A whole-genome sequence and transcriptome perspective on HER2-positive breast cancers.

HER2-positive breast cancer has long proven to be a clinically distinct class of breast cancers for which several targeted therapies are now available. However, resistance to the treatment associated with specific gene expressions or mutations has been observed, revealing the underlying diversity of these cancers. Therefore, understanding the full extent of the HER2-positive disease heterogeneity still remains challenging. Here we carry out an in-depth genomic characterization of 64 HER2-positive breast tumour genomes that exhibit four subgroups, based on the expression data, with distinctive genomic features in terms of somatic mutations, copy-number changes or structural variations. The results suggest that, despite being clinically defined by a specific gene amplification, HER2-positive tumours melt into the whole luminal-basal breast cancer spectrum rather than standing apart. The results also lead to a refined ERBB2 amplicon of 106 kb and show that several cases of amplifications are compatible with a breakage-fusion-bridge mechanism

Power Electronic Converters in Particle Accelerator Applications: A Review

    No full text
International audienceIn particle accelerator (PA) applications, a continuous, reliable, efficient and sustainable power supply solution is required. This supply should guarantee the availability of high power quality through reliable and high energy efficient power electronic stage .This paper presents a review on different power electronic converters for particle accelerator applications and provides an account of research in areas related to main power converters' topologies, operating modes control systems, fault detection and isolation methods and reliability analysis. The paper can be also considered as a guidance to assist in the power converter selection process of the next generations of particle accelerators

    No full text
International audienceA new design of ESRF booster power supply system has been developed and installed. A multiple power supplies control through network including real time control is now operational at ESRF. It manages 4 power supplies to generate 3 waveforms defined with 3x1600 values in a setpoint file. The power supplies states are managed by PLCs. The ramping waveforms are managed by a real time program running on a FPGA board. And a high level control on top of them is assumed by a TANGO multiple classes system. This paper presents how these three levels of controls are interlinked and show the results achieve

Validation of connection details between a UHPFRC ribbed slab and steel girders for a new composite bridge deck type

Within the French R & D project MIKTI focusing on innovative steel-concrete composite bridges, an exhaustive experimental program was carried out to provide an experimental validation of a new ultra-high performance fibre reinforced concrete (UHPFRC) 2 way-ribbed slab made of segments assembled by post-tensioning. Provisions concerning the realization of connection between the UHPFRC ribbed deck and steel profiles, which are critical for ensuring the composite behavior of the full structure, were tested in a specific push-out type experimentation. A safety factor from 1.5 to 2 was demonstrated

Thermal Stability of the New ESRF Extremely Brilliant Source

    No full text
International audienceIn the frame of the Extremely Brilliant Source project (EBS), studies dedicated to disturbances have been more intensively investigated. Engineering instabilities have two origins: mechanical and thermal. Major thermal issues are: - air conditioning presents a temperature ramp up of 2°C along the sector - storage ring requires a warm up period of 4 days for reaching a stable orbit These effects have been observed and corrected for 20 years. With EBS requirements, we need to identify these thermal effects in order to reduce the disturbances, thus improving more systematically the source stability. The study is lead by the comparison between the present and the new thermal system. To do so, it is necessary to evaluate the heat balance in this system, as well as to identify the thermal time constant of each component. FEA models have been performed to reveal sensitivity of these thermal issues. A full scale mock-up cell equipped with a prototype girder is measured with power cables inside. A FEA model was also developed for the present storage ring to analyse the air stream. Although investigations have already been developed, some others remain to be achieved by the end of 2016
