The Antibacterial Activity of Honey Derived from Australian Flora
Chronic wound infections and antibiotic resistance are driving interest in
antimicrobial treatments that have generally been considered complementary,
including antimicrobially active honey. Australia has unique native flora and
produces honey with a wide range of different physicochemical properties. In
this study we surveyed 477 honey samples, derived from native and exotic plants
from various regions of Australia, for their antibacterial activity using an
established screening protocol. A level of activity considered potentially
therapeutically useful was found in 274 (57%) of the honey samples, with
exceptional activity seen in samples derived from marri (Corymbia
calophylla), jarrah (Eucalyptus marginata) and
jellybush (Leptospermum polygalifolium). In most cases the
antibacterial activity was attributable to hydrogen peroxide produced by the
bee-derived enzyme glucose oxidase. Non-hydrogen peroxide activity was detected
in 80 (16.8%) samples, and was most consistently seen in honey produced
from Leptospermum spp. Testing over time found the hydrogen
peroxide-dependent activity in honey decreased, in some cases by 100%,
and this activity was more stable at 4°C than at 25°C. In contrast, the
non-hydrogen peroxide activity of Leptospermum honey samples
increased, and this was greatest in samples stored at 25°C. The stability of
non-peroxide activity from other honeys was more variable, suggesting this
activity may have a different cause. We conclude that many Australian honeys
have clinical potential, and that further studies into the composition and
stability of their active constituents are warranted
Bridging topological and functional information in protein interaction networks by short loops profiling
Protein-protein interaction networks (PPINs) have been employed to identify potential novel interconnections between proteins as well as crucial cellular functions. In this study we identify fundamental principles of PPIN topologies by analysing network motifs of short loops, which are small cyclic interactions of between 3 and 6 proteins. We compared 30 PPINs with corresponding randomised null models and examined the occurrence of common biological functions in loops extracted from a cross-validated high-confidence dataset of 622 human protein complexes. We demonstrate that loops are an intrinsic feature of PPINs and that specific cell functions are predominantly performed by loops of different lengths. Topologically, we find that loops are strongly related to the accuracy of PPINs and define a core of interactions with high resilience. The identification of this core and the analysis of loop composition are promising tools to assess PPIN quality and to uncover possible biases from experimental detection methods. More than 96% of loops share at least one biological function, with enrichment of cellular functions related to mRNA metabolic processing and the cell cycle. Searches for Gravitational Waves from Binary Neutron Stars: A Review
A new generation of observatories is looking for gravitational waves. These
waves, emitted by highly relativistic systems, will open a new window for ob-
servation of the cosmos when they are detected. Among the most promising
sources of gravitational waves for these observatories are compact binaries in
the final min- utes before coalescence. In this article, we review in brief
interferometric searches for gravitational waves emitted by neutron star
binaries, including the theory, instru- mentation and methods. No detections
have been made to date. However, the best direct observational limits on
coalescence rates have been set, and instrumentation and analysis methods
continue to be refined toward the ultimate goal of defining the new field of

Prevalence of Disorders Recorded in Dogs Attending Primary-Care Veterinary Practices in England
Purebred dog health is thought to be compromised by an increasing occurence of inherited diseases but inadequate prevalence data on common disorders have hampered efforts to prioritise health reforms. Analysis of primary veterinary practice clinical data has been proposed for reliable estimation of disorder prevalence in dogs. Electronic patient record (EPR) data were collected on 148,741 dogs attending 93 clinics across central and south-eastern England. Analysis in detail of a random sample of EPRs relating to 3,884 dogs from 89 clinics identified the most frequently recorded disorders as otitis externa (prevalence 10.2%, 95% CI: 9.1-11.3), periodontal disease (9.3%, 95% CI: 8.3-10.3) and anal sac impaction (7.1%, 95% CI: 6.1-8.1). Using syndromic classification, the most prevalent body location affected was the head-and-neck (32.8%, 95% CI: 30.7-34.9), the most prevalent organ system affected was the integument (36.3%, 95% CI: 33.9-38.6) and the most prevalent pathophysiologic process diagnosed was inflammation (32.1%, 95% CI: 29.8-34.3). Among the twenty most-frequently recorded disorders, purebred dogs had a significantly higher prevalence compared with crossbreds for three: otitis externa (P = 0.001), obesity (P = 0.006) and skin mass lesion (P = 0.033), and popular breeds differed significantly from each other in their prevalence for five: periodontal disease (P = 0.002), overgrown nails (P = 0.004), degenerative joint disease (P = 0.005), obesity (P = 0.001) and lipoma (P = 0.003). These results fill a crucial data gap in disorder prevalence information and assist with disorder prioritisation. The results suggest that, for maximal impact, breeding reforms should target commonly-diagnosed complex disorders that are amenable to genetic improvement and should place special focus on at-risk breeds. Future studies evaluating disorder severity and duration will augment the usefulness of the disorder prevalence information reported herein
Regulation of Mouse Small Heat Shock Protein αb-Crystallin Gene by Aryl Hydrocarbon Receptor
The stress-inducible small heat shock protein (shsp)/αB-crystallin gene is expressed highly in the lens and moderately in other tissues. Here we provide evidence that it is a target gene of the aryl hydrocarbon receptor (AhR) transcription factor. A sequence (−329/−323, CATGCGA) similar to the consensus xenobiotic responsive element (XRE), called here XRE-like, is present in the αBE2 region of αB-crystallin enhancer and can bind AhR in vitro and in vivo. αB-crystallin protein levels were reduced in retina, lens, cornea, heart, skeletal muscle and cultured muscle fibroblasts of AhR−/− mice; αB-crystallin mRNA levels were reduced in the eye, heart and skeletal muscle of AhR−/− mice. Increased AhR stimulated αB-crystallin expression in transfection experiments conducted in conjunction with the aryl hydrocarbon receptor nuclear translocator (ARNT) and decreased AhR reduced αB-crystallin expression. AhR effect on aB-crystallin promoter activity was cell-dependent in transfection experiments. AhR up-regulated αB-crystallin promoter activity in transfected HeLa, NIH3T3 and COS-7 cells in the absence of exogenously added ligand (TCDD), but had no effect on the αB-crystallin promoter in C2C12, CV-1 or Hepa-1 cells with or without TCDD. TCDD enhanced AhR-stimulated αB-crystallin promoter activity in transfected αTN4 cells. AhR could bind to an XRE-like site in the αB-crystallin enhancer in vitro and in vivo. Finally, site-specific mutagenesis experiments showed that the XRE-like motif was necessary for both basal and maximal AhR-induction of αB-crystallin promoter activity. Our data strongly suggest that AhR is a regulator of αB-crystallin gene expression and provide new avenues of research for the mechanism of tissue-specific αB-crystallin gene regulation under normal and physiologically stressed conditions
Ireland: Submerged Prehistoric Sites and Landscapes
Evidence of Ireland's drowned landscapes and settlements presently comprises 50 sites spread across the entire island. These comprise mainly intertidal find spots or small collections of flint artefacts. A handful of fully subtidal sites are known, generally from nearshore regions and consisting, with one exception, of isolated single finds. Evidence of organic remains is also sparse, with the exception of Mesolithic and Neolithic wooden fish traps buried in estuarine sediments under Dublin. The relatively small number of sites is probably due to lack of research as much as taphonomic issues, and thus the current evidence hints at the potential archaeological record which may be found underwater. Such evidence could contribute to knowledge of the coastal adaptations and seafaring abilities of Ireland's earliest inhabitants. Nonetheless, taphonomic considerations, specifically relating to Ireland's history of glaciation, sea-level change and also modern oceanographic conditions likely limit the preservation of submerged landscapes and their associated archaeology. Realistically, the Irish shelf is likely characterised by pockets of preservation, which makes detection and study of submerged landscapes difficult but not impossible. A range of potential routes of investigation are identifiable, including site-scale archaeological survey, landscape-scale seabed mapping, archival research and community engagement
Physics, Astrophysics and Cosmology with Gravitational Waves
Gravitational wave detectors are already operating at interesting sensitivity
levels, and they have an upgrade path that should result in secure detections
by 2014. We review the physics of gravitational waves, how they interact with
detectors (bars and interferometers), and how these detectors operate. We study
the most likely sources of gravitational waves and review the data analysis
methods that are used to extract their signals from detector noise. Then we
consider the consequences of gravitational wave detections and observations for

Large-scale features of Last Interglacial climate: Results from evaluating the lig127k simulations for the Coupled Model Intercomparison Project (CMIP6)-Paleoclimate Modeling Intercomparison Project (PMIP4)
Abstract. The modeling of paleoclimate, using physically based tools, is
increasingly seen as a strong out-of-sample test of the models that are used
for the projection of future climate changes. New to the Coupled Model
Intercomparison Project (CMIP6) is the Tier 1
Last Interglacial experiment for 127 000 years ago (lig127k), designed to
address the climate responses to stronger orbital forcing than the
midHolocene experiment, using the same state-of-the-art models as for the future and
following a common experimental protocol. Here we present a first analysis
of a multi-model ensemble of 17 climate models, all of which have completed
the CMIP6 DECK (Diagnostic, Evaluation and Characterization of Klima) experiments. The equilibrium climate sensitivity (ECS) of
these models varies from 1.8 to 5.6 ∘C. The seasonal character of
the insolation anomalies results in strong summer warming over the Northern
Hemisphere continents in the lig127k ensemble as compared to the CMIP6 piControl and
much-reduced minimum sea ice in the Arctic. The multi-model results indicate
enhanced summer monsoonal precipitation in the Northern Hemisphere and
reductions in the Southern Hemisphere. These responses are greater in the
lig127k than the CMIP6 midHolocene simulations as expected from the larger insolation anomalies
at 127 than 6 ka. New synthesis for surface temperature and precipitation, targeted for 127 ka, have been developed for comparison to the multi-model ensemble. The
lig127k model ensemble and data reconstructions are in good agreement for summer
temperature anomalies over Canada, Scandinavia, and the North Atlantic and
for precipitation over the Northern Hemisphere continents. The model–data
comparisons and mismatches point to further study of the sensitivity of the
simulations to uncertainties in the boundary conditions and of the
uncertainties and sparse coverage in current proxy reconstructions. The CMIP6–Paleoclimate Modeling Intercomparison
Project (PMIP4) lig127k simulations, in combination with the proxy record, improve
our confidence in future projections of monsoons, surface temperature, and
Arctic sea ice, thus providing a key target for model evaluation and
optimization.

Chromosome 2p14 Is Linked to Susceptibility to Leprosy
BACKGROUND: A genetic component to the etiology of leprosy is well recognized but the mechanism of inheritance and the genes involved are yet to be fully established. METHODOLOGY: A genome-wide single nucleotide polymorphism (SNP) based linkage analysis was carried out using 23 pedigrees, each with 3 to 7 family members affected by leprosy. Multipoint parametric and non-parametric linkage analyses were performed using MERLIN 1.1.1. PRINCIPAL FINDINGS: Genome-wide significant evidence for linkage was identified on chromosome 2p14 with a heterogeneity logarithm of odds (HLOD) score of 3.51 (rs1106577) under a recessive model of inheritance, while suggestive evidence was identified on chr.4q22 (HLOD 2.92, rs1349350, dominant model), chr. 8q24 (HLOD 2.74, rs1618523, recessive model) and chr.16q24 (HLOD 1.93, rs276990 dominant model). Our study also provided moderate evidence for a linkage locus on chromosome 6q24-26 by non-parametric linkage analysis (rs6570858, LOD 1.54, p = 0.004), overlapping a previously reported linkage region on chromosome 6q25-26.
