CD8+ T cells. We conducted a detailed clonotypic, functional, and phenotypic survey of SIV-specific CD8+ T cells across multiple anatomical sites in chronically infected rhesus macaques with high (>10,000 copies/mL plasma) or low burdens of viral RNA (<10,000 copies/mL plasma). No significant differences in response magnitude were identified across anatomical compartments. Rhesus macaques with low viral loads (VLs) harbored higher frequencies of polyfunctional CXCR5+ SIV-specific CD8+ T cells in various lymphoid tissues and higher proportions of unique Gag-specific CD8+ T cell clonotypes in the mesenteric lymph nodes relative to rhesus macaques with high VLs. In addition, public Gag-specific CD8+ T cell clonotypes were more commonly shared across distinct anatomical sites than the corresponding private clonotypes, which tended to form tissue-specific repertoires, especially in the peripheral blood and the gastrointestinal tract. Collectively, these data suggest that functionality and tissue localization are important determinants of CD8+ T cell–mediated efficacy against SIV

Because performance appraisal is an applied topic, it is useful to periodically consider the current state of performance research and its relation to performance appraisal practice. This review examines the performance appraisal literature published in both academic and practitioner outlets between 1985 and 1990, briefly discusses the current state of performance appraisal practice, highlights the juxtaposition of research and practice, and suggests directions for further research

    Substrate age and tree islands influence carbon and nitrogen dynamics across a retrogressive semiarid chronosequence

    The long-term dynamics of carbon (C) and nitrogen (N) in semiarid ecosystems remain poorly understood. We measured pools and fluxes of surface soil C and N, as well as other soil properties, under tree canopies and in intercanopy spaces at four sites that form a volcanic substrate age gradient in semiarid piñon-juniper woodlands of northern Arizona, United States. Clay content and soil water-holding capacity increased consistently with substrate age, but both soil organic C and N increased only up to the 750,000 year site and then declined at the oldest (3,000,000 year) site. Measures of soil C and N flux displayed a similar pattern to total C and N pools. Pools and fluxes of C and N among the three canopy types became more homogeneous with substrate age up to the 750,000 year site, but disparity between tree and intercanopy microsites widened again at the oldest site. The δ15N of both tree leaves and surface soils became progressively more enriched across the substrate age gradient, consistent with a N cycle increasingly dominated by isotope fractionating losses. Our results point to consistencies in patterns of ecosystem development between semiarid and more humid ecosystems and suggest that pedogenic development may be an important factor controlling the spatial distribution of soil resources in semiarid ecosystems. These data should help both unify and broaden current theory of terrestrial ecosystem development

    Axillary lymph node dissection for breast cancer utilizing Harmonic Focus®

    <p>Abstract</p> <p>Background</p> <p>For patients with axillary lymph node metastases from breast cancer, performance of a complete axillary lymph node dissection (ALND) is the standard approach. Due to the rich lymphatic network in the axilla, it is necessary to carefully dissect and identify all lymphatic channels. Traditionally, these lymphatics are sealed with titanium clips or individually sutured. Recently, the Harmonic Focus<sup>®</sup>, a hand-held ultrasonic dissector, allows lymphatics to be sealed without the utilization of clips or ties. We hypothesize that ALND performed with the Harmonic Focus<sup>® </sup>will decrease operative time and reduce post-operative complications.</p> <p>Methods</p> <p>Retrospective review identified all patients who underwent ALND at a teaching hospital between January of 2005 and December of 2009. Patient demographics, presenting pathology, treatment course, operative time, days to drain removal, and surgical complications were recorded. Comparisons were made to a selected control group of patients who underwent similar surgical procedures along with an ALND performed utilizing hemostatic clips and electrocautery. A total of 41 patients were included in this study.</p> <p>Results</p> <p>Operative time was not improved with the use of ultrasonic dissection, however, there was a decrease in the total number of days that closed suction drainage was required, although this was not statistically significant. Complication rates were similar between the two groups.</p> <p>Conclusion</p> <p>In this case-matched retrospective review, there were fewer required days of closed suction drainage when ALND was performed with ultrasonic dissection versus clips and electrocautery.</p

    A standard protocol to report discrete stage-structured demographic information

    Stage-based demographic methods, such as matrix population models (MPMs), are powerful tools used to address a broad range of fundamental questions in ecology, evolutionary biology and conservation science. Accordingly, MPMs now exist for over 3000 species worldwide. These data are being digitised as an ongoing process and periodically released into two large open-access online repositories: the COMPADRE Plant Matrix Database and the COMADRE Animal Matrix Database. During the last decade, data archiving and curation of COMPADRE and COMADRE, and subsequent comparative research, have revealed pronounced variation in how MPMs are parameterized and reported. Here, we summarise current issues related to the parameterisation and reporting of MPMs that arise most frequently and outline how they affect MPM construction, analysis, and interpretation. To quantify variation in how MPMs are reported, we present results from a survey identifying key aspects of MPMs that are frequently unreported in manuscripts. We then screen COMPADRE and COMADRE to quantify how often key pieces of information are omitted from manuscripts using MPMs. Over 80% of surveyed researchers (n = 60) state a clear benefit to adopting more standardised methodologies for reporting MPMs. Furthermore, over 85% of the 300 MPMs assessed from COMPADRE and COMADRE omitted one or more elements that are key to their accurate interpretation. Based on these insights, we identify fundamental issues that can arise from MPM construction and communication and provide suggestions to improve clarity, reproducibility and future research utilising MPMs and their required metadata. To fortify reproducibility and empower researchers to take full advantage of their demographic data, we introduce a standardised protocol to present MPMs in publications. This standard is linked to www.compa dre-db.org, so that authors wishing to archive their MPMs can do so prior to submission of publications, following examples from other open-access repositories such as DRYAD, Figshare and Zenodo. Combining and standardising MPMs parameterized from populations around the globe and across the tree of life opens up powerful research opportunities in evolutionary biology, ecology and conservation research. However, this potential can only be fully realised by adopting standardised methods to ensure reproducibility

    Efficacy of tissue brushing as measured by the prosthodontic tissue index

    This study was conducted to determine the efficacy of brushing the oral mucosa supporting complete dentures with a soft brush to see if this treatment would reduce inflammation. The oral mucosa health status of 60 patients was monitored for 120 days using the PTI to measure inflammation. For comparison the patients were divided into two groups, and every effort was made to balance the groups for those variables that may effect inflammation. The patients were also compared with themselves. The experiment consisted of three phases; Phase 1 established baseline data, and Phases 2 and 3 were information-gathering sessions. Each patient received brushing instructions at the start of the brushing test period and additional brushing instruction during a reinforcement session after 30 days.
