The Rhetorical Goddess: A Feminist Perspective on Women in Magic
Although female magicians have existed since the rise of entertainment magic, women have faced difficulty in entering the âfraternityâ of the magic community. As an art form largely based around persuasion, it is useful to study the performance of magic as a text. It is additionally useful to study female magicians within this context of rhetoric. Not only will examining the rhetoric of female magicians provide insights on the rhetoric of women in this unique arena, but also of women in a historically gendered and underrepresented field. Research into this area may disclose other details regarding the communicative differences between women and men and how communication is adapted within a gendered communication paradigm
Alix is required for activity-dependent bulk endocytosis at brain synapses
In chemical synapses undergoing high frequency stimulation, vesicle components can be
retrieved from the plasma membrane via a clathrin-independent process called activitydependent bulk endocytosis (ADBE). Alix (ALG-2-interacting protein X/PDCD6IP) is an
adaptor protein binding to ESCRT and endophilin-A proteins which is required for clathrinindependent endocytosis in fibroblasts. Alix is expressed in neurons and concentrates at
synapses during epileptic seizures. Here, we used cultured neurons to show that Alix is
recruited to presynapses where it interacts with and concentrates endophilin-A during conditions triggering ADBE. Using Alix knockout (ko) neurons, we showed that this recruitment,
which requires interaction with the calcium-binding protein ALG-2, is necessary for ADBE.
We also found that presynaptic compartments of Alix ko hippocampi display subtle morphological defects compatible with flawed synaptic activity and plasticity detected electrophysiologically. Furthermore, mice lacking Alix in the forebrain undergo less seizures during
kainate-induced status epilepticus and reduced propagation of the epileptiform activity.
These results thus show that impairment of ADBE due to the lack of neuronal Alix leads to
abnormal synaptic recovery during physiological or pathological repeated stimulations

Between Security and Mobility: Negotiating a Hardening Border Regime in the Russian-Estonian Borderland
This is an Accepted Manuscript of an article published by Taylor & Francis in Journal of Ethnic and Migration Studies on 27th Feb 2015, available online: http://dx.doi.org/10.1080/1369183X.2015.1015408Since the end of the Cold War order post-Soviet borders have been characterised by geopolitical tensions and divergent imaginations of desirable political and spatial orders. Drawing upon ethnographic research in two border towns at the Russian-Estonian border, the article makes a case for a grounded examination of these border dynamics that takes into account how borders as sites of âmobility and enclosureâ are negotiated in everyday life and shaped by the differentiated incorporations of statecraft into peopleâs lives. Depending on their historical memories, people interpret the border either as a barrier to previously free movement or as a security device and engage in correspondingly different relations to the state â privileging local concerns for mobility or adopting the stateâs concerns over security and sovereignty. Analysing these border negotiations and the relations between citizens and the state, articulated in peopleâs expectations and claims, can provide us with a better understanding of how people participate in the making of borders and contribute to the stability and malleability of political orders
Algorithmic deformation of matrix factorisations
Branes and defects in topological Landau-Ginzburg models are described by
matrix factorisations. We revisit the problem of deforming them and discuss
various deformation methods as well as their relations. We have implemented
these algorithms and apply them to several examples. Apart from explicit
results in concrete cases, this leads to a novel way to generate new matrix
factorisations via nilpotent substitutions, and to criteria whether boundary
obstructions can be lifted by bulk deformations.Comment: 30 page
Real-time analysis of the binding of fluorescent VEGFâââ a to VEGFR2 in living cells: Effect of receptor tyrosine kinase inhibitors and fate of internalized agonist-receptor complexes
Vascular endothelial growth factor (VEGF) is an important mediator of angiogenesis. Here we have used a novel stoichiometric protein-labeling method to generate a fluorescent variant of VEGF (VEGFâââ
a-TMR) labeled on a single cysteine within each protomer of the antiparallel VEGF homodimer. VEGFâââ
a-TMR has then been used in conjunction with full length VEGFR2, tagged with the bioluminescent protein NanoLuc, to undertake a real time quantitative evaluation of VEGFR2 binding characteristics in living cells using bioluminescence resonance energy transfer (BRET). This provided quantitative information on VEGF-VEGFR2 interactions. At longer incubation times, VEGFR2 is internalized by VEGFâââ
a-TMR into intracellular endosomes. This internalization can be prevented by the receptor tyrosine kinase inhibitors (RTKIs) cediranib, sorafenib, pazopanib or vandetanib. In the absence of RTKIs, the BRET signal is decreased over time as a consequence of the dissociation of agonist from the receptor in intracellular endosomes and recycling of VEGFR2 back to the plasma membrane
Repercussion of megakaryocyte-specific Gata1 Loss on megakaryopoiesis and the hematopoietic precursor compartment
During hematopoiesis, transcriptional programs are essential for the commitment and differentiation of progenitors into the different blood lineages. GATA1 is a transcription factor expressed in several hematopoietic lineages and essential for proper erythropoiesis and megakaryopoiesis. Megakaryocyte-specific genes, such as GP1BA, are known to be directly regulated by GATA1. Mutations in GATA1 can lead to dyserythropoietic anemia and pseudo gray-platelet syndrome. Selective loss of Gata1 expression in adult mice results in macrothrombocytopenia with platelet dysfunction, characterized by an excess of immature megakaryocytes. To specifically analyze the impact of Gata1 loss in mature committed megakaryocytes, we generated Gata1-Lox|Pf4-Cre mice (Gata1cKOMK). Consistent with previous findings, Gata1cKOMK mice are macrothrombocytopenic with platelet dysfunction. Supporting this notion we demonstrate that Gata1 regulates directly the transcription of Syk, a tyrosine kinase that functions downstream of Clec2 and GPVI receptors in megakaryocytes and platelets. Furthermore, we show that Gata1cKOMK mice display an additional aberrant megakaryocyte differentiation stage. Interestingly, these mice present a misbalance of the multipotent progenitor compartment and the erythroid lineage, which translates into compensatory stress erythropoiesis and splenomegaly. Despite the severe thrombocytopenia, Gata1cKOMK mice display a mild reduction of TPO plasma levels, and Gata1cK-OMK megakaryocytes show a mild increase in Pf4 mRNA levels; such a misbalance might be behind the general hematopoietic defects observed, affecting locally normal TPO and Pf4 levels at hematopoietic stem cell niches. © 2016 Meinders et al. Outcome in patients perceived as receiving excessive care across different ethical climates: a prospective study in 68 intensive care units in Europe and the USA
Purpose: Whether the quality of the ethical climate in the intensive care unit (ICU) improves the identification of patients receiving excessive care and affects patient outcomes is unknown. Methods: In this prospective observational study, perceptions of excessive care (PECs) by clinicians working in 68 ICUs in Europe and the USA were collected daily during a 28-day period. The quality of the ethical climate in the ICUs was assessed via a validated questionnaire. We compared the combined endpoint (death, not at home or poor quality of life at 1 year) of patients with PECs and the time from PECs until written treatment-limitation decisions (TLDs) and death across the four climates defined via cluster analysis. Results: Of the 4747 eligible clinicians, 2992 (63%) evaluated the ethical climate in their ICU. Of the 321 and 623 patients not admitted for monitoring only in ICUs with a good (n = 12, 18%) and poor (n = 24, 35%) climate, 36 (11%) and 74 (12%), respectively were identified with PECs by at least two clinicians. Of the 35 and 71 identified patients with an available combined endpoint, 100% (95% CI 90.0â1.00) and 85.9% (75.4â92.0) (P = 0.02) attained that endpoint. The risk of death (HR 1.88, 95% CI 1.20â2.92) or receiving a written TLD (HR 2.32, CI 1.11â4.85) in patients with PECs by at least two clinicians was higher in ICUs with a good climate than in those with a poor one. The differences between ICUs with an average climate, with (n = 12, 18%) or without (n = 20, 29%) nursing involvement at the end of life, and ICUs with a poor climate were less obvious but still in favour of the former. Conclusion: Enhancing the quality of the ethical climate in the ICU may improve both the identification of patients receiving excessive care and the decision-making process at the end of life
