    Inhibition of interleukin-1β-stimulated collagenase and stromelysin expression in human tendon fibroblasts by epigallocatechin gallate ester

    The medicinal benefits of green tea (Camellia sinensis) consumption have been attributed to bioavailable polyphenols, notably epigallocatechin gallate (EGCG). We have assessed the effects of EGCG and its non-esterified counterpart EGC on the expression of the collagenases, matrix metalloproteinases (MMP)-1 and -13, and the stromelysin, MMP-3, in human tendon-derived fibroblasts. Interleukin (IL)-1ß increased MMP-1, -3 and -13 mRNA and output at least 30-fold. EGCG reduced this stimulation, by 20–30% at 2.5 µM and more than 80% at 25 µM, and had a smaller effect on MMP-2 mRNA expression, which was not stimulated by IL-1ß. In all experiments EGCG was at least 10-fold more potent than EGC. EGCG reduced the stimulation of p54 JNK/SAPK phosphorylation by IL-1ß but did not affect p38 MAPK phosphorylation, the degradation of I?B or the activating phosphorylation of NF?B. We conclude that EGCG reduces the IL-1-stimulated expression of both collagenase and stromelysin mRNA species, an effect which may be mediated by inhibition of the JNK/SAPK pathway. Taken together with previous reports of EGCG effects on the expression and/or activity of gelatinases and aggrecanases, our results underline the importance of extracellular matrix breakdown as a potential target for the actions of green tea polyphenols

    A model to assess customer alignment through customer experience concepts

    Business and Information Technology Alignment (BITA) has been one of the main concerns of IT and Business executives and directors due to its importance to overall company performance, especially today in the age of digital transformation. For BITA has been developed several models which in general has focused in the implementation of alignment strategies for the internal operation of the organizations and in the measurement of this internal alignment, but, there is still a big gap in measurement models of the alignment with the external environment of the organizations. In this paper is presented the design and application of a maturity measurement model for BITA with the customers, where the customers are actors of the external environment of the companies.     Reconstitution of muscle F-actin from Atlantic salmon (Salmo salar L.) with carotenoids – binding characteristics of astaxanthin and canthaxanthin.

    The binding of carotenoids to the myofibrillar protein F-actin purified from the white muscle of Atlantic salmon (Salmo salar L.) was studied using in vitro reconstitution. The binding of astaxanthin and canthaxanthin was saturable, and analysis revealed the presence of a single carotenoid-binding site. The dissociation constants (Kd) for actin prepared from 2.5 Kg FW fish were 1.04 ± 0.13 μg carotenoid mg-1 actin and 0.54 ± 0.11 μgmg-1 for astaxanthin and canthaxanthin, respectively. The saturation binding level (Bmax) for astaxanthin was 1.39 ± 0.07 μgmg-1 and 1.04 ± 0.08 μgmg-1 for canthaxanthin. These values were higher for F-actin prepared from organic and small (~0.5 Kg FW) salmon than for non-organic and larger, mature fish. The structural specificity of carotenoid binding revealed a preference for carotenoids that possess a keto group at C-4 on the end-group of the molecule, but the presence of hydroxyl groups at C-3 or C-4 reduced overall binding efficiency. The study suggests that the ability of myofibrillar proteins to bind carotenoids is not a limiting factor governing the deposition of carotenoids in the muscle of salmonids

    Evaluation of Legacy Forest Harvesting Impacts on Dominant Stream Water Sources and Implications for Water Quality Using End Member Mixing Analysis

    Forests are critical water supply regions that are increasingly threatened by natural and anthropogenic disturbance. Evaluation of runoff-generating processes within harvested and undisturbed headwater catchments provides insight into disturbance impacts on water quality and drinking water treatability. In this study, an extensive hydrologic dataset collected at the experimental Turkey Lakes Watershed (TLW) located on the Canadian Shield was used to quantify sources of stormflow in legacy clear-cut (24-years post harvesting) and forested (control) headwater catchments using an end member mixing analysis (EMMA) model. Stream water, groundwater, soil water, and throughfall water quality were evaluated during spring snowmelt, stormflow, and fall wet-up. Groundwater chemistry was similar to stream water chemistry in both catchments, suggesting that groundwater is a major contributor to stream flow. The water chemistry in small wetlands within the study catchments was comparable to stream water chemistry, suggesting that wetlands are also important contributors to stream flow. Differences in wetland position between the legacy clear-cut and control catchments appeared to have a greater influence on source contributions than legacy harvesting. Results from this study provide insight into runoff-generation processes that reflect event/seasonal flow dynamics and the impacts on water quality

    A consistent analysis of (e,e'p) and (d,3He) experiments

    The apparent discrepancy between spectroscopic factors obtained in (e,e'p) and (d,3He) experiments is investigated. This is performed first for 48Ca(e,e'p) and 48Ca(d,3He) experiments and then for other nuclei. It is shown that the discrepancy disappears if the (d,3He) experiments are re-analyzed with a non-local finite range DWBA analysis with a bound-state wave function that is obtained from (e,e'p) experiments.Comment: 23 pages, 7 figure

    Changing forest water yields in response to climate warming: results from long-term experimental watershed sites across North America

    Climate warming is projected to affect forest water yields but the effects are expected to vary. We investigated how forest type and age affect water yield resilience to climate warming. To answer this question, we examined the variability in historical water yields at long-term experimental catchments across Canada and the United States over 5-year cool and warm periods. Using the theoretical framework of the Budyko curve, we calculated the effects of climate warming on the annual partitioning of precipitation (P) into evapotranspiration (ET) and water yield. Deviation (d) was defined as a catchment’s change in actual ET divided by P [AET/P; evaporative index (EI)] coincident with a shift from a cool to a warm period – a positive d indicates an upward shift in EI and smaller than expected water yields, and a negative d indicates a downward shift in EI and larger than expected water yields. Elasticity was defined as the ratio of inter annual variation in potential ET divided by P (PET/P; dryness index) to inter annual variation in the EI – high elasticity indicates low d despite large range in drying index (i.e., resilient water yields), low elasticity indicates high d despite small range in drying index (i.e., non-resilient water yields). Although the data needed to fully evaluate ecosystems based on these metrics are limited, we were able to identify some characteristics of response among forest types. Alpine sites showed the greatest sensitivity to climate warming with any warming leading to increased water yields. Conifer forests included catchments with lowest elasticity and stable to larger water yields. Deciduous forests included catchments with intermediate elasticity and stable to smaller water yields. Mixed coniferous/deciduous forests included catchments with highest elasticity and stable water yields. Forest type appeared to influence the resilience of catchment water yields to climate warming, with conifer and deciduous catchments more susceptible to climate warming than the more diverse mixed forest catchments
