Variation in the chemical composition, physical characteristics and energy values of cereal grains produced in the Western Cape area of South Africa
Grain samples were produced at 10 different locations in the Western Cape region of South Africa, on 2.1 m x 6 m experimental plots, over a period of three years. Twenty different cereal grain cultivars were used in the study. A randomised square experimental design with four replicates per sample was used. An area of 1.35 m x 5 m from each plot was harvested during 1994, 1995 and 1996 and the yield was determined. Thousand seed mass (TSM) and hectolitre mass (HLM) were also determined. Samples were analysed for dry matter (DM), ash, crude protein (CP), ether extract (EE), neutral detergent fibre (NDF), acid detergent fibre (ADF) and in vitro organic matter digestibility (IVOMD). Digestible energy values (DE) for pigs were determined with a mobile nylon bag technique, while non-structural carbohydrate values (NSC) were calculated. In the first analysis, cultivars were compared by a one-way analysis of variance, followed by pooling of grain type data. Naked oats had the highest DE value, and the respective values (DM basis) for naked oats, wheat, triticale, 2-row brewer's barley, 6-row feed barley and oats were 18.0, 16.0, 15.8, 14.9, 14.4, and 12.6 MJ/kg DM. The high EE value of naked oats (97 g/kg) might be partly responsible for the high DE value. The 6-row and 2-row naked barley cultivars had the highest IVOMD (946 g/kg and 944 g/kg), followed by wheat (910 g/kg), triticale (905 g/kg), naked oats (899 g/kg), 2-row brewer's barley (882 g/kg), 6-row feed barley (844 g/kg) and oats (671 g/kg). Considerable variation was found between samples within a cultivar for DE and IVOMD. Two-row naked barley had the highest mean CP value (159 g/kg) followed by naked oats (159 g/kg), 6-row naked barley (154 g/kg), wheat (148 g/kg), triticale (146 g/kg), oats (143 g/kg), 2-row brewer's barley (136 g/kg) and 6-row feed barley (135 g/kg) on DM basis. Triticale had the highest yield, with naked oats and barley cultivars having the lowest yield.

Single-Cell Analysis Uncovers Osteoblast Factor Growth Differentiation Factor 10 as Mediator of Vascular Smooth Muscle Cell Phenotypic Modulation Associated with Plaque Rupture in Human Carotid Artery Disease
(1) Background: Vascular smooth muscle cells (VSMCs) undergo a complex phenotypic switch in response to atherosclerosis environmental triggers, contributing to atherosclerosis disease progression. However, the complex heterogeneity of VSMCs and how VSMC dediffer-entiation affects human carotid artery disease (CAD) risk has not been clearly established. (2) Method: A single-cell RNA sequencing analysis of CD45 12 cells derived from the atherosclerotic aorta of Apolipoprotein E-deficient (Apoe 12/ 12) mice on a normal cholesterol diet (NCD) or a high cholesterol diet (HCD), respecting the site-specific predisposition to atherosclerosis was performed. Growth Differentiation Factor 10 (GDF10) role in VSMCs phenotypic switch was investigated via flow cytometry, immunofluorescence in human atherosclerotic plaques. (3) Results: scRNAseq analysis revealed the transcriptomic profile of seven clusters, five of which showed disease-relevant gene signature of VSMC macrophagic calcific phenotype, VSMC mesenchymal chondrogenic phenotype, VSMC inflammatory and fibro-phenotype and VSMC inflammatory phenotype. Osteoblast factor GDF10 involved in ossification and osteoblast differentiation emerged as a hallmark of VSMCs undergoing phenotypic switch. Under hypercholesteremia, GDF10 triggered VSMC osteogenic switch in vitro. The abundance of GDF10 expressing osteogenic-like VSMCs cells was linked to the occurrence of carotid artery disease (CAD) events. (4) Conclusions: Taken together, these results provide evidence about GDF10-mediated VSMC osteogenic switch, with a likely detrimental role in atherosclerotic plaque stability
NLRP3 inflammasome activation controls vascular smooth muscle cells phenotypic switch in atherosclerosis
Background: Monocytes and nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome orchestrate lipid-driven amplification of vascular inflammation promoting the disruption of the fibrous cap. The components of the NLRP3 inflammasome are expressed in macrophages and foam cells within human carotid atherosclerotic plaques and VSMCs in hypertension. Whether monocytes and NLRP3 inflammasome activation are direct triggers of VSMC phenotypic switch and plaque disruption need to be investigated. (2) Methods: The direct effect of oxLDL-activated monocytes in VSMCs co-cultured system was demonstrated via flow cytometry, qPCR, ELISA, caspase 1, and pyroptosis assay. Aortic roots of VSMCs lineage tracing mice fed normal or high cholesterol diet and human atherosclerotic plaques were used for immunofluorescence quantification of NLRP3 inflammasome activation/VSMCs phenotypic switch. (3) Results: OxLDL-activated monocytes reduced \u3b1-SMA, SM22\u3b1, Oct-4, and upregulation of KLF-4 and macrophage markers MAC2, F4/80 and CD68 expression as well as caspase 1 activation, IL-1\u3b2 secretion, and pyroptosis in VSMCs. Increased caspase 1 and IL-1\u3b2 in phenotypically modified VSMCs was detected in the aortic roots of VSMCs lineage tracing mice fed high cholesterol diet and in human atherosclerotic plaques from carotid artery disease patients who experienced a stroke. (4) Conclusions: Taken together, these results provide evidence that monocyte promote VSMC phenotypic switch through VSMC NLRP3 inflammasome activation with a likely detrimental role in atherosclerotic plaque stability in human atherosclerosis
Immunodiagnosis of Paracoccidioidomycosis due to Paracoccidioides brasiliensis Using a Latex Test: Detection of Specific Antibody Anti-gp43 and Specific Antigen gp43
BackgroundParacoccidioidomycosis (PCM) is a life-threatening systemic disease and is a neglected public health problem in many endemic regions of Latin America. Though several diagnostic methods are available, almost all of them present with some limitations.Method/Principle FindingsA latex immunoassay using sensitized latex particles (SLPs) with gp43 antigen, the immunodominant antigen of Paracoccidioides brasiliensis, or the monoclonal antibody mAb17c (anti-gp43) was evaluated for antibody or antigen detection in sera, cerebrospinal fluid (CSF), and bronchoalveolar lavage (BAL) from patients with PCM due to P. brasiliensis. the gp43-SLPs performed optimally to detect specific antibodies with high levels of sensitivity (98.46%, 95% CI 91.7-100.0), specificity (93.94%, 95% CI 87.3-97.7), and positive (91.4%) and negative (98.9%) predictive values. in addition, we propose the use of mAb17c-SLPs to detect circulating gp43, which would be particularly important in patients with immune deficiencies who fail to produce normal levels of immunoglobulins, achieving good levels of sensitivity (96.92%, 95% CI 89.3-99.6), specificity (88.89%, 95% CI 81.0-94.3), and positive (85.1%) and negative (97.8%) predictive values. Very good agreement between latex tests and double immune diffusion was observed for gp43-SLPs (k = 0.924) and mAb17c-SLPs (k = 0.850), which reinforces the usefulness of our tests for the rapid diagnosis of PCM in less than 10 minutes. Minor cross-reactivity occurred with sera from patients with other fungal infections. On the upstream mobility scheme for two-phase flow in porous media
When neglecting capillarity, two-phase incompressible flow in porous media is
modelled as a scalar nonlinear hyperbolic conservation law. A change in the
rock type results in a change of the flux function. Discretizing in
one-dimensional with a finite volume method, we investigate two numerical
fluxes, an extension of the Godunov flux and the upstream mobility flux, the
latter being widely used in hydrogeology and petroleum engineering. Then, in
the case of a changing rock type, one can give examples when the upstream

Loss of AQP3 protein expression is associated with worse progression-free and cancer-specific survival in patients with muscle-invasive bladder cancer
Purpose
Urothelial carcinoma has recently been shown to express several aquaporins (AQP), with AQP3 being of particular interest as its expression is reduced or lost in tumours of higher grade and stage. Loss of AQP3 expression was associated with worse progression-free survival (PFS) in patients with pT1 bladder cancer. The objective of this study was to investigate the prognostic value of AQP3 expression in patients with muscle-invasive bladder carcinoma (MIBC).
Methods
Retrospective single-centre analysis of the oncological outcome of patients following radical cystectomy (Cx) due to MIBC. Immunohistochemistry was used to assess AQP3 protein expression in 100 Cx specimens. Expression levels of AQP3 were related to clinicopathological variables. The impact of biomarker expression on progression-free, cancer-specific and overall survival was determined by multivariate Cox regression analysis (MVA).
Results
High expression of AQP3 by the tumour was associated with a statistically significantly improved PFS (75 vs. 19 %, p = 0.043) and CSS (75 vs. 18 %, p = 0.030) and, alongside lymph node involvement, was an independent predictor of PFS (HR 2.871, CI 1.066–7.733, p = 0.037), CSS (HR 3.325, CI 1.204–8.774, p = 0.019) and OS (HR 2.001, CI 1.014–3.947) in MVA.
Conclusions
Although the results of the study would be strengthened by a larger, more appropriately powered, prospective, multi-institutional study, our findings strongly suggest that AQP3 expression status may represent an independent predictor of PFS and CSS in MIBC and may help select patients in need for (neo-)adjuvant chemotherapy
Eurythrematosis as a developmental model of the Diabetes Mellitus type 1 pathological condition: pathophysiological parameters and oxidative stress / Eurytrematose como modelo de desenvolvimento da patologia da Diabetes Mellitus tipo 1: parâmetros fisiopatológicos e stress oxidativo
Eurytrematosis is a helminthic disease caused by trematodes belonging to the genus Eurytrema spp. that parasitize the pancreas of many animals and humans. This parasitosis causes chronic fibrosing pancreatitis, fat infiltration in the pancreatic parenchyma, besides damaging the exocrine pancreas, which is similar to that found in patients with Diabetes Mellitus type 1 (DM1). The current work aimed to evaluate the use of bovine pancreas infected with E. coelomaticum as a model to study DM1 pathophysiology. It was carried out macroscopic analyses, parasite identification, total pancreatic lipid determination and oxidative damage biomarkers levels of pancreas naturally infected with E. coelomaticum. Macroscopically, we observed that the infected pancreas had duct obstruction, organ stiffness due to the visible presence of fibrosis, increased adipose tissue deposition, increased protein and lipid damage, as well as increased antioxidant biomarkers (GSH, CAT and VIT C). Thus, it is possible to show that DM1 may have pancreatic parasitism as a possible primary origin. However, more studies are needed to better investigate this possible primary origin; the results obtained here suggest that the use of pancreas parasitized by E. coelomaticum could be a model to investigate DM1 pathophysiology
