    Autodisplay for the co-expression of lipase and foldase on the surface of E. coli: washing with designer bugs

    Background: Lipases including the lipase from Burkholderia cepacia are in a main focus in biotechnology research since many years because of their manifold possibilities for application in industrial processes. The application of Burkholderia cepacia lipase for these processes appears complicated because of the need for support by a chaperone, the lipase specific foldase. Purification and reconstitution protocols therefore interfere with an economic implementation of such enzymes in industry. Autodisplay is a convenient method to express a variety of passenger proteins on the surface of E. coli. This method makes subsequent purification steps to obtain the protein of interest unnecessary. If enzymes are used as passengers, the corresponding cells can simply be applied as whole cell biocatalysts. Furthermore, enzymes surface displayed in this manner often acquire stabilization by anchoring within the outer membrane of E. coli. Results: The lipase and its chaperone foldase from B. cepacia were co-expressed on the surface of E. coli via autodisplay. The whole cell biocatalyst obtained thereby exhibited an enzymatic activity of 2.73 mU mL-1 towards the substrate p-nitrophenyl palmitate when applied in an OD578 =1. Outer membrane fractions prepared from the same culture volume showed a lipase activity of 4.01 mU mL-1. The lipase-whole cell biocatalyst as well as outer membrane preparations thereof were used in a standardized laundry test, usually adopted to determine the power of washing agents. In this test, the lipase whole cell biocatalyst and the membrane preparation derived thereof exhibited the same lipolytic activity as the purified lipase from B. cepacia and a lipase preparation which is already applied in commercial washing agents. Conclusions: Co-expression of both the lipase and its chaperone foldase on the surface of E. coli yields a lipid degrading whole cell biocatalyst. Therefore the chaperone supported folding process, absolutely required for the lipolytic activity appears not to be hindered by surface display. Furthermore, the cells and the membrane preparations appeared to be stable enough to endure a European standard laundry test and show efficient fat removal properties herein.<br

    Crystallographic structure of ultrathin Fe films on Cu(100)

    We report bcc-like crystal structures in 2-4 ML Fe films grown on fcc Cu(100) using scanning tunneling microscopy. The local bcc structure provides a straightforward explanation for their frequently reported outstanding magnetic properties, i.e., ferromagnetic ordering in all layers with a Curie temperature above 300 K. The non-pseudomorphic structure, which becomes pseudomorphic above 4 ML film thickness is unexpected in terms of conventional rules of thin film growth and stresses the importance of finite thickness effects in ferromagnetic ultrathin films.Comment: 4 pages, 3 figures, RevTeX/LaTeX2.0

    Modelling health and economic impact of nutrition interventions: a systematic review

    Diet related non-communicable diseases (NCDs), as well as micronutrient deficiencies, are of widespread and growing importance to public health. Authorities are developing programs to improve nutrient intakes via foods. To estimate the potential health and economic impact of these programs there is a wide variety of models. The aim of this review is to evaluate existing models to estimate the health and/or economic impact of nutrition interventions with a focus on reducing salt and sugar intake and increasing vitamin D, iron, and folate/folic acid intake. The protocol of this systematic review has been registered with the International Prospective Register of Systematic Reviews (PROSPERO: CRD42016050873). The final search was conducted on PubMed and Scopus electronic databases and search strings were developed for salt/sodium, sugar, vitamin D, iron, and folic acid intake. Predefined criteria related to scientific quality, applicability, and funding/interest were used to evaluate the publications. In total 122 publications were included for a critical appraisal: 45 for salt/sodium, 61 for sugar, 4 for vitamin D, 9 for folic acid, and 3 for iron. The complexity of modelling the health and economic impact of nutrition interventions is dependent on the purpose and data availability. Although most of the models have the potential to provide projections of future impact, the methodological challenges are considerable. There is a substantial need for more guidance and standardization for future modelling, to compare results of different studies and draw conclusions about the health and economic impact of nutrition interventions. © 2022, The Author(s)

    Triggered crustal earthquake swarm across subduction segment boundary after the 2016 Pedernales, Ecuador megathrust earthquake

    Megathrust ruptures and the ensuing postseismic deformation cause stress changes that may induce seismicity on upper plate crustal faults far from the coseismic rupture area. In this study, we analyze seismic swarms that occurred in the north Ecuador area of Esmeraldas, beginning two months after the 2016 Mw_{w} 7.8 Pedernales, Ecuador megathrust earthquake. The Esmeraldas region is 70 km from the Pedernales rupture area in a separate segment of the subduction zone. We characterize the Esmeraldas sequence, relocating the events using manual arrival time picks and a local a-priori 3D velocity model. The earthquake locations from the Esmeraldas sequence outline an upper plate fault or shear zone. The sequence contains one major swarm and several smaller swarms. Moment tensor solutions of several events include normal and strike-slip motion and non-double-couple components. During the main swarm, earthquake hypocenters increase in distance from the first event over time, at a rate of a few hundred meters per day, consistent with fluid diffusion. Events with similar waveforms occur within the sequence, and a transient is seen in time series of nearby GPS stations concurrent with the seismicity. The events with similar waveforms and the transient in GPS time series suggest that slow aseismic slip took place along a crustal normal fault during the sequence. Coulomb stress calculations show a positive Coulomb stress change in the Esmeraldas region, consistent with seismicity being triggered by the Pedernales mainshock and large aftershocks. The characteristics of the seismicity indicate that postseismic deformation involving fluid flow and slow slip activated upper plate faults in the Esmeraldas area. These findings suggest the need for further investigation into the seismic hazard potential of shallow upper plate faults and the potential for megathrust earthquakes to trigger slow-slip and shallow seismicity across separate segments of subduction zones

    The tailings dam failure of 5 November 2015 in SE Brazil and its preceding seismic sequence

    The collapse of a mine tailings dam and subsequent flood in SE Brazil on 5 November 2015 was preceded by a small-magnitude seismic sequence. In this report, we explore the spatiotemporal associations between the seismic events and the accident and discuss their possible connection. We also analyze the signals generated by the turbulent mudflow, as recorded by the Brazilian Seismographic Network (RSBR). In light of our observations, we propose as possible contributing factor for the dam collapse either ground shaking and/or soil liquefaction triggered by the earthquakes. The possibility of such a small-magnitude earthquake contributing to the collapse of a tailings dam raises important concerns regarding safety and related legislation of dams in Brazil and the world. ©2016.     Nanostructured 3D Constructs Based on Chitosan and Chondroitin Sulphate Multilayers for Cartilage Tissue Engineering

    Nanostructured three-dimensional constructs combining layer-by-layer technology (LbL) and template leaching were processed and evaluated as possible support structures for cartilage tissue engineering. Multilayered constructs were formed by depositing the polyelectrolytes chitosan (CHT) and chondroitin sulphate (CS) on either bidimensional glass surfaces or 3D packet of paraffin spheres. 2D CHT/CS multi-layered constructs proved to support the attachment and proliferation of bovine chondrocytes (BCH). The technology was transposed to 3D level and CHT/CS multi-layered hierarchical scaffolds were retrieved after paraffin leaching. The obtained nanostructured 3D constructs had a high porosity and water uptake capacity of about 300%. Dynamical mechanical analysis (DMA) showed the viscoelastic nature of the scaffolds. Cellular tests were performed with the culture of BCH and multipotent bone marrow derived stromal cells (hMSCs) up to 21 days in chondrogenic differentiation media. Together with scanning electronic microscopy analysis, viability tests and DNA quantification, our results clearly showed that cells attached, proliferated and were metabolically active over the entire scaffold. Cartilaginous extracellular matrix (ECM) formation was further assessed and results showed that GAG secretion occurred indicating the maintenance of the chondrogenic phenotype and the chondrogenic differentiation of hMSCs

    Spin-resolved and temperature-dependent appearance-potential spectra of ferromagnetic Nickel are measured and analyzed theoretically. The Lander self-convolution model which relates the line shape to the unoccupied part of the local density of states turns out to be insufficient.     Structural Control on Megathrust Rupture and Slip Behavior: Insights From the 2016 Mw 7.8 Pedernales Ecuador Earthquake

    The heterogeneous seafloor topography of the Nazca Plate as it enters the Ecuador subduction zone provides an opportunity to document the influence of seafloor roughness on slip behavior and megathrust rupture. The 2016 Mw_{w} 7.8 Pedernales Ecuador earthquake was followed by a rich and active postseismic sequence. An internationally coordinated rapid response effort installed a temporary seismic network to densify coastal stations of the permanent Ecuadorian national seismic network. A combination of 82 onshore short and intermediate period and broadband seismic stations and six ocean bottom seismometers recorded the postseismic Pedernales sequence for over a year after the mainshock. A robust earthquake catalog combined with calibrated relocations for a subset of magnitude ≥4 earthquakes shows pronounced spatial and temporal clustering. A range of slip behavior accommodates postseismic deformation including earthquakes, slow slip events, and earthquake swarms. Models of plate coupling and the consistency of earthquake clustering and slip behavior through multiple seismic cycles reveal a segmented subduction zone primarily controlled by subducted seafloor topography, accreted terranes, and inherited structure. The 2016 Pedernales mainshock triggered moderate to strong earthquakes (5 ≤ M ≤ 7) and earthquake swarms north of the mainshock rupture close to the epicenter of the 1906Mw_{w} 8.8 earthquake and in the segment of the subduction zone that ruptured in 1958 in a Mw_{w} 7.7 earthquake

    Seismic signature of mudflow tremor resulted from Brumadinho (Brazil) tailings dam failure

    Mudflow is often associated with seismic activities. The present study applied a seismic based detection of the surface waves generated by the mudflow of Brumadinho dam collapse using records of Brazilian Seismographic Network. The signal envelope and time-frequency spectrograms of the mudflow signals were used in the analysis. As a result, the mudflow signals were successfully detected from the data recorded at a nearby seismic station.
