    Toxic Algae Contamination and Demand for Shellfish: A Case Study of Demand for Mussels in Montreal

    Toxic algae blooms are a worldwide phenomena, which appear to be increasing in frequency and severity. These natural events cause product contaminations that often have significant economic consequences, including supply interruptions due to closed fishing grounds, losses from human illness, and losses due to a decline in demand for the affected products. This paper evaluates the impacts of a toxic algae bloom contamination event on demand for unaffected shellfish. As an empirical example of the economic losses the shellfish industry experiences for these events, demand for mussels in Montreal is estimated using firm-level data and proxies for consumer information, during and after domoic acid contamination of Prince Edward Island mussels. Sales losses due to decreased demand are calculated.     Bilingual, digital, audio-visual training modules improve technical knowledge of feedlot and dairy workers

    Two studies were conducted to determine the effectiveness of digital multimedia modules as training tools for animal care workers. Employees at a commercial feedlot (n = 17) and a commercial dairy (n = 10) were asked to independently complete a 10-question quiz prior to and following viewing of training modules. Module topics in the feedlot were proper handling of non-ambulatory animals and humane methods of euthanasia; modules were administered to the workers, as a group, in either English (n = 7) or Spanish (N = 10), depending on previously indicated worker preference. Modules addressing dairy cattle health practices and dairy cattle handling were presented to the dairy care workers who had a preference for learning in either English (n = 7) or Spanish (n = 3). For feedlot workers, post-test scores were improved by 28% after viewing the modules compared to pre-test scores (74% vs. 58%; P 0.30) between language, topic, and between-test variation, indicating that the modules were equally effective at information delivery to both audiences in both languages. For the dairy workers, test scores improved by 27% from pre-viewing to post-viewing (73% vs. 92%; P < 0.01); there was an interaction between the effect of module and language preference (P < 0.01) indicating that although scores increased for both of the topic areas for the English-speaking workers, only the score for the animal health topic increased for the Spanish-speaking workers. Regardless of nationality, level of formal education, topic, or preferred language, digital media are effective at improving knowledge transfer to animal care professionals

These trends may be grouped into six general categories: (1) the growing tension between federal environmental mandates and their practical implementation through delegated state programs; (2) the continuing debate over the exact definition of interstate commerce and the scope of state authority to regulate in the gray area; (3) the impending deregulation of the electrical energy market; (4) the promotion of sustainable development; (5) the developing schism between state and local land use control; and (6) the evolving nature of administrative law in the environmental context. The following survey discusses changes to the body of environmental law affecting Virginians during 2000-2001 in the context of each of these trends

    AIM: The variable results in clinical trials of adipose tissue-derived stem cells (ASCs) for chondral defects may be due to the different ex vivo culture conditions of the ASCs which are implanted to treat the lesions. We sought to determine the optimal in vitro chondrocyte co-culture condition that promotes infrapatellar fat pad-derived (IFPD) ASC chondrogenic gene expression in a novel co-culture combination. METHODS: In our study, we utilized an in vitro autologous co-culture of IFPD ASCs and articular chondrocytes derived from Kellgren-Lawrence Grade III/IV osteoarthritic human knee joints at ASC-to-chondrocyte seeding log ratios of 1:1, 10:1, and 100:1. Gene expression following in vitro co-culture was quantified by RT-qPCR with a panel comprising COL1A1, COL2A1, COL10A1, L-SOX5, SOX6, SOX9, ACAN, HSPG2, and COMP for chondrogenic gene expression. RESULTS: The chondrogenic gene expression profiles from co-cultures were greater than would be expected from an expression profile modeled from chondrocyte and ASC-only monocultures. Additionally, chondrogenic gene expression decreased with increasing ASC-to-chondrocyte seeding ratios. CONCLUSIONS: These findings provide insight into the mechanisms underlying clinical ASC therapies and signifies that IFPD ASCs pre-conditioned by chondrocyte co-culture may have improved chondrogenic potential for cartilage repair. This model can help further understand IFPD ASCs in chondral and osteochondral repair and the chondrogenic pathways involved. Video Abstract

    A Ship-Based Characterization of Coherent Boundary-Layer Structures Over the Lifecycle of a Marine Cold-Air Outbreak

    Convective coherent structures shape the atmospheric boundary layer over the lifecycle of marine cold-air outbreaks (CAOs). Aircraft measurements have been used to characterize such structures in past CAOs. Yet, aircraft case studies are limited to snapshots of a few hours and do not capture how coherent structures, and the associated boundary-layer characteristics, change over the CAO time scale, which can be on the order of several days. We present a novel ship-based approach to determine the evolution of the coherent-structure characteristics, based on profiling lidar observations. Over the lifecycle of a multi-day CAO we show how these structures interact with boundary-layer characteristics, simultaneously obtained by a multi-sensor set-up. Observations are taken during the Iceland Greenland Seas Project’s wintertime cruise in February and March 2018. For the evaluated CAO event, we successfully identify cellular coherent structures of varying size in the order of 4 × 102 m to 104 m and velocity amplitudes of up to 0.5 m s−1 in the vertical and 1 m s−1 in the horizontal. The structures’ characteristics are sensitive to the near-surface stability and the Richardson number. We observe the largest coherent structures most frequently for conditions when turbulence generation is weakly buoyancy dominated. Structures of increasing size contribute efficiently to the overturning of the boundary layer and are linked to the growth of the convective boundary-layer depth. The new approach provides robust statistics for organized convection, which would be easy to extend by additional observations during convective events from vessels of opportunity operating in relevant areas.publishedVersio

    LAT1 Protein Content Increases Following 12 Weeks of Resistance Exercise Training in Human Skeletal Muscle

    Introduction: Amino acid transporters are essential for cellular amino acid transport and promoting protein synthesis. While previous literature has demonstrated the association of amino acid transporters and protein synthesis following acute resistance exercise and amino acid supplementation, the chronic effect of resistance exercise and supplementation on amino acid transporters is unknown. The purpose herein was to determine if amino acid transporters and amino acid metabolic enzymes were related to skeletal muscle hypertrophy following resistance exercise training with different nutritional supplementation strategies. Methods: 43 college-aged males were separated into a maltodextrin placebo (PLA, n = 12), leucine (LEU, n = 14), or whey protein concentrate (WPC, n = 17) group and underwent 12 weeks of total-body resistance exercise training. Each group\u27s supplement was standardized for total energy and fat, and LEU and WPC supplements were standardized for total leucine (6 g/d). Skeletal muscle biopsies were obtained prior to training and ~72 h following each subject\u27s last training session. Results: All groups increased type I and II fiber cross-sectional area (fCSA) following training (p \u3c 0.050). LAT1 protein increased following training (p \u3c 0.001) and increased more in PLA than LEU and WPC (p \u3c 0.050). BCKDHα protein increased and ATF4 protein decreased following training (p \u3c 0.001). Immunohistochemistry indicated total LAT1/fiber, but not membrane LAT1/fiber, increased with training (p = 0.003). Utilizing all groups, the change in ATF4 protein, but no other marker, trended to correlate with the change in fCSA (r = 0.314; p = 0.055); however, when regression analysis was used to delineate groups, the change in ATF4 protein best predicted the change in fCSA only in LEU (r2 = 0.322; p = 0.043). In C2C12 myoblasts, LAT1 protein overexpression caused a paradoxical decrease in protein synthesis levels (p = 0.002) and decrease in BCKDHα protein (p = 0.001). Conclusions: Amino acid transporters and metabolic enzymes are affected by resistance exercise training, but do not appear to dictate muscle fiber hypertrophy.     The influence of barefoot and barefoot inspired footwear on the kinetics and kinematics of running in comparison to conventional running shoes.

    Barefoot running has experienced a resurgence in footwear biomechanics literature, based on the supposition that it serves to reduce the occurrence of overuse injuries in comparison to conventional shoe models. This consensus has lead footwear manufacturers to develop shoes which aim to mimic the mechanics of barefoot locomotion. This study compared the impact kinetics and 3-D joint angular kinematics observed whilst running: barefoot, in conventional cushioned running shoes and in shoes designed to integrate the perceived benefits of barefoot locomotion. The aim of the current investigation was therefore to determine whether differences in impact kinetics exist between the footwear conditions and whether shoes which aim to simulate barefoot movement patterns can closely mimic the 3-D kinematics of barefoot running. Twelve participants ran at 4.0 m.s-1±5% in each footwear condition. Angular joint kinematics from the hip, knee and ankle in the sagittal, coronal and transverse planes were measured using an eight camera motion analysis system. In addition simultaneous tibial acceleration and ground reaction forces were obtained. Impact parameters and joint kinematics were subsequently compared using repeated measures ANOVAs. The kinematic analysis indicates that in comparison to the conventional and barefoot inspired shoes that running barefoot was associated significantly greater plantar-flexion at footstrike and range of motion to peak dorsiflexion. Furthermore, the kinetic analysis revealed that compared to the conventional footwear impact parameters were significantly greater in the barefoot condition. Therefore this study suggests that barefoot running is associated with impact kinetics linked to an increased risk of overuse injury, when compared to conventional shod running.     Over 95% of pancreatic adenocarcinomas (PDACs), as well as a large fraction of other tumor types, such as colorectal adenocarcinoma, are driven by KRAS activation. However, no direct RAS inhibitors exist for cancer therapy. Furthermore, the delivery of therapeutic agents of any kind to PDAC in particular has been hindered by the extensive desmoplasia and resultant drug delivery challenges that accompanies these tumors. Small interfering RNA (siRNA) is a promising modality for anti-neoplastic therapy due to its precision and wide range of potential therapeutic targets. Unfortunately, siRNA therapy is limited by low serum half-life, vulnerability to intracellular digestion, and transient therapeutic effect.     Part 2: Preparing Entry-Level Occupational and Physical Therapy Students to Promote Healthy Lifestyles Emphasizing Healthy Eating with Individuals with Disabilities

    Rehabilitation professionals including occupational therapists (OT) and physical therapists (PT) are increasingly called upon to incorporate health promotion of lifestyle behaviors including physical activity and healthy eating into routine clinical care. While OTs and PTs may be comfortable promoting activity-related behaviors, many are less comfortable with nutrition behaviors. To address entry-level OT and PT students’ perceived discomfort with discussing diet-related behaviors, faculty developed a healthy eating module for students to use during a community-based service learning program. The purpose of this paper is to describe the formative evaluation process of developing the healthy eating module, and to discuss results of a pilot trial of this module. The formative assessment of the healthy eating module consisted of four steps: focus groups with students, key informant interviews with community partners, expert panel round table, and expert panel review of materials. Students (n=117) completed questionnaires at the end of the service learning program to assess how much they used the new resources, and how useful they found the resources. The final healthy eating module consisted of an on-line training session and a healthy eating toolkit, including resources for assessments, treatment activities and additional nutrition information.
