INTRODUCTION: Activating mutations in exon 3 of the β-catenin gene are involved in the pathogenesis of adamantinomatous craniopharyngiomas. Recently, the interaction between β-catenin and PROP1 has been shown to be responsible for pituitary cell lineage determination. We hypothesized that dysregulated PROP1 expression could also be involved in the pathogenesis of craniopharyngiomas OBJECTIVES: To determine whether dysregulated gene expression was responsible for tumor pathogenesis in adamantinomatous craniopharyngiomas, the β-catenin gene was screened for mutations, and the expression of the β-catenin gene and PROP1 was evaluated. β-catenin gene was amplified and sequenced from 14 samples of adamantinomatous craniopharyngiomas. PROP1 and β-catenin gene expression was assessed by real-time RT-PCR from 12 samples, and β-catenin immunohistochemistry was performed on 11 samples. RESULTS: Mutations in the β-catenin gene were identified in 64% of the adamantinomatous craniopharyngiomas samples. Evidence of β-catenin gene overexpression was found in 71% of the tumors with β-catenin mutations and in 40% of the tumors without mutations, and β-catenin immunohistochemistry revealed a nuclear staining pattern for each of the analyzed samples. PROP1 expression was undetectable in all of the tumor samples. CONCLUSION: We found evidence of β-catenin gene overexpression in the majority of adamantinomatous craniopharyngiomas, and we also detected a nuclear β-catenin staining pattern regardless of the presence of a bcatenin gene mutation. These results suggest that WNT signaling activation plays an important role in the pathogenesis of adamantinomatous craniopharyngiomas. Additionally, this study was the first to evaluate PROP1 expression in adamantinomatous craniopharyngiomas, and the absence of PROP1 expression indicates that this gene is not involved in the pathogenesis of this tumor, at least in this cohort

OBJECTIVE: To compare the different anatomical sites used in the measurement of waist circumference, as well as the effectiveness of these landmarks to predict the percentage of body fat by tetrapolar bioelectrical impedance analysis. METHODS: We evaluated 205 children from 6 to 9 years of age of both sexes. Data on weight, height and waist circumference were collected at three different sites: at the lower abdominal curvature, above the navel and at the midpoint between the last rib and the iliac crest. Nutritional status was assessed through the body mass index (BMI)/age as recommended by the World Health Organization. RESULTS: The sample was considered homogeneous in terms of sex, and the mean age was 7.2±1.2 years. Regarding nutritional status, 6.3% of the children had low weight, 75.1% were eutrophic, 7.3% were overweight, and 11.2% were obese. Among males, there was no statistical difference between the different sites of measurement; in females, the measurement above the navel was statistically higher. In the correlation analysis, the midpoint measurement showed the best correlation with percentage of body fat, with values of 0.50 in boys and 0.62 in girls. CONCLUSION: Waist circumference measured at the midpoint between the last rib and the iliac crest presented the best correlation with percentage of body fat.OBJETIVO: Comparar os diferentes pontos anatômicos de medida da circunferência da cintura, bem como a eficácia desses locais em predizer o percentual de gordura corporal mensurado pela bioimpedância elétrica tetrapolar.

Exploring the correlations between epi indicators of COVID-19 and the concentration of pharmaceutical compounds in Wastewater Treatment Plants in Northern Portugal

The COVID-19 pandemic caused by the SARS-CoV-2 virus led to changes in the lifestyle and human behaviour, which resulted in different consumption patterns of some classes of pharmaceuticals including curative, symptom-relieving, and psychotropic drugs. The trends in the consumption of these compounds are related to their concentrations in wastewater systems, since incompletely metabolised drugs (or their metabolites back transformed into the parental form) may be detected and quantified by analytical methods. Pharmaceuticals are highly recalcitrant compounds and conventional activated sludge processes implemented in wastewater treatment plants (WWTP) are ineffective at degrading these substances. As a results, these compounds end up in waterways or accumulate in the sludge, being a serious concern given their potential effects on ecosystems and public health. Therefore, it is crucial to evaluate the presence of pharmaceuticals in water and sludge to assist in the search for more effective processes. In this work, eight pharmaceuticals from five therapeutic classes were analysed in wastewater and sludge samples collected in two WWTP located in the Northern Portugal, during the third COVID-19 epidemic wave in Portugal. The two WWTP demonstrated a similar pattern with respect to the concentration levels in that period. However, the drugs loads reaching each WWTP were dissimilar when normalising the concentrations to the inlet flow rate. Acetaminophen (ACET) was the compound detected at highest concentrations in aqueous samples of both WWTP (98. 516 g L1 in WWTP2 and 123. 506 g L1in WWTP1), indicating that this drug is extensively used without the need of a prescription, known of general public knowledge as an antipyretic and analgesic agent to treat pain and fever. The concentrations determined in the sludge samples were below 1.65 µg g1 in both WWTP, the highest value being found for azithromycin (AZT). This result may be justified by the physico-chemical characteristics of the compound that favour its adsorption to the sludge surface through ionic interactions. It was not possible to establish a clear relationship between the incidence of COVID-19 cases in the sewer catchment and the concentration of drugs detected in the same period. However, looking at the data obtained, the high incidence of COVID-19 in January 2021 is in line with the high concentration of drugs detected in the aqueous and sludge samples but prediction of drug load from viral load data was unfeasible. This study was supported by the Competitiveness and Internationalisation Operational Programme, Lisbon Regional Operational Programme and Algarve Regional Operational Programme with the support of FEDER, through the Incentive Scheme: research and development activities and investment in testing and optimisation (upscaling) infrastructures in the context of COVID-19, through the Project “SARS CONTROL: Evaluation of the impacts of SARS-CoV-2 on the urban water cycle and the downstream effects on Public Health" (Ref. 070076). This experiment intended to evaluate the initial growth of Sesbania virgata (Cav.) Pers. (sesbânia) seedlings planted with distinct fertilizers in a degraded area after clay extraction. The seedlings were inoculated with Rhizobium and arbuscular mycorrhizal fungi. The experiment was carried out in a randomized complete-block design. Each experimental unit had 4 repetitions with 16 seedlings. The treatments were: 1. control; 2. ferkal (0,6 kg hole-1); 3. urban waste (0,8 kg hole-1); 4. sugar cane filter cake (0,6 kg hole-1) and 5. simple super phosphate (0,1 kg hole-1). Along the first 3 months the rate of surviving plants was evaluated. Height and diameter at the soil level had been bimonthly measured until the tenth month of growth. At this time were also measured the N, P and K nutrient contents of the leaves as well as the biomass production. No difference among the treatments had been observed for these variables. Effects of fertilization on the growth and production of biomass were not found out under the conditions of this study. Lenght and superficial thin root area (diameter larger and smaller than 0,3mm) were evaluated 14 months after planting. Higher values of length and thinner superficial area of the smallest roots were verified in comparison to the larger ones (more than 0,3 mm).

In the extreme south of Bahia State, the Atlantic rainforest has been strongly affected by anthropogenic actions for a long time. Ants are seen as good biological indicators because of their abundance and sensitivity to changes of environmental conditions, thus allowing the evaluation of environmental impacts. The purpose of this study was to evaluate the environmental quality in four Atlantic Forest remnants through the study of Formicidae assemblages. Four reserves sampled during the experiment were: the school of agriculture EMARC (CEPLAC) in Teixeira de Freitas; Vista Alegre Farm in Alcobaça; FIBRASA Farm in Itamarajú; and Riacho das Pedras Farm in Prado. In each area, 50 units of one square meter of litter were sampled at intervals of 50m and 100m of the border using “Winkler” traps. The most frequent ants in EMARC reserve were: Wasmannia auropunctata (60%) and Solenopsis sp.1 (58%); in FIBRASA farm: W. auropunctata (64%) and Solenopsis sp.1 (46%); in Riacho das Pedras farm: Pyramica eggersi (80%) and Solenopsis sp.1 (64%); in Vista Alegre farm: Pheidole sp.1 (34%), Hypoponera sp.8 (32%) and Pyramica denticulata (24%). The highest value of diversity index (Shannon-Weaver) was observed in FIBRASA farm (3.41), followed by Riacho das Pedras farm (3.31), Vista Alegre farm (3.00), and EMARC reserve (2.33). The largest number of species was observed in FIBRASA and in Riacho das Pedras farms (55 species), followed by Vista Alegre (33) and EMARC (22). The area with the highest estimated diversity is the FIBRASA farm, while the reserve of EMARC had the lowest estimate. There were correlations between the level of human impact, the species found and generic richness, as well as with other ecological variables. All the studied areas had a high occurrence of species typical of degraded lands, showing that urgent conservation measures are needed to the increasingly more rare remnants of forest in the extreme South Bahia State.
