Hidden walls: STEM course barriers identified by students with disabilities
Historically, non-disabled individuals have viewed disability as a personal
deficit requiring change to the disabled individual. However, models have
emerged from disability activists and disabled intellectuals that emphasize the
role of disabling social structures in preventing or hindering equal access
across the ability continuum. We used the social relational proposition, which
situates disability within the interaction of impairments and particular social
structures, to identify disabling structures in introductory STEM courses. We
conducted interviews with nine students who identified with a range of
impairments about their experiences in introductory STEM courses. We assembled
a diverse research team and analyzed the interviews through phenomenological
analysis. Participants reported course barriers that prevented effective
engagement with course content. These barriers resulted in challenges with time
management as well as feelings of stress and anxiety. We discuss
recommendations for supporting students to more effectively engage with
introductory STEM courses
On Lagrangian Coherent Structures in Laparoscopy
Laparoscopy is an electrosurgical medical operation often involving an
application of high-frequency alternating current to remove undesired
biological tissue from the insufflated abdomen accessible through inlet and
outlets trocars. One of the main byproducts in this process are the gaseous
particles, called surgical smoke, which is found hazardous for both the patient
and the operating room staff. The elimination of this hazardous material is an
area of active research in the medical community. Thus, understanding dynamics
influenced by the underlying flow inside the abdomen is crucial. In this
article, we propose a computational fluid dynamics model and analyse the
velocity field in an insufflated abdomen shaped domain by identifying the
Lagrangian Coherent Structures (LCS) that are responsible for the
transportation, mixing and accumulation of the material particles in the flow.
By calculating the mixing strength we show that the regions revealed by these
material curves are dependent on the angle, positions and number of the outlets
and inlets. Hence, a novel utility of LCS in medical surgery is presented that
can detail the dynamics of surgical smoke informing better design of effective
smoke removal technologies

International meta-analysis of PTSD genome-wide association studies identifies sex- and ancestry-specific genetic risk loci.
The risk of posttraumatic stress disorder (PTSD) following trauma is heritable, but robust common variants have yet to be identified. In a multi-ethnic cohort including over 30,000 PTSD cases and 170,000 controls we conduct a genome-wide association study of PTSD. We demonstrate SNP-based heritability estimates of 5-20%, varying by sex. Three genome-wide significant loci are identified, 2 in European and 1 in African-ancestry analyses. Analyses stratified by sex implicate 3 additional loci in men. Along with other novel genes and non-coding RNAs, a Parkinson's disease gene involved in dopamine regulation, PARK2, is associated with PTSD. Finally, we demonstrate that polygenic risk for PTSD is significantly predictive of re-experiencing symptoms in the Million Veteran Program dataset, although specific loci did not replicate. These results demonstrate the role of genetic variation in the biology of risk for PTSD and highlight the necessity of conducting sex-stratified analyses and expanding GWAS beyond European ancestry populations
A partial form of inherited human USP18 deficiency underlies infection and inflammation
International audienceHuman USP18 is an interferon (IFN)-stimulated gene product and a negative regulator of type I IFN (IFN-I) signaling. It also removes covalently linked ISG15 from proteins, in a process called deISGylation. In turn, ISG15 prevents USP18 from being degraded by the proteasome. Autosomal recessive complete USP18 deficiency is life-threatening in infancy owing to uncontrolled IFN-I–mediated autoinflammation. We report three Moroccan siblings with autoinflammation and mycobacterial disease who are homozygous for a new USP18 variant. We demonstrate that the mutant USP18 (p.I60N) is normally stabilized by ISG15 and efficient for deISGylation but interacts poorly with the receptor-anchoring STAT2 and is impaired in negative regulation of IFN-I signaling. We also show that IFN-γ–dependent induction of IL-12 and IL-23 is reduced owing to IFN-I–mediated impairment of myeloid cells to produce both cytokines. Thus, insufficient negative regulation of IFN-I signaling by USP18-I60N underlies a specific type I interferonopathy, which impairs IL-12 and IL-23 production by myeloid cells, thereby explaining predisposition to mycobacterial disease
Newborn DNA-methylation, childhood lung function, and the risks of asthma and COPD across the life course
Rationale: We aimed to identify differentially methylated regions (DMRs) in cord blood DNA associated with childhood lung function, asthma and chronic obstructive pulmonary disease (COPD) across the life course.
Methods: We meta-analysed epigenome-wide data of 1688 children from five cohorts to identify cord blood DMRs and their annotated genes, in relation to forced expiratory volume in 1 s (FEV1), FEV1/forced vital capacity (FVC) ratio and forced expiratory flow at 75% of FVC at ages 7-13 years. Identified DMRs were explored for associations with childhood asthma, adult lung function and COPD, gene expression and involvement in biological processes.
Results: We identified 59 DMRs associated with childhood lung function, of which 18 were associated with childhood asthma and nine with COPD in adulthood. Genes annotated to the top 10 identified DMRs were HOXA5, PAOX, LINC00602, ABCA7, PER3, CLCA1, VENTX, NUDT12, PTPRN2 and TCL1A. Differential gene expression in blood was observed for 32 DMRs in childhood and 18 in adulthood. Genes related with 16 identified DMRs were associated with respiratory developmental or pathogenic pathways.
Interpretation: Our findings suggest that the epigenetic status of the newborn affects respiratory health and disease across the life course
The Evolution of Primate Short-Term Memory.
Short-term memory is implicated in a range of cognitive abilities and is critical for understanding primate
cognitive evolution. To investigate the effects of phylogeny, ecology and sociality on short-term memory, we tested the largest and most diverse primate sample to date (421 non-human primates across 41 species) in an experimental delayed-response task. Our results confirm previous findings that longer delays decrease memory performance across species and taxa. Our analyses demonstrate a considerable contribution of phylogeny over ecological and social factors on the distribution of short-term memory performance in primates; closely related species had more similar short-term memory abilities. Overall, individuals in the branch of Hominoidea performed better compared to Cercopithecoidea, who in turn performed above Platyrrhini and Strepsirrhini. Interdependencies between phylogeny and socioecology of a given species presented an obstacle to disentangling the effects of each of these factors on the evolution of short-term memory capacity. However, this study offers an important step forward in understanding the interspecies and individual variation in short-term memory ability by providing the first phylogenetic reconstruction of this trait’s evolutionary history. The dataset constitutes a unique resource for studying the evolution of primate cognition and the role of short-term memory in other cognitive abilities.info:eu-repo/semantics/publishedVersio
