Host Adaptation Through Hybridization: Genome Analysis of Triticale Powdery Mildew Reveals Unique Combination of Lineage-Specific Effectors
The emergence of new fungal pathogens through hybridization represents a serious challenge for agriculture. Hybridization between the wheat mildew (Blumeria graminis f. sp. tritici) and rye mildew (B. graminis f. sp. secalis) pathogens has led to the emergence of a new mildew form (B. graminis f. sp. triticale) growing on triticale, a man-made amphiploid crop derived from crossing rye and wheat, which was originally resistant to the powdery mildew disease. The identification of the genetic basis of host adaptation in triticale mildew has been hampered by the lack of a reference genome. Here, we report the 141.4-Mb reference assembly of triticale mildew isolate THUN-12 derived from long-read sequencing and genetic map-based scaffolding. All 11 triticale mildew chromosomes were assembled from telomere-to-telomere and revealed that 19.7% of the hybrid genome was inherited from the rye mildew parental lineage. We identified lineage-specific regions in the hybrid, inherited from the rye or wheat mildew parental lineages, that harbor numerous bona fide candidate effectors. We propose that the combination of lineage-specific effectors in the hybrid genome is crucial for host adaptation, allowing the fungus to simultaneously circumvent the immune systems contributed by wheat and rye in the triticale crop. In line with this, we demonstrate the functional transfer of the SvrPm3 effector from wheat to triticale mildew, a virulence effector that specifically suppresses resistance of the wheat Pm3 allelic series. This transfer is the likely underlying cause for the observed poor effectiveness of several Pm3 alleles against triticale mildew and exemplifies the negative implications of pathogen hybridizations on resistance breeding. The broad use of the Pm8 resistance gene in wheat resulted in hypermutation of the AvrPm8 gene in the powdery mildew pathogen
Background: Worldwide wheat production is under constant threat by fast-evolving fungal pathogens. In the last decades, wheat breeding for disease resistance heavily relied on the introgression of chromosomal segments from related species as genetic sources of new resistance. The Pm8 resistance gene against the powdery mildew disease has been introgressed from rye into wheat as part of a large 1BL.1RS chromosomal translocation encompassing multiple disease resistance genes and yield components. Due to its high agronomic value, this translocation has seen continuous global use since the 1960s on large growth areas, even after Pm8 resistance was overcome by the powdery mildew pathogen. The long-term use of Pm8 at a global scale provided the unique opportunity to study the consequences of such extensive resistance gene application on pathogen evolution.
Results: Using genome-wide association studies in a population of wheat mildew isolates, we identified the avirulence effector AvrPm8 specifically recognized by Pm8. Haplovariant mining in a global mildew population covering all major wheat growing areas of the world revealed 17 virulent haplotypes of the AvrPm8 gene that grouped into two functional categories. The first one comprised amino acid polymorphisms at a single position along the AvrPm8 protein, which we confirmed to be crucial for the recognition by Pm8. The second category consisted of numerous destructive mutations to the AvrPm8 open reading frame such as disruptions of the start codon, gene truncations, gene deletions, and interference with mRNA splicing. With the exception of a single, likely ancient, gain-of-virulence mutation found in mildew isolates around the world, all AvrPm8 virulence haplotypes were found in geographically restricted regions, indicating that they occurred recently as a consequence of the frequent Pm8 use.
Conclusions: In this study, we show that the broad and prolonged use of the Pm8 gene in wheat production worldwide resulted in a multitude of gain-of-virulence mechanisms affecting the AvrPm8 gene in the wheat powdery mildew pathogen. Based on our findings, we conclude that both standing genetic variation as well as locally occurring new mutations contributed to the global breakdown of the Pm8 resistance gene introgression
Comparison of different CT metal artifact reduction strategies for standard titanium and carbon-fiber reinforced polymer implants in sheep cadavers
BACKGROUND
CT artifacts induced by orthopedic implants can limit image quality and diagnostic yield. As a number of different strategies to reduce artifact extent exist, the aim of this study was to systematically compare ex vivo the impact of different CT metal artifact reduction (MAR) strategies on spine implants made of either standard titanium or carbon-fiber-reinforced-polyetheretherketone (CFR-PEEK).
METHODS
Spine surgeons fluoroscopically-guided prepared six sheep spine cadavers with pedicle screws and rods of either titanium or CFR-PEEK. Samples were subjected to single- and dual-energy (DE) CT-imaging. Different tube voltages (80, DE mixed, 120 and tin-filtered 150 kVp) at comparable radiation dose and iterative reconstruction versus monoenergetic extrapolation (ME) techniques were compared. Also, the influence of image reconstruction kernels (soft vs. bone tissue) was investigated. Qualitative (Likert scores) and quantitative parameters (attenuation changes induced by implant artifact, implant diameter and image noise) were evaluated by two independent radiologists. Artifact degree of different MAR-strategies and implant materials were compared by multiple ANOVA analysis.
RESULTS
CFR-PEEK implants induced markedly less artifacts than standard titanium implants (p < .001). This effect was substantially larger than any other tested MAR technique. Reconstruction algorithms had small impact in CFR-PEEK implants and differed significantly in MAR efficiency (p < .001) with best MAR performance for DECT ME 130 keV (bone kernel). Significant differences in image noise between reconstruction kernels were seen (p < .001) with minor impact on artifact degree.
CONCLUSIONS
CFR-PEEK spine implants induce significantly less artifacts than standard titanium compositions with higher MAR efficiency than any alternate scanning or image reconstruction strategy. Interferon-gamma in the first-line therapy of ovarian cancer: a randomized phase III trial
Intraperitoneal treatment with interferon-γ (IFN-γ) has been shown to achieve surgically documented responses in the second-line therapy of ovarian cancer. To assess its efficacy in the first-line therapy, we conducted a randomized controlled trial with 148 patients who had undergone primary surgery for FIGO stage Ic–IIIc ovarian cancer. In the control arm women received 100 mg m−2cisplatin and 600 mg m−2cyclophosphamide, the experimental arm included the above regimen with IFN-γ 0.1 mg subcutaneously on days 1, 3, 5, 15, 17 and 19 of each 28-day cycle. Progression-free survival at 3 years was improved from 38% in controls to 51% in the treatment group corresponding to median times to progression of 17 and 48 months (P = 0.031, relative risk of progression 0.48, confidence interval 0.28–0.82). Three-year overall survival was 58% and 74% accordingly (n.s., median not yet reached). Complete clinical responses were observed in 68% with IFN-γ versus 56% in controls (n.s.). Toxicity was comparable in both groups except for a mild flu-like syndrome, experienced by most patients after administration of IFN-γ. Thus, with acceptable toxicity, the inclusion of IFN-γ in the first-line chemotherapy of ovarian cancer yielded a benefit in prolonging progression-free survival. © 2000 Cancer Research Campaig

In applying a strategy, structure, ownership and performance (SSOP) framework to three major clearing banks (ABN AMRO, UBS, Barclays), this article debates whether the conclusions generated by Whittington and Mayer about European manufacturing industry can be applied to the financial services sector. While European integration plays a key role in determining strategy, it is clear that global factors were far more important in determining management actions, leading to significant differences in structural adaptation. The article also debates whether this has led to improved performance, given the problems experienced with both geographical dispersion and diversification, bringing into question the quality of decision-making over the long term
Identification of the Orbital Pairing Symmetry in UPt_3
This paper summarizes the results of a comprehensive analysis of the
thermodynamic and transport data for the superconducting phases of UPt_3.
Calculations of the transverse sound attenuation as a function of temperature,
frequency, polarization, and disorder are presented for the leading models of
the superconducting order parameter. Measurements of the specific heat, thermal
conductivity, and transverse sound attenuation place strong constraints on the
orbital symmetry of the superconducting order parameter. We show that the
superconducting A and B phases are in excellent agreement with pairing states
belonging to the odd-parity E_{2u} orbital representation.Comment: 11 pages with 7 figure
The molecular environment of the massive star forming region NGC 2024: Multi CO transition analysis
NGC 2024, a sites of massive star formation, have complex internal structures
caused by cal heating by young stars, outflows, and stellar winds. These
complex cloud structures lead to intricate emission line shapes. The goal of
this paper is to show that the complex line shapes of 12 CO lines in NGC 2024
can be explained consistently with a model, whose temperature and velocity
structure are based on the well-established scenario of a PDR and the Blister
model. We present velocity-resolved spectra of seven CO lines ranging from J=3
to J=13, and we combined these data with CO high-frequency data from the ISO
satellite. We find that the bulk of the molecular cloud associated with NGC
2024 consists of warm (75 K) and dense (9e5 cm-3) gas. An additional hot (~ 300
K) component, located at the interface of the HII region and the molecular
cloud, is needed to explain the emission of the high-J CO lines. Deep
absorption notches indicate that very cold material (20 K) exists in front of
the warm material, too. A temperature and column density structure consistent
with those predicted by PDR models, combined with the velocity structure of a
Blister model, appropriately describes the observed emission line profiles of
this massive star forming region. This case study of NGC 2024 shows that, with
physical insights into these complex regions and careful modeling, multi-line
observations of CO can be used to derive detailed physical conditions in

First observations with CONDOR, a 1.5 THz heterodyne receiver
The THz atmospheric windows centered at roughly 1.3 and 1.5~THz, contain
numerous spectral lines of astronomical importance, including three high-J CO
lines, the N+ line at 205 microns, and the ground transition of para-H2D+. The
CO lines are tracers of hot (several 100K), dense gas; N+ is a cooling line of
diffuse, ionized gas; the H2D+ line is a non-depleting tracer of cold (~20K),
dense gas. As the THz lines benefit the study of diverse phenomena (from
high-mass star-forming regions to the WIM to cold prestellar cores), we have
built the CO N+ Deuterium Observations Receiver (CONDOR) to further explore the
THz windows by ground-based observations. CONDOR was designed to be used at the
Atacama Pathfinder EXperiment (APEX) and Stratospheric Observatory For Infrared
Astronomy (SOFIA). CONDOR was installed at the APEX telescope and test
observations were made to characterize the instrument. The combination of
CONDOR on APEX successfully detected THz radiation from astronomical sources.
CONDOR operated with typical Trec=1600K and spectral Allan variance times of
30s. CONDOR's first light observations of CO 13-12 emission from the hot core
Orion FIR4 (= OMC1 South) revealed a narrow line with T(MB) = 210K and
delta(V)=5.4km/s. A search for N+ emission from the ionization front of the
Orion Bar resulted in a non-detection. The successful deployment of CONDOR at
APEX demonstrates the potential for making observations at THz frequencies from

The APEX-CHAMP+ view of the Orion Molecular Cloud 1 core - Constraining the excitation with submillimeter CO multi-line observations
A high density portion of the Orion Molecular Cloud 1 (OMC-1) contains the
prominent, warm Kleinmann-Low (KL) nebula that is internally powered by an
energetic event plus a farther region in which intermediate to high mass stars
are forming. Its outside is affected by ultraviolet radiation from the
neighboring Orion Nebula Cluster and forms the archetypical photon-dominated
region (PDR) with the prominent bar feature. Its nearness makes the OMC-1 core
region a touchstone for research on the dense molecular interstellar medium and
PDRs. Using the Atacama Pathfinder Experiment telescope (APEX), we have imaged
the line emission from the multiple transitions of several carbon monoxide (CO)
isotopologues over the OMC-1 core region. Our observations employed the 2x7
pixel submillimeter CHAMP+ array to produce maps (~ 300 arcsec x 350 arcsec) of
12CO, 13CO, and C18O from mid-J transitions (J=6-5 to 8-7). We also obtained
the 13CO and C18O J=3-2 images toward this region. The 12CO line emission shows
a well-defined structure which is shaped and excited by a variety of phenomena,
including the energetic photons from hot, massive stars in the nearby Orion
Nebula's central Trapezium cluster, active high- and intermediate-mass star
formation, and a past energetic event that excites the KL nebula. Radiative
transfer modeling of the various isotopologic CO lines implies typical H2
densities in the OMC-1 core region of ~10^4-10^6 cm^-3 and generally elevated
temperatures (~ 50-250 K). We estimate a warm gas mass in the OMC-1 core region

Photon Dominated Regions in NGC 3603
Aims: We aim at deriving the excitation conditions of the interstellar gas as
well as the local FUV intensities in the molecular cloud surrounding NGC 3603
to get a coherent picture of how the gas is energized by the central stars.
Methods: The NANTEN2-4m submillimeter antenna is used to map the [CI] 1-0, 2-1
and CO 4-3, 7-6 lines in a 2' x 2' region around the young OB cluster NGC 3603
YC. These data are combined with C18O 2-1 data, HIRES-processed IRAS 60 and 100
micron maps of the FIR continuum, and Spitzer/IRAC maps. Results: The NANTEN2
observations show the presence of two molecular clumps located south-east and
south-west of the cluster and confirm the overall structure already found by
previous CS and C18O observations. We find a slight position offset of the peak
intensity of CO and [CI], and the atomic carbon appears to be further extended
compared to the molecular material. We used the HIRES far-infrared dust data to
derive a map of the FUV field heating the dust. We constrain the FUV field to
values of \chi = 3 - 6 \times 10^3 in units of the Draine field across the
clouds. Approximately 0.2 to 0.3 % of the total FUV energy is re-emitted in the
[CII] 158 {\mu}m cooling line observed by ISO. Applying LTE and escape
probability calculations, we derive temperatures (TMM1 = 43 K, TMM2 = 47 K),
column densities (N(MM1) = 0.9 \times 10^22 cm^-2, N(MM2) = 2.5 \times 10^22
cm^-2) and densities (n(MM1) = 3 \times 10^3 cm^-3, n(MM2) = 10^3 -10^4 cm^-3)
for the two observed molecular clumps MM1 and MM2. Conclusions: The cluster is
strongly interacting with the ambient molecular cloud, governing its structure
and physical conditions. A stability analysis shows the existence of
gravitationally collapsing gas clumps which should lead to star formation.
Embedded IR sources have already been observed in the outskirts of the
